Cationic liposome-mediated intra-articular delivery of lorecivivint for osteoarthritis treatment.

Osteoarthritis (OA) is the most common joint disease worldwide causing cartilage loss, inflammation and pain. Lorecivivint (LOR) is a new disease-modifying osteoarthritis drug (DMOAD) that targets the Wnt pathway to improve cartilage regeneration and reduce the pro-inflammatory cytokines production. However, its advancement in the clinical evaluation is facing certain challenges due to poor solubility and fast clearance from the joint. Our study is the first to improve LOR delivery using cationic liposomes for intra-articular injection. DSPC-Chol-DOTAP-based liposomes were formulated using mic

Erianin protects chondrocytes against IL-1β-induced oxidative stress and ferroptosis by activating GPX4/STING signaling in osteoarthritis.

BACKGROUND: Erianin (Eri) has been known for its analgesic and antipyretic properties. This research focuses on impact of Eri on chondrocyte viability, inflammatory cytokine production, extracellular matrix (ECM) degradation, and ferroptosis, which are key factors in cartilage diseases. METHODS: The mouse model of osteoarthritis (OA) was induced by destabilization of medial meniscus (DMM). Chondrocytes were treated with different concentrations of Eri and exposed to IL-1β to simulate disease conditions. The chondrocytes were induced to undergo ferroptosis using erastin (Era), and ferroptosis

Dihydrolipoic acid suppresses ferroptosis in chondrocytes to ameliorate the progression of osteoarthritis by modulating the FOXO1/TXNIP signaling pathway.

BACKGROUND: Osteoarthritis (OA), a common degenerative joint disorder, currently lacks effective therapeutic strategies to alleviate its progression. This study aims to investigate the function and underlying mechanisms of dihydrolipoic acid (DHLA) in inhibiting ferroptosis in chondrocytes and alleviating OA progression. METHODS: Mouse primary chondrocytes were exposed to IL-1β to induce ferroptosis and treated with DHLA in vitro, followed by the assessment of ferroptosis-related markers and indicators of chondrocyte anabolism and catabolism. The underlying therapeutic mechanisms of DHLA in O

Artemisia-derived carbon dots with excellent pharmacological activities and ROS scavenging for osteoarthritis treatment.

The pharmacological activities of traditional herbal medicine-derived carbon dots (THM-CDs) render them highly promising for applications in biomedicine. However, the interactions between THM-CDs and intracellular proteins have yet to be fully elucidated. Herein, inspired by the traditional vinegar-processing of Artemisia argyi (A. argyi), acetic acid-treated Artemisia argyi-derived carbon dots (AA-CDs) demonstrate excellent resistance to iron death via efficient ROS scavenging and Fe2+ chelating. Notably, multi-omics analysis uncovers a novel whereby AA-CDs directly target and stabilize heat

Targeting the mitochondrial metabolite-dynamics-MDVs-MitoEVs axis: a new frontier in osteoarthritis management.

BACKGROUND: Osteoarthritis (OA) is characterized by progressive cartilage degradation, osteophyte formation, and synovitis. OA progression is linked to mitochondrial metabolic dysfunction, marked by tricarboxylic acid (TCA) cycle imbalance and impaired oxidative phosphorylation (OXPHOS). KEY SCIENTIFIC CONCEPTS OF REVIEW: Three critical factors regulate mitochondrial metabolism: mitochondrial metabolites, mitochondrial dynamics, and mitochondrial-derived vesicles (MDVs). Mitochondrial metabolites such as fumarate and succinate exacerbate OA pathogenesis by mediating mitochondrial dysfunction,

Basic calcium phosphate crystals aggravate senescence-related osteoarthritis through GPX4-NRF2-mediated ferroptosis.

Osteoarthritis (OA) is a degenerative joint disorder strongly associated with senescence, involving pathological processes such as reactive oxygen species (ROS) accumulation and ferroptosis. Basic calcium phosphate (BCP) crystals are frequently found in the joints of OA patients, yet their role in OA pathogenesis remains poorly understood. Here, we aimed to investigate the role of synthetic BCP crystals in promoting OA progression and to elucidate the molecular mechanisms underlying senescence. In this study, a rat OA model was established by intra-articular injections of BCP crystals into the

Early Unloading After ACL Rupture and Prior to Surgical Restabilization in Mice Slows Post-Traumatic Osteoarthritis Progression.

PURPOSE: People who sustain joint injuries such as anterior cruciate ligament (ACL) rupture often go on to develop post-traumatic osteoarthritis (PTOA). ACL injuries are often treated with ACL reconstruction, but there is typically a gap of several weeks between injury and surgery. However, it is unclear how loading or unloading of the injured joint during the early postinjury period affects the progression of PTOA. The goal of this study was to determine how unloading between noninvasive ACL injury and surgical restabilization of the injured joint affects PTOA progression in mice. FINDINGS:

Structural remodeling impact of osteophytes and Schmorl's nodes on lumbar vertebral morphometry: anatomical basis for spinal instrumentation.

BACKGROUND: Precise anatomical knowledge is fundamental for successful spinal instrumentation, including pedicle screw fixation and interbody fusion. However, degenerative alterations such as osteophytes and Schmorl's nodes (SNs) can distort standard bony landmarks. This study aimed to evaluate how these specific degenerative features influence vertebral morphology and to identify anatomical variations relevant to surgical planning and implant selection. METHODS: Ninety dry lumbar vertebrae were analyzed. Linear and angular morphometric parameters were measured using digital calipers and goni

Osteophyte Bony Balancing in Robotic Total Knee Arthroplasty: A Surgical Technique and Predictive Algorithm for Soft Tissue Laxity.

The removal of osteophytes during total knee arthroplasty (TKA) results in reduced soft tissue tension, which may result in joint laxity. Thus, for gap balancing, a surgeon may try to predict the effect of osteophyte removal on the resulting flexion and extension gap before any bone cuts are made and before those osteophytes are removed. Posterior osteophytes, however, are relatively inaccessible, since their removal can be done only after posterior bone cuts are made on the femur. Any laxity created by posterior osteophyte removal cannot be corrected by adjusting bone cuts because they have a

The relationship between early life and pain among individuals with knee osteoarthritis: findings from the Hertfordshire Cohort Study.

Studies have linked lower birth weight to development of radiographic osteoarthritis (OA). We examined early life factors in relation to subsequent knee pain among individuals with radiographic OA. 143 participants from the UK Hertfordshire Cohort study were included. Birth weight and weight at one year (WA1) were ascertained from health ledgers and used to derive conditional infant weight gain (CIWG). At baseline and follow-up, heath questionnaires (including knee pain) and knee radiographs were collected. Only those with radiographic knee OA at baseline were analysed. Logistic regression was

Functional positioning in robotic lateral unicompartmental knee arthroplasty: a step-by-step technique.

Lateral unicompartmental knee arthroplasty (UKA) represents 1-2% of knee replacement procedures, yet offers distinct advantages including reduced surgical burden, bone stock preservation, and faster functional recovery. However, lateral UKA presents unique technical difficulties due to the surgical complexity of the lateral compartment. Recent advances in image-based robotic systems have demonstrated improved accuracy in implant positioning and promoted more individualized surgical strategies. This article presents a step-by-step surgical technique for lateral UKA using Functional Positioning

Semaglutide ameliorates osteoarthritis progression through a weight loss-independent metabolic restoration mechanism.

Metabolic disorders have been recognized as a major contributor to the occurrence and progression of osteoarthritis (OA). Identifying novel therapeutic agents to ameliorate the progression of OA with metabolic disorder is crucial. In this study, we demonstrate that semaglutide (SG), a glucagon-like peptide-1 receptor (GLP-1R) agonist, exhibits strong chondroprotective effects in an OA mouse model with obesity, as evidenced by reduced pathological changes, including cartilage degeneration, osteophyte formation, synovial lesion, and pain sensitivity. Emerging Role of TRP Channels in Osteoarthritis Pathogenesis.

Osteoarthritis (OA) is a degenerative joint disease characterized by cartilage degradation, synovial inflammation, osteophyte formation, joint space narrowing, and persistent pain. During OA progression, synovial inflammation triggers the release of pro-inflammatory cytokines, including IL-1β, TNF-α, and IL-6, which activate matrix metalloproteinases (MMPs) and aggrecanases, driving extracellular matrix (ECM) degradation. Emerging evidence indicates that transient receptor potential (TRP) channels, via calcium (Ca2+) signaling, function as molecular sensors in joint tissues, including chondroc

Overcoming First Rib Obstruction During Left Internal Thoracic Artery Harvesting in Minimally Invasive-Coronary Artery Bypass Grafting: A Practical Approach Using Ultrasonic Bone Sculpting.

Harvesting the left internal thoracic artery (LITA) during minimally invasive coronary artery bypass grafting (MICS-CABG) can be technically demanding, particularly in patients with a prominent osteophyte at the first costochondral joint. We report the use of an ultrasonic aspirator system, the SONOPET iQ (Stryker), to facilitate safe and effective LITA harvesting in such anatomically challenging cases. The SONOPET iQ was employed when proximal LITA exposure was hindered by the protrusion of the first rib at the costochondral junction. Operating at an ultrasonic frequency of 25 kHz, the device
