Generation of human induced pluripotent stem cell lines derived from Wolf-Hirschhorn syndrome patients with chromosomal 4p deletion.

Wolf-Hirschhorn syndrome (WHS) is a devastating congenital disease caused by deletions on the short arm of chromosome 4 (4p), for which no curative treatments currently exist. To facilitate the development of therapeutic strategies, the development of experimental models of WHS is crucial for investigating its etiology and pathogenesis, which remain elusive. In this study, we successfully generated human induced pluripotent stem cells (hiPSCs) from three fibroblast lines from WHS patients. We then characterized these hiPSCs, along with one hiPSC line previously generated from peripheral blood

Evolution of BA.2.86 to JN.1 reveals that functional changes in non-structural viral proteins are required for fitness of SARS-CoV-2.

UNLABELLED: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of coronavirus disease 2019 (COVID-19), is still circulating among humans, leading to the continuous evolution. SARS-CoV-2 Omicron JN.1 evolved from a distinct SARS-CoV-2 lineage, BA.2.86, and spread rapidly worldwide. It is unclear why BA.2.86 did not become dominant and was quickly replaced by JN.1, which possesses one amino acid substitution in the spike protein (S:L455S) and two in the non-spike proteins NSP6 and ORF7b (NSP6:R252K and ORF7b:F19L) compared to BA.2.86. Here, we utilized recombinant

Craniofacial Microsomia With Tongue Cleft: Embryological Mechanisms and Clinical Implications From Three Rare Cases.

Craniofacial microsomia (CFM) accompanied by a congenital tongue cleft is an exceedingly rare developmental anomaly, with fewer than 10 cases reported globally. This study presents 3 nonsyndromic cases of CFM with concurrent midline tongue cleft, characterized by mandibular hypoplasia (M2B), auricular malformations (E1-E3), soft tissue deficiencies (S1-S3), and hypoglossal/facial nerve dysfunctions. Despite the morphologic presence of tongue clefts, all cases were functionally benign and managed conservatively. Surgical treatment focused on mandibular distraction osteogenesis and auricular rec

Electrocardiogram Diagnosis: ST-Depression With T Wave Inversion in Lead Augmented Vector Left as Harbinger of Inferior ST-Elevation Myocardial Infarction.

A 53-year-old male presented to the emergency department (ED) after waking up with retrosternal chest discomfort. An initial prehospital 12-lead electrocardiogram (ECG) obtained by emergency medical services indicated sinus rhythm with ST-segment depression and T wave inversion in lead augmented vector left (aVL) but without significant ST-segment elevations. Upon ED arrival, a repeat ECG demonstrated minimal ST-segment elevation in the inferior leads, raising concern for an evolving inferior ST-segment elevation myocardial infarction (STEMI) and prompting cardiac catheterization laboratory ac

Case Report: A novel heterozygous variant of the TCOF1 gene in Treacher Collins syndrome.

Treacher Collins syndrome (TCS) is a craniofacial malformation caused by the abnormal development of the first and second pharyngeal arches during embryogenesis. While pathogenic variants in POLR1B, POLR1C, and POLR1D are implicated, the TCOF1 gene represents the primary causative locus. This case report describes a novel heterozygous frameshift variant, TCOF1:NM_001135243.2:c.3559delG (p.Ala1187GlufsTer21), identified in a child with TCS. This truncation disrupts the C-terminal nuclear localization signal (NLS), specifically ablating a phosphorylation site at residues 1,199-1,200. Consequentl

Tracking domestication signals across populations of North American raccoons (Procyon lotor) via citizen science-driven image repositories.

North American raccoons are widespread across the contiguous United States and live in close proximity to humans (i.e. urban) and in rural environments. This makes them an excellent species for comparative work on the effects of human environments on phenotypic traits. We use raccoons as a mammalian model system to test whether exposure to human environments triggers a trait of the domestication syndrome. Our data suggests that urban environments produce reductions in snout length, which are consistent with the domestication syndrome phenotype. These results are crucial for the discussion of t

Lack of Williams syndrome-associated genes alters quantity discrimination in zebrafish.

The ability to discriminate sets of items based on their numerosity is alleged to be an evolutionary conserved mechanism in all vertebrates. People with Williams syndrome (WS), a rare multigenic condition, show altered number and quantity cognition abilities. Assessing the contribution of specific genes to WS using animal models could help understand the basis of numerical impairments. Here, we assessed the quantitative abilities of juvenile zebrafish (Danio rerio) with loss of function of two of the genes affected in WS using a group size preference behavioural assay. The selected genes were:

The PHOX2B c.428A>G missense variant affects post-transcriptional regulation and may explain the absence of neural crest-derived tumors in congenital central hypoventilation syndrome.

INTRODUCTION: Heterozygous mutations in the paired-like homeobox 2b (PHOX2B) gene cause congenital central hypoventilation syndrome (CCHS). While polyalanine expansions are almost exclusively associated with isolated CCHS, missense, nonsense, and frameshift mutations are mainly identified in syndromic CCHS, presenting with Hirschsprung disease (CCHS + HSCR) alone and/or together with neuroblastoma (CCHS + HSCR + NB). CCHS-associated missense mutations occur in the PHOX2B homeodomain, where impaired transcriptional activity has been suggested as their functional effect. However, the molecular p

Severe Osteoporosis in Larsen Syndrome-A Case Report of Bone Morphology and A Novel Filamin B (FLNB) Variant.

Larsen syndrome is a rare genetic condition characterized by facial dysmorphism and skeletal deformities. It is caused by heterozygous pathogenic variants in the Filamin B encoding gene (FLNB). FLNB is a cytoskeletal protein that plays a key role in bone morphogenesis; however, the skeletal phenotype of Larsen syndrome has not been described in detail. Here, we studied the skeletal presentation in two subjects with Larsen syndrome. A case-study including a 63-year-old women and her 33-year-old daughter with Larsen syndrome, both carrying a novel FLNB c.688G > T, p.(Val230Phe) variant. The bone

Distinct trajectories of humoral immune responses after SARS-CoV-2 mRNA vaccination in autoimmune inflammatory rheumatic diseases: A group-based trajectory analysis.

BACKGROUND: Patients with autoimmune inflammatory rheumatic diseases (AIRDs) are at a high risk of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Data on the effects of multiple (>3) vaccine doses and their influencing factors are limited. Understanding antibody dynamics after repeated vaccinations is essential for optimising vaccination strategies. Therefore, this study characterised the antibody response trajectories against SARS-CoV-2 and explored key clinical and immunological determinants. METHODS: This single-centre retrospective cohort study included patients w

Natural History of Metameric Spinal Cord Arteriovenous Malformations.

BACKGROUND: Spinal arteriovenous metameric syndrome (SAMS) is a rare form of spinal cord arteriovenous malformations with a metameric distribution affecting the spinal cord and related structures derived from the same embryological segment. Its natural history and impact on clinical outcomes, compared with nonmetameric spinal cord arteriovenous malformations, remain unclear. METHODS: This retrospective, single-center study included 253 patients with intradural spinal cord arteriovenous malformations between 2002 and 2024, that have either not been considered for embolization or followed up du

Disruption of SPECC1L translation initiation by intragenic deletion: novel pathogenic mechanism in Teebi-hypertelorism syndrome.

Missense and loss-of-function variants in SPECC1L are associated with Teebi Hypertelorism Syndrome 1 (TBHS1). Here, we report a novel SPECC1L intragenic deletion encompassing exon 3, which contains the canonical translation start site, in two related individuals with craniofacial features consistent with TBHS1. We use functional overexpression assays to demonstrate that this deletion leads to alternative start codon usage and protein truncation. This is the first report of a SPECC1L intragenic deletion associated with TBHS1. Our findings suggest that deletion of the canonical N-terminal disord

Rapid-onset obesity, hypothalamic dysfunction, hypoventilation, and autonomic dysregulation as a paraneoplastic syndrome in neuroblastoma: case report.

BACKGROUND: Neuroblastoma, the most common extracranial solid tumour in childhood, is occasionally associated with paraneoplastic syndromes. These syndromes can occur before tumour diagnosis or recurrence. Notably, approximately 40% of patients with rapid-onset obesity, hypothalamic dysfunction, hypoventilation, and autonomic dysregulation (ROHHAD) have underlying neural crest tumours, such as ganglioneuromas or ganglioneuroblastomas. ROHHAD may also manifest as a paraneoplastic syndrome in neuroblastomas. Here, we describe three neuroblastoma cases with ROHHAD to help improve oncologists' rec

ROGDI-Related Disorder Resulting from Disruption of Complex Interactive Neuro-Dental Developmental Networks: A Review and Description of the First Missense Variant.

ROGDI-related neurodevelopmental and dental disorder (ROGDI-RD), also known as Kohlschütter-Tönz syndrome (KTZS, MIM #226750), is a rare condition characterized by developmental abnormalities affecting both the central nervous system (CNS) and the dentition. These phenotypes highlight the role of complex gene-environment interactions and developmental networks shared by the nervous and stomatognathic systems, both of which originate mostly from neural crest-derived cells. In this review, we analyze clinical and genetic data from 54 previously reported ROGDI-RD patients to better define the phe

CHD7 regulates cardiac neural crest cell differentiation through SOX5-mediated self-activation.

CHARGE (coloboma of the eye, heart defects, atresia of the choanae, retardation of growth and development, genital abnormalities, and ear anomalies) syndrome, primarily caused by CHD7 haploinsufficiency, is a complex developmental disorder frequently associated with congenital heart defects. Our comprehensive single-cell RNA sequencing (scRNA-seq) analysis of cardiac neural crest cells (cNCCs) with Chd7 inactivation reveals impaired myocyte differentiation by cNCCs as a major cellular defect; loss of Chd7 disrupts myogenic transcriptional programs, alters cell fate trajectories, and activates

BRD9 depletion-mediated ALOX5 upregulation via chromatin dysregulation induces ferroptosis in SF3B1-mutant hematopoiesis.

SF3B1 mutations are among the most common splicing factor mutations in myeloid malignancies, yet the mechanisms linking aberrant splicing to metabolic phenotypes remain incompletely understood. We previously demonstrated that SF3B1 mutations cause nonsense-mediated decay of BRD9, a core component of the non-canonical BAF chromatin remodeling complex. Here, we investigated how the SF3B1-BRD9 pathway contributes to metabolic reprogramming in hematopoietic cells. Using BRD9-depleted murine models and analyses of SF3B1-mutated samples, we found that BRD9 depletion markedly upregulates ALOX5, which
