Effects of air pollution on disease activity in patients with rheumatoid arthritis.

OBJECTIVES: This study aimed to evaluate the associations of air pollution with disease activity and flare rate in patients with rheumatoid arthritis (RA). METHODS: This prospective cohort study included patients with RA who were treated at a tertiary medical centre in South Korea between January 2021 and December 2024. Air pollution exposure was estimated using monthly mean concentrations of 6 air pollutants (sulfur dioxide, nitrogen dioxide, ozone, carbon monoxide, particulate matter [PM]10, and PM2.5). Disease activity and flares were recorded longitudinally at each outpatient visit. Assoc

Conclusions and Therapeutic Implications of AA.

AA is essential for several tissues in the body, as they are involved in many physiological and pathological processes. Its regulatory role in inflammation, immune response regulation, cell cycle, and membrane structural organization and its ability to control the expression and function(s) of several receptors and convey messages to and from the nucleus are of particular significance. Fine-tuning the metabolism of AA may lead to the development of prevention and management strategies for several diseases, including but not limited to sepsis, inflammatory bowel disease, diabetes mellitus, canc

AA in Rheumatoid Arthritis and Lupus.

Inflammation and immune dysregulation are at the center of various diseases, including autoimmune diseases, especially rheumatoid arthritis (RA) and systemic lupus erythematosus (lupus). In general, corticosteroids, disease-modifying drugs, and monoclonal antibodies against IL-6 and TNF-α, JAK inhibitors, and various other immunomodulators are extensively used in the treatment of RA and lupus. It is known that EFA (essential fatty acid) metabolism, especially that of AA, is altered in RA and lupus. It is proposed that metabolism of AA and of other PUFAs may play a role in predicting disease pr

Global, regional, and national burden of exposure to chewing tobacco, 1990-2023: a systematic analysis of the Global Burden of Disease Study 2023.

BACKGROUND: Exposure to chewing tobacco is a known health risk factor but has received less attention than smoking in global tobacco control efforts. As the evidence base linking chewing tobacco use to poor health grows, it is essential to closely track the changing use of chewing tobacco distinct from other forms of smokeless tobacco, as well as the estimated burden associated with its use. These data are essential to inform policy makers who seek to identify and avert the health burden associated with these dangerous products. METHODS: We estimated the prevalence of chewing tobacco use and

Quantification of Chikungunya Virus RNA by Real-Time RT-PCR Assay.

RT-qPCR is characterized by‌ high sensitivity, strong specificity, excellent reproducibility, and operational simplicity, ‌enabling‌ rapid quantification of viral RNA within a short timeframe. The real-time PCR system relies upon the measurement of a fluorescent reporter during PCR, in which the amount of emitted fluorescence is directly proportional to the amount of the PCR product in a reaction. Here, we describe the use of TaqMan® probe-based one-tube Real-time reverse transcription quantitative polymerase chain reaction (RT-qPCR) for the detection and quantification of the Chikungunya viru

Screening of Antiviral Agents Against CHIKV Using Reporter Virus.

Chikungunya virus (CHIKV) causes a disease characterized by chronic musculoskeletal inflammation for which specific antivirals are not yet available. Currently, a supportive therapy to alleviate fever and pain is used, but it does not limit viral replication or the persistence of chronic arthritis symptoms. Thus, the identification and development of new active molecules against CHIKV is urgently needed. Here, we present a cell-based methodology that enables the implementation of a rapid and cost-effective strategy for high- and medium-throughput screening (HTS) of compounds, including repurpo

Mechanisms underlying rheumatic heart disease immune-mediated pathogenesis: T-cell responses, cytokine networks, and immune dysregulation as drivers of valvular injury.

Acute rheumatic fever is an immune-mediated sequela of Streptococcus pyogenes (group A Streptococcus) infection, presenting with fever, arthritis, chorea, rash, and, crucially, carditis. Without effective secondary prophylaxis and care, acute rheumatic fever can progress to chronic rheumatic heart disease, a leading yet under-recognised cause of cardiovascular morbidity and death that disproportionately affects children and young adults aged 5-24 years in low-resource settings. Robust evidence implicates the role of antigenic mimicry-particularly between streptococcal M protein and cardiac or

Association between preoperative radiographic osteoarthritis severity and patient-reported outcomes after total hip arthroplasty: a systematic review and meta-analysis.

PURPOSE: Although total hip arthroplasty (THA) reliably improves pain and function in patients with hip osteoarthritis (OA), a subset of patients experience limited postoperative benefit. The association between preoperative radiographic OA severity and postoperative patient-reported outcomes (PROMs) remains controversial. METHODS: We performed a systematic review and meta-analysis according to PRISMA guidelines. Electronic databases (MEDLINE, Embase, and Cochrane Library) were searched for studies comparing postoperative PROMs after primary THA between patients with moderate versus severe ra

Simultaneously leveraging individual- and summary-level external data in clinical trials.

With the growing availability of external data (e.g., real-world data (RWD) and historical data), there is increasing interest in integrating external data into Randomized Controlled Trials (RCT), especially for rare diseases with high unmet medical needs where traditional clinical trials would be impractical and/or unethical. However, despite the increasing availability of external data, the level of detail varies across sources, as some provide individual-level patient records while others offer only aggregated summary statistics, often due to privacy and regulatory constraints. Most existin

Identification of human tooth germ ectomesenchymal stem cells.

Cranial neural crest cells (CNCCs) contribute to most cranio-maxillofacial mesenchymal tissues, including the dental ectomesenchyme. However, the developmental relationships between human CNC-derived progenitors and the odontogenic lineage remain incompletely understood. Here, we mapped the single-cell transcriptomic landscape of prenatal human mandibles and reconstructed the putative differentiation trajectories of CNC-derived ectomesenchymal populations. Notably, we identified a distinct population of SLITRK6+ cells enriched in the odontogenic ectomesenchyme. These cells were detected in pre

Site-specific m1A Installation by the MTR1 Ribozyme and Modulation of Mitochondrial mRNA Translation in vitro.

RNA modifications play essential roles in regulating RNA structure, stability, and translation. Among these, m1A is a functionally important modification that influences Watson-Crick base pairing and translational fidelity. However, approaches for site-specific installation and func tional interrogation of m1A in RNA remain limited. Here, we characterize the catalytic properties of the methyltransferase ribozyme MTR1 with a focus on donor compatibility, sequence-dependent kinetics, and activity on structured RNA substrates. We show that MTR1 catalyzes alkyl transfer from O6-methylguanine- deri

Auditory Biofeedback Gait Training in Individuals With Chronic Ankle Instability: Protocol for a Randomized Controlled Trial.

BACKGROUND: Chronic ankle instability (CAI) develops in a large majority of patients with lateral ankle sprain (LAS), who often experience recurring injuries, residual feelings of instability, and abnormal neuromuscular patterns. Specifically, gait alterations are common in those with CAI and considered a contributing factor to poor long-term ankle joint health. Yet, there are no evidence-based gait-training protocols targeted to improve biomechanics, which may improve long-term outcomes related to ankle joint health and patient satisfaction. OBJECTIVE: This study protocol introduces a progre

Non-essential metal mixture and depressive symptoms in chinese older adults: cross-sectional associations and exploratory role of interleukin-6.

Exposure to non-essential metals (NEMs) has been linked to depressive symptoms, but evidence in older adults and the potential role of inflammatory pathways remain limited, particularly for metal mixture. This cross-sectional study aimed to evaluate individual and joint associations of six blood NEMs with depressive symptoms and to exploratorily assess whether interleukin-6 (IL-6) statistically accounts for part of the observed associations. Data were derived from 4076 community-dwelling Chinese older adults in a cross-sectional survey. Six NEMs (barium, cadmium, aluminum, gallium, mercury, th

Interaction between Trichoderma harzianum qt40003 and Bacillus velezensis BV25 Triggers Harzianopyridone Biosynthesis, Enhancing Biological Control Efficacy against Fusarium oxysporum f. sp. Conglutinans.

Cabbage Fusarium wilt, caused by Fusarium oxysporum f. sp. conglutinans (FOC), is a destructive soil-borne disease. Herein, we deciphered how coculture of the bacterium Bacillus velezensis BV25 with the fungus Trichoderma harzianum qt40003 activates expression of the harzianopyridone (HPY) biosynthetic gene cluster, thereby enhancing biocontrol against FOC. Greenhouse assays showed the coculture significantly enhanced biocontrol efficacy compared with single-strain treatments, reducing disease index from 88 to 26. Comparative transcriptomic analysis revealed that coculture specifically trigger

Absolute and Amplified Virus Detection by Isotope Dilution after Catalytic Hairpin Assembly.

The detection of viruses is of great significance for the diagnosis of related diseases. However, current virus detection often faces two challenges: first, due to the complex matrix of physiological samples, detection methods often fail to provide accurate quantitative results; second, conventional PCR-related signal amplification procedures are often challenged by issues such as primer interference and non-specific amplification. Herein, we developed an absolute and amplified virus detection strategy by coupling isotope dilution and catalytic hairpin assembly (CHA). The high-risk cervical ca

Climate Factors, Air Pollutants, and Extreme Events Shape Influenza Risk in China: A Hospital-Based Case-Crossover and Machine-Learning Study.

Although the link between seasonal influenza and environment has been established, the heterogeneous impacts of climate extreme events with their interactions remain poorly understood. Based on 630,120 influenza-positive cases from hospitals in 335 Chinese cities (2005-2019), this case-crossover study adopted distributed lag nonlinear models to quantify exposure-lag-response relationships between environmental exposures and influenza, applied machine learning to rank dominant variables, and performed a Long Short-Term Memory (LSTM) model using Beijing as proof of concept to test the forecastin

Spatial Control of Cleavage Site Accessibility via 12S rRNA Gating on Tetrahedral Nucleic Acid Scaffold Enables Mitochondrial APE1 Imaging for In Vivo Neuroblastoma Risk Stratification.

Mitochondrial apurinic/apyrimidinic endonuclease 1 (APE1) mislocalization is closely linked to tumor progression and prognosis. Yet, its ubiquitous distribution across mitochondria, cytoplasm, and nucleus makes specific in situ imaging of mitochondrial APE1 highly challenging. Herein, we fabricate a sequentially activated biosensor (C-12S-tFNA) with 12S rRNA-gated spatial regulation of APE1 cleavage site (apurinic/apyrimidinic site, AP site) accessibility, where cytochrome c (cyt c) acts as the targeting motif to drive precise mitochondrial enrichment. In the initial state, the AP site is stru
