Placental immunopathology and congenital transmission of Zika virus and Trypanosoma cruzi: comparative mechanisms, immune disturbances, and potential co-infection scenarios.

Congenital transmission of Zika virus (ZIKV) and Trypanosoma cruzi (T. cruzi) remains a major public health concern in the Americas. Both pathogens cross the placental barrier and cause severe neonatal outcomes, yet no confirmed congenital co-infections have been reported, and their combined effects on maternal-fetal health are unknown. Critical gaps persist regarding placental immunopathology, fetal immune programming, and long-term consequences of prenatal exposure. This review synthesizes current evidence on the routes and mechanisms of placental transmission of ZIKV and T. cruzi, their tro

Precancerous cervical lesions and associated factors among HIV-positive women in Southern and Eastern African countries: A systematic review and meta-analysis.

BACKGROUND: Women living with HIV (WLHIV) face a sixfold increased risk of cervical cancer compared to HIV-negative women. Most WLHIV residing in the Southern and Eastern African regions have a substantial HIV-related cervical cancer burden. Studies on precancerous cervical lesions (PCCL) and associated factors in WLHIV from these regions have inconsistent and inconclusive findings. Therefore, we aimed to synthesize the pooled prevalence of PCCL and associated factors among WLHIV in Southern and Eastern Africa. METHODS: This study was registered under PROSPERO (CRD420251123583) and conducted

Utility of recombinant Schistosoma bovis 22.6 kDa antigen for diagnosis of human urogenital schistosomiasis by ELISA: a cross-sectional study in Yewa North LGA, Ogun State, Nigeria.

BACKGROUND: In many endemic regions, accurately diagnosing urogenital schistosomiasis remains challenging, particularly in cases of low-intensity infection and limited laboratory capacity. Exploring antigens that cross-react between related schistosome species provides a promising alternative to conventional diagnostic approaches. Schistosoma bovis-an animal parasite closely related to S. haematobium and sharing the same Bulinus snail hosts in overlapping habitats- is a strong candidate for such studies. This work therefore assessed whether the recombinant S. bovis 22.6 kDa antigen (rSb22.6 kD

Gallopamil exhibits antiplasmodial activity against Plasmodium falciparum and Plasmodium vivax and shows predicted interactions with Plasmodium hexose transporters.

Malaria remains a major parasitic disease caused mainly by Plasmodium falciparum and Plasmodium vivax, and drug-resistant parasites continue to challenge disease control. Drug repurposing may help identify compounds with antimalarial potential. Here, we evaluated gallopamil, an L-type calcium channel blocker, using in vitro, ex vivo, in vivo and in silico approaches. Gallopamil inhibited the chloroquine-resistant P. falciparum W2 strain in vitro (IC₅₀ = 3.65 µM) and showed ex vivo activity against clinical isolates of P. falciparum and P. vivax from CEPEM/RO, with median IC₅₀ values of 6.4 and

Mechanosensation of rice root border cells activates serotonin-mediated pre-invasion behaviors in Meloidogyne graminicola via DEG/ENaC channels.

BACKGROUND: Plant-parasitic nematodes (PPNs) locate host roots primarily through chemosensation, yet the sensory events that convert root contact into the stylet thrusts required for host penetration remain obscure. Root border cells (RBCs), terminally differentiated cells shed from the root cap, constitute the first point of physical contact between the root and invading nematodes, but whether and how RBCs trigger pre-invasion behaviors is unknown. RESULTS: Here we showed that rice (Oryza sativa) RBCs induce preparasitic second-stage juveniles (pre-J2) of the rice root-knot nematode Meloidog

The multifaceted role of mesenchymal stem cells in infectious diseases: antimicrobial properties, immunomodulation and tissue repair.

BACKGROUND: Mesenchymal stem cells (MSCs) have emerged as promising therapeutic agents for infectious diseases owing to their antimicrobial, immunomodulatory, and tissue regenerative properties. Beyond their well-established role in tissue repair, MSCs and their secretome directly influence pathogen clearance through the secretion of antimicrobial peptides, extracellular vesicles, iron-sequestration mechanisms, and modulation of both innate and adaptive immune responses. KNOWLEDGE GAP: Despite extensive preclinical investigations, MSC responses are highly pathogen-dependent, with outcomes tha

Development of a centrifugation-based method for the qualitative molecular detection of Cryptosporidium spp., Giardia duodenalis and Enterocytozoon bieneusi in surface water samples.

Workflow for waterborne parasites DNA detection in surface water. Screening for environmental surveillance in monitoring and resource-limited settings. Zoonotic protozoa and microsporidia are waterborne pathogens with implications for animal and human health. Their resistant stages can persist in freshwater. Standard reference methods for water analysis are often limited to specific parasites, require large water volumes, and depends on expensive equipment. Practical alternatives are needed to facilitate parasite detection in water. This study aimed to develop a pellet-based workflow to allow

Cytomics-driven insights for investigating host-pathogen interactions.

Cytomics, the high-dimensional profiling of immune cells, provides a systems-level view of host responses to infection. Unlike traditional diagnostics focused on pathogen detection or early immune activation, cytomics captures rare and functionally specialized immune subsets linked to disease severity and clinical outcome. In this review, we explore how advances in spectral cytometry, mass cytometry, and multimodal single-cell platforms enable unprecedented resolution of immune complexity during infection by summarizing cytomics evolution, high-dimensional single-cell and multimodal technologi

Transcriptomic, untargeted metabolomic, and gut microbiota studies on the anticoccidial activity of the combination formula in broilers.

UNLABELLED: Coccidiosis, a devastating intracellular parasitic disease, poses a significant threat to the poultry industry. Traditional control methods, which involve the inclusion of anticoccidial drugs in feed, have raised concerns over veterinary drug residues. Consequently, there is a pressing need to identify suitable alternatives or complementary strategies to anticoccidial drugs, with plant-derived natural products emerging as a promising candidate. Our laboratory has previously identified a combination formula that demonstrates good anticoccidial activity. However, the underlying mecha

Implementation of Targeted Selective Treatment (TST) in a Commercial Smallholder Sheep Flock: Reduction in Anthelmintic Expenditure.

The adoption of targeted selective treatment (TST) is an alternative for the sustainable management of gastrointestinal nematodes in sheep. TST mitigates the development of anthelmintic resistance by reducing the number of animals drenched while preserving the parasite refugia population. This study analysed the reduction of anthelmintic (AH) usage and assessed the AH cost reduction derived from a TST program compared to the previous whole-flock treatment regimen on a commercial smallholder sheep farm. Data acquisition spanned a 4.75-year period on a smallholder meat sheep farm in Colombo, Par

Antifibrotic activity of Schistosoma japonicum-derived sja-miR-2a-3p in carbon tetrachloride-induced liver fibrosis.

BACKGROUND: Liver fibrosis is a common consequence of chronic liver injury and is driven largely by persistent activation of hepatic stellate cells and excessive extracellular matrix deposition. We previously identified Schistosoma japonicum-derived sja-miR-2a-3p as a parasite-derived microRNA with antifibrotic activity in schistosomiasis-associated liver fibrosis. Whether this cross-species regulatory molecule also modulates fibrogenesis beyond the context of schistosome infection remains unclear. METHODS AND RESULTS: We investigated the effects of sja-miR-2a-3p in a carbon tetrachloride-ind
