INTRODUCTION: Human Papillomavirus (HPV) is a well-established cause of cancers of the cervix, vulva, and vagina, particularly high-risk types such as HPV-16 and 18. In women living with HIV, impaired immune function reduces viral clearance, leading to persistent infection, faster progression of precancerous lesions, and involvement of multiple sites within the lower genital tract. This phenomenon is often explained by the concept of "field cancerization", where widespread HPV exposure results in synchronous neoplastic changes in adjacent mucosal tissues.

BACKGROUND: The potential for precancerous cervical lesions to develop into cervical cancer over time is a significant public health concern in low and middle-income countries, including Ethiopia. The Ethiopian government is supporting the prevention of cervical cancer by acetic acid screening and vaccinations. However, the determinants of precancerous cervical lesions were not well studied. Therefore, this study aimed to identify determinants of pre-cancerous cervical lesions among women screened for cervical cancer at Debre Markos town public health institution, Ethiopia, 2025.

Background Adolescent pregnancy presents significant health challenges worldwide. Each year, millions of teenage girls in developing regions become pregnant. In Uganda, teenage pregnancy remains common, with marked regional variations. Adolescents in low socioeconomic situations, with limited education, or who face barriers to contraceptive access are more prone to unintended pregnancies. These pregnancies may result in multiple health complications, including premature births, small for gestational age babies, postpartum depression, anaemia, and urinary tract infections (UTIs). Methods A cros

BACKGROUND: Optimal hepatitis C virus (HCV) screening is required to prevent transmission and progression to advanced liver disease and to meet global elimination targets. While administrative health data can highlight trends in testing and treatment, additional information is required to understand barriers in the care cascade. We conducted a real-world analysis documenting the care cascade and the effectiveness of HCV antibody (Ab) screening modalities across different community and clinical settings.
METHODS: This was a cross-sectional study of individuals who completed HCV Ab testing at p

OBJECTIVE: Programmed cell death protein 1 (PD-1) inhibitors have established a cornerstone in cancer therapy; however, there remains a lack of data regarding their use in people living with human immunodeficiency virus (HIV) infection (PWH), especially in patients with low CD4+ T-cell counts or baseline detectable HIV RNA. This study assessed the real-world efficacy and safety of PD-1 inhibitors in PWH presenting with concurrent malignancies, specifically capturing individuals with advanced baseline immunosuppression (CD4+ T-cell count < 100 cells/μL, n = 4) or with detectable HIV RNA (n = 9)

BACKGROUND: Sexually transmitted infections (STIs) continue to pose a significant public health challenge globally, negatively impacting reproductive health and increasing the likelihood of infertility, complications during pregnancy, and mother-to-child transmission. Despite the increasing burden of self-reported (SR) STIs, there is limited evidence on their prevalence and associated factors across Africa. Applying the socio-ecological model (SEM), this study sought to estimate the prevalence and identify the factors associated with SR-STIs among sexually active women in Africa.

Accurate inference of gene regulatory networks (GRNs) from single-cell gene expression data is challenging due to noise, data sparsity, and variability in gene-gene associations across cells. We propose CoReGRN (Contextual Refinement of Gene Regulatory Networks), a nonparametric, context-aware post-processing framework that refines inferred GRNs by reweighting candidate regulatory interactions using Mutual Information based association strength and local network context. The method uses empirical cumulative distribution function (ECDF) based scores to assess how unusual each interaction is rel

BACKGROUND: Malaria remains a major cause of febrile illness in Malawi; however, the epidemiology of non-malarial febrile illnesses (NMFIs) is less well characterized. Understanding the distribution and determinants of NMFIs across demographic groups is essential for improving case management in malaria-endemic settings.
OBJECTIVES: This study aimed to characterized the distribution and predictors of NMFIs among preschool-aged children (PSC), school-aged children (SAC), women of reproductive age (WRA), and men in Malawi.
METHODS: Secondary data from the 2015-2016 Malawi Micronutrient Survey

Adolescent girls and young women (AGYW) in South Africa experience a disproportionate burden of HIV infection. Genital inflammation has been implicated in increased HIV susceptibility through enhanced availability of target immune cells; however, the effects of recent sexual activity and vaginal-stimulating product (VSP) use on mucosal immune responses remain incompletely defined. This study characterised mucosal immune profiles following recent sexual exposure and VSP use in adolescent and adult women. HIV-negative sexually active adolescents (14-19 years) and adult women (25-35 years) were e

BACKGROUND: Cryptococcal meningitis (CM) is a life-threatening central nervous system fungal infection. In adults without human immunodeficiency virus (HIV) infection, early prognostic indicators remain limited. D-dimer reflects activation of coagulation and fibrinolysis and may provide prognostic information in severe infections, but its significance in non-HIV CM is unclear.
METHODS: We retrospectively included consecutive adults with microbiologically confirmed non-HIV CM admitted to a tertiary teaching hospital from January 2012 to December 2023. The primary outcome was all-cause in-hospi

BACKGROUND: The fight against HIV in the Democratic Republic of Congo (DRC) is hindered by systemic challenges; notably, limited access to HIV viral load (VL) testing. Achieving VL suppression is the primary global target for eliminating HIV as a public health threat by 2030. This study aimed to determine the rate of VL suppression and identify associated factors among people living with HIV (PLHIV) receiving antiretroviral therapy (ART) in Bunia.
METHODS: A descriptive and analytical cross-sectional study was conducted among PLHIV receiving care at treatment sites in Bunia over a period of 1

Nucleic acid amplification tests (NAATs) are central to modern virology diagnostics. However, evidence supporting alternative specimen types remains uneven across viral families, especially for emerging viruses. This limits diagnostic flexibility in outbreak and clinically complex settings. We conducted a scoping review of NAAT detectability across key body sites for human RNA viruses. PubMed and Embase were systematically searched for studies reporting NAAT results from urine, blood, fecal, cerebrospinal fluid, or respiratory specimens. Data were independently screened and synthesized to summ

BACKGROUND: HIV-associated tuberculosis is a leading cause of mortality. Mycobacterium tuberculosis bloodstream infection (MTB-BSI) is complicated by diagnostic difficulty and severe illness. We assessed whether MTB-BSI continues to be common in the era of widespread antiretroviral therapy and whether it continues to result in increased risk of early mortality.
METHODS: We enrolled inpatients from medical wards irrespective of tuberculosis symptoms and outpatients with tuberculosis symptoms. All participants had HIV and were ≥18 years old, and all were recruited from 7 countries: Malawi, Sout

BACKGROUND: Tuberculosis (TB) remains a major public health challenge in Africa; however, conventional statistical approaches often fail to capture diagnostic uncertainty, spatial heterogeneity, and complex disease dynamics, limiting evidence-based decision-making. This review synthesizes the application and performance of advanced statistical and computational methods for TB diagnosis and treatment outcomes in Africa.
METHODS: Following the PRISMA 2020 guidelines, we systematically searched PubMed and Scopus for peer-reviewed studies (2010-2025) conducted in Africa that applied advanced meth

Natural killer (NK) cells are key innate effectors during antiviral immune responses, with their activity regulated in part by interactions between polymorphic killer-cell immunoglobulin-like receptors (KIRs) on NK cells and their major histocompatibility complex class I (MHC I) ligands on target cells. In human immunodeficiency virus (HIV) infection, certain KIR and MHC I allelic combinations are associated with enhanced viral control and delayed disease progression. To interrogate the contribution of a common KIR+ NK cell subset to the immune response to simian immunodeficiency virus (SIV) i
