While Campylobacter species are often considered normal gastrointestinal commensal bacteria in many food animals, some species may cause gastrointestinal or reproductive diseases in swine. The U.S. swine industry lacks recent Campylobacter species prevalence estimates, which are useful in animal and public health management recommendations. This study describes the prevalence and characteristics, including antimicrobial resistance (AMR) of Campylobacter species as part of the National Animal Health Monitoring System (NAHMS) Swine 2021 study. Campylobacter species were isolated using culture-ba

Antibiotic resistance has become one of the most significant global health threats, with nearly three million antibiotic-resistant infections occurring per year in the United States alone. This problem is especially acute in Gram-negative bacteria, which possess an outer membrane (OM) that surrounds the aqueous periplasm and cytoplasmic membrane and acts as a permeability barrier capable of excluding many antibiotics. The OM of Enterobacterales also contains a highly conserved, invariant carbohydrate-derived moiety known as enterobacterial common antigen (ECA), which has been shown to play a s

Bacteria of the order Enterobacterales (e.g., Escherichia, Klebsiella, Yersinia) produce an invariant glycan, enterobacterial common antigen (ECA), that plays a role in maintaining outer membrane (OM) impermeability. ECA is found as the headgroup of a phospholipid and attached to LPS in the OM, and as a periplasmic cyclic form (ECACYC). WzyE polymerizes ECA repeat units to form the ECA chain with final chain length regulated by WzzE, a class 1 polysaccharide co-polymerase (PCP1). PCP1 function studies have shown interaction of the PCP1 transmembrane helices with the polymerase and a secondary

This study aimed to isolate and genetically characterize Arcobacter species from broiler chickens sampled at three slaughterhouses in Grenada, West Indies. A total of 126 samples-including cloacal swabs, intestinal contents, and meat-from 42 birds were cultured using a chromogenic agar medium. Arcobacter spp. were detected in 21.4% (9/42) of the birds. Among the sample types, meat exhibited the highest prevalence at 14.3% (6/42), followed by fecal samples at 7.1% (3/42) and cloacal swabs at 2.4% (1/42). Genus- and species-specific polymerase chain reaction assays on 33 isolates identified five

In hospital environments, pathogenic bacteria spread easily and acquire virulence and antibiotic resistance genes. The aim of the study was an evaluation of the genetic diversity of 109 K. pneumoniae isolates recovered from patients of a district hospital in central Poland. The frequencies of genes coding for β-lactamases, efflux pumps, and virulence factors were determined. Genotyping of the isolates was performed with ERIC (Enterobacterial Repetitive Intergenic Consensus) and REP (Repetitive Element Sequence Based) PCR techniques, with 21 and 19 genotypes being identified, respectively. The

Both FeCl3 conventional coagulation and Fe(0) electrocoagulation were highly effective in mitigating the long-tailed somatic phage P1. We targeted enterobacterial coliphages because they are better than fecal indicator bacteria in tracking environmental persistence of viral pathogens and their fate in wastewater unit operations. Cryogenic electron microscopy (cryo-EM) of intact/damaged P1 and enumeration of infective virions by plaque assays demonstrated control via both removal and inactivation. Cryo-single particle analysis was coupled with cryo-electron tomography to generate 3-dimensional

Extrachromosomal and mobile genetic elements, including plasmids, and accessory chromosomes, are prevalent in all life domains. Elements integrated into the host chromosome replicate and segregate via the host life cycle. In contrast, the persistence of autonomously replicating elements relies on their ability to remain within the host population. Here, we compare the evolutionary advantage of different persistence strategies found in prokaryotic plasmids. Through intracellular competitions between plasmid genotypes, we find that the combination of active partitioning during cell division with

Bacteria use sophisticated acid stress response strategies to withstand fluctuating environmental pH, with enterobacterial inducible amino acid decarboxylases playing a major role. The lysine decarboxylase LdcI catalyses lysine-to-cadaverine conversion coupled to proton consumption and carbon dioxide release, thereby buffering cytoplasmic and extracellular pH. Our previous studies showed that Escherichia coli LdcI forms intracellular patches under mild acid stress, and that purified LdcI polymerises into filaments at acidic pH. Here, we investigated the physiological relevance of LdcI filament

Introduction. Nosocomial infections, particularly those caused by Gram-negative bacilli, indeed pose significant challenges in healthcare settings. Hospital staff can act as carriers of these infections, potentially transmitting them to patients and colleagues. Propolis, a natural resinous substance collected by honeybees, has shown promising antibacterial properties against various microorganisms, including Gram-negative bacteria like Escherichia coli and Klebsiella pneumoniae.Hypothesis/Gap Statement. Despite the documented antibacterial properties of propolis, limited research has evaluated

The dissemination of antibiotic resistant bacteria (ARB) and genes is one factor responsible for the increasing antibiotic resistance and the environment plays a role in resistance spread. Animal excreta can contribute to the contamination of the environment with ARBs and antibiotics and in some cases, environmental bacteria under antibiotic pressure may acquire antibiotic resistance genes (ARGs) from ARBs by horizontal gene transfer. In Guadeloupe, a French overseas department, organic amendments derived from human and animal waste are widely used in soil fertilization, but their contribution

BACKGROUND: Prognostic factors significantly associated with postoperative delirium (POD) have been reported discordantly, possibly due to heterogeneous cohorts. Here, bacteria extracellular vesicles (BEVs) were introduced to predict the POD status of a unique patient cohort.
METHODS: One hundred twenty-eight patients who underwent spinal surgery participated in this prospective cohort study. Significant preoperative factors (i.e., baseline characteristics, and sequences of 16s rRNA genes from bloods and stools) between patients with and without delirium were subjected to random forest classi

While lettuce immune responses to enteropathogens have been studied at the molecular and physiological levels, plant secondary metabolite responses have received little attention. We evaluated romaine lettuce phenolic metabolite responses to Escherichia coli O157:H7 and Salmonella enterica Enteritidis infiltrated into the leaf apoplast. Evaluating spectrophotometric profiles of leaf extracts, we detected shifts in overall phenolics and developed a semi-quantitative method to measure representative phenolics absorbing maximally at 255, 273, 280 and 329 nm, based on known standards for quercetin

The Rcs signal transduction system is a phosphorelay responsible for sensing enterobacterial cell envelope stresses. In Escherichia coli, the Rcs system is required to survive treatment with A22 and mecillinam, antibiotics that perturb cell size. To test whether size changes are correlated with envelope damage and thereby sensed by the Rcs system, we tuned E. coli cell size via A22 treatment, mutations in the cell-shape determinant MreB, and mechanically confined growth. In all conditions, cell width was strongly correlated with Rcs activation, and RcsF, the outer-membrane-localized upstream c

The use of metabolic selection markers has advanced stable cell line generation, increasing productivity while simultaneously eliminating the need for antibiotic reagents. This study explores the potential of bacterially derived glutamine synthetases (GS) as a novel generation of metabolic selection markers to further enhance CHO cell culture performance. GS-I proteins were extracted from the genomes of enterobacterial and actinomycetes species. Three of these enzymes demonstrated functionality when stably transfected into GS-deficient CHO cells, leading to a 3- to 4-fold increase in antibody

Segmental duplications play an important role in genome evolution via their contribution to copy-number variation, gene-family diversification, and the emergence of novel functions. The detection of segmental duplications is challenging due to heterogeneous amelioration of sequence similarity among duplicates, which hinders the reconstruction of continuous sequence alignment. Here we introduce SegMantX, a novel approach for the identification of diverged segmental duplications in prokaryote genomes using local alignment chaining. In this approach, local alignments resulting from a preliminary
