Disruption of ALX1 causes frontonasal dysplasia syndrome-3, characterized by extreme microphthalmia, severe midfacial hypoplasia, and orofacial clefting. Recent studies have revealed critical tissue-specific roles of ALX1 in patterning both cranial mesoderm and cranial neural crest-derived facial mesenchyme. However, the molecular mechanisms regulating Alx1 gene expression during craniofacial development are largely unknown. In this study, we have identified a distal enhancer (Alx1-DE1) residing in a large intron of the neighboring Lrriq1 gene and demonstrate that deletion of this enhancer spe

Sox8, Sox9, and Sox10 arose by multiple rounds of genome duplications from a single SoxE gene in ancestral vertebrates. In this review, we will briefly discuss the molecular structure and function of SoxE transcription factors and their evolutionary origin. We will then discuss their expression, function, and developmental disorders. SoxE proteins play critical roles during the development of multiple tissues in vertebrate embryos, including the neural crest, inner ear, cartilages, and glia cells of diverse origins, heart, gonads, and gastrointestinal tract. Because they recognize the same DNA

Mowat Wilson syndrome (MWS) is a rare neurodevelopmental disorder caused by mostly heterozygous loss-of-function variants in ZEB2. Affected individuals show considerable wide variability in clinical presentation. In particular, Hirschsprung disease (HSCR) occurs in only a subset of patients, suggesting that additional genetic factors may modify disease penetrance. To investigate this possibility, we performed whole-genome sequencing of two parent-child trios in which the probands carried pathogenic de novo ZEB2 variants but differed in enteric phenotype: one individual with MWS and long-segmen

Bosma arhinia microphthalmia syndrome (BAMS) is a rare congenital disorder characterized by the absence of a nose, along with eye and reproductive anomalies. BAMS is caused by heterozygous missense variants in SMCHD1, an epigenetic regulator. Despite uncovering its genetic basis, the cellular basis of the disease has remained elusive. The embryonic development of the nose involves direct contributions from both the neural crest and cranial placodes. Here, we differentiated patient-derived induced pluripotent stem cells toward the cranial placode lineage and found that they exhibited significan

BACKGROUND AND OBJECTIVES: Spinal arteriovenous metameric syndrome (SAMS) is a rare, complex vascular disorder characterized by multifocal spinal cord arteriovenous malformations (SCAVMs) affecting structures derived from the same metameric segment. The long-term clinical outcome of SAMS, particularly after embolization, remains poorly understood. This study evaluated the clinical outcomes of embolization for SAMS compared with nonmetameric SCAVMs.
METHODS: This retrospective study included 216 patients with intradural SCAVMs who underwent embolization (62 patients with SAMS and 154 patients

BACKGROUND: Rubinstein Taybi syndrome (RSTS), a rare congenital disease, is caused by mutations in lysine acetyl transferase type 3 (KAT3) genes, EP300 and CREBBP. Many of the tissues affected in RSTS are derived from the neural crest (NC). Hence, we proposed that NC development would be perturbed in RSTS.
RESULTS: Zebrafish RSTS models generated by knocking down or mutating ep300a and crebbpa genes reveal defects in NC migration. This effect on migration is conserved in NC cells generated from human RSTS patient-derived induced pluripotent stem cells (iPSC) cells. The defects in NC migration

Congenital heart disease (CHD) and dermatologic conditions such as lymphedema and acquired melanocytic nevi (AMN) are common in Turner Syndrome (TS). We hypothesized that abnormalities of cranial neural crest cell derivatives drive the skin and heart manifestations of TS. We conducted joint cardiac and skin examinations of volunteers at a 2023 national TS conference. Electrocardiographic abnormalities were classified according to the Minnesota Codes. Aortic diameters and cardiac anatomy were assessed using a standardized echocardiographic protocol. The primary outcome was the association betwe

Long COVID is characterized by persistent symptoms following acute SARS-CoV-2 infection, yet its biological mechanisms remain incompletely understood. Emerging evidence suggests that immune dysregulation, mitochondrial dysfunction, and altered cell survival pathways may contribute to prolonged symptomatology. In this cross-sectional study, peripheral blood mononuclear cells were collected from individuals with Long COVID approximately 10 months post-infection and from recovered individuals without Long COVID symptoms. Symptom burden was assessed using a composite domain-based score. mRNA expre

BACKGROUND: Superior cluneal nerve (SCN) entrapment is an underrecognized cause of chronic low back and gluteal pain and is frequently misdiagnosed as lumbar radiculopathy or sacroiliac joint dysfunction. Radiofrequency ablation (RFA) of the SCN has been previously described only at the level of the iliac crest. No reports exist on proximal SCN ablation at the level of the lumbar transverse processes.
OBJECTIVE: To describe a novel anatomy-based proximal technique for radiofrequency ablation of the superior cluneal nerve at the level of the L1-L3 transverse processes using the Transverse Proc

Cranial sutures are essential for skull growth and tissue homeostasis. Among them, the coronal suture is most frequently affected in syndromic craniosynostosis, yet the mechanisms underlying this preferential involvement remain unclear. Here, we show that the coronal suture mesenchyme undergoes a postnatal lineage transition from mesodermal to cranial neural crest origin, facilitated by dural cell migration into the suture. Mechanistically, this migration is regulated by suture TGFβ signals to Tgfbr2+ dural cells. Loss of dural Tgfbr2 impairs this cell migration into the suture, reduces the Gl

Congenital melanocytic naevi (CMN) are benign melanocytic proliferations originating from neural crest cells and presenting at birth or in early infancy with variable clinical manifestations and malignancy risks. The incidence of CMN varies by sex, with a higher prevalence in female patients but with potentially greater complications in male patients. Genetic studies have highlighted the pivotal role of mutations in BRAF and NRAS in influencing the size and malignancy risk of CMN. Diagnostic challenges necessitate a systematic approach, including histopathology, molecular profiling and risk-st

SIX1 variants underlying branchio-oto-renal syndrome occur in the SIX domain (SD) or homeodomain (HD). We tested whether different variants - V17E (SD), Y129C (HD) - cause distinct developmental phenotypes in Xenopus embryos with reduced Six1 in comparison to wild-type Six1 (Six1WT). In Six1 morphants, Six1WT restored neural crest and preplacodal gene expression; V17E restored foxd3 and irx1 better than Y129C, and Y129C restored sox11 better than V17E. In six1-null otic vesicles, Six1WT partially restored tbx1 and sobp, V17E was less effective and Y129C was least effective; all three restored

BACKGROUND: Hemifacial microsomia (HFM) is a congenital craniofacial malformation characterized by unilateral hypoplasia of structures derived from the first and second pharyngeal arches. Although the phenotype is well described, the underlying etiology remains incompletely understood. Disturbances in embryonic vascular development have long been proposed as a potential mechanism contributing to the characteristic unilateral craniofacial phenotype.
METHODS: A systematic literature review was conducted following PRISMA guidelines. Searches were performed in PubMed, Embase, Web of Science, and

BACKGROUND: The present video article demonstrates the use of Evans and Cotton osteotomies with porous titanium wedges for the treatment of pediatric flatfoot. Pediatric flatfoot is not commonly painful or disabling and usually resolves as the arch elevates spontaneously. However, in cases of flatfoot that persist despite nonoperative treatment, symptoms such as severe activity-related pain and medial foot calluses are an indication for surgical intervention3. The aims of treatment in pediatric patients are to correct anatomical deformity and reduce pain while maintaining mobility.

Background: Congenital central hypoventilation syndrome (CCHS) is a rare genetic disorder that causes alveolar hypoventilation because of impaired chemoreceptor response to hypercapnia and hypoxia occurring due to a dysfunctional central autonomic drive. CCHS is a result of pathogenic variants of the PHOX2B gene, which affects neural crest cell development. Consequently, CCHS can be accompanied by dysregulation of the autonomic system, Hirschsprung's disease and other gastrointestinal disorders, cardiac arrhythmias, and neural cell-derived tumors. Disease severity correlates with mutation type

Discordant ventriculo-arterial (VA) connection characterizes dextro-transposition of the great arteries (d-TGA) and congenitally corrected transposition of the great arteries (ccTGA). These distinct defects involve overlapping developmental processes related to cardiac looping, left-right patterning, and outflow tract (OFT) morphogenesis. Recent genetic studies in sporadic and familial cases support second-heart field and neural crest involvement and an oligogenic architecture shaped by gene-environment interactions. Consistent with this framework, environmental perturbations, including retino

Housekeeping genes maintain robust expression across cell types despite dynamic transcription factor fluctuations, yet their haploinsufficiency is associated with many tissue-specific developmental disorders. To understand this paradox, we focus on TCOF1, a broadly expressed regulator of rRNA synthesis, whose haploinsufficiency causes Treacher Collins syndrome (TCS). We show that transitional cranial neural crest cells (tCNCC) undergoing mesenchymal specification exhibit extreme dosage-sensitivity to TCOF1, but not to RNA polymerase I, explaining both cellular origins of TCS and prevalence of
