Proteomic analysis of oxidative modifications in human atherosclerotic plaques and control arteries.

Cardiovascular disease is a major cause of worldwide mortality. A prime underlying cause is atherosclerotic plaque formation at specific arterial locations. Considerable evidence links plaques with chronic inflammation and leukocyte activation, with this potentially resulting in enhanced oxidant-mediated damage to arterial proteins. In this study, human carotid artery plaques and control tissue (superior thyroid artery) were analyzed by liquid chromatography-mass spectrometry (LC-MS/MS) for the presence of post-translational modifications (PTMs) generated by redox reactions on proteins. Of 715

Levodopa-induced dyskinesia provokes inflexible head-pelvis coordination during standing in Parkinson's disease.

BACKGROUND: To achieve postural control, individuals must orient their body segments to each other and maintain equilibrium with gravity. However, levodopa, the standard and most effective medication for the symptomatic treatment of Parkinson's disease (PD), also increases postural sway and head instability in people with levodopa-induced dyskinesia (LID). LID is characterized by involuntary, uncontrollable movements occurring after levodopa intake. This study investigated the effect of LID on body segment coordination while standing in PD. METHODS: People with PD (13 with LID, 12 without LID

Assessment of the posterior pelvic ring during examination under anesthesia changes surgeons' classification and treatment decisions of anterior posterior compression pelvic ring injuries.

PURPOSE: Traditionally, examination under anesthesia (EUA) focuses on displacement of the anterior pelvic ring and may fail to detect or localize sacroiliac (SI) joint diastasis in patients with anterior-posterior compression (APC) pelvic injuries. This study evaluates the effect of directly observing sacroiliac (SI) diastasis during EUA, compared to observing only the anterior pelvic ring (AP EUA), on changes in classification and treatment plan. METHODS: Four hundred international pelvic surgeons were invited to participate in an online survey, with one month between two surveys. All survey

Design and biomechanical investigation of a patient-specific total talar prosthesis for total talar replacement: A finite element analysis.

BACKGROUND: Total talar replacement (TTR) is an emerging treatment for end-stage talar diseases, although concerns persist regarding ankle stability due to essential ligament transection. Further investigation is necessary regarding the biomechanical impact of patient-specific total talar prosthesis (TTP) on ankle stability and bone-implant mechanics. OBJECTIVE: The purpose of this study is designing a patient-specific TTP and assess its biomechanical performance on ankle stability and stress distribution under various loading conditions using finite element analysis. METHODS: A CT scan is u

Patella Alta and Trochlear Dysplasia as a Determinant of Patellar Tendon Lateralization and Inclination: A Cross-sectional MRI Study in Adolescents With Patellofemoral Instability.

BACKGROUND: Patellofemoral instability (PFI) is traditionally assessed using tibial tubercle-trochlear groove (TT-TG) distance, patellar height indices, and trochlear morphology. However, soft tissue-based measures of patellar tendon orientation may provide complementary insight into patellar containment and extensor mechanism malalignment. Patellar tendon-lateral trochlear ridge distance (PT-LTR) has been proposed as a tendon-based marker of lateralization. The authors introduce the patellar tendon inclination angle (PTIA): a magnetic resonance imaging (MRI)-based parameter designed to quanti

Androgen Receptor-Immune Crosstalk in Metastatic Castration-Resistant Prostate Cancer: From Context-Dependent Remodeling to Biomarker-Guided Translation.

BACKGROUND: Metastatic castration-resistant prostate cancer (mCRPC) rarely responds to immune checkpoint blockade despite clear evidence that androgen receptor (AR) signaling intersects with tumor immunity. The view that androgen deprivation uniformly enhances antitumor immunity does not explain the repeated failure of immunotherapy combinations in biomarker-unselected populations. METHODS: This narrative review synthesizes mechanistic, translational, and clinical evidence and organizes it by cellular compartment, treatment-associated immune state, clinical trial strategy, and biomarker readi

Cancer-Associated Fibroblasts Drive Papillary Thyroid Cancer Progression through ROS/NF-κB/IL-6 Signaling in Hypoxic Microenvironment.

Cancer-associated fibroblasts (CAFs) are key components of the tumour microenvironment (TME) in papillary thyroid carcinoma (PTC) and play a critical role in disease progression. We previously reported that interleukin-6 (IL-6) promotes PTC progression through activation of MAPK signalling. However, whether CAFs serve as a major source of IL-6 and the underlying regulatory mechanisms remain incompletely understood. By integrating single-cell RNA-seq data from PTC patients, we identified fibroblasts as the predominant cellular source of IL-6, with IL-6 expression positively correlated with the

Differential somatostatin sensitivity of corticotropic and somatotropic responses following amino acid infusion.

PURPOSE OF STUDY: A new clinical test named "The shoulder anterior jerk test" is proposed to confirm the diagnosis of chronic shoulder anterior instability. MATERIAL: The test combines of a compression force and a translation force, applied along the arm between the humeral head and the glenoid cavity. In so doing, a subluxation of the humeral head is provoked and it is accompanied with a jerk recognised by the patient as his instability. METHODS: Three different populations of patients had been studied: Population A: 28 patients operated on for chronic anterior dislocation, uni or bilateral

Shape Matters: Few-Shot Object Classification From High-Information Contour Features.

INTRODUCTION: Acromioclavicular joint dislocation (AC) - Rockwood III (RIII) is a controversial topic with a wide range of therapeutic approaches. Operative therapy offers dozens of stabilization methods, which only confirms the absence of a "gold standard". The currently available literature tends to favor conservative therapy, involving several consecutive phases of physiotherapeutic care after the pain has subsided. The aim is to gradually improve the mobility of the shoulder and subsequently strengthen and stabilize the entire shoulder girdle. METHODS: A study was conducted between 01/201

Targeted Therapy for Advanced Gastrointestinal Stromal Tumors: Evolution and Future Directions.

Gastrointestinal stromal tumor (GIST) is the most common mesenchymal tumor of the gastrointestinal tract, with its pathogenesis primarily linked to activating mutations in the KIT or platelet derived growth factor receptor alpha (PDGFRA) genes. Surgical resection remains the standard curative treatment for localized GIST; however, ~50% of patients eventually develop recurrence or metastasis. Since the introduction of imatinib in the early 21st century, the management of metastatic GIST has shifted from solely surgical intervention to a systemic, chronic disease management model centered on tyr

Flexible Triple Fixation for Chronic Syndesmotic Instability: A Technical Tip.

BACKGROUND: Chronic syndesmotic instability alters normal ankle biomechanics and impairs load distribution, potentially leading to degenerative arthritis. The aim of this study was to describe a technical tip for the treatment of chronic tibiofibular instability using a triple-fixation construct, combining a suture button and FiberTape. METHODS: We described the triple-fixation technique and reviewed the cases of 3 patients, aged 22, 25, and 53 years, all diagnosed with chronic syndesmotic instability associated with incongruence of the distal tibiofibular joint. RESULTS: The mean preoperati

Comparing the Effects of Lateral Opening Wedge Distal Femoral Osteotomy and Medial Closing Wedge High Tibial Osteotomy on Tibial Tubercle-Trochlear Groove Distance: A 3D Computed Tomography Simulation Study.

BACKGROUND: Genu valgum is a known risk factor for recurrent patellar instability, and surgical correction of deformity can be utilized as part of the management strategy to improve tracking and optimize outcomes. Tibial tuberosity-trochlear groove (TT-TG) distance is a widely used objective measurement of the lateral quadriceps force vector in patients with patellar instability. The evidence documenting the effect of lateral opening wedge distal femoral osteotomy (LOWDFO) and medial closing wedge high tibial osteotomy (MCWHTO) on TT-TG is limited, with minimal data directly comparing the biom

Engineering a Wnt-activated osteomimetic microenvironment with iPSC-derived MSCs for synergistic bone regeneration, angiogenesis, and neurogenesis.

To advance the bioactivity of artificial bone grafts, human induced pluripotent stem cell (hiPSC)-derived mesenchymal stem cells (iMSCs) are ideal seed cells. However, the instability in iMSC generation and their limited osteogenic capacity challenge their clinical application. In this study, we applied a novel strategy that combines a stable iMSC induction method that mimics the developmental process with an osteocytic Wnt-based osteogenic microenvironment, with the aim of creating a scalable, mechanistically defined platform to enhance bone regenerative efficiency. The small-molecule Wnt act

[Knee osteotomies-Indication, planning and technical considerations].

Knee osteotomies used to correct lower limb malalignment, help to preserve the knee joint and to correct ligament or patellar instability. With recent advancements in osteotomy research, decision making on whether to perform a single level, double level or multiplanar osteotomy focuses on patient's specific anatomy and demands. In knee osteotomy safety, reproducibility, precision and meticulous planning are paramount. Anticipation and management of possible complications and preventive strategies for complications, such as unintentional changes of alignment in other planes are essential skills

User needs, prosthetic design priorities, and functional movement assessments for children with above/through-knee limb absence in Cambodia: a mixed methods interactive interview.

PURPOSE: Prosthetic user needs for children with above/through-knee (A/TK) limb absence in low-resource environments have not been defined at a discrete component level. This knowledge gap hinders design innovation. Proxy-reports from other stakeholders are insufficient. This study aims to define these user needs through interactive interviews with children in Cambodia. MATERIALS AND METHODS: Children were recruited across three centres in Cambodia. A novel interactive interview guide including card games provided quantitative and qualitative data. A functional movement assessment provided in
