BACKGROUND: Post-Kala-Azar Dermal Leishmaniasis (PKDL), a dermal sequel of apparently cured visceral leishmaniasis (VL), presents with macular or polymorphic lesions. A relationship, if any, between immune cell infiltration and cytokines potentially causing tissue-remodelling remains poorly defined, and was the focus of this study.
METHODOLOGY/PRINCIPAL FINDINGS: Skin biopsies along with plasma from PKDL patients and healthy controls were analyzed. Tissue sections were assessed for immune cell subsets (CD8 ⁺ T cells, CD68 ⁺ macrophages, CD20 ⁺ B cells, CD66b⁺ neutrophils), associated cytokine

BACKGROUND: For lymphatic filariasis (LF) elimination, the World Health Organization recommends multiple rounds of mass drug administration (MDA) to interrupt transmission. While LF antigen (Ag) is routinely used to monitor progress, recent evidence suggests more time-sensitive indicators are needed during the immediate post-MDA period. In Samoa, triple-drug MDA was distributed in 2018 and 2023. This study aimed to evaluate the impact of Samoa's second round of triple-drug MDA on human and mosquito-based indicators of transmission.
METHODOLOGY: Surveys were conducted in eight primary sampling

Background/Objectives: Healthcare-associated infections (HAIs) disproportionately burden low- and middle-income countries, but no such description has been published for Uzbekistan. We describe national trends in reported HAIs over 2015-2024 with their regional and clinical distribution and review the country's infection prevention and control (IPC) regulations against the eight World Health Organization (WHO) Core Components. Methods: We analysed the complete national series of reported HAI cases for 2015-2024 (ten annual counts; 817 cases in 2024, disaggregated by region and HAI type), with

Malaria exhibits pronounced sexual dimorphism, with males experiencing greater disease severity and mortality than females do. Although these differences have been partially attributed to the immunosuppressive effects of testosterone, the underlying mechanisms remain poorly understood. Here, we investigated the role of testosterone in sex-specific immune responses by administering a combination of letrozole and testosterone (LT) to male and female mice infected with Plasmodium berghei ANKA. LT treatment increased testosterone concentrations in both sexes and was associated with higher parasite

Giardiasis is a parasitic disease for which there is a growing need to develop new therapeutic alternatives due to the adverse effects associated with first-line treatments such as metronidazole and the increasing emergence of resistant strains. In this study, the in vitro antigiardial activity of three quassinoids, chaparrin (1), amarolide (2), and chaparrinone (3), isolated from the stems of Castela tortuosa Liebm., was evaluated. The compounds were obtained from the methanolic extract through fractionation and chromatographic purification, and their antigiardial activity was determined agai

Trypanosoma cruzi, Trypanosoma brucei and Leishmania spp. are the causative agents of Chagas disease, human African trypanosomiasis, and leishmaniasis, respectively. These three neglected tropical diseases affect millions of individuals globally, and current therapeutic options remain severely limited. The major cysteine proteases (CPs) of these parasites play critical roles in their infectivity and survival, making them subjects of prolonged research. Therefore, cruzain, TbrCATL and CPB from Leishmania spp. have been recognized as the primary targets of most drugs under investigation. Among t

Hepatic echinococcosis is a rare parasitic disease, with the incidence of the alveolar form increasing in Central Europe in recent years. Cystic and alveolar echinococcosis differ substantially in their biological behavior and management. The aim of this study is to provide an overview of the current literature and to present the experience from a tertiary center with the surgical treatment of this disease. The study combines a review from the literature with a retrospective analysis of patients who underwent surgical treatment at the University Hospital Hradec Králové between January 2022 and

We describe the case of a 16-year-old female patient from Bolivia, with a family history of pulmonary tuberculosis, who presented with a two-month history of ulcerated lesions on the dorsum of her left hand. A skin biopsy revealed amastigotes consistent with Leishmania spp., and antiparasitic treatment was initiated. Subsequently, Mycobacterium tuberculosis was also isolated, and antituberculosis treatment was started, resulting in a favorable outcome.

Toxoplasma gondii persists in the central nervous system and may cause severe toxoplasmic encephalitis, particularly in immunocompromised hosts. Our previous studies demonstrated that T. gondii-induced neuropathology involves oxidative and nitrosative stress, apoptosis, glial activation, blood-brain barrier injury, and GMF-β- and SAP/NF-κB-associated responses. This study investigated the temporal immunoreactivity of CD68, S100B, Foxp3, and IL-5 during early and chronic experimental infection. Twenty-four female Swiss albino mice were assigned to one uninfected control group and three ME49-inf

Primary amoebic meningoencephalitis (PAM), caused by Naegleria fowleri, is a rapidly progressive and fatal infection, and inflammation of the olfactory bulb plays a key role in disease pathogenesis. Although inflammatory and oxidative mediators have been studied in vitro, their production during the progression of PAM remains poorly understood. This study evaluated the temporal production of TNF-α, IL-1β, IL-6, nitric oxide (NO), and reactive oxygen species (ROS) in the olfactory bulbs of BALB/c mice from 1 to 7 days after intranasal infection with N. fowleri trophozoites. RT-PCR detected incr

BACKGROUND AND OBJECTIVES: Despite a low national prevalence of 0.20%, India's absolute burden of 2.544 million people living with human immunodeficiency virus (HIV) presents clinicians with varied neurological complications. While combination antiretroviral therapy (ART) has extended survival, the neuro-HIV profile in India, characterized by the predominance of HIV-1 clade C, remains distinct from that reported in Western cohorts. This scoping review aimed to identify patterns of neurological manifestations in India across the pre-ART, ART, and "treat-all" eras and to support the establishmen

INTRODUCTION: Over 300,000 babies are born annually with sickle cell anemia (SCA) in sub-Saharan Africa. Despite malaria chemoprevention and early penicillin prophylaxis, these children develop febrile illnesses. The effect of malaria chemoprevention regimens on the incidence of bacterial infections and antimicrobial resistance is unclear. We assessed incidence of bacterial infections and resistance patterns in children with SCA receiving weekly DP or monthly SP.
METHODS: This study was nested in a randomized, double-blind clinical trial comparing weekly DP with monthly SP. Participants were

BACKGROUND: In Vietnam, an upper-middle-income country, the majority of people living with HIV smoke. Vietnam has established quitline services to support this population, but they remain underused. Text messaging is widely used in Vietnam, and studies in other countries have shown that text messaging interventions can promote smoking cessation. Our goal was to conduct a formative study to inform the development of a text messaging intervention to increase access to quitline services and promote smoking cessation among people living with HIV who smoke in Vietnam.
OBJECTIVE: This study aimed t

BACKGROUND: Malaria causes immune perturbations and affects immunogenicity to vaccines. It is unknown, however, how repeated malaria infections affect the duration of vaccine protection. This study assessed variability in tetanus antitoxin immunoglobulin G (IgG) antibody kinetics including by malaria exposure history at district and individual levels.
METHODS: We analyzed tetanus antitoxin IgG antibody concentrations in 3064 plasma samples from 2011 to 2017 in 597 children aged 6 months to 10 years residing in the Tororo (high malaria transmission) and Jinja (low transmission) districts in Ug

BACKGROUND: A subset of people living with HIV (PLHIV), known as immunological non-responders (INR), fails to achieve adequate CD4⁺ T cell recovery despite viral suppression with antiretroviral therapy (ART). These individuals face an increased risk of adverse clinical outcomes. This study aims to explore the intracellular mechanisms and molecular signatures underlying this incomplete immune recovery using a multi-omics approach.
METHODS: The study analysed CD4⁺ and CD8⁺ T cells from 100 ART-naïve PLHIV. Participants were classified into controls (baseline CD4⁺ > 200 cells/µL, n = 49) and cas

Fasciolosis, a disease induced by liver fluke infection, poses a major threat to the livestock industry. The emergence of drug resistance and the lack of commercial vaccines necessitate the development of novel control strategies. Studies on helminth immunoregulatory molecules have become a key research focus. Bone morphogenetic proteins (BMPs), ligand members of the TGF-β superfamily, have been identified in Fasciola gigantica and designated FgBMPs, and are hypothesized to regulate host immune responses. To explore the effects of FgBMP on hosts, the mechanistic actions of FgBMP-1 on buffalo p

Introduction: HIV pre-exposure prophylaxis (HIV PrEP) and HIV post-exposure prophylaxis (HIV PEP) are established preventive strategies against HIV, while doxycycline post-exposure prophylaxis (doxy-PEP) is used to prevent bacterial sexually transmitted infections (STIs). Awareness, access, and use of these methods may differ between heterosexual persons and sexual minority individuals (SMIs). This study aimed to assess awareness of HIV PrEP, HIV PEP, and doxy-PEP among adults in Poland recruited through social media and to compare awareness according to self-reported sexual orientation. Progress in malaria control has stagnated since the early 21st century in many countries, requiring new approaches such as the use of spatially-targeted interventions. However, evidence on the effectiveness of spatially-targeted interventions is mixed, depending on both the setting and intervention. In addition, the potential role of community health programs in implementing spatial targeting is underexplored, with models focusing on coarser spatial scales than those needed to guide programmatic activities. We developed a two-age-class, spatially-explicit model of malaria at the community-scal

BACKGROUND: Malaria remains a major health burden, with an estimated 282 million cases and approximately 610,000 deaths recorded in 2024. The increasing prevalence of insecticide-resistant Anopheles vectors and drug-resistant Plasmodium strains has substantially eroded the efficacy of conventional control strategies, underscoring the critical need for innovative approaches. Accumulating evidence suggest that the mosquito midgut microbiome exerts a significant influence on Plasmodium sporogonic development; however, no scoping investigation has fully mapped the metagenomic evidence base in this

BACKGROUND: Tuberculosis (TB) and HIV co-infection are characterized by dysregulated inflammation and increased risk of immune reconstitution inflammatory syndrome (IRIS) following antiretroviral therapy (ART) initiation. Inflammasomes are key regulators of innate immune responses, but their role in TB-HIV co-infection and IRIS remains incompletely understood. We investigated inflammasome-related gene expression and cytokine profiles in TB, HIV-1, and TB-HIV individuals, and their association with IRIS development.
METHODS: Fifty-nine participants were stratified into four groups: TB-HIV (n =
