MBD2 suppresses SFRP1 expression and promotes colorectal cancer development by blocking MED19 binding to its methylated promoter.

BACKGROUND: The Wnt signaling pathway antagonist SFRP1 is frequently silenced by promoter DNA hypermethylation in colorectal cancer (CRC). MBD2, a DNA methylation reader, is known to contribute to SFRP1 epigenetic silencing. Previous work showed that MBD2 critically suppresses SFRP1 expression without altering promoter methylation, though the underlying mechanism remained unclear. Elucidating how DNA methylation silences tumor suppressor genes, such as SFRP1, could reveal novel therapeutic targets with significant clinical potential. METHODS: MBD2 was inhibited in CRC models using either siRN

Targeting RBPMS selectively eliminates FOXO1-mediated stem cell signatures in mouse models of acute myeloid leukemia.

Leukemia is a malignant tumor with a high recurrence rate and poor prognosis for patients. Thus, there is an urgent need to explore new therapeutic targets that play critical roles in leukemogenesis but have little effect on normal hematopoietic cells. Here, we show that RNA binding protein with multiple splicing (RBPMS), which is highly expressed in acute myeloid leukemia (AML) and associated with poor prognosis of AML, plays critical roles in leukemogenesis. Our study shows that inhibition of RBPMS inhibits self-renewal of leukemia-initiating cells (LICs) and leukemia development but has lit

Programmable Circular Single-Stranded DNA Acts as Recyclable Anti-miRNA Nucleotides to Inhibit Colorectal Cancer.

MicroRNA (miRNA) therapy represents an attractive approach for the treatment of colorectal cancer (CRC). However, the efficacy of therapy targeting a single oncomiRNA (oncomiR) is limited, as multiple oncomiRs often act simultaneously in promoting CRC development. In addition, current anti-miRNA nucleotide (AMiN) design faces challenges, including high synthetic error rate and difficulty in synthesizing long oligonucleotides. Circular single-stranded DNA (CssDNA), an ancient form of DNA that has gained increasing attention recently, is known for its easy synthesis, high stability, low error ra

Human mesenchymal stem cell-derived miR-3614-5p exosomes suppress progression of colorectal cancer by targeting IL7Rα.

BACKGROUND: Colorectal cancer (CRC) represents a growing global health burden, particularly in developing countries. Mesenchymal stem cells (MSCs) have been reported to suppress tumor progression partly through the release of exosomes that deliver therapeutic molecules. However, the therapeutic potential of engineered MSC-derived exosomal microRNAs (Exo-miRNAs) in CRC remains largely unexplored. METHODS: Plasma exosome miRNA sequencing was performed to identify dysregulated miRNAs in CRC. Gain- and loss-of-function assays were conducted to evaluate the biological effects of candidate miRNAs i

JAK-STAT signaling pathway in cancer: from molecular mechanisms to clinical intervention.

The Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway is a conserved signaling network that mediates communication between extracellular cytokine stimulation and nuclear transcriptional responses. Beyond its established role in regulating cell proliferation, differentiation, immune responses, and tissue homeostasis, dysregulated JAK/STAT signaling contributes to tumor progression, immune evasion, therapeutic resistance, and remodeling of the tumor microenvironment (TME). Although canonical ligand-dependent JAK/STAT activation has been extensively charac

A Non-Canonical Role of SMAD4 in Regulating 3D Genome Architecture to Inhibit Lung Squamous Cell Carcinoma Development.

Lung squamous cell carcinoma (LUSC) lacks clearly defined key drivers and effective targeted therapies, reflecting an incomplete understanding of its molecular pathogenesis. Here, we identify SMAD4 as a critical regulator of three-dimensional (3D) genome organization in LUSC and uncover a mechanistic link between tumor suppressor loss and oncogenic transcriptional activation. By integrating clinical datasets, genetically engineered mouse models, human and murine LUSC cell lines, and multi-omics analyses, we demonstrate that SMAD4 deficiency promotes LUSC progression by unleashing EP300-mediate

ABCC2 as a novel therapeutic target in lung adenocarcinoma: a machine learning-driven discovery linking ammonia metabolism to prognosis and drug resistance.

BACKGROUND: Ammonia, long regarded as a metabolic waste product, has recently been recognized as a pivotal oncometabolite in the tumor microenvironment, contributing to cancer progression and immune evasion. However, its prognostic value and therapeutic relevance in lung adenocarcinoma (LUAD) remain insufficiently characterized. METHODS: Transcriptomic data from multiple LUAD cohorts were obtained from public databases. Ten machine learning algorithms were integrated into 101 combinations to construct predictive models and identify key genes associated with prognosis. The tumor immune microen

A locoregional program of photothermal debulking, stemness suppression, and dendritic cell activation for durable glioblastoma immunotherapy.

Glioblastoma (GBM), especially in large or unresectable tumors, remains difficult to treat because of the lack of effective debulking, the persistence of therapy-resistant glioma stem cells (GSCs), and a profoundly immunosuppressive tumor microenvironment. Here, a locoregional hydrogel platform is developed to integrate photothermal debulking, post-ablation GSC suppression, and dendritic-cell-mediated immune activation for GBM therapy. The brain-compliant injectable hydrogel (MIN-PPIC@iGel) co-encapsulates indocyanine green/napabucasin-loaded micelles and poly(I:C)-loaded polymersomes, enablin

Unveiling VARS1: a key driver of colorectal cancer progression and immune modulation.

BACKGROUND: Colorectal cancer (CRC) is a leading cause of cancer-related mortality worldwide. Valyl-tRNA synthetase 1 (VARS1), an aminoacyl-tRNA synthetase, has been implicated in various cancers, but its role in CRC remains unclear. This study investigated the expression pattern, clinical significance, biological functions, and potential molecular associations of VARS1 in CRC. METHODS: VARS1 expression was analyzed in CRC tissues using TCGA/GEO databases, immunohistochemistry, and Western blotting. Prognostic models were built via Cox regression and nomograms. In vitro, VARS1 was knocked dow

PYCR1 promotes glutamine metabolism and the progression of lung adenocarcinoma by regulating the expression of OPLAH.

Lung adenocarcinoma (LUAD) is the most common subtype of lung cancer. Glutamine plays a critical role in the progression of LUAD. However, the function of pyrroline-5-carboxylate reductase 1 (PYCR1) and its regulatory role in glutamine metabolism remain unclear. Transcriptomic and clinical data for LUAD were obtained from The Cancer Genome Atlas (TCGA) and validated using Gene Expression Omnibus (GEO) datasets (GSE19188, GSE13213). Glutamine metabolism-related genes were analyzed for differential expression and prognostic significance. Functional enrichment was performed via gene ontology (GO)

TFPI2 promotes NK cell-mediated glioblastoma killing through adhesion and checkpoint control.

Immune-mediated killing triggers dynamic transcriptional adaptations in tumor cells that can reciprocally regulate the cytolytic process. Unraveling such feedback mechanisms is crucial for advancing cancer immunotherapy. Here, we identified tissue factor pathway inhibitor 2 (TFPI2) as a central node in natural killer (NK)-glioblastoma cross talk. Using transcriptomic and functional approaches, we demonstrated that NK cell attack induces TFPI2 expression in glioblastoma cells via IL1β- and TNFα-driven activation of NFκB signaling. TFPI2 not only restrains tumor proliferation by suppressing the

RUNX1-regulated ITGB1-enriched extracellular vesicles drive pancreatic cancer liver metastasis via fibrotic pre-metastatic niche formation.

BACKGROUND: Liver metastasis (LM) is the predominant distant metastatic target of pancreatic ductal adenocarcinoma (PDAC) and a major contributor to poor patient outcomes, with pre-metastatic niche (PMN) formation as its critical prerequisite. However, its mechanisms remain incompletely elucidated. METHODS: RNA sequencing was performed on primary tumor tissues and paired adjacent non-tumor tissues from PDAC patients, combined with the analysis of a GEO dataset related to PDAC liver metastasis (PDAC-LM), followed by validation using in vitro assays and mouse orthotopic liver metastasis models.

piR-26681 suppresses ovarian cancer progression by enhancing METTL3/METTL14-mediated m6A modification of FBXO16 and impairing DNA repair via MORF4L1 degradation.

BACKGROUND: Survival rates for ovarian cancer drop sharply at late-stages due to late diagnosis. Although PARP inhibitors are effective in homologous recombination-deficient (HRD) tumors, their efficacy in homologous recombination (HR)-proficient ovarian cancer remains limited, highlighting the need for novel molecular targets to inhibit tumor progression and improve patient outcomes. METHODS: Differentially expressed piRNAs were screened using ovarian cancer tissues from early- and advanced-stage patients. Functional studies were performed in ovarian cancer cell lines, xenograft models, and

Comprehensive multi-omics analysis reveals a fatty acid metabolism gene signature for prognostic assessment and immunotherapy in nasopharyngeal carcinoma, and identifies ABCC1 as a potential novel therapeutic target.

BACKGROUND: Nasopharyngeal carcinoma (NPC) is a subtype of head and neck squamous cell carcinoma characterized by high recurrence and metastasis rates and poor prognosis. Although immune checkpoint inhibitors have emerged as a promising treatment strategy for recurrent/metastatic nasopharyngeal carcinoma (R/M NPC), only a few patients have benefitted significantly from them. Lipid metabolism reprogramming plays a crucial role in NPC progression and its interaction with the immune microenvironment. This study aims to establish a prognostic model for NPC based on lipid metabolism-related factors

PMVK drives hepatocarcinogenesis via SP1 phosphorylation-mediated lipid metabolic reprogramming.

Metabolic reprogramming is a hallmark of cancer, yet the critical drivers and mechanisms in hepatocellular carcinoma (HCC) remain incompletely understood. Through a metabolism-focused CRISPR screen, we identified phosphomevalonate kinase (PMVK), a mevalonate pathway enzyme, as a key regulator of HCC stemness and progression. PMVK directly phosphorylates the transcription factor SP1 at Thr355, which enhances SP1's DNA binding affinity and promotes its interaction with the master lipid regulator SREBP1/2, driving a transcriptional program essential for de novo cholesterol and fatty acid synthesi

The role of the WD40-repeat protein family in cancer.

WD40-repeat (WDR) proteins constitute one of the largest and most functionally diverse scaffold families in eukaryotes. By folding tandem WD repeats into β-propeller domains, they provide modular interaction surfaces that assemble multi-protein complexes governing transcription and epigenetic control, ubiquitin-dependent proteostasis, RNA metabolism, and cell-cycle progression. Despite their pervasive involvement in oncogenic signaling and hallmark cancer phenotypes, the cancer field still lacks an integrated framework that connects WDR structural logic to context-dependent mechanisms and, cri
