Mouse Models for Use in Cryptosporidium Infection Studies, Quantification of Parasite Burden Using Flow Cytometry, qPCR and Histopathology, and Confocal Imaging of Oocysts.

Cryptosporidiosis threatens the lives of young children in developing countries and newborn calves around the world. No vaccine or therapy can prevent or cure this diarrhea-inducing enteric disease caused by Cryptosporidium spp. protozoan parasites. There is an essential need to discover new therapeutic drugs efficient in reducing parasite burden in infected individuals. Research therefore relies on reliable small animal models of cryptosporidiosis. Here, we present excellent mouse models which can efficiently mimic pathogenesis of human and bovine cryptosporidiosis. We also describe methods t

Calf Clinical Model of Cryptosporidiosis for Efficacy Evaluation of Therapeutics.

Cryptosporidiosis, caused by the apicomplexan parasite Cryptosporidium parvum, is a moderate-to-severe diarrheal disease, now recognized as one of the leading causes of morbidity and mortality in livestock globally and in humans living in resource-limited parts of the world, particularly those with AIDS or who are malnourished. This recognition has fueled efforts for the discovery of effective therapeutics. While recent progress in drug discovery has been encouraging, there are presently no acceptably effective parasite-specific drugs for the disease. The urgent need for new drug discovery or

High-Content Screening for Cryptosporidium Drug Discovery.

High-content screening (HCS) is a cell-based type of phenotypic screening that combines multiple simultaneous readouts with a high level of throughput. A particular benefit of this form of screening for drug discovery is the ability to perform the interrogation in a biologically relevant system. This approach has greatly advanced the field of drug discovery for cryptosporidiosis, a diarrheal disease caused by protozoan parasites of Cryptosporidium spp. These parasites are obligate intracellular parasites and cannot be cultured in vitro without the support of a host cell, limiting the options f

Phenotypic High-Throughput Screening and Drug Efficacy Test Against the Growth of Cryptosporidium parvum In Vitro by qRT-PCR.

An effective method to quantify parasite loads is key to evaluating anti-cryptosporidial drug efficacy in vitro. However, high-throughput screening of drugs against Cryptosporidium parvum was previously infeasible using labor-intensive traditional assays. Here, we describe a simplified quantitative RT-PCR assay that directly uses cell lysates as templates, enabling high-throughput quantification of parasite loads. This assay can also be used to evaluate drug efficacy against the growth of C. parvum in vitro.

In Vitro Culture of Cryptosporidium spp. Using Stem Cell-Derived Intestinal Epithelial Monolayers.

Cryptosporidium parvum has a complex life cycle consisting of asexual and sexual phases that culminate in oocyst formation in vivo. The most widely used cell culture platforms to study C. parvum only support a few days of growth and do not allow the parasite to proceed past the sexual stages to complete oocyst formation. Additionally, these cell culture platforms are mostly adenocarcinoma cell lines, which do not adequately model the parasite's natural environment in the small intestine. We present a method for generating mouse primary intestinal epithelial cell monolayers that support long-te

Two- and Three-Dimensional Human Intestinal Tissue Models for Cryptosporidium Infection.

Conventional cell cultures utilizing transformed or immortalized cell lines or primary human epithelial cells have played a fundamental role in furthering our understanding of Cryptosporidium infection. However, they remain inadequate with respect to their inability to emulate in vivo conditions and support long-term growth and completion of the parasite's life cycle. Previously, we developed a three-dimensional (3D) silk scaffold-based model using transformed human intestinal epithelial cells (IECs). This model supported C. parvum infection for up to 2 weeks and resulted in completion of the

Cryptosporidium Oocyst Purification Using Discontinuous Gradient Centrifugation.

Many laboratory studies in cryptosporidial research require a source of purified oocysts. Sources can include experimentally infected laboratory animals or from samples collected from naturally infected animals and from clinical cases of human cryptosporidiosis. Purification of oocysts can be accomplished with readily available laboratory equipment including table-top centrifuges and microcentrifuges. Following purification, oocysts can be stored in antibiotic-supplemented buffers or in 2.5% aqueous potassium dichromate for over 6 months. Ultimately, oocyst viability and infectivity declines t

Transfection of the free-living alga Chromera velia enables direct comparisons with its parasitic apicomplexan relative, Toxoplasma gondii.

Chromera velia is a photosynthetic, free-living alga closely related to the apicomplexan parasites, a phylum of intracellular pathogens responsible for many devastating diseases, including malaria, cryptosporidiosis and toxoplasmosis. With molecular and cellular landmarks clearly related to but distinguishable from those found in apicomplexan parasites, Chromera provides an opportunity to investigate the evolutionary origin of the structures and processes needed for intracellular parasitism. However, tools for defining localization and functions of gene products do not exist for Chromera, whic

Stimulation of growth in Piper betle L. via induced systemic resistance (ISR) under combined pathogenic stress through priming of Jasmonic acid.

BACKGROUND: Choosing an orthopedic surgeon is a pivotal decision shaped by clinical competence and multiple non-clinical influences. While surgical expertise and outcomes are fundamental considerations, patients and their families also weigh communication quality, cultural alignment, institutional reputation, financial constraints, and increasingly, digital information sources. Understanding these determinants is crucial to delivering patient-centered orthopedic care. METHODS: A scoping review was conducted following Arksey and O'Malley's framework and reported in accordance with PRISMA-ScR g

Isospora habiae n. sp. (Chromista: Apicomplexa: Eimeriidae) from Red-crowned Ant-tanagers Habia rubica (Vieillot, 1817) (Aves: Passeriformes: Cardinalidae) in South America.

The Red-crowned Ant-tanager Habia rubica (Vieillot, 1817) is a forest-dependent passerine that inhabits the understory of humid forests and is widely distributed throughout Central and South America, extending northward into Mexico. Wild birds may host a broad diversity of parasites, among which coccidia constitute one of the most species-rich and taxonomically complex groups. Within this context, the present study reports a new species of Isospora Schneider, 1881 parasitizing Red-crowned Ant-tanagers captured in Itatiaia National Park, a protected area of high ecological vulnerability in sout

Cystoisospora belli infection in an immunocompetent patient.

Cystoisospora belli, also known as Isospora belli, is an opportunistic pathogen which predominantly affects individuals with compromised immunity, including transplant recipients and patients with AIDS, HTLV-1 infections, or hematologic malignancies. The primary symptom of C. belli infection is diarrhea, but the rarity of C. belli infection makes C. belli an uncommon etiology in patients with diarrhea. Only sporadic cases of C. belli infection have been documented in the past century. Diagnosis of C. belli infection in non-endemic regions poses a challenge and delays can escalate mortality ris

Phylogeography and genetic structure of avian isospora based on mitochondrial COI gene sequences from 17 countries.

Isosporiasis is caused by apicomplexan protozoa of the genus Isospora, transmitted primarily via the fecal-oral route. While Isospora infects various vertebrates, infections are particularly notable in birds. However, the phylogeography and genetic structure of avian Isospora remain poorly documented. In total, 273 selected mitochondrial cytochrome c oxidase subunit I (COI) sequences belonging to avian Isospora species were retrieved from NCBI and subsequently analyzed to understand their phylogeographic patterns. Overall, 119 haplotypes, predominated by Hap_2, were identified across 17 countr

Isospora atticorae n. sp. (Apicomplexa: Eimeriidae) from the white-thighed swallow Atticora tibialis (Cassin, 1853) (Passeriformes: Passeri: Hirundinidae) in Brazil.

BACKGROUND: The diversity of coccidians in Neotropical wild avifauna remains significantly underestimated, particularly within the family Hirundinidae. While cosmopolitan and widely distributed hosts like Hirundo rustica Linnaeus, 1758 dominate the literature with three described Isospora spp., ecological traits of other swallows may drive different parasitic dynamics. This study provides the first integrative characterization of a coccidian parasite from Atticora tibialis (Cassin, 1853), a resident swallow strictly associated with complex and preserved Neotropical forest biomes. METHODS: Fec

Detection of Cryptosporidium and Giardia duodenalis in Reptiles in Thailand.

Cryptosporidium spp. and Giardia duodenalis are significant intestinal protozoan parasites affecting humans and animals worldwide. These infections are transmitted through the faecal-oral route, by contaminated water, food or close contact with infected hosts. The zoonotic risk of transmission from reptiles is considered low; yet, limited research exists regarding Cryptosporidium and Giardia infections in these animals. This study investigated the presence of Cryptosporidium spp. and G. duodenalis in reptile faecal samples from a breeding farm in the Chonburi Province, Eastern Thailand. The te

Gastrointestinal Pathogens Are Highly Prevalent in Endangered Amargosa Voles (Microtus californicus scirpensis).

Disease can threaten endangered species by affecting fitness and survival, suppressing population growth and species recovery. Herein, we investigated three gastrointestinal pathogens-Giardia, Cryptosporidium, and Helicobacter-that could negatively affect the endangered Amargosa vole (Microtus californicus scirpensis). We used PCR testing to determine the presence of the target parasites in 76 fecal samples from 71 Amargosa voles collected during summer 2021 in four marshes in Tecopa, California, USA. Overall, 51 (72%) of 71 voles were PCR positive for Giardia microti, 48 (68%) of 71 for Crypt

Single Oocyst Selection and Whole-Genome Amplification of Cryptosporidium parvum.

Cryptosporidium parvum is a zoonotic parasite of the intestine and poses a threat to human and animal health. However, it is difficult to obtain a large number of oocysts for genome sequencing using in vitro culture. To address this challenge, we describe a procedure for purification of C. parvum from a clinically positive sample and a method for isolation of a single oocyst of C. parvum using a three-axis hydraulic micromanipulator. We employ the strategy of whole-genome amplification (WGA) of 10 oocysts, and purify the whole-genome DNA after amplification, and finally meet the basic conditio

Anti-cryptosporidial efficacy of tricyclic carbazole aminoalcohols in vitro and in vivo.

Cryptosporidiosis is a major cause of diarrheal disease in humans and animals, yet there are no fully effective drugs, particularly for immunocompromised patients. Carbazole aminoalcohols (CAAs) are a relatively unexplored chemical class with reported broad-spectrum antiparasitic activities. Here, we systematically evaluated 36 CAA derivatives for anti-cryptosporidial efficacy. In vitro screening using a 44-h Cryptosporidium parvum infection assay identified eight compounds with low-micromolar EC50 values (1.53-6.86 μM). The three most potent hits (H1402 at 25 mg/kg/d, and YFM1 and YFM3 at 15

New tools for exploring parasite biology and elucidating host-pathogen interactions in cryptosporidiosis.

SUMMARYCryptosporidiosis is a major public health concern, the extent of which has only truly been appreciated within the last decade. Cryptosporidium research has undergone a renaissance, with new insights into population structure, species diversity, and evolution of the parasite driven by the advent of genetic transformation techniques and novel models for culture in vitro and in vivo. Here, we summarize the impact of these advances on our understanding of this important parasite. In the initial section, we focus on what we have learned about host range and infectivity from comparative geno
