Integrated in vivo and in silico analysis of immune gene expression in cattle infected with Brucella abortus.

Brucella abortus is an intracellular pathogen that causes infection in cattle, leading to reproductive losses and posing a zoonotic risk to humans. Understanding host immune responses at the molecular level is essential for developing targeted diagnostics and control strategies. This study aimed to investigate the expression of immune-related genes in Bos taurus naturally infected with Brucella abortus using integrated in vivo and in silico approaches. The infection was characterized as chronic based on persistent seropositivity and historical herd records of infection for 12–18 months prior t

Genomic analysis of Brucella Ovis in Italy reveals high genetic diversity and a novel lineage.

BACKGROUND: Brucella ovis, the causative agent of ovine brucellosis, poses an underestimated threat to sheep farming worldwide due to its impact on reproductive health and flock productivity. Although B. ovis is non-zoonotic, the disease is globally distributed and often excluded from mandatory control programs, especially in Europe, resulting in scarce prevalence data and limited genomic information that hinder effective management strategies. This study investigated the global and regional phylogenetic structure of B. ovis using Multilocus Variable-Number Tandem Repeat (MLVA) analysis, core

BvrR From Brucella abortus Induces Neuroinflammation Through IRE1-Mediated Activation of ATF2 and NF-κB.

Brucella-induced neuroinflammation represents a key mechanism in the development of neurobrucellosis. The objective of this investigation was to clarify the molecular pathways through which the BvrR contributes to neuroinflammation and cognitive dysfunction. Human microglial clone 3 (HMC3) cells were transfected with pcDNA3.1-BvrR-His to examine the effects of BvrR from Brucella abortus on endoplasmic reticulum (ER) function and the activation of activating transcription factor 2 (ATF2) and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) p65. The role of phosphorylated i

WITHDRAWN: BvrR of Brucella initiates microglial inflammation through the activation of the IRE1 pathway.

Источник пометил публикацию как отозванную

Brucella-induced microglial inflammation plays a pivotal role in the neuropathology of neurobrucellosis. The virulence factor BvrR, critical for Brucella replication and survival within the host cell's endoplasmic reticulum (ER), has an undefined role in this inflammatory process. To elucidate this, HMC3 cells were transfected with pcDNA3.1-BvrR-His to investigate the impact of Brucella abortus BvrR on the ER and the activation of ATF2 and NF-κB p65 proteins. To support the hypothesis that BvrR mediates activation of the ATF2/IL-6 and NF-κB p65/TNF-α pathways via p-IRE1, the effects of the IRE

Identification and Characterization of Outer Membrane Proteins and Membrane Spanning Protein Complexes in Brucella melitensis.

Brucellosis (Malta fever) is a zoonotic disease that affects both humans and animals, including cattle, sheep, and goats. Brucella melitensis is the most virulent and clinically significant species in humans. It is a gram-negative bacterium with three groups of outer membrane proteins (OMPs): minor OMPs (Group 1) and major OMPs (Groups 2 and 3). OMPs with β-barrel architecture play important roles in nutrient transport, efflux, adhesion, and membrane biogenesis. Despite their importance, the structure, function, and interaction dynamics of several B. melitensis β-barrel OMPs and associated pro

Spatiotemporal dynamics of Brucella immune evasion across infection stages.

Brucella, a major zoonotic pathogen, poses a significant threat to global public health and causes substantial economic losses in the livestock industry. It employs diverse and sophisticated immune evasion strategies to circumvent host surveillance, establishing and maintaining chronic infections that are difficult to treat and prone to relapse. While previous reviews have catalogued individual virulence factors-such as the VirB type IV secretion system-and their actions on pathways like TLR4 signaling, most analyses focus on isolated stages or mechanisms, overlooking the integrated, dynamic r

Brucella, Coxiella, and Theileria Species DNA in Haemaphysalis qinghaiensis Ticks Collected from Goats and Sheep in Qinghai Province, Northwest China.

Haemaphysalis qinghaiensis is an endemic tick species distributed in the western plateau areas of China. Although they are three-host ticks, infesting multiple animals (including humans), the occurrence of various tick-borne agents has barely been investigated. In this study, we collected 136 H. qinghaiensis specimens from sheep and goats in Menyuan County in Qinghai Province, northwest China. The Brucella, Coxiella, and Theileria/Babesia species' DNA were detected by nested or hemi-nested PCR and further identified by amplifying their key genes. Brucella abortus and B. melitensis DNA were det

TANK-binding Kinase 1 regulates the intracellular survival of Brucella.

Brucella, the causative agent of brucellosis, is a globally important intracellular pathogen. TANK-binding kinase 1 (TBK1), a key component of the type I interferon induction pathway, is known to play a critical role in controlling intracellular bacterial infections. However, the function of TBK1 during Brucella infection remains poorly understood. In this study, we investigated the role of TBK1during Brucella infection. We observed that Brucella infection upregulates TBK1 expression in macrophages. Using TBK1-targeting small interfering RNAs (siRNAs) and overexpression approaches, we examined

Vegetation composition shapes denitrifier community structure and enhances nitrogen removal in estuarine wetlands: evidence from Reed-Willow Mix promoting nirK-dominated guilds.

Estuarine wetlands are critical biogeochemical hotspots where vegetation and soil properties jointly regulate microbial processes such as denitrification. This study investigated soil physicochemical properties and denitrifying bacterial communities (harboring nirS and nirK genes) across different vegetation types (Reed, Zhongshanshan, and Reed-Willow Mix) and soil depths (0-20 cm, 20-40 cm, and 40-60 cm) in the mudflat of the Paihe River estuary, Chaohu Lake. Soil nutrient availability and pH varied significantly with vegetation, with mixed vegetation supporting higher organic matter, nitrate

Brucella abortus histidine auxotrophs are copper sensitive.

Despite decades of investigation into bacterial pathogens, the conditions met by intracellular bacteria are still unclear. These conditions can include access to nutrients, such as amino acids, and exposure to toxic compounds, like copper. To investigate the ability of Brucella abortus, a facultative intracellular pathogen responsible for a major zoonosis, to cope with copper, we performed a Tn-seq analysis to identify copper-sensitive mutants. Unexpectedly, we realized that classical copper resistance systems (involving CopA and CueO homologs) do not appear to be robustly needed, while histid

Structure-guided identification of a PrPC-binding peptide derived from Brucella abortus Hsp60 for receptor-mediated mucosal targeting.

Here, we report the structure-guided identification pipeline of a novel PrPC-binding peptide, PEP1, derived from Brucella abortus Hsp60 through computational docking. Computational modeling predicted that PEP1 engages the PrPC binding interface with favorable binding energetics, motivating its experimental evaluation as a receptor-targeting ligand. Fusion of PEP1 to a model protein enabled receptor-mediated targeting to intestinal M-cell-associated regions and resulted in increased mucosal IgA and systemic IgG responses relative to the untargeted protein. Together, these findings establish PEP

Study on the detection method of Brucella differential type PCR.

Currently, the prevention and control of zoonotic diseases such as brucellosis in some developing countries still primarily rely on "vaccination + quarantine and culling." However, the inability to differentiate between natural infection and vaccine-induced antibodies in immunized livestock has become a new challenge in veterinary clinical diagnosis. To address the issue of distinguishing between wild-type Brucella strains and attenuated vaccine strains in vaccinated animals, this study developed singleplex and multiplex PCR assays targeting six differential loci (NC7250, GL2189, wbkF-wbkD, WP

Seroprevalence and Associated Factors of Camel and Human Brucellosis in Dire District, Southern Ethiopia: A One-Health Perspective.

INTRODUCTION: The public health and economic impact of brucellosis remain a significant concern in Ethiopia. The high seroprevalence rates observed in both camels and humans indicate the potential for cross-species transmission, highlighting the risk of brucellosis spreading. However, there is limited evidence concerning the relationship between brucellosis prevalence in humans and camels in the pastoralist regions of Ethiopia. METHODS: A cross-sectional study was conducted among 390 camels and 390 camel farmers in the Dire district from November 2023 to March 2024, using multistage sampling.
