BACKGROUND: A key feature of the autoimmune conditions adult and juvenile dermatomyositis (DM/JDM) is the correlation of interferon (IFN) beta induction with disease initiation and activity. Our aim was to establish a short interfering RNA (siRNA) approach to knockdown IFNB1 expression as a therapeutic strategy for DM/JDM.
METHODS: A library of 74 siRNAs targeting IFNB1 was designed and screened for IFNB1 knockdown by qRT-PCR. Lead siRNAs were analysed for secreted IFN beta protein reduction and acute cytotoxicity. IFNB1 expression was induced for efficacy screening in vitro, including in imm

Background/Objectives: Idiopathic inflammatory myopathies (IIMs) are very rare, chronic, immune-mediated diseases that clinically manifest with myositis symptoms and extramuscular manifestations. The rarity of the disease makes it difficult to conduct double-blind studies, and large population-based studies are needed to improve our knowledge of them. This study aims to analyze the hospitalization rates and trends of IIMs between 2010 and 2024. Additionally, a characterization of the IIM patient population in Poland was conducted. Methods: A retrospective study using data from the Nationwide G

Flagellate erythema is an exceptionally rare cutaneous manifestation of idiopathic inflammatory myopathies (IIM) with poorly understood clinical significance. We report two patients who developed zebra-like flagellate erythema: one with immune-mediated necrotising myopathy (IMNM) and one with anti-Mi-2-positive cancer-associated myositis (CAM). To place these observations in broader context, we performed a thorough literature search across PubMed, Scopus and Embase and identified 34 previously published cases of IIM-associated flagellate erythema. The most frequent underlying diagnosis was der

Juvenile dermatomyositis (JDM) is an autoimmune myopathy with skin manifestations, yet biomarkers reflecting disease activity and organ involvement at presentation remain incompletely defined. This study aimed to identify clinical and immunologic indicators at diagnosis, focusing on myositis-specific autoantibodies (MSAs) and lymphocyte subsets. We retrospectively analyzed 36 patients with JDM treated at a tertiary center in Japan between 2003 and 2024. MSAs (anti-TIF1-γ, anti-MDA5, anti-NXP2, anti-Mi-2, anti-Jo-1, anti-ARS, and anti-SAE) were assessed at diagnosis. Clinical features, laborato

PURPOSE: Persistence of skin and wound infections is nowadays accepted being linked to bacterial biofilms, which are highly recalcitrant to treatments and contribute to maintain a constant inflammation state and prevent a correct healing. Topical antimicrobials are the most common first-line self-medications; however, treatment failure is not uncommon and emerging resistance to antibiotics is alarming. Chlorquinaldol is an antimicrobial with a wide spectrum of activity and desirable characteristics for topical application. Aim of this study was to evaluate the efficacy of chlorquinaldol to pre

Calcinosis is a disabling and difficult-to-treat complication of adult dermatomyositis (DM), juvenile dermatomyositis (JDM), and systemic sclerosis (SSc), for which no standardized therapy is currently available. Growing evidence implicating JAK/STAT signaling in dermatomyositis has prompted interest in Janus kinase inhibitors (JAKi) as a therapeutic option. We describe a 52-year-old woman with long-standing juvenile-onset dermatomyositis and extensive, treatment-refractory calcinosis who received off-label baricitinib after multiple previous therapies. After one month, the patient reported im

BACKGROUND: Serum tumor markers such as CA 125, CA 19-9, carcinoembryonic antigen (CEA), alpha-fetoprotein (AFP), and CA 15-3 are widely available, inexpensive, and easily measured, which has led to their inappropriate use for cancer screening in asymptomatic individuals. Despite guideline recommendations against this practice, these tests remain common, contributing to false positives, unnecessary anxiety, and avoidable invasive procedures.
METHODS: We conducted a systematic review of PubMed, Cochrane Library, and LILACS from inception to May 10, 2025, identifying studies evaluating the use

BACKGROUND AND PURPOSE: Postpartum depression (PPD) is a common childbirth complication that impairs the mother-infant relationship and contributes to long-term developmental challenges among children. Although PPD treatment reduces depressive symptoms, it is not necessarily sufficient to improve maternal-infant interaction. In addition, several barriers preclude timely access to care including mental health stigma, limited specialized providers, and challenges with navigating the healthcare system and securing necessary childcare and transportation. Our team developed a 12-session patient- an

OBJECTIVES: We assessed the validity of Patient-Reported Outcome Measurement Information System (PROMIS) for juvenile and adult-onset idiopathic inflammatory myopathies (JIIM/IIM) and determined associations with disease characteristics and assessments.
METHODS: PROMIS-49 or -57 questionnaires were completed by parents (n = 70) and pediatric (n = 49) or adult patients with IIM (n = 61). Construct validity was evaluated using legacy health-related quality of life (HRQoL) instruments and associations with disease assessments and clinical characteristics.
RESULTS: All groups reported decreased

OBJECTIVE: This study aimed to establish the role of myositis-specific antibodies (MSAs) in the association between type I interferon (IFN-I) plasma levels and disease activity in juvenile dermatomyositis (JDM).
METHODS: We prospectively obtained 400 samples from 101 JDM patients from two independent cohorts. Autoantibody levels were determined for all patients. Muscle activity was assessed using the Childhood-Myositis Assessment Scale (CMAS). Two characterized homebrew digital ELISAs measuring, respectively, all 12 IFN-α subtype protein and IFN-β were used to quantify IFN-I in patient plasma

OBJECTIVE: Juvenile dermatomyositis (JDM) is a rare autoimmune condition. The treat-to-target strategy has garnered interest in pediatric rheumatology. It is based on defining clear therapeutic targets, with frequent disease activity monitoring, and adjustment of the treatments if targets are not met within a defined time frame. Recently, an international task force of experts launched an initiative aimed at the development of recommendations for the adoption of treat-to-target strategy in JDM. This study was done to support those recommendations. We aimed to determine the time-to-treatment re

OBJECTIVES: To compare the severity at diagnosis and disease activity during follow-up between juvenile (jDM) and adult-onset (aDM) dermatomyositis.
METHODS: Patients with DM fulfilling the 2017 ACR/EULAR criteria and tested for myositis-specific antibodies were included. Overall severity at diagnosis was defined by at least one of the following: severe muscle impairment, symptomatic interstitial lung disease (ILD), gastrointestinal (GI) vasculitis, myocarditis, severe skin ulceration or admission to an intensive care unit. Clinically inactive disease and remission were defined according to P

Juvenile dermatomyositis (JDM) is a rare, idiopathic autoimmune disorder characterized by inflammation of both muscle and skin, with a significant contribution from the interferon (IFN) pathway in its pathogenesis. Here, we present the case of a 4-year-old boy with JDM who tested positive for Mi2-α and MDA5 antibodies and showed combined muscle and skin involvement. In view of his markedly elevated IFN signature, the Janus kinase (JAK) inhibitor baricitinib was introduced very early as a targeted steroid-sparing agent in addition to standard immunosuppressive therapy. The patient experienced m
