STING inhibition by H-151 is associated with amelioration of psoriasis severity via modulation of pathogenic T helper cell subsets.

Psoriasis is a chronic immune-mediated skin disorder characterized by dysregulated activation of T helper (Th) cells. While Th1 and Th17 subsets are well-established contributors to disease pathology, the role of Th9 cells remains poorly understood. To investigate the relationship between disease severity and Th cell dynamics, we established a time-course imiquimod (IMQ)-induced psoriasis mouse model. Prolonged IMQ application resulted in progressive worsening of psoriatic inflammation, accompanied by increased populations of Th1, Th17, and Th9 cells, along with elevated levels of their associ

Mechanistic insights into psoriasis and type 2 diabetes mellitus comorbidity - Implications for treatment: A review.

Psoriasis is a chronic systemic inflammatory disease primarily affecting the skin, yet it is increasingly recognized for its systemic implications, particularly its strong association with type 2 diabetes mellitus (T2DM). This review synthesizes recent mechanistic and clinical evidence to elucidate the shared pathways linking psoriasis and T2DM, as well as to explore therapeutic strategies for this comorbidity. We conducted a narrative review of studies published between January 2020 and October 2025, encompassing preclinical models, clinical trials, and high-quality reviews that address patho

The path to interception in psoriatic disease: from conceptual clarity to clinical translation.

Psoriatic arthritis develops in up to one-third of individuals with psoriasis, typically following a prolonged subclinical phase. Diagnostic delays are common, often exceeding 2 years, and can result in irreversible joint damage. The growing recognition of this latent period has fuelled interest in earlier identification and interception. However, efforts are hampered by inconsistent definitions of early or subclinical psoriatic arthritis, insufficient prognostic tools, and an absence of consensus on the outcome for interception studies. This Review synthesises a rapidly evolving field, offeri

Advancements in psoriasis classification using custom transfer learning algorithms.

Psoriasis is a skin disorder which mainly occurs as a rash, scaly areas and an itchy skin. The symptoms usually occur on the chest, elbows, and the scalp. World Health Organization (WHO) reports that about 125 million individuals in the world, that is about two to three% of the world population, live with psoriasis. Moreover, approximately 30% of psoriasis patients can have psoriatic arthritis. Psoriasis can have a detrimental effect on the quality of life of the affected people. They can also deal with several chronic diseases, such as depression, metabolic syndrome, cardiovascular diseases,

Building the foundations for an international patient-centred outcomes set for psoriasis: A scoping study.

BACKGROUND: Value-based healthcare emphasizes outcomes that matter to patients, and patient-centred outcomes sets are vital to its success. For psoriasis, initial work proposed a patient-centred outcomes set in Belgium, but it requires further validation to ensure international applicability. OBJECTIVES: This scoping study aimed to refine the outcomes set in collaboration with patient representatives and dermatologists, in preparation for international validation through a Delphi consensus process. METHODS: An international Working Group of patient representatives and dermatologists was esta

Dual targeted therapy with biologic agents and small molecules in refractory inflammatory arthritis: clinical outcomes and safety profile.

Biologic agents and small molecules have transformed the management of rheumatoid arthritis (RA), psoriatic arthritis (PsA), and spondyloarthritis (SpA). Nevertheless, some patients experience persistent, uncontrolled inflammation despite multiple targeted therapies. Evidence for dual targeted therapy (DTT) remains scarce. This study aims to describe the clinical characteristics, therapeutic rationale, and outcomes of patients with severe, multi-drug-refractory inflammatory arthritis and autoinflammatory disease treated with DTT. This clinical case series included patients with RA, PsA, FMF-as

Cycling or Swapping in Spondyloarthritis? Current Knowledge.

Psoriatic arthritis (PsA) and axial Spondyloarthritis (axSpA) share some common therapeutic options, namely TNF-inhibitors, anti-IL-17 and JAK-inhibitors. Despite the wealth of choices, often patients do not respond to the first b/tsDMARDs, but they have to switch to another one, either of the same therapeutic category (cycling) or of another (swapping). Which of the abovementioned strategies is better is ill-defined thus far. In this narrative review, we aimed to critically present the relevant literature. PubMed was searched from inception to June 2025 using the following keywords: "Psoriati

The Pathophysiological Significance of Dual Inhibition of IL-17A and IL-17F in Psoriatic Arthritis and Axial Spondyloarthritis.

OBJECTIVE: Spondylarthritides (SpA), including psoriatic arthritis (PsA) and axial spondyloarthritis (axSpA) are immune-mediated diseases in which the IL-23/IL-17 axis plays a central role. IL-17A inhibition is already a well-established and effective treatment strategy. Emerging evidence suggests that IL-17F, a cytokine closely related to IL-17A but less potent, is not a passive bystander but a key participant in chronic sustained inflammation and therapeutic escape, offering a compelling concept for dual inhibition. This review aims to examine the pathophysiological significance of dual inhi

Topical delivery of high-drug-loading nanoparticle gels for psoriasis treatment.

Psoriasis is a chronic, immune-mediated skin disease characterised by epidermal hyperplasia and compromised barrier integrity, which significantly complicates effective drug delivery. Topical drug delivery (TDD) offers a promising, non-invasive, and patient-centric alternative therapy for its management. However, the efficacy of TDD is constrained by the markedly thickened stratum corneum in psoriatic lesion skin, which acts as a formidable barrier to effective drug penetration. In this work, we developed a high-drug-loading TDD system for the highly effective topical delivery of apremilast (A

Attritional Carpal Dislocation and Flexor Tendon Rupture as a Rare Complication of Conservatively Managed Distal Radius Fracture in a Rheumatoid Patient.

Distal radius malunions are known in rare instances to present a risk of tendon rupture. Rheumatoid arthritis and other inflammatory arthropathies also place patients at-risk of attritional tendon rupture as a sequelae of tendon degeneration from the underlying disease process. In this case report, we present a patient with psoriatic arthritis who sustained a distal radius fracture malunion after initially electing to pursue nonsurgical management, subsequently sustaining attritional dorsal carpal dislocation and ruptures of multiple flexor tendons 3 years after her initial injury resulting in

Discovery of Potent PDE4 Inhibitors with 3(2H)-Pyridazinone Scaffold: Synthesis, In Silico Studies and In Vitro/Vivo Evaluation.

Phospodiesterase 4 (PDE4) has long been an attractive target not only for the anti-inflammatory therapy in respiratory diseases, but also for other pathologies such as psoriatic arthritis and atopic dermatitis. In this study, we report the synthesis of 5-acetyl-2-ethyl-6-phenyl-3(2H)-pyridazinones differently substituted at position 4 with a variety of aryl/alkylamines, which act as potent PDE4B1 inhibitors in the low nanomolar range. The selectivity toward PDE4A4, PDE4D3 and HARBS, as well as the ability to inhibit TNFα production in human whole blood (hWB), was also evaluated for the most po
