Toxoplasmosis in South America: a hybridization-driven disease burden?

Human-mediated migrations have reshaped Toxoplasma gondii in South America through massive hybridizations between Old World domestic lineages and wild native populations. We propose that this convergence of domestic adaptation and virulence traits from wild strains underlies the continent's unusually severe disease burden, particularly ocular toxoplasmosis, with major public health implications.

Eosinophil CD48 interactions with Candida albicans Als6 is protective in vitro and in mouse systemic candidiasis.

Eosinophils are innate immune cells with central roles in allergy, parasitic diseases and multiple inflammatory conditions. Moreover, their role in host-pathogen interactions has been well characterized. However, the role of eosinophils during fungal infection is poorly defined. In this study, we delineate the importance of eosinophils during C. albicans systemic infections. C. albicans is promptly phagocytosed by human eosinophils, but growing hyphae escape this mechanism by releasing the fungal toxin candidalysin, which causes eosinophil membrane damage and cell death. Concomitantly, eosinop

DANet a lightweight dilated attention network for malaria parasite detection.

Malaria remains a critical global health challenge, requiring accurate and efficient diagnostic tools, particularly in developing countries with limited medical expertise. Detecting malaria parasites from red blood cell (RBC) blood smear images is challenging due to subtle color variations, indistinct demarcation lines, and diverse parasite morphologies. While numerous deep learning models address these issues, their high parameter counts often hinder practical deployment. We propose DANet, a lightweight Dilated Attention Network with approximately 2.3 million parameters, designed for robust m

Computational insights into harmala alkaloids as novel antimalarial candidates against Plasmodium falciparum.

Malaria, caused by the Plasmodium falciparum, remains a significant global health challenge, accounting for approximately 50% of all cases and posing the greatest threat. Despite advances in control measures, malaria continues to cause an estimated one million deaths annually. The complex lifecycle of P. falciparum, involving both vertebrate hosts and anopheline mosquitoes, complicates eradication efforts. The parasite's resistance to existing antimalarial drugs, along with medication toxicity, necessitates innovative therapeutic approaches. Recent research has revealed that harmane, an alkalo

Malaria knowledge and preventive practices among caregivers of under-five children in Southwest Nigeria.

The knowledge and practice of malaria prevention and treatment among caregivers of children under the age of five is critical to reducing the morbidity and mortality caused by malaria in the under-five population. This study, therefore, assessed the knowledge and practices of caregivers of children under five years in Southwestern Nigeria towards malaria prevention. A hospital-based, cross-sectional study was conducted using a pretested, semi-structured, interviewer-administered questionnaire. Data were analyzed using multiple logistic regression analysis. Out of the 797 caregivers surveyed, 6

MicroRNA expression dynamics in mouse liver at different stages of Echinococcus multilocularis infection.

BACKGROUND: Alveolar echinococcosis (AE) is a globally widespread zoonotic disease caused by the larval stage of the tapeworm Echinococcus multilocularis, posing a high fatality rate and poor prognosis if not properly managed. Currently, effective vaccines or drugs for echinococcosis remain elusive. MicroRNAs (miRNAs) play crucial roles in various biological processes and are closely linked with parasite infection and pathogenicity. To date, there is limited knowledge about host miRNA profiles in E. multilocularis infection at different stages. Hence, exploring host miRNA expression patterns a

Identification of Cdc45-interacting partners uncovers two Leishmania donovani proteins involved in DNA replication.

BACKGROUND: Among fatalities due to parasitic diseases, deaths due to Leishmaniases rank second only to those caused by malaria. Endemic to 90 countries, more than a billion people are at risk of Leishmania infection. While there are no vaccines against Leishmaniases, treatments are available. However, the complexities of the treatment regimens, their high costs and toxic side effects, and most importantly the emerging problem of drug resistance, have continued to thrust forward research into this parasite's cellular processes. The parasite is digenetic, reproducing by binary fission in both h

The November-2023-March-2024 malaria epidemic in Zanzibar: a spatiotemporal epidemiological analysis.

BACKGROUND: In Zanzibar, malaria epidemiology has changed within the past 10 years, from focal, near-elimination transmission to recurrent and more widespread outbreaks. The observed changes culminated in a large-scale epidemic between November 2023 and March 2024 including 23,569 confirmed malaria cases. METHODS: This study investigated the epidemiological characteristics of the 21-week outbreak by characterizing the risk profile of affected individuals, identifying malaria hotspots across space and time, and determining the association between malaria incidence and precipitation and tempera

The second-round evaluation of financial incentives on malaria prevalence in the Lake Victoria Basin, Kenya: updated study protocol for a cluster-randomized controlled trial.

BACKGROUND: Despite significant progress in global malaria reduction since 2000, driven primarily by supply-side interventions including improvements in bed nets and medication availability, recent years have seen a stagnation in this decline. In certain regions, cases have even increased ([World Health Organization], [World Malaria Report], [2023]). This trend is particularly concerning in high-burden areas such as Suba South Sub-county in Homa Bay County, Kenya, where malaria remains a persistent public health challenge. However, demand-side barriers-such as lack of knowledge of the disease

Multiplex long-amplicon sequencing for comprehensive molecular surveillance of Plasmodium falciparum resistance to artemisinin and partner drugs in artemisinin-based combination therapies (ACTs).

BACKGROUND: Antimalarial resistance, especially artemisinin resistance, in Plasmodium falciparum threatens global malaria control. Molecular surveillance of Plasmodium falciparum drug resistance is critical for monitoring the efficacy of artemisinin-based combination therapies (ACTs). Current molecular surveillance tools for Plasmodium falciparum artemisinin resistance are restricted to predefined polymorphism hotspots in limited loci (Pfk13 domain), failing to capture novel mutations in emerging resistance genes (Pfcoronin, Pfubp1, Pfap2μ) or cover complete coding regions. This study addresse

Loss of NcBPK1 impairs bradyzoite differentiation and enhances virulence in Neospora caninum.

BACKGROUND: Neospora caninum is an apicomplexan parasite responsible for bovine neosporosis, a disease that leads to substantial economic losses in cattle due to abortion and reduced productivity. The pathogenesis of N. caninum is shaped by complex host-parasite interactions, and virulence is known to vary between strains. BPK1 (Bradyzoite pseudokinase 1), a pseudokinase previously identified as a potential virulence factor in Toxoplasma gondii, has not yet been functionally characterized in N. caninum. METHODS: To investigate the role of NcBPK1 in parasite virulence, a knockout strain (NcΔBP

Tuberculosis treatment outcomes after transfer or release from incarceration: a retrospective cohort study from Brazil.

BACKGROUND: Tuberculosis (TB) disproportionately affects people deprived of liberty (PDL). Prior studies have shown higher TB treatment completion rates among PDL compared to the general population. However, little is known about how incarceration-related movements such as transfers between facilities and releases to the community affect TB treatment outcomes. METHODS: We linked person-level incarceration data with TB notifications data from the Notifiable Disease Information System for the Brazilian state of Mato Grosso do Sul between January 2006 and December 2018. We constructed a cohort o

Chyluria With Coexisting Minimal Change Disease: An Uncommon Clinical Association.

Chyluria is a rare clinical entity defined by the presence of chyle in the urine, classically presenting as milky or cloudy urine. While typically associated with parasitic infections such as Wuchereria bancrofti in endemic regions, non-parasitic causes are less common and diagnostically challenging. We report the case of a 56-year-old male presenting with milky urine, dysuria, urinary hesitancy, and rectal discomfort. Urine triglyceride level was found to be >525 mg/dL (reference range <10 mg/dL). An extensive infectious and parasitic workup was negative. Renal biopsy revealed minimal change

Oxymatrine alleviates Echinococcus multilocularis infection by remodeling the liver immune microenvironment and intestinal flora homeostasis.

BACKGROUND: Alveolar echinococcosis (AE) is a zoonotic parasitic disease that poses a grave threat to human health. Recent studies have indicated that the gut microbiota plays a significant role in the pathogenesis of alveolar echinococcosis. Consequently, the quest for innovative microbial targeted modulators is anticipated to address the treatment of alveolar echinococcosis. Oxymatrine, an alkaloid extracted from the legume plant Sophora flavescens, has been demonstrated in research studies to regulate gut microbiota, thus treating various diseases, including rheumatoid arthritis and autoimm

Prevalence of lice infestation in alpaca crias from the southern Peruvian Andes.

Lice are very common ectoparasites of humans and other animals. These ectoparasites cause various health problems in the hosts, like alpacas, such as intense itching, skin irritation, hair or wool loss, anaemia, and, in severe cases, reduced milk and meat production due to the stress they cause in the host. Epidemiological studies in alpacas are necessary to determine the magnitude and distribution of lice infestation, which would assist in the implementation of control measures. The aim of this study was to determine the prevalence of lice infestation in newborn and young alpacas (crias) from
