The pervasive socio-economic and psychological burden of onchocerciasis in endemic communities of Bo District, Sierra Leone: an explanatory sequential mixed-methods study.

BACKGROUND: Onchocerciasis (river blindness) persists in Bo District, Sierra Leone, despite decades of ivermectin-based mass drug administration (MDA). Programmatic efforts have prioritized parasitological and ocular outcomes, so the socio-economic and psychological burden of the disease within affected communities remains poorly quantified. We aimed to determine the magnitude, structure and distribution of this non-clinical burden. METHODS: We used an explanatory sequential mixed-methods design (QUAN → qual). In the quantitative phase, 1,500 adults across five endemic chiefdoms of Bo Distric

Efficacy of Simparica Trio® (sarolaner, moxidectin, and pyrantel chewable tablets) against circulating microfilariae in dogs infected with Dirofilaria immitis.

BACKGROUND: Canine heartworm disease remains a global threat despite the availability of multiple preventive options, a challenge compounded by the emergence of macrocyclic lactone-resistant heartworm strains, especially in the USA. Circulating microfilariae play a pivotal role in heartworm transmission, serving as a continuous infection reservoir for mosquito vectors. Therefore, targeting this life cycle stage is critical in the framework of a comprehensive and multimodal heartworm prevention strategy. METHODS: A randomised, blinded, negative-controlled laboratory study evaluated the efficac

Coccidiosis in South American camelids: A review of Eimeria species, diagnostic limitations and research gaps.

Protozoan parasites of the genus Eimeria represent important enteric pathogens of South American camelids (SACs), particularly affecting juvenile alpacas and llamas. Despite their veterinary and economic significance, current knowledge regarding camelid coccidiosis remains limited, especially in relation to species taxonomy, endogenous development, molecular diagnostics, and species-specific treatment strategies. This review summarizes current knowledge on Eimeria species infecting SACs, including their morphology, life cycle, epidemiology, pathogenicity, diagnosis, treatment, and prevention.

Evaluating indoor residual spraying in Côte d'Ivoire: the role of routine malaria data quality.

OBJECTIVES: Indoor residual spraying (IRS) was introduced in Côte d'Ivoire in 2020. We seek to identify potential factors that could compromise the quality of routine malaria incidence and determine how impact quantification of IRS on malaria incidence depends on data quality by emphasizing robust counterfactuals with contemporaneous covariates. METHODS: Using mixed-effect models and correlation analysis we compare data from two sources (routine surveillance and re-digitized registries). To quantify decrease in incidence post intervention, we apply Bayesian structural time series analysis wit

A qualitative study exploring factors associated with the accuracy of recording malaria rapid diagnostic test results in two high-burden regions of Côte d'Ivoire.

BACKGROUND: While the expanded use of malaria rapid diagnostic tests (RDTs) in Sub-Saharan Africa has improved testing rates, healthcare workers do not always adhere to results or report them accurately. Because misrecording RDT results compromises malaria surveillance, this study identified factors associated with providers' guideline adherence and accurate RDT reporting in in two high-burden regions in Côte d'Ivoire. METHODS: This qualitative study was embedded within a mixed-methods evaluation of RDT reporting and recording accuracy called the Malaria RDT Capture and Reporting Assessment (

Pentalinonsterol at the Host-Parasite Interface: Antileishmanial Activity, Host Immunomodulation, and Unresolved Mechanisms.

Leishmaniasis remains difficult to treat because available therapies are constrained by toxicity, administration burden, variable efficacy, cost, and drug resistance. This critical narrative review evaluates the preclinical evidence for pentalinonsterol (PEN), a plant-derived cholestane-type steroidal ketone originally isolated from the Mayan medicinal plant Pentalinon andrieuxii. Experimental studies demonstrate PEN activity against Leishmania promastigotes and intracellular amastigotes, together with marked parasite ultrastructural injury; however, no parasite molecular target has been valid

Molecular survey of zoonotic protozoa in free-ranging marmosets (Callithrix penicillata) from Central Brazil: implications for One Health surveillance.

Urban expansion and environmental fragmentation have intensified contact among wildlife, domestic animals, and humans, reshaping infectious disease dynamics at the human-wildlife interface. Non-human primates (NHP), particularly the black-tufted marmoset (Callithrix penicillata), readily adapt to anthropogenic landscapes and may serve as sentinel hosts for zoonotic pathogens. This study investigated the occurrence of Leishmania spp., Plasmodium spp., Toxoplasma gondii, and Trypanosoma cruzi. in 232 free-ranging C. penicillata found dead in urban and peri-urban areas of the Federal District, ce

Performance of recombinase polymerase amplification assay for cutaneous leishmaniasis detection: a systematic review and meta-analysis.

BACKGROUND: Sensitive and robust point-of-care (PoC) assays are urgently needed for the diagnosis of cutaneous leishmaniasis (CL). Recombinase polymerase amplification (RPA), an isothermal molecular assay, has shown promise for CL diagnosis using either a lateral-flow assay (LFA) or a mobile-suitcase-laboratory (MSL) platform. However, its diagnostic performance has been inconsistent across studies. This systematic review and meta-analysis aimed to determine the pooled sensitivity and specificity of RPA for CL diagnosis. METHODS: A comprehensive search was conducted in Google Scholar, EMBASE,

Accessing invasion-competent Plasmodium vivax merozoites reveals antibody-dependent NK cell activation.

Malaria pathogenesis and clinical symptoms arise from blood-stage replication of Plasmodium parasites, which depends on rapid invasion of red blood cells by merozoites. Despite the central importance of this stage, direct functional studies of Plasmodium vivax (P. vivax) merozoites remain limited by the lack of continuous in vitro culture and restricted access to viable, invasion-competent merozoites from clinical isolates. Consequently, key aspects of P. vivax merozoite biology and immune recognition remain poorly defined. Here, we establish a robust platform enabling synchronized release and

Molecular Prevalence and Genotype Detection of Babesia microti in Multiple Wild Rodent Species and Suncus murinus From Subtropical Mountainous Regions of South-Central Zhejiang, China.

Human babesiosis, an emerging zoonosis with ongoing geographic expansion, is primarily caused by Babesia microti, a tick-borne apicomplexan parasite that resides within host erythrocytes. While small mammals serve as its recognized reservoir hosts, the relative contribution of sympatric wild rodents and shrews in subtropical mountain endemic areas of Eastern China remains poorly characterized. In South-central Zhejiang, an endemic focus of human babesiosis, systematic data on local small-mammal hosts remain limited. In this study, 630 small mammals, comprising six rodent species and the shrew

Sevuparin in children with severe malaria: a phase I safety and dose-finding trial.

Mortality rates for African children with severe malaria remain high. Sevuparin, a heparin-like compound, as an adjunctive treatment, may improve outcomes due to its effects on Plasmodium falciparum-infected erythrocytes. A Phase I dose-finding trial in Kenyan and Zambian children with severe malaria aimed to identify the maximum tolerated dose (MTD) of sevuparin, given as three infusions (at 0, 8 and 16 h post-enrolment) using the continual reassessment method (ISRCTN32271864). The primary endpoint was dose limiting toxicity (DLT) defined as Activated Partial Thromboplastin Time (APTT) > 95 s
