BACKGROUND: Mild cervical ossification of the posterior longitudinal ligament (OPLL) is often asymptomatic or minimally symptomatic; however, its impact on physical function remains unclear. Locomotive syndrome (LS), a condition involving a decline in mobility due to musculoskeletal disorders, may help predict early functional impairments beyond neurological deficits. This study aimed to assess the severity of LS in patients with mild OPLL using the locomotive syndrome risk test battery.
METHODS: This cross-sectional study included 37 patients with mild OPLL (Japanese Orthopaedic Association

Trigger thumb is a condition caused by incongruity in diameter between the swollen flexor tendon and its sheath at the metacarpal head, often due to thickening of the A1 pulley. This results in a clicking or popping sensation during thumb extension. Treatment options include splinting, physical therapy, steroid injections, and surgical release of the A1 pulley. In Ayurveda, trigger thumb can be attributed to Snayugata Vata (vitiated Vata dosha in tendons/ligaments). Agnikarma is an Ayurvedic procedure, effective for treating Snayugata Vata and other joint disorders. A 56-year-old male patient,

INTRODUCTION: Anterior tibial subluxation (ATS) measured on weight-bearing extended lateral knee radiographs is being increasingly used to evaluate anterior cruciate ligament (ACL) injury. However, standardized measurement methods and their clinical relevance are yet to be established. This study aimed to evaluate the reliability and clinical utility of ATS measurements obtained from weight-bearing extended lateral knee radiographs for assessing knee instability in ACL-deficient knees.
METHODS: This retrospective multicenter ACL reconstruction cohort study included patients who underwent preo

OBJECTIVES: Mechanical compression of the periodontal ligament (PDL) is a key trigger of orthodontically induced root resorption (OIRR). Although PDL compression during the early phase of orthodontic force application correlates strongly with OIRR severity, associated cellular events remain insufficiently defined. This study aimed at elucidating mechanisms underlying the initiation of OIRR, focusing on PDL compression and associated cellular responses, and identifying cellular targets through which lithium exerts inhibitory effects.
MATERIALS AND METHODS: Thirty-seven 10-week-old male Wistar

Median arcuate ligament syndrome (MALS) is a rare vascular disorder. We report the case of a 72-year-old man with MALS who was found to have an anomalous right inferior phrenic artery originating from the celiac artery (CA), which was not detected on preoperative computed tomography angiography. During laparoscopic decompression, this vessel was encountered unexpectedly. A temporary clamping test was performed, confirming no diaphragmatic or hepatic ischemia, after which the vessel was safely ligated. The CA was then fully decompressed. Postoperatively, the patient's symptoms resolved complete

Periodontitis is an inflammatory disorder that leads to the destruction of periodontal tissues. Pyroptosis, related to the Nod-like receptor protein 3 (NLRP3) inflammasome, is implicated in the pathogenesis of periodontitis. ELAVL1 (embryonic lethal, abnormal vision, Drosophila-like 1), an N6-methyladenine (m6A) reader, is associated with inflammatory responses; however, its role in periodontitis requires further clarification. Our research elucidated the regulatory functions of ELAVL1 in periodontitis and explored the underlying mechanisms. Expression of ELAVL1 in periodontitis mice and Porph

Closed modeling methods for lateral ankle sprain (ALAS) avoid surgical drawbacks but lack standardization due to reliance on the operator's subjective force, leading to variable outcomes. This study aimed to refine a closed ALAS rat model by quantifying manipulation parameters and establishing optimal ranges for different injury grades. Ninety rats were randomly assigned to groups receiving manual ankle inversion under different combinations of force (0-8 N or 8-16 N) and plantar flexion angle (100°-130° or 130°-160°), with control groups. A flexible thin-film pressure sensor and a goniometer

Periodontitis is a complex, multifactorial inflammatory condition characterized by progressive destruction of the periodontal supporting structures. It profoundly affects oral health, esthetics, and masticatory function and is increasingly recognized as a contributing risk factor for systemic disorders. Deep periodontal pockets establish a severely hypoxic microenvironment induced by periodontal pathogens, increased oxygen consumption of infiltrated inflammatory cells, and accompanying periodontal vascular changes. The cellular response to hypoxia is centrally regulated by hypoxia-inducible fa

BACKGROUND: Post-traumatic Osteoarthritis (PTOA) is a common secondary joint disease characterized by cartilage degradation, subchondral bone remodeling, and synovial inflammation following joint trauma. Current therapeutic options are limited in reversing pathological progression. Tetramethylpyrazine (TMP), an active alkaloid extracted from traditional Chinese medicine Ligusticum chuanxiong, exhibits anti-inflammatory, antioxidant, and cartilage protective properties. This study aimed to investigate the therapeutic effects of TMP in a rat model of PTOA.
METHODS: Sixty-three male Sprague-Dawl

Osteoarthritis (OA) is a degenerative joint disorder characterized by cartilage degradation and synovial inflammation, with its pathogenesis involving intricate interactions between cellular metabolic processes and inflammatory pathways. In this study, we established OA models in both wild-type and histone deacetylase 6 (HDAC6) knockout mice via anterior cruciate ligament transection (ACLT) to investigate the role of HDAC6 in OA progression. Histological and immunohistochemical analyses revealed that the absence of HDAC6 significantly mitigated ACLT-induced cartilage damage and synovitis in mi

BACKGROUND: Soft tissue calcifications are inconspicuous entities seen on orthopantomograms as incidental findings, often overlooked, which may be a result of underlying disorders. Hence, it is important to evaluate their location, extent, and cause, to determine their clinical significance which may warrant further investigations and appropriate interventions to avoid any complications.
PURPOSE: To determine the prevalence of soft tissue calcifications on digitally acquired panoramic radiographs.
MATERIALS AND METHODS: This cross-sectional, retrospective study analyzed panoramic radiographs

Ossification of the posterior longitudinal ligament (OPLL) is a degenerative spinal disorder characterized by heterotopic ossification of ligamentous tissue. Its pathogenesis is multifactorial and complex, involving genetic susceptibility, chronic inflammation, mechanical stress, and metabolic dysregulation. In recent years, accumulating evidence has demonstrated that angiogenesis not only supplies essential nutrients and metabolic support to ossified ligament regions but also actively regulates the differentiation of mesenchymal stem cells toward osteogenic and chondrogenic lineages through s

The Peritoneal Cancer Index (PCI) is widely used to quantify disease extent and guide therapeutic decisions for cytoreductive surgery (CRS) and hyperthermic intraperitoneal chemotherapy (HIPEC) across multiple primary tumors. While PCI is a validated prognostic indicator, the use of a single total surgical PCI threshold as a determinant of eligibility for CRS-HIPEC presents significant conceptual and practical limitations. Surgical PCI calculation is subject to inter-surgeon variability, particularly at anatomical region boundaries, where lesion allocation may alter the total score sufficientl

The periodontium hosts diverse populations of mesenchymal stem and progenitor cells that are essential for maintaining homeostasis and driving regeneration. These include cells derived from the periodontal ligament, gingiva, and apical papilla. In health and disease, the fate and function of these stem cell populations are shaped by their microenvironment, particularly by inflammatory signals and their resolution. Chronic inflammation, such as that observed in periodontitis, disrupts the regenerative capabilities, impairing stem cell function and biasing differentiation pathways. Inflammation

Tendinopathy is a painful overuse disorder marked by a progressive functional decline. Although chronic disease mechanisms have been described, early molecular changes remain poorly defined. Here, we performed temporal proteomic profiling of human patellar tendon biopsies across one, two, and three months of symptom duration and evaluated proteome differences between symptomatic and nonsymptomatic contralateral tendons. Biopsies were collected from participants with unilateral patellar tendinopathy and symptoms lasting three months or less for analysis by data-independent acquisition liquid ch

BACKGROUND: By analyzing the single-cell RNA-Seq libraries we established of OA joints during exercise therapy, we found cartilage intermediate zone might participate in early OA exercise therapy. METHODS: Early OA rat model was established by 4-week anterior cruciate ligament transection (ACLT). The radiomics was used to evaluate the relative damaged and undamaged area in OA patients’ cartilage. We overexpressed and knocked down CILP in early OA chondrocyte to explore its potential mechanism. The quantitative proteomics was used to examine the protein profiles of the CILP-treated chondorcyte.

Ossification of the posterior longitudinal ligament (OPLL) is a debilitating spinal disorder characterized by heterotopic bone formation and severe neurological deficits, yet its underlying pathogenic mechanisms remain poorly understood. Here, we report that glycolytic reprogramming orchestrated by the ADAM12/IGF1 axis is a critical driver of OPLL pathogenesis. Through integrated multi-omics analysis of human OPLL tissues, we uncovered a profound metabolic shift featuring enhanced glycolysis and suppressed oxidative phosphorylation as ligament progenitor cells differentiated into osteoblasts.
