Optimized Expression and Purification of Outer Membrane Protein 25 (Omp25) of Brucella melitensis.

Our previous immunoproteomic studies on patients with acute brucellosis identified outer membrane protein 25 (Omp25) of Brucella melitensis as one of the immunogenic antigens, indicating its potential to be used as a diagnostic marker and a tool to differentiate acute and chronic human brucellosis. Expression and purification of Omp25 are technically challenging using conventional approaches owing to its membrane association. Existing expression and extraction protocols frequently lead to host cell toxicity, low protein yields, and poor reproducibility. To address these limitations, we develop

The evolution of Brucella as a model intracellular pathogen.

Brucella species are intracellular bacterial pathogens that cause brucellosis, one of the most common zoonoses worldwide. This review traces how Brucella, first discovered in the 19th century through investigations of human and bovine disease, became a modern experimental model system. Work on Brucella has illuminated how intracellular bacteria evade and adapt to host defenses and how they remodel host cell biology to establish a replicative niche. It has also yielded insights that extend beyond pathogenesis into bacterial cell envelope architecture and development. Though long regarded as a s

Evaluation and Outcomes of Obstetric and Surgical Healthcare-associated Brucella Exposures.

BACKGROUND: Exposure to Brucella spp. in healthcare settings is rarely reported, and optimal exposure assessment and management remain poorly defined. We describe Brucella melitensis exposure in obstetric/surgical and laboratory settings and our exposure management approach. METHODS: Seventy-four potentially exposed employees were identified; exposure risk was assessed using four categories adapted from the CDC Brucellosis Reference Guide for laboratory exposures. RESULTS: Twenty (27%) were minimal risk requiring no intervention, 17 (23%) minimal risk requiring 24-week symptom monitoring, 2

Serologically supported pediatric brucellosis presenting with marked conjugated hyperbilirubinemia and marked systemic inflammation: A case report.

Brucellosis is a multisystem zoonosis that can mimic complicated bacterial infection in children. A 10-year-old girl presented with prolonged high-grade fever, jaundice, abdominal pain, anorexia and weight loss. The initial syndrome was managed empirically as an undifferentiated complicated bacterial illness because enteric fever and a hepatobiliary or intra-abdominal source were considered. CRP was 333.2 mg/L and total/direct bilirubin 7.24/5.21 mg/dL despite only mild aminotransferase elevation; ultrasonography showed hepatomegaly and nonspecific gallbladder-wall edema without biliary dilata

Colorimetric/chemiluminescent vertical flow immunoassay based on multifunctional cobalt-gold bimetallic nanoclusters for Brucella antibody detection and deep learning-assisted semiquantitative analysis.

A colorimetric/chemiluminescent vertical flow immunoassay (VFI) for Brucellosis was developed using glutathione-protected cobalt-gold nanoclusters conjugated with staphylococcal protein A (GSH@Co-AuNCs-SPA) as multifunctional catalytic immunoprobes. The nanocluster probe integrates antibody recognition with peroxidase-like catalytic activity, enabling three complementary signal readouts within a common immunorecognition architecture: direct colorimetric visualization, TMB-H₂O₂-amplified colorimetry, and luminol-H₂O₂ chemiluminescence. The prepared GSH@Co-AuNCs exhibited H₂O₂-mediated catalytic

BR-bodies link RNA homeostasis to stress tolerance and intracellular fitness in Brucella.

Bacterial gene expression depends on the coordinated regulation of RNA synthesis, processing, and decay. The conserved RNA degradosome scaffold, ribonuclease E (RNase E), assembles into phase-separated bacterial ribonucleoprotein bodies (BR-bodies) through its C-terminal intrinsically disordered region (IDR). This IDR also scaffolds recruitment of degradosome client proteins that carry out post-transcriptional gene regulation. Whether BR-bodies regulate virulence programs to support infection, however, is largely undefined. We show that RNase E of the intracellular pathogen Brucella ovis forms

Metagenomic Next-Generation Sequencing in Brucellar Spondylitis with Discordant Serologic Findings in a Non-Endemic Region: A Case Report and Literature Review.

BACKGROUND: Brucellar spondylitis (BS) is a focal osteoarticular manifestation of brucellosis with nonspecific clinical and imaging features. Diagnosis may be difficult in areas where brucellosis is not traditionally considered endemic, particularly when epidemiologic exposure is not identified and serologic tests yield discordant results. We report a case in which metagenomic next-generation sequencing (mNGS) of vertebral lesion tissue provided additional microbiologic evidence for the diagnosis. CASE PRESENTATION: A 71-year-old man presented with a 2-month history of fatigue, anorexia, nigh

Liquid versus solid culture media for the microbiological diagnosis of eye infections: a retrospective comparative study.

BACKGROUND: Ocular infections are an important cause of preventable vision loss, yet culture yield is limited by small specimen volumes and low bacterial loads. Comparisons of liquid and solid media in ocular infection are scarce. We evaluated four bedside-inoculated media: two liquid (Brucella broth, thioglycolate broth) and two solid (chocolate agar, 5% sheep blood agar). METHODS: We retrospectively analysed 102 ocular specimens collected between January 2023 and August 2025, each inoculated simultaneously onto all four media at the bedside. Because solid media were incubated for 7 days and

Brucellosis in Southeast Asia: A Systematic Review of Epidemiological Evidence, Surveillance Gaps, and cgMLST-Based Insights Into Genetic Relatedness.

Brucellosis is an important zoonosis affecting public health and livestock production in Southeast Asia, but its regional epidemiology remains incompletely characterized. In this study, PubMed, Embase, Web of Science, and Scopus were searched for primary studies from the 11 Southeast Asian countries that reported serological findings, molecular detection, bacterial isolation, or Brucella species identification in human or animal populations. Publicly available PubMLST records were also analyzed using the 1764-locus Brucella core-genome multilocus sequence typing scheme. Across the eligible stu

Brucella prosthetic joint infection following total knee arthroplasty: a rare case report.

BACKGROUND: Brucellosis is a zoonotic infection with a high prevalence in endemic regions and a strong predilection for osteoarticular involvement. Brucella prosthetic joint infection (PJI) is an exceptionally rare but challenging complication of total knee arthroplasty (TKA), often characterized by delayed presentation, indolent symptoms, and diagnostic difficulty. There is no consensus regarding optimal management, particularly concerning surgical strategy and duration of antimicrobial therapy. METHODS: A narrative literature review was conducted using PubMed and Scopus, searched on 1 March

Culture-confirmed Brucellosis Presenting with Acute Kidney Injury and Multi-organ Dysfunction.

Brucellosis is a widespread zoonosis caused by Gram-negative Brucella coccobacilli. Renal involvement, particularly acute kidney injury (AKI), is uncommon. A 47-year-old construction worker with livestock exposure and raw milk consumption presented with intermittent fever, pedal edema, hematuria, and AKI. He subsequently developed presumed Brucella endocarditis, rapidly progressive glomerulonephritis, and multiorgan dysfunction. Brucella melitensis (B. melitensis), bacteremia was confirmed by prolonged blood culture and PCR, supported by positive Brucella serology. Despite targeted antimicrobi

Scrub Typhus Meningoencephalitis With Concurrent Brucellosis in Rural Nepal: Diagnostic Challenges With Negative PCR in a Low-Resource Setting.

Scrub typhus is an important cause of acute febrile illness in Nepal and can rarely present with meningoencephalitis and multiorgan involvement. We report a 32-year-old female farmer from rural Nepal who presented during the monsoon season with high-grade fever, myalgia, vomiting, and an eschar following an unidentified forest bite. She developed meningismus, hypotension, mild acute respiratory distress syndrome, and transaminitis. Cerebrospinal fluid showed lymphocytic pleocytosis and elevated protein, while bacterial culture, acid-fast staining, and GeneXpert were negative. Serum scrub typhu

Consumption of Unpasteurized (Raw) Milk and Milk Products by Pregnant People and Children: Policy Statement.

Sales of unpasteurized (raw) milk and milk products remain legal in at least 27 US states. Unpasteurized milk and milk products from cows, goats, and sheep continue to be a source of infections attributable to several virulent pathogens, including Listeria monocytogenes, Campylobacter jejuni, Salmonella species, Brucella species, and Escherichia coli O157. These infections can occur in both healthy and immunocompromised individuals, including older adults, infants, young children, and pregnant people and their fetuses, potentially resulting in life-threatening illness and fetal miscarriage. Lumbar spinal brucellosis presenting as an atypical epidural abscess.

Brucellosis is a zoonotic infection that affects the spine, typically presenting as spondylodiscitis. Predominantly, epidural involvement without significant concurrent disc or vertebral involvement is rare and may closely mimic non-infectious pathologies. We report a man in his 50s presenting with low back pain and lower limb radiculopathy. Initial MRI demonstrated an epidural-predominant ventral epidural collection at L4-L5 associated with a posterior annular fissure, without significant vertebral or disc involvement, mimicking a sequestered disc fragment. Diffusion-weighted imaging showed p

Retrospective case-control study of haematological indices and inflammatory ratios in the diagnosis of paediatric brucellosis.

BACKGROUND: Brucellosis is a common zoonotic infection in Turkey and other Mediterranean countries. In children, symptoms are usually non-specific, including fever, fatigue, arthralgia, and myalgia, which may complicate diagnosis. Haematological indices and inflammatory ratios have recently been studied as possible supportive markers in infectious diseases, but findings in brucellosis remain inconsistent. This study investigated red cell distribution width (RDW), plateletcrit (PCT), platelet indices, neutrophil-to-lymphocyte ratio (NLR), and platelet-to-lymphocyte ratio (PLR) in paediatric bru

Re-emergence of Brucella species: epidemiology and pathogenesis.

SUMMARYBrucella spp. are highly adapted intracellular bacterial pathogens that remain a substantial global cause of zoonotic disease. The globally distributed species Brucella melitensis, Brucella abortus, Brucella suis, and Brucella canis account for the majority of human infections, reflecting their efficient transmission across animal hosts, and their exceptional capacity to survive and replicate within the intracellular niche. Although animal infections are often clinically silent, human brucellosis can exhibit persistent intracellular infection and relapsing fever. Growing genomic data hi
