The presence of multiple variants of IncF plasmid alleles in a single genome sequence can hinder accurate replicon sequence typing using in silico pMLST tools.

UNLABELLED: IncF plasmids are mobile genetic elements found in bacteria from the Enterobacteriaceae family and often carry critical antibiotic and virulence gene cargo. The classification of IncF plasmids using the plasmid Multi-Locus Sequence Typing (pMLST) tool from the Center for Genomic Epidemiology (CGE; https://www.genomicepidemiology.org/) compares the sequences of IncF alleles against a database to create a plasmid sequence type (ST). Accurate identification of plasmid STs is useful as it enables an assessment of IncF plasmid lineages associated with pandemic enterobacterial STs. Our i

Enterobacterial common antigen repeat-unit flippase WzxE is required for Escherichia coli growth under acidic conditions, low temperature, and high osmotic stress conditions.

Colanic acid and enterobacterial common antigen (ECA) are cell-surface polysaccharides that are produced by many Escherichia coli isolates. Colanic acid is induced under acidic, low temperature, and high-salt conditions and is important for E. coli resistance to these stresses; however, the role of ECA in these stresses is less clear. Here, we observed that knockout of flippase wzxE, which translocates lipid-linked ECA repeat units from the cytoplasmic side of the inner membrane to the periplasmic side, resulted in the sensitivity of E. coli BW25113 to acidic conditions. The wzxE-knockout muta

Unraveling the mechanisms of propofol-induced psychological dependence: a multi-omics approach linked to gut microbiota in hippocampal function.

INTRODUCTION: Drug abuse is becoming a global public health crisis. According to the United Nations, the number of drug users worldwide has increased dramatically over the past decade, with a surge in the number of drug abusers. The problem was exacerbated by the expanding market for illicit drugs and the increasing availability of synthetic drugs such as fentanyl. Clinical drug abuse is a problem that requires particular attention, and the potential addictive properties of some drugs and their mechanisms of action are currently unknown, which limits the development and implementation of drug

Genomic diversity and tracing of Paenibacillus larvae in Australia: implications for American foulbrood outbreak surveillance in low-diversity populations.

Paenibacillus larvae is the causative agent of American foulbrood (AFB) in honeybees (Apis mellifera) and a devastating pathogen for honey and pollination industries worldwide. Despite this threat, a genomic survey of P. larvae has not been attempted within Australia. To examine the diversity of Australian populations, we sequenced 368 P. larvae genomes sourced primarily from south-eastern Australia. Multilocus sequencing typing analysis identified only 4 sequence types across all 368 samples, with 2 sequence types (ST18 and ST5) representing 96% of all isolates. In comparison to European-sour

Identification of SanA as a novel regulator of peptidoglycan biogenesis in Escherichia coli.

Gram-negative bacterial cell envelope consists of a surface-exposed lipid bilayer (outer membrane or OM) that serves as a permeability barrier to maintain the cellular integrity. Beneath the OM is the periplasmic space that harbours peptidoglycan (PG), a highly cross-linked mesh-like glycan polymer closely encasing the inner membrane (IM). During growth of a bacterium balanced synthesis of the envelope components is required to maintain the cellular integrity, of which little is known. In this study, we identify sanA, an ORF of unknown function encoding a predicted IM-anchored protein as a fac

Coexistence of PMQR and ESBL genes among clinical Escherichia coli isolates from community-acquired UTI in Mexicali, on the US-Mexico border.

Escherichia coli is an opportunistic pathogen and a leading cause of Community-Acquired Urinary Tract Infections (CA-UTIs). E. coli can harbor multiple genetic resistant determinants, such as Extended Spectrum Beta-lactamases (ESBL) and Plasmid-Mediated Quinolone Resistance (PMQR) genes, complicating the empirical treatment of UTIs with β-lactams and quinolones. The aim of his study was to characterize ESBL and PMQR genes among E. coli isolates from CA-UTI in Mexicali, Mexico Isolates were collected from January to December 2023. Identification was performed by MALDI-TOF, and ESBL-producing de

Repurposing of Furin Inhibitors to Reduce Pathogenic E. coli- and Shigella flexneri-Induced Cytotoxicity, Oxidative Stress and Inflammation in Mammalian Epithelial Cells.

BACKGROUND/OBJECTIVES: Enterobacteriaceae, including pathogenic Shigella (S.) flexneri and Escherichia (E.) coli, cause severe gastrointestinal infections through toxins like Shiga and Shiga-like toxins. Antibiotic use is often discouraged due to its potential to increase toxin effects or contribute to the development of resistance. The host protease furin is capable of activating several viral glycoproteins and bacterial toxins, thus enhancing pathogen infectivity. METHODS: To assess the therapeutic potential of furin inhibitors, cultured epithelial cell models (IPEC-J2 and MDCK) were used.

Dynamics and Interactions of OmpF Porin in an Asymmetric Bacterial Outer Membrane including LPS, ECA, and CPS.

Outer membrane (OM) proteins play a vital role in the physiology of Gram-negative bacteria, and outer membrane protein F (OmpF) is one of the most studied porins in Escherichia coli. In this study, we have developed a comprehensive E. coli OM model with lipopolysaccharides (LPS), enterobacterial common antigen (ECA), and capsular polysaccharides (CPS) in the outer leaflet and with phospholipids in the inner leaflet. Using extensive all-atom molecular dynamics simulations of OmpF in this realistic asymmetric OM environment, we have investigated the structure and dynamics of OmpF within the OM a

Functional loss of rffG and rfbB, encoding dTDP-glucose 4,6-dehydratase, alters colony morphology, cell shape, motility and virulence in Salmonella Typhimurium.

Lipopolysaccharide (LPS) O-antigen and enterobacterial common antigen (ECA) play crucial roles in maintaining the outer membrane in Gram-negative bacteria. Mutations in the biosynthetic pathways of LPS and ECA may lead to accumulation of intermediates, resulting in morphological changes and activation of stress responses. However, the functional consequences of abrogation of both O-antigen and ECA synthesis in Salmonella enterica serovar Typhimurium (S. Typhimurium) are not well investigated. In this study, we generated single and double-deletion mutants of rfbB and rffG, encoding dTDP-glucose

Simulated microgravity triggers a membrane adaptation to stress in E. coli REL606.

Investigating the evolution of Escherichia coli in microgravity offers valuable insights into microbial adaptation to extreme environments. Here the effects of simulated microgravity (SµG) on gene expression and genome evolution of E. coli REL606, a strain evolved terrestrially for 35 years, is explored. The transcriptomic changes for glucose-limited and glucose-replete conditions over 24 h illustrate that SµG increased the expression of genes involved in stress response, biofilm, and metabolism. A greater number of differentially expressed genes related to the general stress response (GSR) an

Molecular assessment of carbapenem-resistant and ESBL Klebsiella pneumoniae clinical isolates to decipher the correlation of antimicrobial resistance with virulence traits.

Klebsiella pneumoniae is a nosocomial pathogen that poses a serious concern due to the high prevalence of extended-spectrum beta-lactamases (ESBL) and carbapenem-resistant K. pneumoniae (CRKP) strains. However, the data on the prevalence of contributing virulence determinants are limited in the Pakistani population. The study aims to characterize clinical isolates of K. pneumoniae to understand clonal relationships and determine the relationship of antibiotic resistance with different virulence factors (i.e., biofilm, capsular polysaccharide, hemolysis, efflux pump, and outer membrane porins).
