A Conductive, Photothermal and Antioxidant ε-Poly-L-Lysine/Carbon Nanotube Hydrogel as a Candidate Dressing for Chronic Diabetic Wounds.

BACKGROUND: Chronic diabetic wounds, particularly diabetic foot ulcers (DFUs), are prone to recurrent infection and delayed healing, resulting in substantial morbidity, mortality, and economic burden. Multifunctional wound dressings that combine antibacterial, antioxidant, conductive, and self-healing properties may help to address the complex microenvironment of chronic diabetic wounds. METHODS: In this study, ε-poly-L-lysine and amino-terminated polyethylene glycol were grafted onto carboxylated single-walled carbon nanotubes (SWCNTs) via amide coupling to obtain ε-PL-CNT-PEG. Aminated chon

Transcriptomic Profiling of Diabetic Porcine Wound Healing Model Identifies Key Metabolic, Inflammatory, and Oxidative Stress Pathways.

Diabetic foot ulcers ( DFU s) remain a major clinical challenge as diabetes prevalence rises, emphasizing the need for improved therapeutics and relevant preclinical models. Common rodent wound-healing models poorly recapitulate human skin anatomy and repair. Although porcine skin is comparable to human skin, many studies employ young, healthy pigs that do not reflect typical chronic human wounds. Here, we evaluated wound healing in full-thickness skin wounds in non-diabetic and diabetic Yucatan minipigs. RNA sequencing identified key transcriptional differences in wounds of diabetic versus no

Neonatal Behçet's syndrome: a case report and literature review.

Behçet's syndrome is a rare, chronic inflammatory disorder characterized by recurrent oral and genital ulcers, skin lesions, and eye inflammation. While most children born to mothers with Behçet's syndrome remain asymptomatic, some newborns may develop symptoms due to unclear pathological mechanisms. A male newborn, born at 39 weeks to a mother with a 14-year history of Behçet's syndrome, developed oral lesions on the 10th day of life. The lesions progressed, becoming ulcerated and causing painful feeding. Additional findings included a few facial pustules, pathergy, leukocytosis, and elevated

Biomimetic hydrogel dressings: An integrated "perception-response-repair" strategy for chronic diabetic wounds.

With the dramatic increase in the global incidence of diabetes, the incidence of its complications such as chronic wounds and ulcers has also increased significantly. How to more effectively treat diabetic wounds has always been a huge challenge for clinicians and even global health systems. Diabetic wound coverage and rapid healing require multifunctional skin-mimicking wound dressings, including sufficient mechanical strength, antibacterial, antioxidant, anti-inflammatory and tissue adhesion properties. This minireview discusses in detail the current and emerging preparation ideas of hydroge

Multifunctional natural herbal extracts Lamiophlomis rotata (Benth.) Kudo composite gel for infected wounds healing.

Wounds are susceptible to persistent infections and chronic inflammation, which can significantly impeding the healing process.Lamiophlomis rotata (Benth.) Kudo(LR), a traditional Tibetan medicine, is known for its efficacy in treating injuries, contusions, and skin ulcers. Existing LR-related formulations such as capsules and patches are primarily used for pain relief and cannot provide an optimal environment for infected wounds, thereby limiting their healing potential. In this study, we successfully extracted an extract of LR and confirmed its strong antibacterial activity, supporting its u

Polyglycerol-b-polyoxazoline: A next-generation biocompatible agent for diabetic wound treatment.

Diabetic wounds remain a major clinical challenge due to persistent inflammation and impaired tissue regeneration. In this study, we report the scalable synthesis of hyperbranched polyglycerol-b-poly(2-ethyl-2-oxazoline) star copolymers bearing terminal arginine groups (hPG-b-PEO-Arg), designed to self-assemble into nanoscale hydrocolloids (70-200 nm) in aqueous media, driven by the distinct hydrophilicity of the polymer blocks. These hydrophilic nanoparticles penetrate damaged wound tissue, facilitating in vivo healing of diabetic wounds in rats (n = 10). The copolymers were synthesized on a

Ultra-microskin as cytokine reservoirs promote diabetic wound healing: experimental research.

BACKGROUND: Chronic wounds, such as diabetic foot ulcers, are challenging to heal due to persistent inflammatory responses and imbalanced growth factors. Conventional microskin grafting techniques face limitations, including insufficient viability, orientation-dependent survival, and restricted paracrine functions. This study investigates the feasibility of using ultra-microskin grafts (approximately 0.3 mm thick, 0.4 × 0.4 mm in surface dimensions) at expansion ratios of 1:10 to enhance diabetic wound repair. MATERIALS AND METHODS: Ultra-microskins were prepared using an automated skin-cutti

Topical application of Clostridium butyricum by an anaerobic hydrogel for accelerated diabetic wound healing through selective bacteria inhibition and ROS scavenging.

Selective inhibition of harmful bacteria without affecting skin symbiotic probiotics and selective scavenging of highly toxic reactive oxygen species (ROS) such as hydroxyl radicals (•OH) and peroxynitrite anions (ONOO-) are new requirements for more precise treatment of diabetic ulcer wounds. Achieving either is challenging, simultaneous achievement remains unreported. Clostridium butyricum (C. butyricum) inherently offers selective antibacterial action and produces hydrogen, specifically scavenging •OH and ONOO-, showing great potential for diabetic wound treatment. However, as an anaerobic

Skin-mimetic bilayer hydrogel normalizes diabetic wound healing by orchestrating inflammatory cell dynamics: An early intervention strategy.

Diabetic ulcers, characterized by persistent chronic inflammation and impaired healing, pose a significant clinical challenge. Inspired by the physiological healing process observed in healthy wounds, we develop an early intervention strategy by the spatiotemporal modulation of inflammatory cell dynamics to restore inflammation homeostasis in diabetic wounds. To achieve this, a skin-mimetic bilayer hydrogel that mimics the layered structure, composition, and pore size distribution of human skin is developed to allow for the sequential release of neuropeptide substance P and interleukin-10. Our

Human ISG15 deficiency unveils impaired healing of ulcerations via type I interferon-mediated fibrosis.

ISG15 deficiency is a type I interferonopathy characterized by elevated circulating type I interferon (IFN-I), intracranial calcifications, fibrotic skin lesions, and occasionally inflammatory lung disease. However, the mechanisms driving immune-mediated fibrosis remain poorly understood. In this study, we combined molecular biology approaches with spatial transcriptomics in ex vivo patient samples and in vitro models to characterize a novel loss-of-function ISG15 variant and to elucidate disease-associated molecular pathways. Analysis of patient skin biopsies revealed increased IFN-I-dependen

Galectins: Role and Therapeutics in Diabetes and Diabetic Foot Ulcers.

Diabetes is a chronic inflammatory disease due to decreased insulin release or insulin resistance. Diabetes complications stem from high blood sugar damaging blood vessels and nerves, leading to issues like heart disease, stroke, kidney failure, nerve damage, vision loss, foot ulcers, gum disease, skin infections, and digestive/bladder issues. Galectins, especially galectin-3, are emerging as key players in diabetes complications, promoting fibrosis, inflammation, and vascular damage. This suggests that galectins may be potential therapeutic targets in diabetes and its complications, and a nee

Traumatic Marjolin's ulcer in sheep: Squamous cell carcinoma arising from chronic dorsal fence-wire injuries.

Marjolin's ulcer (MU) is a rare but aggressive squamous cell carcinoma arising from chronic cutaneous injury and has been only exceptionally reported in domestic animals. Chronic skin wounds are common in small ruminants raised under semi-arid conditions and may predispose to malignant transformation; however, MU has not previously been documented in sheep. Here, we report five cases in Santa Inês ewes from a confined flock in the semi-arid region of Northeastern Brazil. The affected 36-month-old ewes developed chronic dorsal wounds after repeatedly crawling under galvanised wire fencing, resu

Predictive modeling of chronic foot ulcer outcomes using longitudinal photoacoustic imaging.

This study reports the first clinical longitudinal photoacoustic imaging (PAI) of chronic foot ulcers, a major complication in patients with peripheral vascular disorders. Compared to traditional methods such as ABI or near-infrared spectroscopy, the photoacoustic imaging approach provides non-invasive, high-resolution, and quantitative monitoring of vascular dynamics over time. Our system provided dorsal-side imaging of vascular structures with an expanded field of view and incorporated a skin artifact suppression algorithm to improve visualization of subdermal vasculature. From the acquired

The Barriers and Facilitators Healthcare Professionals' Experience When Assessing the Cutaneous Manifestations of Chronic Venous Insufficiency and Peripheral Arterial Disease in People With Dark Skin Tones: A Qualitative Descriptive Study.

Recognising peripheral arterial disease and chronic venous insufficiency early and managing them appropriately improves patient outcomes and prevents ulceration. However, few studies consider the influence patient skin tone has on this assessment. The study aimed to explore the experiences of healthcare professionals when assessing the cutaneous manifestations of chronic venous insufficiency and peripheral arterial disease in people with dark skin tones. We conducted semi-structured online interviews with healthcare professionals working in the United Kingdom between July and December 2023. Th
