Spatially organized chemical defences shape microbial communities in honey bee colonies.

The success of social insects relies on dense colony living, but these conditions also favour pathogen transmission. Colonies mitigate this risk through social immunity, a coordinated system of behavioural, chemical, and immunological defences. Here, we investigate how chemical defences contribute to colony-level immunity using the European honey bee (Apis mellifera) as a model system. Integrating microbiome profiling, antimicrobial assays, and chemical characterization across hive components, we show that antimicrobial activity exhibits clear distance decay across compartments, closely parall

Mechanistic Landscape and Therapeutic Opportunities of Non-Coding RNAs in Modulating Fibroblast-Like Synoviocytes in Rheumatoid Arthritis.

Current disease-modifying antirheumatic drugs (DMARDs) for rheumatoid arthritis (RA) predominantly target systemic immune-inflammatory pathways, yet fail to effectively reverse progressive articular structural damage driven by fibroblast-like synoviocytes (FLS), representing a core unmet clinical need in the field. Non-coding RNAs (ncRNAs) are key epigenetic regulators of pathological FLS activation; however, the field is constrained by three core limitations: fragmented evidence, mechanistic homogenization, and a persistent basic-to-clinical translational gap. Adopting a structured evidence-g

How I Do It: An Imaging Approach and Review of Autoimmune Encephalitis.

Encephalitis is a medical emergency characterized by inflammation of the brain parenchyma due to an infective cause or an immune-mediated process. Autoimmune encephalitis (AE) is characterized by a misdirected immune attack against neuronal structures, resulting in an inflammatory response. Triggers for the development of antibodies are often paraneoplastic or parainfectious. AE may occur in children or adults with symptoms including subacute onset of altered mentation, cognitive issues, drug-resistant epilepsy, movement disorders, and/or autonomic dysfunction. Limbic encephalitis is a common

Distinct immune signatures define hepatitis D- versus hepatitis B-associated hepatocellular carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) is a leading cause of cancer-related mortality worldwide. Hepatitis D is the most severe form of viral hepatitis. However, the clinical, virological, and systemic inflammatory characteristics of HDV-associated HCC remain insufficiently characterized. METHODS: In this single-center retrospective study, we identified 162 patients with HCC due to chronic hepatitis D (HDV-HCC; n=54) or chronic hepatitis B (HBV-HCC; n=108). Plasma levels of 92 soluble inflammatory mediators (SIMs) were quantified using proximity extension assay in HCC patients [HBV-HCC (n

A tape-based intestinal chip co-culture model for assessing anti-inflammatory activity of chitosan oligosaccharides.

Intestinal inflammation is a central pathological process in gastrointestinal diseases, yet conventional in vitro models poorly reproduce epithelial-immune interactions, and accessible platforms for rapid anti-inflammatory screening remain lacking. Here, a low-cost, high-throughput tape-based intestinal chip co-culturing Caco-2 epithelial cells and RAW264.7 macrophages under dynamic flow was applied to approximate the epithelial-macrophage interface of the intestinal wall and to evaluate the anti-inflammatory activity of chitosan oligosaccharides (COS). Three COS fractions (COS1, COS2, COS3; 3

Comparison of the safety and 24-36-month efficacy of medial unicompartmental knee arthroplasty versus total knee arthroplasty in elderly patients (≥ 75 years) with isolated medial-compartment knee osteoarthritis.

The number of very elderly patients presenting with end-stage knee osteoarthritis (OA) is rising rapidly, yet the comparative merits of medial unicompartmental knee arthroplasty (UKA) and total knee arthroplasty (TKA) in this group remain insufficiently characterised. We compared the perioperative safety and short-term clinical and patient-reported efficacy of the two procedures in patients aged 75 years and older with isolated medial-compartment OA. This single-centre retrospective cohort study compared patients aged ≥ 75 years undergoing primary medial UKA or TKA for isolated medial-compartm

Adult-Onset Still Disease Triggered by Recent Cytomegalovirus Infection: A Diagnosis of Exclusion and a Review of the Literature.

Adult-onset Still disease (AOSD) is a rare systemic autoinflammatory disorder characterized by quotidian fever, inflammatory arthritis, evanescent rash, neutrophilic leukocytosis, and hyperferritinemia. Because no pathognomonic diagnostic test exists, diagnosis relies on characteristic clinical features and exclusion of infectious, malignant, and autoimmune disorders. Viral infections, including cytomegalovirus (CMV), have been proposed as potential immunologic triggers in genetically susceptible individuals, although causality remains unproven. We report a 47-year-old Hispanic woman with hype

Additive effects of active vitamin D3 or vitamin E administration on therapeutic lower-limb home exercise for knee osteoarthritis: A pilot prospective randomized controlled trial.

BACKGROUND: Quadriceps weakness, inflammation, and reduced physical function are common in knee osteoarthritis (OA), and lower limb exercise therapy is an established non-pharmacological treatment. Vitamin D3 (vit D3) and vitamin E (vit E) have been proposed as adjunctive therapies; however, clinical findings are inconsistent. We aimed to examine whether combining lower limb home exercises with oral active vit D3 or vit E supplementation provides additive benefits compared with home exercise alone in women with symptomatic knee OA. METHODS: Eighty-eight postmenopausal women with knee OA (Kell

Musculoskeletal Manifestations of Light Chain Amyloidosis: A Systematic Review.

INTRODUCTION: Diagnosing amyloid arthropathy remains challenging because its manifestations often mimic those of other inflammatory musculoskeletal diseases. This review aimed to describe the clinical, biological, and imaging characteristics of musculoskeletal involvement in light-chain (AL) Amyloidosis. METHODS: We conducted a review in accordance with the PRISMA guidelines. We searched SCOPUS and MEDLINE for case reports and series published between January 2000 and December 2023 that detailed musculoskeletal manifestations of AL amyloidosis. To minimize the risk of omitting relevant articl

Identification of Akkermansia muciniphila as a candidate gut commensal involved in the pathogenesis of rheumatoid arthritis.

OBJECTIVE: The "mucosal origins hypothesis" posits that mucosal sites, such as the gut, are initial sites where immune responses first develop that may, months to years later, result in clinical rheumatoid arthritis (RA). Our goal here was to conduct a discovery-based, hypothesis-generating study using samples from a relatively small number of patients to implicate previously unrecognized highly IgA-coated commensal flora that may have a role in RA pathogenesis (primary outcome). The secondary supporting outcomes were to determine whether HLA-DR-presented microbial peptides from such flora cou

Association of phenotypic and functional characteristics of regulatory T cells from minor salivary glands with expression of the delta 7 FoxP3 splice variant in patients with Sjögren's disease.

BACKGROUND: Sjögren's disease (SjD) is a systemic autoimmune disorder characterized by lymphoplasmacytic infiltration of exocrine glands, including minor salivary glands (MSG), with potential formation of ectopic germinal centers. Regulatory T cells (Tregs) are critical suppressors of autoimmune responses, and their dysfunction has been implicated in SjD pathogenesis. FoxP3, the master transcription factor of Tregs, undergoes alternative splicing to produce four variants: full-length (FL), Δ2, Δ7, and Δ2Δ7. While FoxP3 splice variants are known to influence Treg proliferation and suppressive a

Cystic pulmonary amyloidosis with lymphoid interstitial pneumonia in Sjögren's disease: a case report.

BACKGROUND: Interstitial lung diseases are commonly seen in association with Sjögren's disease, especially lymphoproliferative disorders, such as lymphoid interstitial pneumonia. In occasional and rare cases, pulmonary amyloidosis with cysts, nodules, and/or ground-glass opacites as radiological features, can also become a complication of Sjögren's disease. On the other hand, diffuse cystic lung disease, without nodules, concomitant with chylous fluid accumulation in female leads to initial suspicion of lymphangioleiomyomatosis. Interestingly, it's challenging to differentiate if diffuse cysti

In silico prioritization of potential matrix metalloproteinase-13 inhibitors for cartilage degeneration in knee injury-associated osteoarthritis.

Knee trauma is an important risk factor for post-traumatic osteoarthritis (PTOA), a progressive joint disease characterized by articular cartilage degeneration and limited disease-modifying treatment options. Matrix metalloproteinase-13 (MMP-13/collagenase-3) contributes to cartilage extracellular matrix degradation through cleavage of type II collagen, making it a relevant molecular target for cartilage-protective drug discovery. This study aimed to identify and prioritize potential small-molecule MMP-13 inhibitors using an integrated in silico workflow. The workflow included MMP-13 sequence

Lipid metabolism remodeling in human cardiomyocyte differentiation and maturation.

Cardiac development involves extensive metabolic remodeling, yet dynamic changes of endogenous lipid species during human cardiomyocyte (CM) differentiation remain poorly characterized. In this study, we systematically performed high-coverage lipidomic profiling across five consecutive CM differentiation stages, using high-performance liquid chromatography. MYL2 expression confirmed ventricular lineage specification in late-stage CMs. Ceramide levels peaked at the cardiac progenitor cell (CPC) stage, suggesting a stage-specific association with the mesendoderm (MES)-to-CPC transition. FAK Mediated Coordination of Extracellular Matrix Remodeling and Cellular Mechanics Impairs Nanomedicine Delivery in Solid Tumors.

Tumor extracellular matrix (ECM) remodeling and cell-intrinsic mechanical properties critically limit nanoparticle (NP) delivery, yet how these multiscale biophysical cues are coordinately regulated remains unclear. Here, we identify focal adhesion kinase (FAK/PTK2) as a central mechanobiological regulator that couples ECM remodeling with cellular mechanics to control NP transport and cellular uptake. The ECM derived from patients' tumors exhibited increased stiffness and collagen density, which severely restricted NP penetration, while PTK2 emerged as a key integrin-associated signaling hub l

Isolation of the symbiotic bacterium Brevundimonas aurantiaca and its potential role in modulating reproductive traits in Culex pipiens pallens.

BACKGROUND: Culex pipiens pallens is a significant vector of mosquito-borne diseases, and its autogenous trait exerts a critical impact on population expansion and disease transmission risks. Understanding the regulatory effect of gut-specific symbiotic bacteria on the autogeny of Cx. pipiens pallens will provide a microbial perspective to elucidate the mechanisms underlying the formation of autogenous reproductive strategies and provide a theoretical basis for the development of novel mosquito-borne disease control. METHODS: Autogenous Cx. pipiens pallens were utilized to isolate and culture
