Dual molecular diagnosis of CEP290 and GLI3 mutations identified in an infant with leber congenital amaurosis and postaxial polydactyly, a Bardet-Biedl syndrome phenocopy.

BACKGROUND: Leber congenital amaurosis (LCA) is one of the earliest-onset and most severe forms of inherited retinal disease, and Bardet-Biedl syndrome (BBS) is a rare non-motile primary ciliopathy with a diverse multi-organ phenotype. To our knowledge, we present the first reported genetically confirmed case of LCA10 co-occurring with isolated postaxial polydactyly type A1 harboring CEP290 and GLI3 mutations, which was initially misdiagnosed as BBS. CASE PRESENTATION: A 6-month-old female infant presented with no visual tracking, nystagmus, and postaxial polydactyly. Targeted gene panel test

A study on the RUS-CHN bone age assessment method in adolescents and children from the Suzhou region.

BACKGROUND: The RUS-CHN method, part of the China-05 standard, is widely used in China. However, its performance may vary across regions due to socioeconomic and nutritional factors. This study aimed to clinically validate the RUS-CHN method against the Greulich-Pyle (G-P) atlas in a cohort from the developed Suzhou region. METHODS: We conducted a cross-sectional study of 271 children. Left-hand radiographs were independently assessed by two experienced raters using both the G-P and RUS-CHN methods. Statistical analyses compared the distributions, correlations, and specific bone scores derive

Temporal deep neural network for tactile sensing in artificial finger pulp skin.

Tactile perception plays a vital role in artificial finger pulp skin, especially in regions responsible for grasping and touching tasks, where precise sensing of deformation position and applied force is critical. Conventional demodulation methods often fail to fully leverage temporal correlations in data during the pressing process, limiting the accuracy of tactile demodulation. To address this, we propose a tactile sensing system based on quasi-distributed Fiber Bragg Gratings (FBGs) integrated into artificial finger pulp skin, along with a two-stage hybrid LSTM-Transformer neural network (T

Reassessing Benign ASXL1 Variants in Bohring-Opitz Syndrome: The Role of Population Databases in Variant Reinterpretation.

Background/Objectives: ASXL1 is a chromatin-associated gene implicated in both hematologic malignancies and neurodevelopmental disorders, including Bohring-Opitz syndrome (BOS). Although many ASXL1 variants are well classified, a substantial proportion remain variants of uncertain significance (VUS), complicating molecular diagnosis and genetic counseling. The objective of this study was to evaluate whether structural context can inform the interpretation of selected ASXL1 missense variants in a clinical setting. Methods: We describe a 17-year-old female with clinical features consistent with

A unilateral accessory belly of the extensor indicis muscle: morphological presentation and its clinical implications.

PURPOSE: The extensor indicis muscle is an important muscle of the deep layer of the posterior compartment of the forearm, as it allows independent extension of the index finger. Numerous forms of variations of the extensor indicis muscle have been reported. This case report aims to describe a rare unilateral presence of an accessory muscle belly of the extensor indicis. CASE REPORT: An incidental finding during the routine dissections of the posterior compartments of the forearm and the dorsum of the hand is presented. The spindle-shaped accessory muscle of the extensor indicis originated fro

The role of skin mechanics in contact force variation under different friction conditions.

When grasping objects, humans actively adjust grip force in response to surface slipperiness and motion. Previous studies have showed that corrective actions occur after tactile afferents signal surface friction or slip events. However, the influence of the mechanical behavior of the skin on the development of contact forces is poorly understood. In this study, using contact kinematics derived from a natural reach-and-grasp task, we applied a glass surface onto restrained fingers via a robotic manipulator under low- and high-friction conditions. Contact forces were measured with a force sensor

MGRN1 is linked to recessive heart and laterality defects: the first genotype-phenotype report in humans.

Recurrent idiopathic severe fetal structural anomalies present major challenges for reproductive decision-making and genetic counselling. A non-consanguineous healthy Estonian couple had experienced two electively terminated pregnancies at 12-13 weeks' gestation due to unexplained major fetal malformations and one early miscarriage. Their three pregnancies had resulted in unaffected newborns. Exome sequencing of fetal tissues from both terminated pregnancies identified a homozygous rare missense variant in MGRN1 (mahogunin ring finger 1), c.881G>A, p.(Arg294His) (Genome Aggregation Database V.

Trapeziometacarpal joint arthroplasty: avoiding imbalance and optimizing outcomes.

INTRODUCTION: Trapeziometacarpal joint osteoarthritis is common and can lead to pain, loss of pinch strength, and progressive deformity, including the classic Z-deformity. Understanding the pathomechanics of the disease progression can help surgeons effectively balance trapeziometacarpal joint arthroplasty.Biomechanics:The disease process begins with resorption of the anterior beak of the thumb metacarpal, weakening the anterior oblique ligament. This alters joint contact mechanics, leading to thumb metacarpal flexion and dorsoradial subluxation. As deformity progresses, the effective excursio

HOXD12 a candidate gene for a novel form of synpolydactyly.

Synpolydactyly is a rare limb deformity characterized by a unique combination of syndactyly and polydactyly. Synpolydactyly has an autosomal dominant mode of inheritance often with reduced penetrance. Variants in HOXD13, FBLN1, GLI3, and TTC30B have been reported to underlie the etiology of synpolydactyly. Here, we describe a three-generational Pakistani family segregating a form of synpolydactyly which has not previously been reported that includes, severe hand involvement characterized by bilateral syndactyly of the third, fourth, and fifth fingers, accompanied by preaxial polydactyly in all

Dual PDL 595 nm-Nd:YAG 1064 nm Laser as an Alternative to Surgery in Acquired Digital Arteriovenous Malformations: A 26-Patient Retrospective Cohort.

IMPORTANCE: Acquired digital arteriovenous malformations (ADAVMs) are rare vascular anomalies traditionally managed with surgery, which may lead to morbidity, recurrence, and functional impairment. Evidence regarding laser therapy-historically considered contraindicated in arteriovenous malformations-is extremely limited, and no large cohort has been reported to date. OBJECTIVES: To report the largest cohort of ADAVMs treated with laser therapy and to determine whether dual sequential pulsed dye (595 nm) and Nd:YAG (1064 nm) laser therapy is an effective and safe alternative to surgery for Sc

Rapid Gelation of Antihydrolyzing Imine Hydrogels with a Stable Cross-Linked Structure for Flexible Sensors and In Situ Assembled Flexible Supercapacitors.

Hydrogels based on Schiff base chemistry are widely used for in situ applications due to their rapid gelation property. However, the inherent hydrolytic instability and dynamic nature of imine bonds limit their use in scenarios requiring long-term structural stability and stress relaxation resistance, especially in non-neutral pH environments. Herein, we report an imine hydrogel with a stable cross-linked network that exhibits enhanced resistance to hydrolysis. The hydrogel forms rapidly (<3 min) through condensation between aldehyde groups from glutaraldehyde (GA) and primary amines (-NH2) ge

RIM Hand: A Robotic Hand with an Accurate Carpometacarpal Joint and Nitinol-Supported Skeletal Structure.

This article presents the flexible RIM Hand, a biomimetic robotic hand that precisely replicates the carpometacarpal joints and employs superelastic Nitinol wires throughout its skeletal framework. By modeling the full carpal-to-metacarpal anatomy, the design enables realistic palm deformation through tendon-driven fingers while enhancing joint restoration and supports skeletal structure with Nitinol-based dorsal extensors. A flexible silicone skin further increases contact friction and contact area, enabling stable grasps for diverse objects. Experiments show that the palm can deform up to 28

A Hybrid-Frequency Sampling Tactile Sensing System Based on a Flexible Piezoresistive Sensor Array: Design and Dynamic Loading Validation.

A Hybrid-Frequency Sampling Tactile Sensing System Based on a Flexible Piezoresistive Sensor Array is presented for reliable and real-time tactile perception under dynamic loading conditions. While recent studies have developed multi-channel tactile arrays, most systems remain limited by time-dependent drift in channel responses, inconsistent dynamic behavior, or insufficient temporal resolution under simultaneous loading. In this work, a system-level design integrating a flexible piezoresistive sensor array with a real-time data acquisition module is developed, incorporating a hybrid-frequenc

A Flexible Piezoresistive Sensor Based on ZnO/MWCNTs/PDMS Composite Foam with Overall Performance Trade-Offs.

The flexible foam piezoresistive sensor demonstrates significant potential for wearable strain-sensing applications due to its substantial deformation capacity, excellent flexibility, and cost effectiveness. However, conventional flexible foam piezoresistive sensors often struggle to simultaneously achieve high sensitivity, a wide pressure detection range, fast response and long-term stability. This paper employed a glucose-based sugar-templating method to fabricate a fine-pore (50 μm) foam structure complemented by a dual-filler strategy to enhance overall performance. A robust porous conduct

Thumb Fractures and Associated Injuries: An Evidence Based Review to Guide Treatment.

Fractures of the thumb encompass a diverse spectrum of injury patterns, each requiring nuanced consideration of various treatment strategies. Associated conditions include injuries to the ulnar collateral ligament and radial collateral ligament of the metacarpophalangeal joint, which can significantly impact thumb stability and function. Although extra-articular fractures of the thumb metacarpal and phalanges are more tolerant of residual deformity due to compensatory movement of the carpometacarpal joint, intra-articular fractures, including Bennett and Rolando fractures, often benefit from s
