Modulation of autophagy and inflammation in human periodontal ligament fibroblasts by the LncRNA-KAT7/miR-455-5p/NRG1 axis in response to LPS.

Autophagy plays a pivotal role in the development of periodontitis, and long non-coding RNA (lncRNA) significantly influences the regulation of autophagy during the progression of this disease. However, the precise regulatory mechanisms remain elusive. Our prior research indicated that lncRNA-KAT7 is critically involved in periodontitis development, yet the specific molecular mechanisms by which lncRNA-KAT7 impacts periodontitis are still undefined. In this study, we discovered that both lncRNA-KAT7 and Neuroregulin-1 (NRG1) are downregulated in periodontitis. Alterations in lncRNA-KAT7 levels

FSCN1 induces subchondral bone sclerosis in osteoarthritis via modulating actin cytoskeleton dynamics and YAP signaling.

BACKGROUND: Subchondral bone is a dynamic tissue maintained by bone remodeling and responded rapidly to mechanical loading, which is strongly associated with cartilage degradation in osteoarthritis (OA), but the underlying mechanisms of which is still blurring. METHODS: Experimental OA mice models were generated by mechanical overload with 60 cycles of 14 N axial compressive loads twice a week or anterior cruciate ligament transection surgery. FSCN1 expression was evaluated in subchondral bone from experimental OA mice and human OA. Mice with FSCN1 conditional knockout in pre-osteoblasts were

Mechanisms and Therapeutic Potential of Stem Cell-Derived Extracellular Vesicles as Nanocarriers in Musculoskeletal Disorders.

The musculoskeletal system consists of bones, joint cartilage, tendons, ligaments, muscles, and their associated nerves and blood vessels. These components work together to maintain human movement and mechanical stability, serving as the critical foundation for sustaining life activities. In recent years, with the acceleration of population aging, the incidence of musculoskeletal-related diseases such as sarcopenia, muscle atrophy, osteoporosis (OP), osteoarthritis (OA), and tendon injuries has continued to rise, significantly impacting patients' quality of life and imposing a heavy social hea

Technetium-99m-labeled pyrophosphate uptake in the rectum of a patient with wild-type transthyretin cardiac amyloidosis.

Wild-type transthyretin amyloidosis is a disease characterized by deposition of transthyretin amyloid protein in systemic organs and tissues, especially the heart, lungs, tenosynovium, and ligaments. Transthyretin amyloid is also frequently deposited in the gastrointestinal tract. Technetetium-99m-labeled pyrophosphate facilitates imaging of its depositions in the myocardium and extracardiac organs and tissues, such as subcutaneous fat and skeletal muscles. However, few reports have described the uptake of bone-seeking radiotracers in the gastrointestinal tract. This report presents the case o

Supramolecular helicity dependent osteogenesis and angiogenesis crosstalk of periodontal ligament stem cell.

The two pillars supporting effective tissue regeneration are multipotent stem cells and matrix materials that direct differentiation. The chirality of the extracellular matrix is a key structural characteristic that affects stem cell fate. However, little is known about the effects of either molecular chirality or supramolecular helicity on the differentiation of periodontal ligament stem cells (PDLSCs), a safe and easily accessible stem cell source. Here, we constructed fibrils through the co-assembly of chiral amino acid derivative enantiomers (l/d-GC18) and a bridging pyrazine molecule. The

Arginine Methylation Antagonizes TEAD3-Mediated Repression to Promote Osteogenic Differentiation by Disrupting RUNX2-Sequestrating Condensates.

Osteogenic differentiation is essential for bone remodeling and repair. Protein arginine methyltransferases (PRMTs) regulate this process; however, their key substrates and mechanisms remain elusive. Here, we identify TEAD3, a TEA domain transcription factor mediating Hippo signaling output, as an arginine-methylated regulator of periodontal ligament stem cells (PDLSCs) osteogenesis. Mechanistically, TEAD3 is methylated at arginine 55 (R55), a conserved residue within its DNA-binding TEA domain. Disruption of R55 methylation via R55K mutation enhances formation of TEAD3 homodimer condensates,

Neisseria bacilliformis is a periodontal pathogen exacerbating periodontitis by inducing nitric oxide production.

Periodontitis, a chronic inflammatory disease, often causes alveolar bone loss. Neisseria bacilliformis is a Gram-negative bacterium that has been identified in periodontal patients, but its role in periodontitis remains unclear. In the present study, we examined whether N. bacilliformis exacerbates periodontitis in a mouse ligature-induced periodontitis (LIP) model and investigated its underlying molecular mechanism. Topical treatment with N. bacilliformis on maxillary second molar exacerbated alveolar bone loss and worsened epithelial and periodontal ligament damage. Histological analyses sh

Exogenous parathyroid hormone (1-34) attenuates knee osteoarthritis in mice through activating the TGF-β/Smad signaling pathway.

OBJECTIVE: Parathyroid hormone (PTH) has been reported to delay the progression of osteoarthritis (OA), while the mechanism remains unclear. The TGF-β/Smad signaling pathway plays a key role in chondrocyte regulation and OA development. We aimed to explore the effect and mechanism of PTH on knee OA (KOA) in mice. METHODS: Anterior cruciate ligament transection (ACLT) was performed in 40 mice to induce KOA. They were randomly divided into the ACLT + PTH and ACLT + vehicle groups. Another 20 mice that underwent only arthrotomy and normal saline were allocated to the sham-vehicle group. The knee

BMP4 dose dictates lineage specification bias in human periodontal ligament stem cells.

INTRODUCTION: Periodontal ligament stem cells (PDLSCs) represent a promising therapeutic cell source for regenerating periodontal tissues impaired by disease. While exogenous BMP4 has been used to induce human PDLSC differentiation into distinct periodontal cell lineages, its dose-dependent effects on lineage specification remain poorly understood. METHODS: Here, we systematically investigated how BMP4 dosage modulates human PDLSC differentiation toward osteogenic and tenogenic lineages using single-cell RNA sequencing and in vitro functional assays. RESULTS: Single-cell transcriptomic analy
