INTRODUCTION/AIMS: Previous ultrasound (US)-based assessments of median nerve (MN) displacement within the carpal tunnel have shown inconsistent results due to methodological variability. Quantitative data on how different upper-limb movements affect MN displacement and shear strain at the wrist remain scarce. This study aimed to quantify MN longitudinal displacement and shear strain during finger, wrist, and elbow movements in healthy individuals to establish normative patterns of nerve gliding and deformation.
METHODS: Twenty healthy subjects (13 females; mean age: 31.9 years, range: 27-36

Craniosynostosis is one of the most common craniofacial anomalies, resulting from premature fusion of one or more cranial sutures. While most cases are sporadic, a significant number have a genetic etiology, including monogenic mutations. Coronal synostosis, in particular, is frequently associated with genetic variants. Mutations in the zinc finger protein of cerebellum 1 (ZIC1) gene have recently been recognized as a rare cause of coronal craniosynostosis. We report an 11-month-old female infant with a ZIC1 mutation presenting with unilateral left coronal craniosynostosis, microcephaly, and m

Rehabilitation is crucial for stroke survivors to restore motor functions and improve the quality of life. However, existing rehabilitation systems are relatively large and require professional assistance, which limits their accessibility. This study develops small patch-type sensors using biocompatible and flexible single-crystalline piezoelectric thin films to monitor the accuracy and response time of finger, hand, and forearm movements. The sensors attached to the skin of joints and muscles exhibit distinct output voltages in response to the directions and degrees of movement, with high sen

OBJECTIVE: Kirner’s deformity is a progressive deformity of the distal phalange of the little finger in a volar and radial direction. The FGFR2 gene plays a critical role in skeletal development and growth, with abnormal expression and activation of FGFR2 being associated with skeletal abnormalities. This study presents a family with Kirner’s deformity involving multiple patients. METHODS: Whole-exome sequencing was performed. SIFT, MutationTaster and Gerp+, while Clustal Omega was used for conservative analysis. In addition, I-TASSER was used to predict the 3D protein model. PyMOL was used fo

INTRODUCTION: Buratti-Harel syndrome (BURHAS) is a rare autosomal dominant neurodevelopmental disorder caused by SIAH1 (Siah1 E3 ubiquitin ligase) variants, characterized by infantile hypotonia, global developmental delay, and variable multisystem involvement. To date, 13 pathogenic SIAH1 variants have been reported in 13 patients, but the functional and phenotypic implications of mutations in the SIAH1 zinc finger domain remain poorly characterized. This study included a Chinese pediatric patient with unexplained neurodevelopmental and multisystem abnormalities.
METHODS: Trio whole-exome seq

Human fingers are one of the most remarkable organs for handling complex tasks or manipulating unknown objects, not only due to its dexterous and powerful movement capabilities but also its rich kinematic sense at joints and tactile sensing at skins. Shape and tactile sensing are crucial for soft pneumatic fingers to achieve embodied intelligence. Reliable tactile sensing of soft pneumatic-driven robots is particularly challenging due to its large deformation and adaptability. Here, we propose a distributed local curvature sensing-based solution for simultaneous shape and tactile perceptions i

SATB2-associated syndrome (SAS) is a multisystemic disorder characterized by developmental delay, moderate to profound intellectual disability, speech delay and/or absent speech, behavioral issues such as autistic tendencies, agitation or aggressive outbursts, self-injury, impulsivity, hyperactivity, anxiety and sleeping difficulties. Alterations in the SATB2 gene have been identified as pathogenic causes of SAS. No formal clinical diagnostic criteria have been established for SAS, and molecular disruption of SATB2 is necessary to confirm the diagnosis. To investigate the molecular pathogenesi

Cohen syndrome is a rare autosomal recessive disorder caused by biallelic pathogenic variants in the VPS13B gene, classically presenting with developmental delay, distinctive craniofacial features, neutropenia, truncal obesity, and progressive retinal dystrophy. Metabolic abnormalities, including insulin resistance and diabetes mellitus, have been increasingly recognized, but early-onset diabetes with diabetic ketoacidosis remains uncommon. We report a 28-year-old Chinese woman with early-onset insulin-resistant diabetes complicated by recurrent diabetic ketoacidosis and nephropathy. She prese

PURPOSE: Skeletal abnormalities are common in Turner Syndrome (TS), yet data on objective radiographic markers are limited. We aimed to establish normative reference ranges for phalangeal length ratios and assess their utility in detecting skeletal abnormalities in TS.
METHODS: We analyzed 4,082 female bone age X-rays (<18 years) from the Radiological Society of North America (RSNA) database after quality screening and outlier exclusion as a reference cohort. Phalangeal length ratios-4th to 3rd metacarpal (4:3 MC), 5th to 3rd metacarpal (5:3 MC), and 5th to 3rd middle phalanx (5:3 MP)-were me

There is currently no consensus on which existing treatment for chronic Boutonniere deformity is most effective. We describe a method of central slip reconstruction with an autologous "V" shaped tendon graft folded on itself through a bone tunnel for the treatment of Boutonniere deformity. All 3 patients healed well with an average active range of motion of the distal interphalangeal joint 10 to 70 degrees, proximal interphalangeal joint 15 to 90 degrees, and metacarpophalangeal joint 0 to 90 degrees with a follow-up duration between 4 years and 5 years. This is a straightforward technique tha

INTRODUCTION: The prevalence of bone disease in peritoneal dialysis patients has been recently shown to exceed 54%, including patients with parathormone (PTH) levels within the theoretical adequate target, yet demonstrating low bone turnover on histomorphometry. Moreover, bone disease is often associated with abnormalities in calcium and phosphate metabolism, leading to tissular deposits such as extraosseous calcifications.
CASE PRESENTATION: We present a 22-year-old female patient managed on peritoneal dialysis with persistent swelling of all four extremities, including the fingers, hands, a

The surgical treatment of carpometacarpal (CMC) arthritis of the thumb is widely performed, and provides good pain relief and restoration of function. This condition may be successfully treated using one of many surgical procedures. While the incidence of failure following these procedures is low, potential complications include persistent pain, deformity, and weakness. Identifying the cause of persistent symptoms requires a careful and methodical approach. This review aims to highlight the common reasons for failure, provide a comprehensive discussion of the literature surrounding failed surg

Radioulnar synostosis with amegakaryocytic thrombocytopenia type 2 (RUSAT-2) is a rare inherited bone marrow failure syndrome characterized by congenital or progressive thrombocytopenia, frequent radioulnar synostosis, and variable multisystem involvement. It is caused by heterozygous germline pathogenic variants in the MDS1 and EVI1 Complex Locus (MECOM) gene, which encodes transcription factors essential for hematopoietic stem cell regulation and embryonic development. Disruption of highly conserved zinc finger domains within MECOM impairs long-term hematopoietic stem cell maintenance, leadi

Hypospadias is a common congenital malformation of the male external genitalia, with severe cases presenting considerable surgical and long-term challenges. Despite the clinical importance of severe hypospadias demonstrated by prolonged hospital stays, repeated surgeries, and substantial costs, the genetic etiology of severe hypospadias remains incompletely understood, particularly in diverse populations. To determine the molecular basis, we performed whole-exome sequencing (WES) on 30 Chinese patients from southeastern China with confirmed 46,XY karyotypes. Our analysis identified clinically
