Native coronal patellar tracking demonstrates three distinct patterns associated with coronal alignment in robotic-assisted total knee arthroplasty: a preliminary study.

OBJECTIVES: This study aimed to objectively identify and describe intraoperative patellar tracking patterns in osteoarthritic knees and examine their relationship with coronal alignment according to the coronal plane alignment of the knee (CPAK) classification. METHODS: A retrospective analysis was performed on 252 knees that underwent robotic-assisted primary total knee arthroplasty (TKA). Intraoperative patellar tracking was assessed prior to any bone cuts. Patellar tracking was recorded using a previously described technique with the CORI robotic system. Preoperative full-length standing r

Macrophages in rotator cuff injuries and tendon-to-bone healing: Heterogeneity, pathophysiology, and therapeutic strategies.

Rotator cuff tears (RCTs) are a leading cause of shoulder disability, with high postoperative failure rates primarily attributed to poor tendon-to-bone healing. Macrophages act as central regulators of the inflammatory and regenerative microenvironment in the injured enthesis, tendon stump, subacromial bursa, and rotator cuff musculature. This review highlights the spatiotemporal heterogeneity and phenotypic plasticity of macrophages revealed by single-cell and spatial transcriptomics. Rather than following a simple M1-to-M2 transition, macrophages adopt context-dependent inflammatory, resolvi

Anti-Inflammatory Effects of Vicatia thibetica De Boiss. in Rheumatoid Arthritis and Associated Multi-Pathway Modulation In Vivo.

Vicatia thibetica de Boiss. (VTDB) is not only used as an important Tibetan medicine but is also consumed as a common wild vegetable. Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent synovial inflammation and progressive destruction of cartilage and bone. VTDB is recorded in the Four Medical Classics for treating "Huang-Shui," a condition resembling RA. To preliminarily investigate its potential therapeutic basis, VTDB and its polarity-based fractions were evaluated for their effects against RA. The chemical constituents of VTDB-TE were profiled using UHPL

Circulating angiogenic T cells as potential immune-vascular biomarkers associated with disease activity in axial spondyloarthritis.

OBJECTIVE: Axial spondyloarthritis (axSpA) is a chronic inflammatory autoimmune disease associated with persistent systemic inflammation and endothelial dysfunction. Angiogenic T cells (Tang cells), defined as CD3+CD31+CD184+ T cells, are specialized immune cells involved in vascular repair and endothelial homeostasis, but their clinical relevance in axSpA remains unclear. This study aimed to characterize circulating Tang cells in axSpA and evaluate their potential as immune-vascular biomarkers of disease activity. METHODS: Peripheral blood samples were obtained from patients with axSpA strat

Polypharmacological Target-Aware Molecule Generation.

MOTIVATION: Designing compounds capable of modulating multiple target proteins simultaneously (i.e., polypharmacology) is critical for treating complex diseases like cancer and neurodegenerative disorders. However, AI-driven multi-target drug design remains severely hindered by the scarcity of multi-target 3D structural data and the inefficiency of traditional "generate-then-filter" pipelines. There is a pressing need for scalable methods that can satisfy diverse binding constraints across multiple proteins without relying on scarce structural priors. RESULTS: To overcome these bottlenecks, w

Refining Home-Based Squat Training: The Role of Hip Adduction in Improving Neuromuscular Control for Clinically Diagnosed Patellofemoral Pain Syndrome.

BACKGROUND Patellofemoral pain syndrome (PFPS) often stems from neuromuscular imbalances, particularly vastus medialis obliques (VMO) dysfunction impairing patellar tracking. Research is shifting toward integrated rehabilitation strategies, such as combining hip adduction with squats to enhance VMO recruitment. MATERIAL AND METHODS Thirty participants with PFPS were randomized to traditional squatting (SQU, n=15) or squatting with hip adduction (SQU-HA, n=15) for an 8-week home-based program with identical stretching. SQU-HA squeezed a Swiss ball during squats. Fifteen healthy individuals serv

Proteomic landscape of IGF-I gene transfer in osteoarthritis-derived human bone marrow stromal cells undergoing chondrogenesis.

BACKGROUND: Osteoarthritis (OA) is a progressive joint disease marked by the breakdown of cartilage and impaired repair capacity. Gene therapy applied to human bone marrow-derived adherent stromal cells (BM-MSCs) with recombinant adeno-associated virus (rAAV) vectors represents a promising strategy for cartilage regeneration. However, the molecular mechanisms underlying its effects remain largely unexplored. In this study, we investigated the proteomic changes induced by rAAV-mediated insulin-like growth factor I (IGF-I) gene transfer during the chondrogenic differentiation of BM-MSCs derived

Trained immunity in chronic, relapsing and low-grade joint inflammation.

Trained immunity is a form of innate immune memory mediated by metabolic and epigenetic reprogramming that promotes sustained inflammatory responses. Accumulating evidence implicates a role for maladaptive trained immunity in sustaining inflammation and promoting disease progression across distinct forms of inflammatory joint disease, including rheumatoid arthritis, gout, calcium pyrophosphate deposition disease and osteoarthritis. Inflammatory cytokines, damage-associated patterns and metabolic signals can induce long-lasting inflammatory programmes in both immune cells, such as monocytes and

Immune profiling of anti-SSA-positive at-risk individuals reveals increased type I IFN activity preceding lymphocyte abnormalities in Sjögren's disease.

OBJECTIVES: Identifying immune features that distinguish asymptomatic autoantibody-positive at-risk individuals from patients with established autoimmune disease is key to understanding the evolution from autoreactivity to established autoimmune disease. This study investigated immune characteristics of anti-SSA-positive at-risk individuals, including pregnant women, compared with patients with Sjögren's disease (SjD) and healthy donors (HDs). METHODS: Peripheral blood samples were obtained from asymptomatic anti-SSA-positive women (at-risk individuals; n = 7, including two ongoing pregnancie

Sanguinarine ameliorates rheumatoid arthritis by activating the Hippo-YAP/TAZ pathway to induce apoptosis and suppress inflammation in fibroblast-like synoviocytes.

INTRODUCTION: Rheumatoid arthritis (RA) is a chronic autoimmune disease with progressive joint damage, functional disability and systemic comorbidity. Available therapies are not satisfactory, including low rates of remission and about 30% of patients with poor response to first-line treatment. Thus, more effective therapeutic strategies are urgently needed. Sanguinarine, a benzophenanthridine alkaloid primarily isolated from Sanguinaria canadensis and other members of the Papaveraceae family, has been documented to possess notable anti-bacterial, anti-inflammatory, and anti-neoplastic propert

Biocontrol efficacy of Bacillus albus SSR3 for controlling postharvest fungal pathogens and mycotoxin contamination.

Sweetpotato black rot, caused by Ceratocystis fimbriata, is a major postharvest disease that leads to substantial storage losses worldwide. In this study, a salt-tolerant rhizobacterial strain, Bacillus albus SSR3, was isolated from the rhizosphere of sweetpotato grown in saline-alkali soil, with broad-spectrum antagonistic activity against postharvest fungal pathogens. LC-MS/MS analysis revealed diverse bioactive metabolites associated with its antifungal activity. Integrated transcriptomic and metabolomic analyses showed that SSR3 bioactive metabolites extensively reprogrammed fungal metabol

Dislocation analysis on bio-inspired open hip joint with active-and-passive stabilizers.

This study presents a simulator framework to replicate joint dislocation that can be used to evaluate the contributions of various joint stabilizers. Compared to conventional axial joint configurations, biologically inspired open joints allow for a large range of motion with fewer rotational components and no singularities. Despite these advantages, open joints have few practical applications due to joint dislocation. To facilitate the development of bio-inspired open joints, our study evaluates the performance of biomimetic joint stabilizers involving (1) the muscle actuators as the active st

Structural Validity and Measurement Properties of the Tampa Scale for Kinesiophobia in Knee Osteoarthritis: Comparison of Candidate Factor Structures.

PURPOSE: Kinesiophobia is clinically relevant in knee osteoarthritis (KOA), but the appropriate Tampa Scale for Kinesiophobia (TSK) structure remains uncertain. We compared candidate TSK factor structures and examined the selected measurement properties of the solution with the most favourable relative fit in this sample. METHODS: Among 119 participants with KOA, ordinal confirmatory factor analysis tested three a priori structures: one-factor 17-item, one-factor 11-item, and two-factor 11-item. After three items in the two-factor 11-item structure had loadings below 0.20, a secondary explora

Concomitant Abdominoplasty and Repair of Midline Abdominal Wall Defects.

BACKGROUND: Diastasis recti and midline abdominal wall hernias are frequently encountered in patients undergoing abdominoplasty, particularly postpartum women and individuals following significant weight loss. The associated musculoaponeurotic laxity results in functional abdominal wall weakness and contour deformities. METHODS: We describe a surgical technique combining abdominoplasty with midline fascial repair, performed without mesh reinforcement. The procedure utilizes a modified overlapping Mayo double-suturing method to restore midline strength and contour. This paper presents a 39-yea

Genome-wide analysis of common structural variants in Alzheimer's disease identifies a risk-associated regulatory CCG repeat in FAM193B.

Although Alzheimer's disease (AD) has a polygenic architecture, with numerous associated single-nucleotide polymorphisms (SNPs) identified, the contribution of structural variants (SVs) is unclear. In this study, using long-read (n = 11) and short-read (n = 3,190) sequencing data from the Chinese population, we show that common SVs contribute to AD heritability beyond common SNPs. We identify an expanded CCG repeat in the FAM193B promoter that shows a genome-wide significant association with increased AD risk (odds ratio = 2.20 per 10-repeat increase, P = 5.5 × 10-13). The expanded FAM193B-CCG
