Lipoid proteinosis (LP) is a rare autosomal recessive disorder caused by loss-of-function mutations in the extracellular matrix protein 1 (ECM1) gene. While the clinical features of LP are well documented, the cellular mechanisms underlying its oral manifestations, particularly the direct consequences of ECM1 deficiency in oral keratinocytes, remain poorly understood. This study investigated the role of ECM1 in human oral keratinocytes (HOK) using an in vitro gain- and loss-of-function model. Functional assays were performed to evaluate proliferation, migration, keratinization and epithelial-m

BACKGROUND: Variants in STAR, encoding Steroidogenic Acute Regulatory (StAR) protein, and CYP11A1, encoding the cholesterol Side-Chain Cleavage (SCC) enzyme, result in proximal steroidogenic defects. These are classically described as presenting with neonatal-onset primary adrenal insufficiency, with the additional finding of sex reversal in 46,XY newborns. However, the phenotypic spectrum is much wider. The present study aimed at describing the phenotypic heterogeneity and genotypes of patients with STAR and CYP11A1 variants followed up at a tertiary care centre in India.
METHODS: Hospital r

Exogenous lipoid pneumonia (ELP) is a rare pulmonary disorder resulting from the aspiration or inhalation of exogenous lipid substances. While most documented cases follow an indolent, chronic course and are associated with favorable outcomes, fulminant postoperative ELP remains exceedingly uncommon-often presenting with rapid respiratory decompensation that may clinically and radiologically mimic severe community-acquired pneumonia or acute respiratory distress syndrome (ARDS). We report a 61-year-old man with occult fulminant postoperative ELP after repeated oral liquid paraffin use for bowe

INTRODUCTION: Minimal Change Disease (MCD) represents a prevalent pathological cause of nephrotic syndrome. While podocyte injury is recognized as the direct cause of proteinuria in MCD, aberrant T-cell activation is a key driver of this podocyte damage. However, the absence of T-cell infiltration in renal biopsies and ineffectiveness of immunosuppressive therapy in a subset of patients suggest the existence of undiscovered key mechanisms underlying MCD immunopathogenesis.
OBJECTIVES: This study investigates the role of podocyte-derived exosomes (Pdo-Exos) in T cell activation via antigen pre

INTRODUCTION: The primary objective of this study was to assess urinary vitamin D binding protein (uVDBP) levels in idiopathic nephrotic syndrome. Secondary objectives were to evaluate the ability of uVDBP to differentiate between steroid resistant nephrotic syndrome (SRNS) and steroid sensitive nephrotic syndrome (SSNS), and between minimal change disease (MCD) and focal segmental glomerulosclerosis (FSGS) of SRNS.
METHODS: Sixty-three cases of idiopathic nephrotic syndrome were included; 23 were first episode nephrotic syndrome (FENS), 24 were relapsing nephrotic syndrome (RNS), and 16 were

Lipid droplets (LDs) dynamically interact with other organelles, such as mitochondria, in surveillance of cellular metabolic homeostasis. The transient nature of LDs, however, poses technical challenges to snapshot molecular information underlying these interactions. Herein, we present a small-molecule-based photocatalytic protein proximity labeling method (named LipoID) to enable in situ labeling, capturing and profiling of the LD-interacting proteome. This method is enabled by a set of LD-targeting probes designed to catalyze protein modifications nearby LDs using nucleophilic substrates. Pr

Lipid nanoparticle (LNP) formulations have emerged as a versatile platform for the delivery of therapeutics. However, achieving long-term stability and effective delivery of water-soluble small molecule drugs remains a challenge. In this study, we demonstrate a cryopreservable LNP formulation incorporating a hydrophobically modified bis-prodrug of lamivudine. By systematically varying the surfactant composition by combining a PEGylated surfactant (Brij S20) with an unPEGylated, zwitterionic lipid (Lipoid S100), we identify formulations that maintain colloidal stability following freeze-drying

IntroductionLupus podocytopathy (LP) is an under-recognized pathological manifestation in patients with systemic lupus erythematosus (SLE). Despite being a distinct entity, current American College of Rheumatology (ACR)/European Alliance of Associations for Rheumatology (EULAR) guidelines do not include specific recommendations about LP, contributing to uncertainty regarding its diagnosis and treatment. This systematic review aims to synthesize the available literature on LP from case reports, case series and retrospective cohort studies to better characterize its clinical course, thereby info

This case report describes a patient with exogenous lipoid pneumonia (ELP) presenting as a solitary pulmonary nodule. ELP is a rare chronic pulmonary disorder characterized by nonspecific clinical symptoms and imaging findings. When manifesting as a mass or nodule, features such as lobulation and spiculation on imaging can closely resemble lung cancer, frequently leading to misdiagnosis and unnecessary surgical intervention. We present a retrospective analysis of a pathologically confirmed case of ELP to improve recognition of this condition among both clinicians and pathologists.

Lipoid proteinosis is a rare, autosomal recessive inherited metabolic disorder. Its low prevalence contributes to a relatively high rate of clinical misdiagnosis. A 5-year-old female with progressive hoarseness over 4 years and the development of multiple papules on the eyelids over the past 6 months. Physical examination revealed pale, sesame-sized papules densely distributed along both eyelids; irregular, pale white patches on the oral mucosa; and clustered pale patches without evident scaling on the left forearm. Histopathologic analysis demonstrated deposition of lightly stained, homogeneo

The etiology of primary podocytopathies including childhood nephrotic syndrome, minimal change disease, primary FSGS as well as recurrent FSGS had long remained elusive. The development of robust anti-nephrin autoantibody detection methods, the identification of these antibodies in idiopathic nephrotic syndrome, and the demonstration of their causal role in podocytopathy have led to a paradigm shift in our understanding of these diseases, offering new insights into pathophysiology and promising improved diagnostic, prognostic, and therapeutic approaches. Prognostication and risk stratification

Bioactive compounds in green coffee beans, such as caffeine (CF) and cafestol (CA), possess potential pharmaceutical benefits, including antioxidant and anti-inflammatory properties, but face challenges in skin penetration. This study developed a novel liposomal system combining CF and CA for the first time, formulated with either Lipoid S 75 or Phospholipon® 90 G and cholesterol (80:20 ratio), aiming to enhance skin penetration of CF and CA by encapsulating them into liposomes dermal application. Formulations with 1% CF and either 0.13% or 0.07% CA were developed and tested for skin permeatio

A lupus-like condition induced by intraperitoneal administration of pristane (2,6,10,14-tetramethylpentadecane) in mice is widely used as a model of systemic lupus erythematosus (SLE). Due to their phylogenetic distance from humans, murine models are not always suitable tool for studying the specific activity of therapeutic agents and the pathogenesis of SLE. In order to overcome species-specific limitations of murine models, this approach was tested in non-human primates-cynomolgus monkeys (Macaca fascicularis). Two intraperitoneal injections at a dose of 3.5 mL/kg, administered at weeks 1 an

Cutaneous candidiasis, a widespread opportunistic fungal infection primarily caused by Candida albicans, accounts for 1% of outpatient and 7% of inpatient dermatology consultations. The rise in antifungal resistance due to genetic mutations in Candida species has complicated therapeutic outcomes and increased mortality rates. This study aimed to develop a more effective topical therapy by formulating Luliconazole (LCZ), a potent imidazole antifungal limited by poor solubility and skin penetration, into a Transethosomal gel (LCZ-TEG) for enhanced delivery and efficacy. LCZ-loaded Transethosomes

Minimal change disease (MCD) and focal segmental glomerulosclerosis (FSGS) are two key nephrotic syndrome types with significant clinical implications. MCD predominantly affects children, while FSGS is more common in adults, often leading to irreversible kidney dysfunction. Despite shared features like podocyte injury and immune dysregulation, their pathological and clinical presentations differ. Understanding gene expression changes in these diseases could reveal new therapeutic targets. Single-cell transcriptomic datasets (GSE213030 and GSE176465) were analyzed to investigate cellular intera

Minimal change disease (MCD) is a common cause of nephrotic syndrome and typically responds well to glucocorticoid therapy. Although spontaneous remission is considered rare, its true frequency may be underestimated, possibly due to the early initiation of treatment in most cases. We report a case of a 45-year-old male who developed nephrotic syndrome following an influenza infection. A kidney biopsy revealed findings consistent with minimal change disease, including diffuse foot process effacement, and proteinuria resolved spontaneously without immunosuppressive therapy. The patient remained
