Effects of air pollution on disease activity in patients with rheumatoid arthritis.

OBJECTIVES: This study aimed to evaluate the associations of air pollution with disease activity and flare rate in patients with rheumatoid arthritis (RA). METHODS: This prospective cohort study included patients with RA who were treated at a tertiary medical centre in South Korea between January 2021 and December 2024. Air pollution exposure was estimated using monthly mean concentrations of 6 air pollutants (sulfur dioxide, nitrogen dioxide, ozone, carbon monoxide, particulate matter [PM]10, and PM2.5). Disease activity and flares were recorded longitudinally at each outpatient visit. Assoc

Conclusions and Therapeutic Implications of AA.

AA is essential for several tissues in the body, as they are involved in many physiological and pathological processes. Its regulatory role in inflammation, immune response regulation, cell cycle, and membrane structural organization and its ability to control the expression and function(s) of several receptors and convey messages to and from the nucleus are of particular significance. Fine-tuning the metabolism of AA may lead to the development of prevention and management strategies for several diseases, including but not limited to sepsis, inflammatory bowel disease, diabetes mellitus, canc

AA in Rheumatoid Arthritis and Lupus.

Inflammation and immune dysregulation are at the center of various diseases, including autoimmune diseases, especially rheumatoid arthritis (RA) and systemic lupus erythematosus (lupus). In general, corticosteroids, disease-modifying drugs, and monoclonal antibodies against IL-6 and TNF-α, JAK inhibitors, and various other immunomodulators are extensively used in the treatment of RA and lupus. It is known that EFA (essential fatty acid) metabolism, especially that of AA, is altered in RA and lupus. It is proposed that metabolism of AA and of other PUFAs may play a role in predicting disease pr

Global, regional, and national burden of exposure to chewing tobacco, 1990-2023: a systematic analysis of the Global Burden of Disease Study 2023.

BACKGROUND: Exposure to chewing tobacco is a known health risk factor but has received less attention than smoking in global tobacco control efforts. As the evidence base linking chewing tobacco use to poor health grows, it is essential to closely track the changing use of chewing tobacco distinct from other forms of smokeless tobacco, as well as the estimated burden associated with its use. These data are essential to inform policy makers who seek to identify and avert the health burden associated with these dangerous products. METHODS: We estimated the prevalence of chewing tobacco use and

Screening of Antiviral Agents Against CHIKV Using Reporter Virus.

Chikungunya virus (CHIKV) causes a disease characterized by chronic musculoskeletal inflammation for which specific antivirals are not yet available. Currently, a supportive therapy to alleviate fever and pain is used, but it does not limit viral replication or the persistence of chronic arthritis symptoms. Thus, the identification and development of new active molecules against CHIKV is urgently needed. Here, we present a cell-based methodology that enables the implementation of a rapid and cost-effective strategy for high- and medium-throughput screening (HTS) of compounds, including repurpo

Mechanisms underlying rheumatic heart disease immune-mediated pathogenesis: T-cell responses, cytokine networks, and immune dysregulation as drivers of valvular injury.

Acute rheumatic fever is an immune-mediated sequela of Streptococcus pyogenes (group A Streptococcus) infection, presenting with fever, arthritis, chorea, rash, and, crucially, carditis. Without effective secondary prophylaxis and care, acute rheumatic fever can progress to chronic rheumatic heart disease, a leading yet under-recognised cause of cardiovascular morbidity and death that disproportionately affects children and young adults aged 5-24 years in low-resource settings. Robust evidence implicates the role of antigenic mimicry-particularly between streptococcal M protein and cardiac or

Role of the GAPDH-mediated glycolysis-senescence axis in osteoarthritis chondrocytes and its modulation by semaglutide.

OBJECTIVES: This study aimed to identify key regulatory genes involved in chondrocyte senescence in osteoarthritis (OA) and to evaluate the therapeutic potential of the GLP-1 receptor agonist semaglutide. METHODS: Transcriptomic data of chondrocytes from OA and control groups were collected. Batch effects were corrected using Harmony, and differential expression analysis was performed with DESeq2 to identify significantly altered genes. Functional enrichment and pathway involvement were assessed using KEGG and Gene Set Enrichment Analysis (GSEA). Weighted gene co-expression network analysis (

Transcutaneous electrical nerve stimulation changed the relative abundance of intestinal microbiota in rats with knee osteoarthritis: a potential treatment for KOA.

BACKGROUND: Knee osteoarthritis (KOA) is a common degenerative bone disease, and transcutaneous electrical nerve stimulation (TENS) is an alternative and complementary therapy (ACM). This study revealed the role of TENS in regulating the intestinal microbiota in KOA rats. METHODS: This study concentrated on the intestinal microbiota of KOA rats which were treated with TENS for 1, 2, and 3 weeks. Three intensities of TENS were used to treat KOA rats, and the expressions of IL6/8, PI3K-AKT were measured. The intestinal microbiota was analyzed by 16 S rDNA sequencing. RESULTS: Compared with the

Hip range of motion is differentially associated with pain sensitivity in individuals with and without knee osteoarthritis.

BACKGROUND: Knee osteoarthritis (KOA) is a multifactorial condition influenced by both mechanical and neurophysiological factors. Hip joint mobility may be associated to knee joint loading and pain processing; however, the relationships between hip range of motion (ROM), pain sensitivity, and muscle torque in individuals with KOA remain unclear. This study aimed to investigate the association between knee pain sensitivity and hip rotator ROM in individuals with and without KOA. Additionally, it examined the association between hip rotation ROM and muscle torque and compared hip ROM between gro

Screening active small-molecule components from wutou decoction for rheumatoid arthritis treatment via intestinal barrier repair.

Disruption of intestinal barrier integrity is increasingly recognized as an important contributor to the initiation and progression of rheumatoid arthritis (RA), making intestinal barrier protection a potential therapeutic strategy. Wutou Decoction (WTD), a traditional Chinese medicine prescription, has been reported to exert anti-inflammatory and immunoregulatory effects and to ameliorate RA-associated intestinal barrier dysfunction. However, the small-molecule constituents responsible for its intestinal barrier-protective effects remain largely unclear. In this study, the intestine was consi
