BACKGROUND: Hepatitis C virus (HCV) diagnosis usually requires detection of antibody followed by HCV RNA. The requirement for 2 tests leads to major drop-offs in the cascade of care. Existing near-care HCV RNA tests have slow turnaround time and are expensive with limited availability. We aim to develop a cost-effective, rapid, and sensitive test for detection of HCV RNA to enhance screening, particularly in marginalized and remote populations.
METHODS: After RNA extraction from plasma, HCV RNA is reverse-transcribed and amplified using loop-mediated isothermal amplification with HCV-specific

The host-hepatitis C virus (HCV) interaction determines whether the acute phase of HCV infection will undergo complete resolution or progress to the development of viral persistence and, ultimately, chronic HCV infection. The host cell mechanism that fights against the virus culminates in the production of interferons (IFNs), IFN-stimulated genes, and cytokines as well as the induction of autophagy and apoptosis. Innate immune responses modulate adaptive immune responses. T cells often fail and the virus persists as a result of T-cell exhaustion and the emergence of viral escape mutations. Exh

Hepatitis E virus (HEV), a leading cause of acute and chronic viral hepatitis, poses a persistent global health challenge. A deeper mechanistic understanding of virus-host interactions is critical for identifying therapeutic targets to mitigate HEV-associated disease. In this study, we employ a systems biology framework to comprehensively map metabolic and bioenergetic alterations induced by HEV genotypes 1 and 3 in HepG2/C3a-MAVS-KD cells, a robust model of HEV infection, enabling reliable assessment of virus- and host-driven cellular changes. Our analyses reveal extensive remodelling of host

This study investigates the impact of the indirect bilirubin percentage-to-albumin ratio (iBAR) on the prognosis of patients with acute-on-chronic liver failure (ACLF), as defined by the Chinese Group on the Study of Severe Hepatitis B-ACLF (COSSH ACLF) criteria, who were treated with an artificial liver support system (ALSS). In a retrospective cohort of 258 eligible patients, restricted cubic splines, linear regression, and Cox proportional hazards models were used to analyse the association of iBAR with disease severity and 28-day and 90-day outcomes. The 28-day transplant-free and overall

Hepatitis E virus (HEV) is a leading cause of acute viral hepatitis, yet the virus has been associated with extrahepatic complications, including neurological and renal disorders. These extrahepatic manifestations pose challenges for the diagnosis and treatment of HEV and are considered a reservoir for disease relapse. In addition to recent in vivo patient data, several animal and in vitro models have been established in the last few years, confirming the broad HEV tissue tropism. However, it remains unclear whether extrahepatic complications arise from viral replication within the tissue or f

Swine acute diarrhea syndrome coronavirus (SADS-CoV) is an emerging bat-origin alphacoronavirus causing severe disease in neonatal piglets, with significant economic losses to the swine industry. The virus exhibits a broad species tropism, infecting cells derived from pigs, humans, and mice, highlighting its potential for cross-species transmission. Due to drawbacks associated with the use of young piglets, there is a need for an appropriate small animal model to study SADS-CoV biology. Here we established a mouse infection model based on a murinized mutant of the virus, mSADS-CoV, in which th

Male infertility poses a substantial healthcare challenge and severely impacts the lives of patients. We aimed to investigate the risk factors for infertility and abnormal semen parameters. We conducted a comprehensive search of the articles published in Web of Science, MEDLINE, and Embase databases from January 2000 to February 2025. Infertility, semen volume, sperm concentration, sperm count, sperm morphology, sperm motility, and sperm progressive motility were used as endpoints to evaluate the relevance of risk factors. A total of 43 studies were included, covering 67 risk factors associate

Despite the success of vaccination and small-molecule antivirals in significantly reducing mortality from acute viral infections, chronic viral infections such as human immunodeficiency virus (HIV), hepatitis B virus (HBV), cytomegalovirus (CMV), and Epstein–Barr virus (EBV) remain persistent global health burdens. Latent reservoirs, drug resistance mutations, and immune evasion mechanisms enable these viruses to persist, highlighting the need for novel therapeutic approaches. Chimeric antigen receptor T-cell (CAR-T) therapy is an emerging immunotherapeutic strategy, initially developed to tre

Hepatitis E, caused by the hepatitis E virus (HEV), represents an emerging global public health problem, constituting one of the main causes of acute viral hepatitis. Since its first description, it has been considered a disease with an epidemiological pattern linked to the consumption of contaminated water and food, and with a higher prevalence in geographical areas with poor socio-sanitary conditions. According to WHO estimates, around 2 billion people, representing a quarter of the world's population, live in areas where HEV is endemic and are therefore at risk of infection. Improvements in

Hepatitis B virus (HBV) infection remains a significant public health challenge in Florida despite advancements in prevention and treatment. This review analyses the current epidemiology, disease burden and management strategies for hepatitis B in Florida as of 2025, drawing on state surveillance data and recent public health reports. Florida continues to experience higher rates of both acute and chronic hepatitis B compared to national averages, with pronounced racial, ethnic and geographic disparities. Rural areas face disproportionately high rates of acute infections, while chronic infectio

Acute-on-chronic liver failure (ACLF) represents a rapidly progressive deterioration in chronic liver disease (CLD) marked by multi-organ dysfunction and high short-term mortality. Although consensus on a single definition remains elusive, the clinical phenotype is increasingly recognized across diverse populations, with regional differences in underlying liver disease and precipitants influencing diagnostic criteria and reported incidence. Bacterial infections, alcohol-associated hepatitis (AH), viral hepatitis flares, and drug-induced liver injury (DILI) are among the most common intrahepati

Massive hepatic necrosis (MHN), characterised by extensive loss of hepatocytes, represents the most severe pathological lesion in acute liver failure (ALF). MHN-associated ALF primarily occurs in patients with acute viral hepatitis A, B or E infections. Additionally, MHN-associated ALF can develop in patients with autoimmune hepatitis or in those taking idiosyncratic drugs. MHN-associated ALF is associated with significantly higher mortality than that caused by other aetiologies. In contrast to other forms of ALF, liver regeneration following MHN depends on liver progenitor cells (LPCs)-the sm

Most common adverse effect causing cessation of anti-tubercular treatment (ATT) is drug-induced liver injury (DILI) which is unpredictable due to its idiosyncratic nature. ATT is the most common cause of DILI and drug-induced acute liver failure (ALF) in South East Asia. Spectrum of ATT-DILI ranges from asymptomatic raised transaminases to acute hepatitis to acute liver failure (ALF). ALF due to ATT has a more aggressive course with up to 70% mortality. Both modifiable and non-modifiable risk factors are involved. Increasing age, female gender, genetic predisposition, poor nutrition, underlyin

BACKGROUND: Hepatitis B virus-related acute-on-chronic liver failure (HBV-ACLF) is associated with dyslipidemia and inflammatory responses. However, the association between a novel comprehensive lipid parameter, the non-high-density lipoprotein cholesterol to high-density lipoprotein cholesterol ratio (NHHR), and clinical outcomes in HBV-ACLF remains unclear. This study aimed to investigate the relationship between NHHR and 28- and 90-day transplant-free (TF) mortality among patients with HBV-ACLF. METHODS: This study retrospectively enrolled 452 patients with HBV-ACLF from the First Affiliate

Hepatitis E virus (HEV) is an emerging zoonotic pathogen and the leading cause of acute viral hepatitis worldwide. Nevertheless, HEV remains largely underestimated and poorly controlled. HEV infects a broad range of host species, transmits through diverse routes and can cause chronic infections in immunocompromised individuals and severe hepatitis in pregnant women. Currently, there is no HEV-specific antiviral treatment available and the only licensed vaccine is restricted to a few countries. Taken together, this highlights its significance as a global health treat and underscores the urgent

BACKGROUND: Epstein-Barr virus infection is common. However, to our knowledge, it has never been reported to cause synchronous acute acalculous cholecystitis and acute pancreatitis alongside hepatitis. This case highly suggests the potential for novel multiorgan involvement.
CASE PRESENTATION: A 32-year-old Asian male patient presented with high-grade fever, abdominal pain, and jaundice. Comprehensive diagnostic evaluation confirmed acute hepatitis, acute acalculous cholecystitis, and acute pancreatitis. Histopathological examination of lymph node and liver biopsy specimens identified Epstein
