Sanguinarine ameliorates rheumatoid arthritis by activating the Hippo-YAP/TAZ pathway to induce apoptosis and suppress inflammation in fibroblast-like synoviocytes.

INTRODUCTION: Rheumatoid arthritis (RA) is a chronic autoimmune disease with progressive joint damage, functional disability and systemic comorbidity. Available therapies are not satisfactory, including low rates of remission and about 30% of patients with poor response to first-line treatment. Thus, more effective therapeutic strategies are urgently needed. Sanguinarine, a benzophenanthridine alkaloid primarily isolated from Sanguinaria canadensis and other members of the Papaveraceae family, has been documented to possess notable anti-bacterial, anti-inflammatory, and anti-neoplastic propert

HDAC4 Promotes Cancer Stemness in Nonsmall Cell Lung Cancer by Upregulating SLC38A2 Expression via HIF-1α to Enhance Glutamine Metabolism.

BACKGROUND AND PURPOSE: The malignant progression of nonsmall cell lung cancer (NSCLC) is closely related to cancer stemness. Histone deacetylase 4 (HDAC4) plays a regulatory role in lung cancer, but its effect on NSCLC stemness remains unclear. This study aimed to investigate the role and mechanism of HDAC4 in NSCLC stemness. METHODS: In this study, a tumor-bearing model was established by subcutaneously injecting A549 cells into the right dorsal side of nude mice. Reverse transcription quantitative polymerase chain reaction (RT-qPCR), Western blotting, and immunohistochemistry were used to

Analysis of Alternative Splicing Events in Melanoma and the Implications for Prognosis.

INTRODUCTION: Alternative splicing (AS) events significantly affect melanoma progression. Therefore, understanding their effect on prognosis is important for developing new treatments. METHODS: Univariate Cox regression analysis and LASSO regression were carried out to identify key AS events, build an AS risk model, and classify sample risk levels. Pearson correlation analysis was also performed to analyze the relationship between AS events and RNA-binding protein (RBP) genes or indicators of immune infiltration. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were

Epitranscriptomic silencing of STK25 promotes ferroptosis evasion via the METTL3-YAP-GPX4 axis in osteosarcoma.

Osteosarcoma (OS) is an aggressive bone malignancy with poor outcomes in recurrent and metastatic disease. Although ferroptosis represents as a promising therapeutic strategy, its regulation in OS remains incompletely understood. Here, we identified STK25 as a regulator of ferroptosis susceptibility and tumor growth in OS. Transcriptomic analysis of paired parental and multidrug-resistant OS cell lines identified lower STK25 expression in the resistant derivatives. In a retrospective tissue microarray, low tumor STK25 expression was associated with a lower histological tumor necrosis rate afte

Shaping a pro-carcinogenic hepatic microenvironment by TCDD: An integrated approach combining network toxicology, machine learning, molecular docking, molecular dynamics and experimental validation.

The increasing prevalence of environmental contaminants has raised concerns regarding their potential contribution to hepatic dysfunction and associated diseases. 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), classified as a Group 1 carcinogen and the most toxic congener of dioxins, has been implicated in adverse hepatic outcomes. However, the molecular mechanisms by which TCDD-driven signaling cascades establish a pro-carcinogenic microenvironment in the liver remain insufficiently elucidated. By integrating network toxicology with machine learning, CYP1A2, CYP2C9, and HSP90AB1 were identified

Non-invasive screening in hereditary cancer: a randomized controlled trial to test cell-free DNA-based early detection in the CHARM consortium.

Individuals with hereditary cancer syndromes are born with germline genetic variants that significantly increase their lifetime risk of developing multiple cancers. Cancer rates and overall mortality can be reduced with intensive surveillance to facilitate early cancer detection. However, participating in diagnostic imaging and endoscopy surveillance programs is often time-consuming, overwhelming, inconvenient, and anxiety-inducing. To improve this, multi-cancer early detection tests are being developed using cell-free DNA (cfDNA) sequencing analysis to detect cancers with more sensitivity tha

SLC46A1 deficiency-mediated folate restriction suppresses colorectal cancer progression through epigenetic-transcriptional reprogramming.

The association between folate metabolism abnormalities and the development of colorectal cancer (CRC) remains controversial. Here, we report that the folate exerts a tumor-suppressive role in CRC; however, the manifestation of this effect is restricted by the expression level of folate transporter SLC46A1 in CRC cells. Multi-cohort profiling revealed significant downregulation of SLC46A1 in CRC tissues compared to adjacent normal tissues, where low expression independently predicted poor overall survival. Functional studies demonstrated that SLC46A1-mediated folate uptake suppressed tumor pro

Transmembrane proteins in cancer: insights from mechanism to clinical impact.

Transmembrane proteins (TMEMs) constitute a large family of proteins that span biological membranes and are distributed across various cellular organelles, playing key roles in maintaining cellular homeostasis. Increasing evidence has revealed that dysregulation of TMEMs is closely associated with cancer development and progression. Therefore, a deeper understanding of the relationship between TMEMs and cancer is essential. Different TMEMs can function either as oncogenes or tumour suppressors, depending on the context. In this review, we explore the involvement of TMEMs in cancer, categorizin

Overexpression, clinical significance and potential mechanisms of protein kinase D1 in hepatocellular carcinoma: multi-omic analyses and pharmacological insights.

Protein kinase D1 (PRKD1), a serine/threonine kinase of the PKD family, has been implicated in tumor biology, but its role in hepatocellular carcinoma (HCC) remains unclear. We explored PRKD1 expression and function using immunohistochemistry, bulk and single-cell RNA sequencing, and in vitro functional assays. PRKD1 protein levels were significantly elevated in 339 HCC tissues compared to corresponding adjacent non-tumorous samples (11.390 ± 1.560 vs. 6.277 ± 2.357, P < 0.0001). Multicenter bulk transcriptomic data confirmed consistent PRKD1 mRNA overexpression (SMD = 0.26, 95% CI = 0.14-0.39

A monocyte-derived blood transcriptomic signature reveals systemic immunosuppression in HCC and partial reversal following curative therapy.

BACKGROUND: Liver ablation or resection can cure early-stage hepatocellular carcinoma (HCC), yet late diagnosis and high relapse rates hinder long-term survival. We sought to delineate how tumor burden-and its removal-reshape the systemic immune transcriptome and extract blood-based signatures with diagnostic and prognostic potential. METHOD: Peripheral blood mononuclear cells (PBMCs) from six early-stage HCC patients were subjected to single-cell RNA sequencing (scRNA-seq) both prior to and 1-3 months following curative therapy, alongside six age-matched healthy controls. The data were integ

Lumbar Pseudogout Mimicking Pyogenic Spondylitis in an Older Patient: A Diagnostic Pitfall in Emergency Care.

Lumbar pseudogout, or calcium pyrophosphate dihydrate deposition disease (CPPD), is an uncommon manifestation of a crystal-induced arthropathy that typically affects peripheral joints. Spinal involvement, particularly in the lumbar region, is rare and may closely mimic infectious conditions such as pyogenic spondylitis. An 85-year-old man was hospitalized for pneumococcal pneumonia and subsequently developed severe low back pain on day 7 of admission. Although his pneumonia improved with antimicrobial therapy, inflammatory markers remained elevated, raising suspicion of pyogenic spondylitis. C

Molecular Insights Into Canine Hepatocellular Carcinoma: A Cross-Species Transcriptomic Comparison With Human HCC.

Canine hepatocellular carcinoma (HCC) requires further molecular characterization to identify diagnostic and therapeutic targets, and to establish whether dogs with this condition can model the human disease. Accordingly, we aimed to identify differentially expressed genes (DEGs) in canine HCC and evaluate cross-species transcriptomic dysregulation in canine and human HCC. Liver tissue samples from three dogs with HCC and three healthy dogs were subjected to next-generation sequencing, followed by RT-qPCR validation. Identified DEGs were then targeted in bioinformatics analyses (pathway enrich

Establishment of insulin resistance-related ten-gene signature in endometrial cancer and identification of ACTL8 as a prognostic and immunological biomarker.

BACKGROUND: Endometrial cancer (EC) is a common gynecological tumor. Insulin resistance (IR) increases the risk of EC. However, the common molecular basis between the two remains unclear. This study aims to screen the common differential expression genes (DEGs) between the two diseases and construct a prognostic risk model. METHODS: We obtained gene expression profiles and clinical information of patients with IR and EC from GEO and TCGA datasets. We performed differential analysis to discover the shared DEGs between IR and EC. Subsequently, the interactions among overlapping DEGs, along with

Integration of Dual Functions of Cytosensing and Highly Efficient Photothermal Therapy Using a Cascaded Ω-Shaped Fiber Optic Localized Surface Plasmon Resonance Sensor.

Elimination of circulating tumor cells (CTCs) is a promising way to inhibit tumor metastasis using antitumor devices. The current devices lack the functions for sensing the captured CTCs in situ, which causes potential toxicity and off-target effects in the therapy for CTCs. In this work, a novel cascaded Ω-shaped fiber optic functionalized with gold nanoparticles/gold nanobipyramids (AuNPs/AuNBPs) was prepared for the realization of cytosensing and photothermal therapy. The incorporation of a no-core fiber (NCF) segment in the Ω-shaped fiber optic enhanced the evanescent field via bending-ind

Role of MED15 in Enhancing EMT and Metastasis in Bladder Cancer Through YAP1 Stabilization.

Bladder cancer (BLCA) remains a highly prevalent and aggressive malignancy of the urinary tract, characterized by frequent recurrence and metastasis. Mediator complex subunit 15 (MED15) functions as a key transcriptional coactivator bridging transcription factors and RNA polymerase II, yet its role in BLCA progression has not been elucidated. Here, we identify MED15 as markedly upregulated in BLCA tissues. Gene set enrichment analysis (GSEA) revealed that MED15 expression correlates with epithelial-mesenchymal transition (EMT)-related pathways. Functionally, loss- and gain-of-function assays d

International Multidisciplinary Consensus Report on Definitions, Diagnostic Criteria, and Management of Fatty Pancreas: A Joint Statement Endorsed by EPC, APA, EASD, EASL, ESGAR, ESGE, ESP, ESPCG, ESPEN, ESPGHAN, IAP, JPS, KPBA, LAPSG, and UEG.

This international, multidisciplinary consensus report represents the first effort to systematically define and characterize fatty pancreas. A key outcome of this endeavor was the recommendation to adopt "fatty pancreas" as the standardized and inclusive term to describe all forms of fat accumulation in the pancreas. This terminological consensus provides a critical foundation for unified reporting and clinical communication. Another major contribution of the report is the consensus on diagnostic imaging findings, which was based on radiological and endoscopic modalities. The proposed criteria
