Targeting EP2 Receptor Improves Muscle and Bone Health in Dystrophin-/-/Utrophin-/- Double-Knockout Mice.

Duchenne muscular dystrophy (DMD) is a severe genetic muscle disease occurring due to mutations of the dystrophin gene. There is no cure for DMD. Using a dystrophin-/-utrophin-/- (DKO-Hom) mouse model, we investigated the PGE2/EP2 pathway in the pathogenesis of dystrophic muscle and its potential as a therapeutic target. We found that Ep2, Ep4, Cox-2, 15-Pgdh mRNA, and PGE2 were significantly increased in DKO-Hom mice compared to wild-type (WT) mice. The EP2 and EP4 receptors were mainly expressed in CD68+ macrophages and were significantly increased in the muscle tissues of both dystrophin-/-

Retropharyngeal effusion secondary to calcific tendinitis of the longus colli muscle: A case report and brief review of the literature.

Calcification or ossification of the longus colli tendon is a rare and often underdiagnosed cause of acute neck pain, typically seen in middle-aged adults, especially women. This condition results from the deposition of calcium hydroxyapatite crystals in the tendon, causing an inflammatory or granulomatous response. Common symptoms include neck pain, painful swallowing, and fever. CT imaging is the preferred diagnostic tool, showing retropharyngeal and prevertebral effusions with minimal or no peripheral enhancement, often accompanied by calcifications in the superior fibers of the longus coll

Fulminant Heterotopic Ossification following COVID-19 associated Systemic Inflammatory Response Syndrome: Manifestations in Radiology, Nuclear Medicine, and Clinical Application.

Heterotopic ossification (HO) is an unclear etiological trigger that results in diverse extra-skeletal bone formation in muscles and soft tissues. This often results in morbidity and reduced quality of life with pain, contractures, and mobility impairment. We present two patients with HO with a history of severe COVID-19 infection requiring 1-month-long mechanical ventilation. The first patient was found to have progressive stiffening of the right knee and left elbow, with clear demonstration of radiographic findings and active dystrophic calcification by nuclear medicine three-phase bone scan

Senescence: An Overlooked VSMC Phenotype and Therapeutic Opportunity?

Vascular smooth muscle cells (VSMCs) modulate their phenotype from a quiescent, contractile cell to a dedifferentiated, synthetic fibroproliferative cell in response to injury and cardiovascular risk factors. Senescence is a recognized phenotypically distinct cellular state characterized by cell cycle arrest and activation of the p16 and p53 damage response pathway and expression of the senescence-associated secretory phenotype. Low levels of senescence in healthy arteries contribute to vascular homeostasis by ensuring that only healthy VSMCs compose the artery, but they are not intended to be

Myocardial calcification revealed by postmortem computed tomography in a case of infantile myocardial ischemia caused by anomalous origin of the left coronary artery from the pulmonary artery.

Bland-Garland-White syndrome, or an anomalous origin of the left coronary artery from the pulmonary artery (ALCAPA), is a rare congenital coronary anomaly that can cause myocardial ischemia, congestive heart failure, and sudden death. In this study, we aimed to present a case of sudden infant death caused by ALCAPA. Postmortem computed tomography (PMCT) showed a collapsed left lung, enlarged heart volume, and severe calcification of the left ventricular papillary muscles and posterior left ventricular wall. Macroscopic examination during the autopsy revealed marked cardiac hypertrophy and an a

Skeletogenesis and acupuncture points.

The formation of the embryo begins with a "calcium wave" that contributes to the creation of heterogeneities, and then some of them solidify. The further development and a collective information architecture of the embryo is controlled by the so-called morphogenetic field, whose nature is mainly associated with a physical inerton field that is a substructure of the quantum mechanical matter waves, which plays the role of information field in biological systems. The inerton field takes on the function of organising the geometry and topology of the developing system, which we associate with the

Expansion and Temporal Expression Regulation of Cuticle-Associated Gene Families: Molecular Basis of Adaptive Evolution of Moulting in Decapods.

Moulting is fundamental to decapod growth and development, yet its molecular regulatory mechanisms and relationship with environmental adaptive evolution remain incompletely elucidated. Through comparative genomic and transcriptomic analyses, we identified six cuticle-associated gene families (CP8, CP AMP1A, pro-resilin, CP AM1199, CP575, and CP1246) that underwent significant expansion in decapods, displaying species-specific distribution patterns. Notably, the CP1246 family, specifically expanded in crabs and enriched with calcification-related cuticle domains, likely represents a key evolut

Notch signaling in osteoblast progenitor cells is required for BMP-induced bone formation.

Notch signaling is active during bone formation and prior studies have shown that it is required for both robust intramembranous and endochondral bone regeneration. Particularly, the systemic blockade of Notch signaling has been shown to inhibit BMP-induced bone formation in a murine calvarial defect model. In this study, we genetically disrupted the expression of both the dominant Notch receptor, Jagged-1, and the essential Notch signaling transcription factor Rbpj in osteoblast progenitors during calvarial bone healing. We found that Jagged-1 (and Jagged-2) expression by alpha Smooth Muscle

37-Day microgravity exposure in 16-Week female C57BL/6J mice is associated with bone loss specific to weight-bearing skeletal sites.

Exposure to weightlessness in microgravity and elevated space radiation are associated with rapid bone loss in mammals, but questions remain about their mechanisms of action and relative importance. In this study, we tested the hypothesis that bone loss during spaceflight in Low Earth Orbit is primarily associated with site-specific microgravity unloading of weight-bearing sites in the skeleton. Microcomputed tomography and histological analyses of bones from mice space flown on ISS for 37 days in the NASA Rodent Research-1 experiment show significant site-specific cancellous and cortical bone

Evaluation of individual and composite CT-based markers in frailty diagnostics in older patients with cancer undergoing major abdominal surgery.

BACKGROUND: Frailty, a multifactorial syndrome associated with age-related decline in physiological reserves, is a critical determinant of surgical outcomes in the elderly. Comprehensive Geriatric Assessment (CGA) is the gold standard for diagnosing frailty but is resource-intensive. This study investigates the relationship between CT parameters and frailty syndrome based on CGA. Additionally, it aims to evaluate the effectiveness of the proposed composite radiological score in diagnosing frailty and predicting postoperative complications. METHODS: The study was a retrospective analysis of pr

Malignant Ossifying Fibromyxoid Tumor With Lung Metastasis: A Case Report and Literature Review.

Ossifying fibromyxoid tumor (OFMT) is a rare mesenchymal neoplasm of uncertain differentiation, typically exhibiting shell-like ossification and an indolent clinical course. However, some cases demonstrate aggressive behavior with local recurrence or metastasis. A 48-year-old Japanese man presented with a painless right thigh mass. He had been aware of it for 20 years and it had been enlarging slowly for the past year. Pathologic examination revealed that the removed 55-mm tumor chiefly consisted of spindle tumor cells proliferating, in the central area in a hypocellular fashion with scattered

Calciphylaxis diagnosis, management and future directions: a comprehensive update on behalf of the European Renal Association CKD-MBD Working Group.

Calciphylaxis, or calcific uraemic arteriolopathy, is a rare and life-threatening condition predominantly affecting people receiving dialysis. Characterized by painful necrotic skin lesions due to arteriolar calcification and thrombosis, calciphylaxis is associated with high morbidity and mortality. Diagnosis is frequently delayed due to misdiagnosis and an absence of specific diagnostic tests. Current treatment approaches are largely based on registry data and small uncontrolled studies. This update brings together the latest understanding of calciphylaxis pathogenesis, diagnostic approaches

Balancing benefits and risks of aerobic exercise for aging and musculoskeletal health.

BACKGROUND: Long-term exercise is recognized as one of the most effective means of maintaining health after aging, but the relationship between moderate exercise and health in the older population is often overlooked. OBJECTIVE: The present study was conducted to investigate the effects of excessive endurance exercise on the old mice musculoskeletal system. RESULTS: The 8 weeks of normal endurance exercise significantly improved skeletal muscle mitochondrial biogenesis and increased femoral osteogenesis in young and old mice. However, the continued accumulation of total exercise volume as th

A Less Common Anatomical Variant of Bilateral Multiple Lesser Metatarsal Sesamoids With Radiologic and Clinical Correlation.

Accessory or supernumerary bones are bones that are not normally present in the body but can be found as an anatomical variant. While hallucal sesamoids are common, lesser metatarsal sesamoids are uncommon and often underreported. These ossicles develop through endochondral ossification and aid biomechanics by enhancing muscle function and reducing tendon friction. This report presents the first documented case of bilateral multiple lesser metatarsal sesamoids involving several metatarsophalangeal joints, supported by radiologic and clinical correlation. A 48-year-old woman presented with bila

Toll-like receptor 9 promotes aortic chondrogenesis and calcification in apolipoprotein E-deficient mice.

Vascular calcification represents a significant clinical challenge, leading to cardiovascular disease, though its underlying mechanisms remain incompletely understood. Recent studies indicate that Toll-like receptor 9 (TLR9), a key element of innate immunity, plays a pathogenic role in vascular inflammation and atherogenesis. Therefore, we hypothesized that TLR9 signaling promotes vascular chondrogenesis and calcification. We compared apolipoprotein E-deficient (ApoE-/-) mice and Tlr9-/- ApoE-/- mice after 24 -weeks high-cholesterol diet feeding. There were no differences between the groups in
