Autoantibodies against interleukin-1 receptor antagonist in Still's disease in adult patients, data from a multicenter cohort study.

OBJECTIVES: Emerging evidence supports the presence of autoantibodies targeting interleukin-1 receptor antagonist (IL-1Ra), a regulator of IL-1 signaling, in adult-onset Still's disease (AOSD). However, existing data are limited by small cohorts and potential confounding due to prior exposure to IL-1 inhibitors. This study aimed to develop a simple and reproducible indirect ELISA for detecting anti-IL-1Ra antibodies and to assess their frequency and immunological features in IL-1 inhibitor-naïve AOSD patients. METHODS: Serum samples from IL-1 inhibitor-naïve AOSD patients (n=46) and healthy c

NTDB: a curated database of natural transformation machinery and regulatory genes in prokaryotes.

Natural transformation, genetically controlled by specialized machinery and regulatory genes, drives evolution and adaptation in prokaryotes. This process facilitates the spread of clinical traits such as antibiotic resistance. The DNA uptake specificity is mediated by conserved motifs found in certain naturally transformable species. Despite its evolutionary and clinical significance, dedicated resources for natural transformation have been lacking. Here, we present the Natural Transformation DataBase (NTDB) (https://bioinfo-mml.sjtu.edu.cn/NTDB/), which compiles information on machinery and

Association between ankylosing spondylitis and hearing loss: A Mendelian randomization study.

Observational studies have suggested a potential link between ankylosing spondylitis (AS) and hearing loss. However, this association remains controversial due to potential drug-induced ototoxicity from treatments. Therefore, we employed a Mendelian randomization (MR) approach to evaluate the association between AS and hearing loss from a genetic perspective. This MR study utilized summary data from genome-wide association studies on AS and hearing loss, including sensorineural hearing loss (SNHL) and conductive hearing loss (CHL). The primary analysis was conducted using the inverse-variance

Noninvasive ultrashort wave neuromodulation enhances glymphatic clearance to alleviate traumatic brain edema and Alzheimer's pathology.

The glymphatic system clears cerebral waste, yet strategies to enhance its function remain elusive. Here, we show that 40.68 MHz ultrashort wave (USW) rapidly enhances glymphatic bulk flow in healthy mice within 10 minutes. USW potentiates both glymphatic influx and efflux, concomitant with increased Delta wave neuronal synchronization, as confirmed by local field potentials and electroencephalograms. Chemogenetic silencing confirms that USW-driven glymphatic enhancement depends on neuronal activity, while pharmacological blockade of aquaporin-4 (AQP4) abolishes the effects, revealing dual rel

Divergent approaches to rheumatoid arthritis-associated interstitial lung disease: a comparative analysis of international clinical practice guidelines.

Rheumatoid arthritis-associated interstitial lung disease (RA-ILD) is associated with substantial morbidity and mortality. We compared international and regional clinical practice guidelines to identify agreement, differences, and gaps in RA-ILD care. We conducted a narrative comparative review of five guideline groups: ERS/EULAR, ACR/CHEST, EULAR, PANLAR, and ALAT. Recommendations were assessed across screening, diagnosis, monitoring, and treatment, together with their methodological approaches. Diagnostic recommendations showed the greatest agreement: ILD-focused guidelines emphasized high-r

Synovial inflammation and immune-stromal crosstalk in osteoarthritis-related synovial fibroblast research, 2006-2025: a bibliometric analysis.

BACKGROUND: Osteoarthritis (OA) is increasingly understood as a whole-joint organ disease involving cartilage, synovium, subchondral bone, and the local immune-stromal microenvironment. Synovial fibroblasts and fibroblast-like synoviocytes (FLS) participate in synovitis, matrix remodeling, cartilage-synovium crosstalk, pain-associated microenvironments, and disease heterogeneity. However, the knowledge structure, main knowledge sources, and frontier evolution of OA-related synovial fibroblast research remain incompletely mapped. METHODS: We retrieved publications on OA-related synovial fibrob

Single-cell mass cytometry cartography highlights systemic lupus erythematosus and Sjögren's disease immune differences.

OBJECTIVES: Autoimmune diseases such as primary Sjögren's disease (pSjD) and systemic lupus erythematosus (SLE) share clinical and serological features, including type I interferon signatures, autoantibody formation, and chronic inflammation, but the underlying immunopathology is not the same. To support the development of targeted therapies, we aimed to systematically characterize disease-specific alterations in peripheral immune cell subsets of pSjD and SLE compared to healthy donors (HD). METHODS: We analyzed peripheral blood mononuclear cells from 15 HD, 12 pSjD and 21 SLE patients by mas

Effects of microtrauma on joint structural changes in the Curdlan-injected SKG mouse model of ankylosing spondylitis.

Although microtrauma is considered a potential environmental factor in the development of ankylosing spondylitis (AS), there is a lack of experimental research regarding its precise contribution to disease pathogenesis. To induce physiological microtrauma, the right ankles of SKG mice were immobilized for a week using surgical plaster and a pipette tip to stimulate compensatory mechanical overload in the contralateral limb. Following the removal of the immobilization, the mice were intraperitoneally injected with 3 mg of Curdlan. In this model, the non-immobilized left ankle served as the micr

Damage-Associated Molecular Patterns and Trained Immunity of Monocyte-Macrophage Populations in Osteoarthritis: Potential Molecular Links and Therapeutic Opportunities.

Osteoarthritis (OA) is now widely discussed as a whole-joint immunometabolic disease involving cartilage, synovium, and subchondral bone, rather than as cartilage degeneration driven only by mechanical stress. Tissue injury, cell death, mitochondrial dysfunction, and extracellular matrix degradation can lead to the persistent release of damage-associated molecular patterns (DAMPs), which activate innate immune responses through pattern recognition receptors (PRRs). A key unresolved issue is why inflammation can persist or recur when there is no continuous external stimulus. Trained immunity pr

Differential Expression of Cytotoxic-Associated Markers in Circulating CD4+CD28- T Cells and NK Cells in Rheumatoid Arthritis.

Rheumatoid arthritis (RA) involves alterations in cytotoxic lymphoid differentiation, yet the precise phenotypic remodeling of circulating helper T cells and natural killer (NK) cells remains incompletely understood. We evaluated circulating CD4+CD28+ and CD4+CD28- T-cell subsets and NKG2A+CD56+ NK cells, alongside granule-mediated cytotoxic markers (granzyme B and perforin) and the surface-bound death receptor Fas ligand (FasL), in 28 female patients with RA and 12 age-matched healthy female controls using multiparameter flow cytometry. CD4+CD28- cells were increased in RA compared with contr

The Gut-Lung-Joint Axis: A Conceptual Framework for Microbiome-Mediated Crosstalk Between Respiratory and Rheumatic Diseases in Childhood.

There is growing interest in the role of the gut microbiome in immune development and homeostasis during childhood. Increasing evidence suggests that early alterations in microbial composition might influence susceptibility to both respiratory and rheumatic diseases through interconnected pathways. While the gut-lung and gut-joint axes have been investigated separately, their integration into a biologically specific and testable model remains largely unexplored. In this narrative review, the literature was searched in PubMed/MEDLINE and Scopus for evidence syntheses published from 2020 to 2026
