Effects of air pollution on disease activity in patients with rheumatoid arthritis.

OBJECTIVES: This study aimed to evaluate the associations of air pollution with disease activity and flare rate in patients with rheumatoid arthritis (RA). METHODS: This prospective cohort study included patients with RA who were treated at a tertiary medical centre in South Korea between January 2021 and December 2024. Air pollution exposure was estimated using monthly mean concentrations of 6 air pollutants (sulfur dioxide, nitrogen dioxide, ozone, carbon monoxide, particulate matter [PM]10, and PM2.5). Disease activity and flares were recorded longitudinally at each outpatient visit. Assoc

AA in Rheumatoid Arthritis and Lupus.

Inflammation and immune dysregulation are at the center of various diseases, including autoimmune diseases, especially rheumatoid arthritis (RA) and systemic lupus erythematosus (lupus). In general, corticosteroids, disease-modifying drugs, and monoclonal antibodies against IL-6 and TNF-α, JAK inhibitors, and various other immunomodulators are extensively used in the treatment of RA and lupus. It is known that EFA (essential fatty acid) metabolism, especially that of AA, is altered in RA and lupus. It is proposed that metabolism of AA and of other PUFAs may play a role in predicting disease pr

Quantification of Chikungunya Virus RNA by Real-Time RT-PCR Assay.

RT-qPCR is characterized by‌ high sensitivity, strong specificity, excellent reproducibility, and operational simplicity, ‌enabling‌ rapid quantification of viral RNA within a short timeframe. The real-time PCR system relies upon the measurement of a fluorescent reporter during PCR, in which the amount of emitted fluorescence is directly proportional to the amount of the PCR product in a reaction. Here, we describe the use of TaqMan® probe-based one-tube Real-time reverse transcription quantitative polymerase chain reaction (RT-qPCR) for the detection and quantification of the Chikungunya viru

Association between preoperative radiographic osteoarthritis severity and patient-reported outcomes after total hip arthroplasty: a systematic review and meta-analysis.

PURPOSE: Although total hip arthroplasty (THA) reliably improves pain and function in patients with hip osteoarthritis (OA), a subset of patients experience limited postoperative benefit. The association between preoperative radiographic OA severity and postoperative patient-reported outcomes (PROMs) remains controversial. METHODS: We performed a systematic review and meta-analysis according to PRISMA guidelines. Electronic databases (MEDLINE, Embase, and Cochrane Library) were searched for studies comparing postoperative PROMs after primary THA between patients with moderate versus severe ra

Strain in the transverse plane of the patellar tendon depends on both local composition and structure.

Ligaments and tendons are heterogeneous structures composed of two distinct materials at the mm-length scale: fascicles and the interfascicular matrix (IFM). Many studies model the mechanics of ligaments and tendons using a spatially homogeneous, transversely isotropic mechanical model. However, recent studies have demonstrated that these two structures likely need to be modeled separately to explain failure behavior. Additionally, quantification of IFM mechanics would help to better understand the mechanical environment around cells in ligaments and tendons. While much work has been done to q

Consumer Grade Wearable Sensor Metrics Are Associated with Internal Forces at Common Running Injury Locations.

INTRODUCTION: Inexpensive consumer-grade wearable sensors have expanded possibilities to assess, monitor, or modify a runner's gait. However, it is unclear which sensor-measured gait metrics are strongly associated with internal loading at common injury locations. PURPOSE: To measure the association between gait metrics measured by consumer-grade wearable sensors and internal forces at the Achilles tendon and the patellofemoral joint. METHODS: 55 healthy runners wearing consumer-grade chest straps foot-pods completed a 38-minute run on an instrumented treadmill at 12 speeds from 70%-125% of

Influences of joint dynamics on take-off velocity in elite male 110-m hurdlers.

OBJECTIVE: To study the relationship between lower limb joint moments, joint stiffness, and take-off velocity in high-level hurdlers. METHODS: This study employed a Qualisys three-dimensional infrared motion-capture system synchronized with a force plate. Based on inverse dynamics principles, kinematic and kinetic data were collected during the clearance phase over the fourth hurdle in 16 elite male 110-m hurdlers. RESULTS: During the landing phase, Group B had significantly higher peak joint moments at the hip, knee, and ankle compared to Group A (P < 0.01). During the propulsive phase, Gro

Delayed decision, harder landing: how mental fatigue alters reactive step biomechanics and joint loading in table tennis players.

OBJECTIVE: Mental fatigue (MF) is known to impair psychobiological performance, yet its cross-dimensional impact on the neuromechanical control of reactive athletic movements remains underexplored. This study investigated the effects of MF on the cognitive-temporal and lower-limb biomechanical characteristics of table tennis athletes executing a reactive chasse-step. METHODS: Twenty-four collegiate table tennis athletes (12 males, 12 females) completed a randomized, counterbalanced crossover study. Participants performed a choice-reaction chasse-step forehand topspin task following either a 4

Heterologous contralateral repeated bout effect: eccentric knee extensor conditioning enhances neuromuscular fatigue resistance and attenuates muscle stiffness in contralateral elbow flexors in young women.

BACKGROUND: The repeated bout effect (RBE) protects skeletal muscles from exercise-induced muscle damage (EIMD). While homologous contralateral RBE is well-documented, the existence of heterologous RBE-specifically whether lower limb conditioning can protect upper limb muscles-remains debated. PURPOSE: This study investigated whether high-intensity eccentric exercise of the knee extensors attenuates eccentric exercise-induced neuromuscular fatigue and muscle stiffness and preserves neuromuscular control in the contralateral elbow flexors. METHODS: Twenty-four sedentary young women were rando

MagGami: An Electromagnetic Origami Actuator With High Stiffness-Switching Density for Lightweight Joint-Space Wearable Haptics.

Kinesthetic feedback gloves play a central role in immersive virtual and augmented reality (VR/AR) by enabling the perception of object shape, stiffness, and spatial constraints. However, key challenges remain: 1) achieving a large stiffness contrast between free-space and constrained-space states, 2) enabling independent actuation on finger joints, and 3) maintaining lightweight, low-power wearable operation. Here, we introduce MagGami, a miniaturized origami-based variable-stiffness actuator weighing 1.2 g for distributed joint-space integration. By combining origami geometric interlocking w

The usefulness of a newly developed ultrasonographic measurement method for predicting post-operative stiffness after arthroscopic rotator cuff repair.

BACKGROUND: Post-operative shoulder stiffness (POSS) is one of the most common complications following arthroscopic rotator cuff repair (ARCR) and is thought to be caused by pathologic changes in the anterosuperior soft-tissue structures of the glenohumeral joint. The aim of this study was to determine whether objective ultrasonographic evaluation can predict the occurrence of POSS at 1 year after surgery and to investigate the reliability of a newly developed ultrasonographic measurement method. METHODS: This retrospective cohort study included patients who underwent ARCR at a single institu

Novel prefabricated joints for underground silos with steel cavity and concrete-filled cavity: Axial compression tests and numerical simulations.

In terms of new materials, innovative technologies, and advanced construction techniques, the prefabricated steel plate-concrete composite underground silo offers an effective solution to the key challenges of cost, construction duration, and waterproofing associated with conventional reinforced concrete underground silos. However, joints represent critical weak components in prefabricated structures, and the development of simple and efficient joint configurations is essential for the further advancement and application of novel prefabricated underground silos. To address this issue, two nove

Immobilisation versus early active motion following primary flexor pollicis longus tendon zones 1-2 repair: protocol for a registry-based multicentre randomised controlled trial in Sweden.

INTRODUCTION: Flexor pollicis longus (FPL) tendon injuries in the thumb are surgically challenging and require careful postoperative rehabilitation to avoid complications such as tendon rupture, stiffness and impaired hand function. Rupture rates after FPL repairs are substantially higher than those reported for finger flexor tendons. Although controlled early active motion (EAM) is widely used following FPL repairs, rehabilitation protocols developed for finger flexor tendons may not be directly transferable to the thumb because of differences in anatomy, biomechanics and functional demands.

Novel image analysis method for estimating muscle strain from ultrasound B-mode images.

BACKGROUND AND OBJECTIVE: Accurate quantification of muscle strain is essential for understanding musculoskeletal mechanics. Ultrasound-based methods for estimating muscle strain commonly rely on muscles with clearly visible fascicles and aponeuroses. We aimed to develop a novel ultrasound image analysis method for estimating muscle strain, particularly in muscles with indistinct aponeuroses. METHODS: Ultrasound B-mode images of the forearm flexor muscles were acquired during passive wrist extension in 16 healthy adults. Muscle strain was estimated using a feature-based tracking approach impl

Simultaneously leveraging individual- and summary-level external data in clinical trials.

With the growing availability of external data (e.g., real-world data (RWD) and historical data), there is increasing interest in integrating external data into Randomized Controlled Trials (RCT), especially for rare diseases with high unmet medical needs where traditional clinical trials would be impractical and/or unethical. However, despite the increasing availability of external data, the level of detail varies across sources, as some provide individual-level patient records while others offer only aggregated summary statistics, often due to privacy and regulatory constraints. Most existin

Identification of human tooth germ ectomesenchymal stem cells.

Cranial neural crest cells (CNCCs) contribute to most cranio-maxillofacial mesenchymal tissues, including the dental ectomesenchyme. However, the developmental relationships between human CNC-derived progenitors and the odontogenic lineage remain incompletely understood. Here, we mapped the single-cell transcriptomic landscape of prenatal human mandibles and reconstructed the putative differentiation trajectories of CNC-derived ectomesenchymal populations. Notably, we identified a distinct population of SLITRK6+ cells enriched in the odontogenic ectomesenchyme. These cells were detected in pre

Site-specific m1A Installation by the MTR1 Ribozyme and Modulation of Mitochondrial mRNA Translation in vitro.

RNA modifications play essential roles in regulating RNA structure, stability, and translation. Among these, m1A is a functionally important modification that influences Watson-Crick base pairing and translational fidelity. However, approaches for site-specific installation and func tional interrogation of m1A in RNA remain limited. Here, we characterize the catalytic properties of the methyltransferase ribozyme MTR1 with a focus on donor compatibility, sequence-dependent kinetics, and activity on structured RNA substrates. We show that MTR1 catalyzes alkyl transfer from O6-methylguanine- deri

Auditory Biofeedback Gait Training in Individuals With Chronic Ankle Instability: Protocol for a Randomized Controlled Trial.

BACKGROUND: Chronic ankle instability (CAI) develops in a large majority of patients with lateral ankle sprain (LAS), who often experience recurring injuries, residual feelings of instability, and abnormal neuromuscular patterns. Specifically, gait alterations are common in those with CAI and considered a contributing factor to poor long-term ankle joint health. Yet, there are no evidence-based gait-training protocols targeted to improve biomechanics, which may improve long-term outcomes related to ankle joint health and patient satisfaction. OBJECTIVE: This study protocol introduces a progre
