BACKGROUND: Breast implant-associated squamous cell carcinoma (BIA-SCC) has emerged as a rare but aggressive malignancy with a mortality rate of 25% at 1 year. Its pathogenesis remains uncharacterized.
METHODS: Breast implant capsules from women 18 to 80 years old were systematically sampled in six locations at a single institution. Samples were analyzed for squamous marker expression including p63, p40, and cytokeratin 5 (CK5) using PCR. Immunohistochemistry was used to assess presence of proteins and squamous metaplasia.
RESULTS: 23 breast implant capsules were analyzed for squamous-relate

Diabetic foot ulcers (DFUs) are among the most serious and costly complications of diabetes, characterised by delayed healing, frequent infections, and a high risk of recurrence. Despite advances in wound care, many current therapies fail to address the multifactorial pathophysiology of diabetic wounds, including vascular dysfunction, immune dysregulation, chronic inflammation, and microbial imbalance. In this context, topical probiotics have emerged as a promising microbiome-based strategy aimed at restoring microbial balance while promoting tissue repair. This review summarises current evide

Chronic wound healing is a complex pathological process driven by multiple factors, presenting a significant global healthcare challenge. It not only severely compromises patients' quality of life but also imposes a substantial socioeconomic burden. In recent years, with deepening insights into the wound microenvironment, composite therapeutic strategies combining natural herbal medicines and their active components with modern biomaterials have offered novel approaches to overcoming refractory wounds caused by diabetic ulcers, vascular lesions, burns, and infections. This paper first outlines

Solid organ transplant recipients are living longer due to advances in immunosuppressive therapy, but chronic immunosuppression carries a high burden of cutaneous adverse effects. This clinical review provides a practical, agent-specific overview of cutaneous toxicities associated with commonly used immunosuppressants in solid organ transplant recipients, including induction agents, calcineurin inhibitors, antimetabolites, mammalian target of rapamycin inhibitors, corticosteroids, and co-stimulation blockers. Cutaneous adverse effects range from early hypersensitivity reactions, alopecia, and

Chronic granulomatous disease can affect multiple organ systems and is associated with poor wound healing, which may preclude a patient from undergoing a bone marrow transplant. We present a patient with chronic granulomatous disease and extensive nonhealing full-thickness wounds who was successfully treated surgically with split-thickness skin grafts and spray-on autologous skin cell suspension. This treatment paradigm may be useful for other patients with immunologic dysfunction and chronic wounds.

Scleroderma, or systemic sclerosis (SSc) usually affects the gastrointestinal (GI) tract (up to ~90%), but severe lower GI dysmotility and intestinal pseudo- obstruction are uncommon, occurring in approximately 5. 4% of patients with SSc. We report a 25-year-old Palestinian male with a three- year history of profound unintentional weight loss (67 kg) and an eight- month history of postprandial diffuse abdominal pain, bloating, vomiting, diarrhea, and loss of appetite. Examination revealed cachexia, digital ulcers, and generalized skin thickening with hyperpigmentation. Inflammatory markers wer

Chronic ulceration following snake-bite envenoming is a severely disabling and therapeutic challenge, often refractory to multiple treatment modalities. We report the successful management of a non-healing pedal ulcer, 30 years in duration, on the left lower limb of a 53-year-old man, secondary to a viper bite. The ulcer had a three-decade history of recurrence despite various treatments, including a failed surgical skin graft 15 years earlier. Upon presentation, extensive workup ruled out vascular insufficiency, infection, osteomyelitis, and malignancy. A wound biopsy confirmed chronic inflam

Chronic non-healing skin ulcers are one of the most important clinical challenges, usually refusing conventional treatments. The applications of autologous platelet concentrates include platelet-rich plasma and platelet-rich fibrin, which enhance healing properties by accelerating wound healing by growth factor release. This randomised, prospective clinical trial included 36 patients aged between 18 and 65 years suffering from chronic non-healing ulcers. They were then randomly allocated into three treatment groups: Group A (PRP), Group B (PRF) and Group C (Conventional treatment). PRP and PRF

INTRODUCTION: Cutaneous squamous cell carcinoma (cSCC) is a common nonmelanoma skin cancer primarily driven by chronic sun exposure and advanced age. Standard treatments include wide local excision, Mohs surgery, and radiotherapy, with advanced cases often managed with chemotherapeutics. PD1 inhibitors like cemiplimab and pembrolizumab are FDA-approved treatments that have shown efficacy in locally advanced or metastatic cSCC not amenable to surgery or radiation.
OBJECTIVES: This study evaluated clinical responses to these inhibitors using RECIST 1.1 criteria through direct visual examination

Chronic wounds are expensive and burdensome to the health care system, with some acute wounds, such as wounds after Mohs micrographic surgery (MMS), contributing to this problem. The rate of MMS performed is increasing, and if they become nonhealing, they can be challenging to treat and necessitate proper care. Both acellular dermal grafts and negative-pressure wound therapy are therapies that can be used to treat complicated wounds, but the combination of these 2 therapies has not been extensively studied. The study authors report the successful use of an acellular dermal graft in combination

OBJECTIVE: Local management of chronic or difficult-to-heal wounds often involves pain. Effective pain management remains challenging for practitioners, as existing treatments may fall short in providing comprehensive relief. Sevoflurane has emerged as a potential local analgesic in wound care. This systematic review sought to evaluate the safety and efficacy of using sevoflurane as a local analgesic for managing pain in chronic or hard-to-heal wounds.
DATA SOURCES: The search encompassed PubMed, Scopus, SciELO, and Cochrane databases.

OBJECTIVE: Chronic skin wounds caused by diabetes, peripheral artery disease, pressure ulcers, and venous insufficiency do not fully recover anatomically and functionally. We previously found that topical application of 3% WIN-1001X cream reduces skin inflammation. In this study, we investigated whether 3% WIN-1001X cream alleviates chronic skin wounds exhibiting prolonged and extensive inflammation using streptozotocin-induced diabetic mouse.
METHODS: WIN-1001X contained 20% ethanol extracts of three botanical drugs: Polygala tenuifolia Willd., Angelica tenuissima Nakai, and Dimocarpus longa

Livedoid vasculopathy (LV) is a chronic thrombotic microangiopathy characterized by painful ulcerations that significantly impair quality of life. Management of refractory LV remains challenging as conventional therapies have limited efficacy. We report two female patients with chronic, painful ulcers unresponsive to multiple intravenous immunoglobulin cycles who achieved complete ulcer healing and rapid pain resolution following a combined regimen of CHAP therapy, comprising nifedipine, hydroxychloroquine, aspirin and pentoxifylline, compression bandaging and punch grafts. Patient 1 is a 60-y

Electrospun nanofibers have emerged as a promising platform for drug delivery, particularly in the treatment of skin disorders, chronic wounds, and oral ulcers. Their high surface area, porosity, and structural resemblance to the extracellular matrix (ECM) offer excellent drug-loading capacity, controlled release, and tissue integration. This review uniquely emphasises the integration of plant-derived bioactive compounds, such as curcumin, quercetin, hesperidin, and Aloe vera, into multifunctional electrospun nanofiber systems. Unlike existing reviews that focus on either nanofiber fabrication

Mitochondria are nanoscale organelles essential for cellular metabolism and redox regulation, making them a compelling target for regenerative therapeutics. Analysis of wound-edge tissues from pediatric patients with chronic non-healing ulcers revealed marked metabolic insufficiency and impaired regenerative signaling, underscoring an unmet clinical need for mitochondrial-based interventions. Here, we show that topically applied mesenchymal stem cell-derived mitochondria (MSC-mt), functioning as naturally derived nanoscale organelles, markedly accelerate wound closure in a murine full-thicknes

The healing process of diabetic foot ulcers (DFUs) presents a slow pattern with an increased risk of infections, ischemia, and thrombosis correlated with high levels of reactive oxygen species production. Vascular injury is one of the factors contributing to the difficulty of wound healing in diabetic patients. Although the understanding of the pathophysiology of DFUs has significantly increased in recent years, associated treatments still have a high level of failure, leading to high morbidity rates, mortality, and amputations. Three-dimensional (3D) cell culture platforms offer a new approac

Diabetic ulcers (DUs) are a main category of nonhealing chronic wounds that tend to be vulnerable and prone to recurrence, posing a significant clinical challenge for DUs healing. DUs are frequently plagued by bacterial infections, oxidative stress, persistent inflammation, suppressed angiogenesis, and reduced growth factor expression, ultimately leading to persistent barriers to tissue regeneration and even amputation. Herein, we developed a multifunctional polysaccharide-based conductive hydrogel microneedle patch (MN-EP) integrating hyaluronic acid (HA) needle shafts, carboxymethyl chitosan

BACKGROUND: Prolidase deficiency is a rare autosomal recessive metabolic disorder resulting from mutations in the PEPD gene, leading to impaired collagen turnover and a broad spectrum of clinical features. Its manifestations include recurrent ulcerative skin lesions, recurrent infections, and hematological abnormalities. Celiac disease, another multisystem disorder, presents with chronic diarrhea, malabsorption, and iron deficiency anemia. The coexistence of both conditions is exceedingly rare and may lead to diagnostic delays.
CASE PRESENTATION: We report the case of an 18-year-old male indi

Globally, the prevalence of diabetes mellitus (DM), a chronic metabolic disease, is at its peak. Type 2 diabetes (T2D) is more common worldwide, which raises the risk of ulcers and chronic healing, or non-healing wounds. Overall 15% to 20% of people with diabetes develop diabetic foot ulcers (DFUs), which is a significant consequence. Managing and treatment of wounds require an advancement of novel healing therapies with the use of biological dressings. The development of DFU wound dressings and the biomanufacturing of composite 3D skin substitutes are examples of 3D bioprinting technology for
