Molecular Insights Into Canine Hepatocellular Carcinoma: A Cross-Species Transcriptomic Comparison With Human HCC.

Canine hepatocellular carcinoma (HCC) requires further molecular characterization to identify diagnostic and therapeutic targets, and to establish whether dogs with this condition can model the human disease. Accordingly, we aimed to identify differentially expressed genes (DEGs) in canine HCC and evaluate cross-species transcriptomic dysregulation in canine and human HCC. Liver tissue samples from three dogs with HCC and three healthy dogs were subjected to next-generation sequencing, followed by RT-qPCR validation. Identified DEGs were then targeted in bioinformatics analyses (pathway enrich

Establishment of insulin resistance-related ten-gene signature in endometrial cancer and identification of ACTL8 as a prognostic and immunological biomarker.

BACKGROUND: Endometrial cancer (EC) is a common gynecological tumor. Insulin resistance (IR) increases the risk of EC. However, the common molecular basis between the two remains unclear. This study aims to screen the common differential expression genes (DEGs) between the two diseases and construct a prognostic risk model. METHODS: We obtained gene expression profiles and clinical information of patients with IR and EC from GEO and TCGA datasets. We performed differential analysis to discover the shared DEGs between IR and EC. Subsequently, the interactions among overlapping DEGs, along with

Integration of Dual Functions of Cytosensing and Highly Efficient Photothermal Therapy Using a Cascaded Ω-Shaped Fiber Optic Localized Surface Plasmon Resonance Sensor.

Elimination of circulating tumor cells (CTCs) is a promising way to inhibit tumor metastasis using antitumor devices. The current devices lack the functions for sensing the captured CTCs in situ, which causes potential toxicity and off-target effects in the therapy for CTCs. In this work, a novel cascaded Ω-shaped fiber optic functionalized with gold nanoparticles/gold nanobipyramids (AuNPs/AuNBPs) was prepared for the realization of cytosensing and photothermal therapy. The incorporation of a no-core fiber (NCF) segment in the Ω-shaped fiber optic enhanced the evanescent field via bending-ind

Role of MED15 in Enhancing EMT and Metastasis in Bladder Cancer Through YAP1 Stabilization.

Bladder cancer (BLCA) remains a highly prevalent and aggressive malignancy of the urinary tract, characterized by frequent recurrence and metastasis. Mediator complex subunit 15 (MED15) functions as a key transcriptional coactivator bridging transcription factors and RNA polymerase II, yet its role in BLCA progression has not been elucidated. Here, we identify MED15 as markedly upregulated in BLCA tissues. Gene set enrichment analysis (GSEA) revealed that MED15 expression correlates with epithelial-mesenchymal transition (EMT)-related pathways. Functionally, loss- and gain-of-function assays d

International Multidisciplinary Consensus Report on Definitions, Diagnostic Criteria, and Management of Fatty Pancreas: A Joint Statement Endorsed by EPC, APA, EASD, EASL, ESGAR, ESGE, ESP, ESPCG, ESPEN, ESPGHAN, IAP, JPS, KPBA, LAPSG, and UEG.

This international, multidisciplinary consensus report represents the first effort to systematically define and characterize fatty pancreas. A key outcome of this endeavor was the recommendation to adopt "fatty pancreas" as the standardized and inclusive term to describe all forms of fat accumulation in the pancreas. This terminological consensus provides a critical foundation for unified reporting and clinical communication. Another major contribution of the report is the consensus on diagnostic imaging findings, which was based on radiological and endoscopic modalities. The proposed criteria

Development and validation of a succinylation-related prognostic model for esophageal squamous cell carcinoma based on multi-omics bioinformatics analysis.

BACKGROUND: Esophageal squamous cell carcinoma (ESCC), an aggressive malignancy with poor prognosis, requires reliable prognostic biomarkers. Protein succinylation, a critical post-translational modification implicated in cancer biology, has not yet been systematically investigated for its prognostic significance in ESCC. METHODS: Transcriptomic data from GSE53624 (n = 119) and The Cancer Genome Atlas (TCGA)-ESCC (n = 95) cohorts were analyzed in this study. Succinylation-related genes (SRGs) were identified via weighted gene co-expression network analysis (WGCNA), a curated SRG set, and diffe

NAD(P)H: quinone oxidoreductase 1-mediated oxidative stress resistance and tumor microenvironment remodeling in glioblastoma.

BACKGROUND: Glioblastoma (GBM) is a highly aggressive brain tumor, with glioblastoma stem cells (GSCs) occupying the pinnacle of a complex tumor microenvironment (TME), conferring therapeutic resistance. The TME plays a role in tumor development by creating a niche rich in reactive oxygen species (ROS) through oxidative stress (OS). Here, we identified NAD(P)H: quinone oxidoreductase 1 (NQO1) as an essential regulatory factor in antioxidant stress response, which is key to maintaining GSCs and the immunosuppressive TME. METHODS: Proteomics analysis and epigenetic profiling using H3K27ac ChIP-

Cognitive changes associated with chemotherapy in breast cancer: an assessment of social cognition and executive functions in Peruvian patients.

BACKGROUND: Cognitive impairment related to chemotherapy-commonly referred to as "chemo brain"-is a well-documented phenomenon among breast cancer patients. These impairments affect memory, attention, executive function, and social cognition, yet remain understudied in low- and middle-income countries. In Peru, where populations present a high proportion of Amerindian ancestry and distinct sociocultural factors, evidence is scarce. METHODS: We conducted a longitudinal study of 143 Peruvian women aged 28-64 years, newly diagnosed with early-stage breast cancer and naïve to chemotherapy, treate

MCM10, a novel YAP1/TEAD4 target, drives gastric cancer progression by bridging DNA replication to stemness acquisition.

OBJECTIVES: Gastric cancer (GC) remains a major global health challenge, with chemotherapy resistance significantly hindering treatment efficacy. A significant proportion of chemotherapeutics impact DNA replication, yet the mechanisms by which tumors evade this lethality remain incompletely understood. Notably, minichromosome maintenance 10 replication initiation factor (MCM10) is pivotal in initiating DNA replication, holding promise in mediating acquired chemotherapy resistance. This work aims to elucidate the driving roles of MCM10 GC pathogenesis and chemotherapeutic resistance. METHODS: T

Oncogenic role of PHF23 in gastric cancer progression.

Gastric cancer (GC) poses a persistent global health burden, largely attributed to its asymptomatic early stages and limited efficacy of existing therapies. PHF23, which has been primarily studied in hematologic malignancies and degenerative joint diseases, has not been explored in gastric cancer. In this study, PHF23 expression was assessed in GC tissues through immunohistochemistry and public dataset analysis. Functional assays, including proliferation, migration, invasion, and apoptosis, were performed following PHF23 knockdown in GC cell lines. Tumorigenic effects were evaluated in nude mo

Ferroptosis as a therapeutic nexus: traditional Chinese medicine interventions in rheumatoid arthritis.

Ferroptosis is an iron-dependent, lipid peroxidation-driven form of programmed cell death that plays an important role in neurodegenerative, neoplastic, and autoimmune diseases. Recent studies in Rheumatoid arthritis (RA) have shown that iron metabolism disorders caused by iron overload and impaired transferrin function lead to the production of reactive oxygen species; ferroptosis-associated pathways, such as dysregulation of the System Xc-/GPX4 axis dysregulation, NCOA4-mediated ferritin autophagy, endoplasmic reticulum stress and ferroptosis pathway crosstalk; as well as ferroptosis plays a

The multifaceted functions of selective autophagy in cancer: molecular basis, consequences, and clinical prospects.

Selective autophagy is a critical cellular process in eukaryotic cells, characterized by the targeted degradation of specific organelles and proteins. This intricate mechanism plays a vital role in maintaining cellular homeostasis and has significant implications in the pathogenesis and progression of various diseases, notably cancer. Recent studies have highlighted the dual role of selective autophagy in cancer, acting both as a suppressor in the early stages and potentially facilitating progression in later stages. The complexity of selective autophagy, particularly in the context of oncogen

Targeting MUC16 suppresses malignant progression and chemoresistance in large-duct type intrahepatic cholangiocarcinoma.

Intrahepatic cholangiocarcinoma (iCCA), a highly lethal subtype of liver cancer with increasing incidence and poor prognosis, displays profound molecular heterogeneity that limits therapeutic efficacy. Combining multi-omics with pathological subtyping in iCCA, we identified a strong association between the most aggressive molecular subtype and the large-duct pathological type, characterized by aberrant mucin overexpression and prominent myeloid cell infiltration. Through integrative analysis, MUC16 was identified as a key molecular marker specifically enriched in large-duct type intrahepatic c

ANXA5: A Multi-Omics-Derived, UPR-Associated Dual-Function Biomarker for Prognosis and Immunotherapy Response Prediction in Glioma.

Glioma, a highly aggressive brain cancer, thrives in an immunosuppressive tumor microenvironment (TME). The Unfolded Protein Response (UPR) is a key adaptive stress pathway implicated in therapy resistance. To discover clinically actionable biomarkers, we employed an integrated multi-omics strategy, combining single-cell and bulk transcriptomic data from multiple glioma cohorts. Using hdWGCNA network analysis, we identified a UPR-associated gene module that defined two molecular subtypes with stark survival differences. Through a rigorous machine learning pipeline (Random Survival Forest, LASS

Phase separation of β-catenin assembles active loop hubs in colorectal cancer.

BACKGROUND: Aberrant three-dimensional genome organization is a hallmark of cancer. However, the underlying regulatory mechanisms remain elusive. RESULTS: Here, we identify β-catenin associated loop hubs in colorectal cancer HCT116 cells using HiChIP profiling. These hubs exhibit an active transcriptional environment marked by elevated gene expression and H3K4me3 enrichment. Disruption of loop hub formation by artificial DNA methylation suppresses hub gene expression and inhibits colorectal cancer growth, indicating its functional importance in cancer. Mechanistically, β-catenin forms phase-s
