Glacial forelands in Maritime Antarctica are rapidly expanding due to deglaciation, thereby increasing ecological connectivity among previously isolated microbial reservoirs. From a One Health perspective, this process is relevant because environmental microorganisms represent long-standing reservoirs of antibiotic resistance and virulence-associated traits. However, current knowledge is mainly limited to dominant, fast-growing taxa recovered by conventional cultivation, leaving the functional potential of slow-growing and rare Antarctic bacteria largely unexplored. Here, we compared tradition

Soil salinity limits rice growth through osmotic stress, ionic imbalance, and oxidative damage. In this study, halotolerant plant growth-promoting bacteria were isolated from Peganum harmala collected at the Khewra Salt Mine, Pakistan, and one of the best isolates, Brucella intermedia PM63, was evaluated for its ability to improve salt tolerance in rice. Ten bacterial isolates were screened for salt tolerance and plant growth-promoting (PGP) traits, and three selected strains were characterized by phenotypic tests, FTIR, SEM, 16S rRNA sequencing, and PCR detection of functional genes. PM63 tol

Agri-food systems are increasingly recognised as drivers of zoonotic disease emergence, yet the mechanisms by which modern food production, trade, and distribution reshape pathogen transmission remain poorly quantified. Here we integrate two decades of nationwide surveillance, epidemiological modelling, and phylogenomic analyses of 4215 Brucella genomes from 79 countries and regions to elucidate the drivers of brucellosis re-emergence in China. Nationwide, human cases increased by 282.6% between 2005 and 2023, far exceeding the 8.1% increase in sheep inventory. This decoupling indicates a fund

Brucellosis, a major global zoonosis causing over 2.1 million human infections annually, poses a significant threat to public health and livestock economies. Conventional vaccines face substantial limitations, including residual virulence and diagnostic interference in live attenuated vaccines, and poor immunogenicity in subunit vaccines. To address these issues, we developed BP26-OMP16Es, a self-assembling nanoparticle vaccine constructed by fusing immunodominant B- and T-cell epitopes from OMP16 to the self-assembling antigen BP26 via flexible linkers, yielding single-copy (1×) and double-co

Brucella ceti has a global distribution and is associated with neurologic disease, systemic infection, and reproductive losses in cetaceans. B. ceti has been associated with perinate mortality and sporadic systemic infections in bottlenose dolphins (BNDs) Tursiops truncatus from strandings both during and outside of unusual mortality events on the southern and eastern coasts of the USA. In 2014-2015, 2 BND from the waters of Texas and Florida stranded on separate occasions, were admitted to rehabilitation facilities, and were later placed under permanent professional care. Both dolphins had su

BACKGROUND: Brucellosis is one of the most widespread zoonotic diseases globally. Of the twelve known Brucella species that affect mammals, Brucella melitensis is the most significant cause of human infection. Others include B. abortus, B. suis, and B. canis, with transmission occurring via unpasteurized dairy, direct animal contact, or occupational exposure (veterinarians, farmers, and laboratory workers). Although hemophagocytic lymphohistiocytosis (HLH) and neurological complications are known sequelae of brucellosis, their simultaneous occurrence with multiorgan failure is rarely reported.

Brucella is a genus of zoonotic, intracellular alphaproteobacteria known to cause disease in marine mammals as well as livestock and companion animals. Specifically, Brucella ceti may be associated with severe neurological, respiratory, and reproductive disease in small cetaceans. Nematodes, particularly members of the Pseudaliidae, are common in small cetaceans and may harbor Brucella ceti, although the nature of this relationship remains unclear. This study investigated the prevalence of Brucella spp. in nematodes collected at necropsy from 64 small cetaceans that stranded on the central Cal

BACKGROUND: Brucellar arthritis is the most common complication of brucellosis, and its chronic progression is closely associated with immune cell dysfunction. Natural killer (NK) cells play a central role in anti-infective immunity; however, their functional status and regulatory mechanisms in brucellar arthritis remain poorly understood.
METHODS: Peripheral blood and synovial tissue samples were collected from healthy controls, as well as from patients with acute-phase and chronic-phase brucellar arthritis. Single-cell RNA sequencing (scRNA-seq), flow cytometry, quantitative real-time PCR (

WHAT IS ALREADY KNOWN ABOUT THIS TOPIC?: Brucellosis is a zoonotic disease historically concentrated in pastoral regions of China; however, an increasing number of cases have been reported in large urban centers in recent years.
WHAT IS ADDED BY THIS REPORT?: Analysis of 191 human-derived Brucella isolates collected in Shenzhen between 2005 and 2024 showed that Brucella melitensis predominated and that Shenzhen isolates were interspersed with strains from multiple provinces, suggesting repeated introductions from other regions. Most cases occurred in working-age adults, and both imported case

BACKGROUND: Brucellosis, caused by bacterial pathogens of the genus Brucella, is a chronic zoonotic disease affecting small ruminants such as sheep and goats. This study evaluated and compared the seroprevalence of Brucella antibodies among apparently healthy sheep and goats in Vwang District, Jos South Local Government Area, Plateau State, Nigeria.
METHODOLOGY: A cross-sectional study design was employed from May to June 2024. A total of 140 serum samples collected from goats (n = 77) and sheep (n = 63) using systematic random sampling across four administrative wards were screened for Bruce

The zinc import system ZnuABC is essential for the virulence of smooth (bearing O-polysaccharide chains) zoonotic Brucella species but remains uncharacterized in the non-zoonotic rough (lacking O-chains) species Brucella ovis, the causative agent of a type of ovine brucellosis of veterinary and economic importance. In this study, we constructed a panel of B. ovis mutants for genes of the znuACB locus that were characterized by evaluating growth, gene expression, protein abundance, metal content, and virulence in mice. We identified a ZigA ortholog (uncharacterized in the genus Brucella but lin

OBJECTIVE: The most common genitourinary complication of brucellosis is Brucella epididymo-orchitis (BEO), which, if not promptly diagnosed and treated, may lead to severe testicular complications. The present study aimed to evaluate the frequency, clinical characteristics, laboratory findings, and treatment outcomes of BEO in an endemic region.
METHODS: This retrospective cohort study included male patients diagnosed with brucellosis between August 2017 and October 2022 at a tertiary referral center. We identified patients with BEO based on clinical, laboratory, and ultrasonographic findings

Brucellosis remains an important zoonotic disease for livestock production and public health, creating a need for rapid molecular screening methods with limited equipment requirements. We developed a visual recombinase polymerase amplification (RPA)-CRISPR/Cas12a assay targeting a 642-bp region of the Brucella omp25 gene. Comparative sequence analysis was used to assess target conservation, after which four RPA primer pairs and two CRISPR RNAs (crRNAs) were experimentally screened. RPA3-crRNA1 produced the strongest fluorescence response and was used for assay optimization. The final Cas12a re

Brucella mastitis represents a rare infectious disease that is frequently misdiagnosed as metastatic breast carcinoma, particularly when older women present with systemic manifestations. Here, we report a 60-year-old woman presenting with a neck pain and left upper extremity numbness. Initial contrast-enhanced magnetic resonance imaging (CEMR) and 18F-FDG PET-CT highly suggested breast malignancy with osseous metastasis, while contrast-enhanced ultrasound (CEUS) provided critical microvascular perfusion insights favoring an inflammatory process. The diagnosis was ultimately confirmed via blood

Our previous immunoproteomic studies on patients with acute brucellosis identified outer membrane protein 25 (Omp25) of Brucella melitensis as one of the immunogenic antigens, indicating its potential to be used as a diagnostic marker and a tool to differentiate acute and chronic human brucellosis. Expression and purification of Omp25 are technically challenging using conventional approaches owing to its membrane association. Existing expression and extraction protocols frequently lead to host cell toxicity, low protein yields, and poor reproducibility. To address these limitations, we develop

Brucella species are intracellular bacterial pathogens that cause brucellosis, one of the most common zoonoses worldwide. This review traces how Brucella, first discovered in the 19th century through investigations of human and bovine disease, became a modern experimental model system. Work on Brucella has illuminated how intracellular bacteria evade and adapt to host defenses and how they remodel host cell biology to establish a replicative niche. It has also yielded insights that extend beyond pathogenesis into bacterial cell envelope architecture and development. Though long regarded as a s

BACKGROUND: Exposure to Brucella spp. in healthcare settings is rarely reported, and optimal exposure assessment and management remain poorly defined. We describe Brucella melitensis exposure in obstetric/surgical and laboratory settings and our exposure management approach.
METHODS: Seventy-four potentially exposed employees were identified; exposure risk was assessed using four categories adapted from the CDC Brucellosis Reference Guide for laboratory exposures.
RESULTS: Twenty (27%) were minimal risk requiring no intervention, 17 (23%) minimal risk requiring 24-week symptom monitoring, 2
