Loss of Trp53 results in a hypoactive T cell phenotype accompanied by reduced pro-inflammatory signaling in a syngeneic orthotopic mouse model of ovarian high-grade serous carcinoma.

Contrast-enhanced spectral mammography (CESM) is one of the new diagnostic modalities implemented in clinical practice. In the case of these techniques, there are two major issues to be addressed: (1) their diagnostic usefulness, and (2) the relation between parameters assessed using these techniques and well-known diagnostic/prognostic/predictive markers (histological, clinical, and molecular). Therefore, we studied the relationship between the tumour margin assessed on CESM and (1) tumour borders defined on the basis of macroscopic and microscopic examination, (2) pT, (3) pN, and (4) tumour

Functional protein biomarkers based on distributions of expression levels in single-cell imaging data.

MOTIVATION: The intra-tumor heterogeneity of protein expression is well recognized and may provide important information for cancer prognosis and predicting treatment responses. Analytic methods that account for spatial heterogeneity remain methodologically complex and computationally demanding for single-cell protein expression. For many functional proteins, single-cell expressions vary independently of spatial localization in a substantial proportion of the tumor tissues, and incorporation of spatial information may not affect the prognostic value of such protein biomarkers. RESULTS: We dev

Utilization of weight management treatment and subsequent cardiovascular events among patients with breast cancer.

PURPOSE: Overweight and obese breast cancer (BC) survivors face higher risks of recurrence and all-cause mortality, including from cardiovascular disease (CVD). Weight management therapy (WMT) may reduce cardiovascular events (CVE). We assessed trends in WMT in BC survivors and evaluated rates of CVE. METHODS: We conducted a retrospective cohort study using the MarketScan Database, including overweight and obese patients (18-95 years) with invasive BC (2009 -2021), who underwent breast surgery. Exclusions were prior bariatric surgery or secondary cancers. Patients were categorized by weight s

Integrated, Cross-Entity Information on Preventive Measures for Bowel, Breast, and Prostate Cancer: Evaluation Study of the Web Application "Prevent-Take-Up".

BACKGROUND: About 40% of cancers are preventable through evidence-based interventions; however, uptake remains suboptimal. Knowledge and acceptance of primary and secondary preventive measures in the general population is not sufficient. We hypothesized that a web-based tool providing comprehensive, easily accessible, and individualized information on preventive strategies for multiple tumor entities could support informed decisions. OBJECTIVE: This study aimed to evaluate an interactive web application offering guideline-based, risk-adapted information on preventive measures for colorectal,

Bidirectional Risk Association Between Hidradenitis Suppurativa and Inflammatory Bowel Disease.

Hidradenitis suppurativa is an inflammatory skin disease of the pilosebaceous unit encompassing painful, recurrent, and malodorous abscesses, inflammatory nodules, dermal sinus tracts, and cicatrices. Hidradenitis suppurativa has been linked to several comorbidities, including inflammatory bowel disease, though evidence is limited by small sample sizes and possible bidirectional associations remain uncertain. A large-scale, propensity-score matched retrospective cohort study was performed using the TriNetX platform to investigate bidirectional risks between hidradenitis suppurativa and inflamm

Dysplastic nevi and malignant melanoma: insights into pre-biopsy diagnostic accuracy and the critical role of dermatology consultation.

A dysplastic nevus (DN) is an acquired melanocytic nevus that displays clinical and histological features similar to malign melanoma (MM). This study aims to evaluate the demographic and clinical characteristics of DN and primary cutaneous MM, as well as assess the accuracy of pre-biopsy diagnoses. We retrospectively analyzed 206 histopathologically confirmed cases of DN (n = 185) and MM (n = 21) from 159 patients at a tertiary care center. The biopsy samples had been sent from both dermatology and other surgical departments. The study included 21 patients with MM and 139 with DN; one patient

Pediatric Giant Cell Fibroblastoma: A Review of Treatment and Outcomes.

Giant cell fibroblastoma (GCF) is a rare predominantly pediatric tumor with frequent local recurrence that has clinical and histological similarities with DFSP. We conducted a literature review of published pediatric GCF cases to review demographics, management and outcomes. We identified 104 pediatric GCF cases from 52 articles, of which 81 were treated with excision of unspecified margins (EUM), 17 with wide local excision (WLE), two with Mohs micrographic surgery (MMS), and one with chemotherapy. Local recurrence was noted in 38/104 (36.5%); given the high recurrence rates of GCF following

Patterns and prognostic implications of distant metastasis in breast Cancer based on SEER population data.

Distant metastasis remains the leading cause of mortality in breast cancer, yet comprehensive population-based evaluations of metastatic site combinations and their survival implications are limited. This study aimed to explore the clinicopathological determinants and prognostic outcomes of site-specific and multi-organ metastases in breast cancer using SEER data. A total of 200,558 female breast cancer patients diagnosed between 2014 and 2023 were extracted from the SEER database. Logistic regression was used to assess associations between clinicopathological features and metastatic patterns.

From neglected to notable: A growing public health challenge driven by hospitalization for sporotrichosis in Pernambuco, Northeast Brazil (2016-2024).

Opportunistic and endemic mycoses represent a significant global health concern, particularly in Brazil. In the northeastern state of Pernambuco (PE), sporotrichosis has become a rapidly escalating public health challenge, evidenced by a notable increase in hospital admissions, despite its mostly outpatient nature. The present study, therefore, aimed to investigate the epidemiological and clinical profile of mycosis-related hospitalizations in PE from 2016 to 2024, with a primary focus on the burden of sporotrichosis. This statewide surveillance study used data from Brazil's Unified Health Sys

The causal effects between autoimmune thyroid disease and urticaria: evidence from a Mendelian randomization study.

Previous studies have suggested a comorbidity between autoimmune thyroid disease (AITD) and urticaria. However, the causal effects between AITD and urticaria remain unclear. This study employed genome-wide association study (GWAS) data to analyze the associations between AITD and urticaria through bidirectional Mendelian randomization (MR), meanwhile, examined the role of 731 immune traits as mediators between the two diseases through mediation analysis. The IVW results indicated that Graves' disease (GD) and Hashimoto's thyroiditis (HT) have a potential risk effect on urticaria, particularly

An Investigation of Mucosal Leishmaniasis in the Military Health System.

Leishmaniasis is a protozoal infection with an increased risk of transmission to those serving in the U.S. Military due to theaters of operation in endemic regions. There has, in recent decades, been robust experience with old-world leishmaniasis in the Military Health System (MHS); however, new-world leishmaniasis, which may result in mucosal leishmaniasis, has been less studied. A total of 88 patients from 2012 to 2022 with diagnosis codes for "mucocutaneous leishmaniasis" or "leishmaniasis, unspecified" were identified in the Military Data Repository and reviewed. Within this cohort, upon m
