Host IFN-γ is dispensable for anti-arthritic effects of Plasmodium yoelii 17XNL in mouse collagen-induced arthritis.

We previously reported that Plasmodium yoelii 17XNL (Py) protected mice from collagen-induced arthritis (CIA). However, the host factors required for this protection are not fully understood. As interferon-gamma (IFN-γ) is often induced by protozoan infections and is also known as an anti-arthritic cytokine in CIA, we here investigated the requirement of IFN-γ for the anti-arthritic effects of Py infection. CIA was induced by immunization with collagen type II (CII) emulsified with Freund's complete adjuvant in IFN-γ knockout (IFN-γKO) and wild-type (WT) mice. At 2-week post-immunization, mice

Construction of a recombinant attenuated Salmonella enterica serovar Choleraesuis vaccine expressing three antigens of Streptococcus suis and evaluation of its safety and immunogenicity.

Streptococcus suis serotype 2 (SS2) is a major cause of meningitis and septicemia in piglets, prompting a pressing need for safe and effective vaccines amid reduced antibiotic use. In this study, we developed a novel recombinant attenuated Salmonella Choleraesuis vaccine vector, rSC0148, incorporating triple safety systems (delayed lysis, attenuation, and balanced lethality), to deliver a fusion antigen of three SS2 immunogens (SaoA, Eno, and 6PGD), yielding the candidate strain rSC0148(pS-SE6). Our results demonstrated that compared to the wild-type strain C78-3, rSC0148(pS-SE6) showed reduce

Apolipoprotein E Deficiency Attenuates Neuroinflammatory Responses and Demyelination in Experimental Autoimmune Encephalomyelitis.

The precise etiology of multiple sclerosis (MS) remains incompletely understood, despite a well-established genetic predisposition. Clinical and epidemiological studies have identified an association between apolipoprotein E (ApoE) and MS progression; however, the underlying mechanisms remain largely unresolved. In this study, we investigated the role of ApoE in MS progression using an experimental autoimmune encephalomyelitis (EAE) model in ApoE knockout mice. EAE was induced in female C57BL/6 wild-type (WT) and ApoE knockout (ApoE KO) mice via immunization with myelin oligodendrocyte glycopr

Functionalized screen-printed electrodes for non-invasive detection of vascular-endothelial cadherin in extracellular vesicles.

In this study, we developed a biosensor using a gold screen-printed electrode (Au-SPE) functionalized with mercaptoundecanoic acid (MUA) and an antibody for detecting the vascular-endothelial cadherin (CD144) as a endothelial biomarker protein on extracellular vesicles (EVs) isolated from saliva. The MUA functionalization provides a stable platform for immobilizing the CD144 antibody, ensuring the detection of the target protein. This biosensor combines Au-SPE technology with an immunoassay, offering a rapid, sensitive, and non-invasive method for detection of CD144 carried by EVs. Characteriz

Recombinant AAV vaccine expressing PEDV S1 enhances immunity with superior neonatal protection.

To address suboptimal antibody induction by commercial PEDV vaccines, we constructed a recombinant adeno-associated virus (rAAV-CMV-PEDV S1) expressing regions of the PEDV S1 subunit based on Adeno-associated virus serotype 2 (AAV-2) vector. In mice, rAAV-CMV-PEDV S1 induced significantly more remarkable and persistent serum-specific IgG compared with the inactivated vaccine and accompanied by robust CD3+CD8+ T cells activation within 4 weeks post-immunization. In pregnant sows, compared with commercially available inactivated and attenuated vaccines, the rAAV-CMV-PEDV S1 induced significantly

LmCen-/- based vaccine is protective against canine visceral leishmaniasis following three natural exposures in Tunisia.

Dogs are the main reservoir host of Leishmania infantum, etiological agent of zoonotic visceral leishmaniasis (ZVL). An effective vaccine against Canine Visceral Leishmaniasis (CVL) will help the control and elimination of ZVL. In this study, we evaluated in dogs the safety, immunogenicity, and efficacy of a live attenuated Leishmania major Centrin gene-deleted (LmCen-/-) as a vaccine. Two doses (106 or 107) of LmCen-/- vaccine were administered intradermally in a prime-boost regimen. Both vaccine doses induced equally high level of IgG anti-Leishmania and exhibited strong antigen-specific cel

Roles of non-structural protein 1 in dengue virus replication and immunomodulation in vivo.

Dengue virus (DENV) is a vector-borne positive-sense RNA virus causing the dengue epidemic in tropical and subtropical regions. Non-structural viral protein 1 (NS1) is a key modulator of host-pathogen interactions. We investigated the in vivo effect of NS1 on DENV infection using an NS1-expressing recombinant vaccinia virus DIs strain (rDIs-DENV2C-NS1). NS1 expression and anti-NS1 antibody induction were observed. To assess its functional role, rDIs-DENV2C-NS1 was administered to AG129 mice lacking type I and II interferon receptors. Two weeks post-immunization, the mice were challenged with D

Jellyfish collagen ameliorates collagen-induced arthritis CIA in Sprague Dawley rats by modulating inflammatory mediators by inhibiting the NF κB and JAK STAT pathways.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by joint inflammation, synovial hyperplasia, and systemic complications. Current treatment options have side effects and limitations, leading to a growing interest in alternative therapies. AIM: This study investigates the therapeutic potential of jellyfish collagen (JC) on collagen-induced arthritis in a rat model. METHODS: Twenty-eight male Sprague Dawley rats were randomly assigned to four groups: negative control (G1), positive control with RA but no treatment (G2), and two experimental groups treated wit

Boosting PRRSV-Specific Cellular Immunity: The Immunological Profiling of an Fc-Fused Multi-CTL Epitope Vaccine in Mice.

The continuously evolving PRRSV has been plaguing pig farms worldwide for over 30 years, with conventional vaccines suffering from insufficient protection and biosecurity risks. To address these challenges, we identified 10 PRRSV-specific CTL epitopes through enzyme-linked immunospot assay (ELISPOT) and constructed a multi-epitope peptide (PTE) by linking them in tandem. This PTE was then fused with a modified porcine Fc molecule to create the recombinant protein pFc-PTE. Our findings indicate that pFc-PTE effectively stimulates PRRSV-infected specific splenic lymphocytes to secrete high level

IL-10 is not required for the alleviation of collagen-induced arthritis by non-lethal malarial infection in mice.

We previously reported that Plasmodium yoelii 17XNL (Py), a non-lethal rodent malarial parasite, could suppress collagen-induced arthritis (CIA) and increase the production of T cell-derived interleukin (IL)-10. However, it remained unclear whether IL-10 is essential for the Py-induced suppression of CIA. Male IL-10 knockout (KO) DBA/1 J mice were immunized with bovine type II collagen (CII) and subsequently infected with Py at one week post-immunization. The development of arthritis was evaluated by an arthritis score up to 6 weeks post-immunization. At 3 weeks post-immunization, cytokine pro

Downregulation of IRF7-mediated type-I interferon response by LmCen-/- parasites is necessary for protective immunity.

Leishmaniasis is a tropical disease caused by Leishmania parasites and currently has no licensed vaccines. We developed a dermotropic Leishmania major centrin gene-deleted strain (LmCen-/-) as a live attenuated vaccine. Recent studies have shown that type I interferons (IFNs) play important roles in immunity to parasitic and viral pathogens. However, their relevance in protective immunity following vaccination is not understood. We found that immunization with LmCen-/- induces a transient increase in type I IFN response along with its regulatory factor IRF7 that is downregulated 7-21 days post

Inhibitory effect of baricitinib on microglia and STAT3 in a region with a weak blood-brain barrier in a mouse model of rheumatoid arthritis.

OBJECTIVES: In patients with RA, baricitinib not only improves arthritis symptom severity, but also patients' neuropsychological symptoms, such as depression and fatigue. However, the cellular mechanisms through which baricitinib can affect neural activity is unexplored. While the blood-brain barrier (BBB) permeability of this drug remains unclear, Janus kinase inhibitors (JAKi) might reach the area postrema, which is a unique brain region with a weak BBB function. Our recent study demonstrated microglial activation during experimental arthritis in the area postrema. Therefore, we sought to as

Inclusion of a dual signal sequence enhances the immunogenicity of a novel viral vectored vaccine against the capsular group B meningococcus.

BACKGROUND: Disease caused by the capsular group B meningococcus (MenB) is the leading cause of infectious death in UK infants. A novel adenovirus-based vaccine encoding the MenB factor H binding protein (fHbp) with an N-terminal dual signal sequence induces high titres of protective antibody after a single dose in mice. A panel of N-terminal signal sequence variants were created to assess the contribution of components of this sequence to transgene expression kinetics of the encoded antigen from mammalian cells and the resultant effect on immunogenicity of fHbp. RESULTS: The full-length sign

Intervention of Tyrosine Hydroxylase Expression Alters Joint Inflammation and Th17/Treg Imbalance in Collagen-Induced Arthritis.

BACKGROUND/AIMS: Neuroendocrine dysregulation has been associated with rheumatoid arthritis (RA). Tyrosine hydroxylase (TH), a rate-limiting enzyme for synthesis of neuroendocrine hormones such as epinephrine, is also expressed in T lymphocytes and regulates balance between helper T (Th) 17 cells and regulatory T (Treg) cells. Herein, we aimed to show that TH expression in joints alleviates joint inflammation and Th17/Treg imbalance in collagen-induced arthritis (CIA), an animal model of RA, and these effects may be implemented by the mechanism of epinephrine action on α1-adrenoreceptor (α1-AR

The shared susceptibility epitope of HLA-DR4 binds citrullinated self-antigens and the TCR.

Individuals expressing HLA-DR4 bearing the shared susceptibility epitope (SE) have an increased risk of developing rheumatoid arthritis (RA). Posttranslational modification of self-proteins via citrullination leads to the formation of neoantigens that can be presented by HLA-DR4 SE allomorphs. However, in T cell-mediated autoimmunity, the interplay between the HLA molecule, posttranslationally modified epitope(s), and the responding T cell repertoire remains unclear. In HLA-DR4 transgenic mice, we show that immunization with a Fibβ-74cit69-81 peptide led to a population of HLA-DR4Fibβ-74cit69-

Driving β2- While Suppressing α-Adrenergic Receptor Activity Suppresses Joint Pathology in Inflammatory Arthritis.

OBJECTIVE: Hypersympathetic activity is prominent in rheumatoid arthritis, and major life stressors precede onset in ~80% of patients. These findings and others support a link between stress, the sympathetic nervous system and disease onset and progression. Here, we extend previous research by evaluating how selective peripherally acting α/β2-adrenergic drugs affect joint destruction in adjuvant-induced arthritis. METHODS: Complete Freund's adjuvant induced inflammatory arthritis in male Lewis rats. Controls received no treatment. Arthritic rats then received vehicle or twice-daily treatment

Sustained microglial activation in the area postrema of collagen-induced arthritis mice.

BACKGROUND: Central nervous system (CNS)-mediated symptoms, such as fatigue, depression, and hyperalgesia, are common complications among patients with rheumatoid arthritis (RA). However, it remains unclear how the peripheral pathology of RA spreads to the brain. Accumulated evidence showing an association between serum cytokine levels and aberrant CNS function suggests that humoral factors participate in this mechanism. In contrast to the well-known early responses of microglia (CNS-resident immune cells) in the area postrema [AP; a brain region lacking a blood-brain barrier (BBB)] to experim

Arsenic trioxide ameliorates experimental autoimmune encephalomyelitis in C57BL/6 mice by inducing CD4+ T cell apoptosis.

BACKGROUND: Multiple sclerosis (MS) is an immune-mediated disease of the central nervous system characterized by severe white matter demyelination. Because of its complex pathogenesis, there is no definite cure for MS. Experimental autoimmune encephalomyelitis (EAE) is an ideal animal model for the study of MS. Arsenic trioxide (ATO) is an ancient Chinese medicine used for its therapeutic properties with several autoimmune diseases. It is also used to inhibit acute immune rejection due to its anti-inflammatory and immunosuppressive properties. However, it is unclear whether ATO has a therapeut
