Warning from a pediatric patient with severe myositis ossificans combined with femoral fracture: a case report.

Myositis ossificans (MO) is a self-limiting benign ossification disorder, characterized by heterotopic ossification within skeletal muscles. Pediatric MO cases are rarer, easy to be misdiagnosed. The diagnosis of traumatic MO is based on a thorough history, physical examination, and imaging studies. Early and proactive conservative treatment can often be effective. We report a case of a 6-year-old patient with a femoral fracture combined with severe MO. He was misdiagnosed at a community hospital as having a bone tumor combined with a pathological femoral fracture and was referred to our hospi

Myositis Ossificans of the Temporalis Muscle Following Neurosurgical Intervention: A Report of a Rare Case and Literature Review.

Myositis ossificans (MO) is a rare benign condition characterized by heterotopic bone formation within soft tissues, with masticatory muscle involvement being exceptionally uncommon. This case report describes a 45-year-old male patient who developed progressive trismus and a preauricular mass six months after right frontotemporal craniotomy for meningioma resection. Imaging revealed an ossified lesion in the temporalis muscle, consistent with post-traumatic MO. Surgical management involved zygomatic arch osteotomy and coronoidectomy, restoring intraoperative mouth opening to 23 mm. Histopatho

Leptin receptor signaling mediates the distinct tendon-bone interface reconstruction in rotator cuff tears and osteoporosis-comorbid rotator cuff tears.

BACKGROUND: The intact tendon-bone interface (TBI) consists of four histological layers-tendon, fibrocartilage, calcified fibrocartilage, and bone-that gradually merge into each other, making complete structural restoration after injury challenging. Osteoporosis poses a significant risk for rotator cuff tears (RCTs) and re-tears after arthroscopic rotator cuff repair (ARCR). Activating Leptin receptor (Lepr) mediated Stat3 signaling transduction facilitates the transcription of Runx2 and Sox9, respectively, and promotes osteogenesis and chondrogenesis. MATERIALS AND METHODS: Sixty-five female

Membrane Palmitoylated Protein 7 is Required for Osteogenesis and is Linked with Bone Mineralization and Osteoporosis: The Functional Evaluation of GEFOS GWAS Hit.

Genome-wide association studies have identified multiple loci associated with bone mineral density, a major determinant of osteoporotic fracture risk. At one such locus, genetic, bioinformatic, and zebrafish knockout data strongly prioritize membrane palmitoylated protein 7 (MPP7) as a candidate gene, although its precise role in bone biology remains poorly defined. MPP7 encodes a member of the p55 Stardust family of membrane-associated guanylate kinase proteins, which are key regulators of epithelial cell polarity and junctional organization. Here, we investigated the functional role of MPP7

Focal Adhesion Kinase Promotes Calcification of Vascular Smooth Muscle Cells via Regulation of Histone Deacetylase 4 and 5.

BACKGROUND: Vascular calcification is an active process driven by osteogenic phenotypic transition of vascular smooth muscle cells (VSMCs) and regulated by a bone-related gene regulatory network. Recent studies showed that FAK (focal adhesion kinase) regulates bone formation by affecting the cellular localization of HDAC (histone deacetylase) 4 and HDAC5. However, it is not known whether FAK exerts effects on vascular calcification in VSMCs through regulating HDACs. METHODS: We used perturbational assays to assess the role of HDAC4, HDAC5, and FAK in VSMC calcification. Pharmacological inhibi

Low-Temperature Three-Dimensional Bioprinted Dual-Factor rhBMP-2/VEGF-165 Biomimetic Scaffolds for Synergistic Bone-Vascularization Repair in Large Segmental Bone Defect.

The reconstruction of large segmental bone defects remains a formidable challenge in orthopedic surgery. Benefiting from the rapid advancement of three-dimensional (3D) printing technology, growth factor-loaded artificial bone scaffolds have been extensively studied and have emerged as one of the pivotal strategies for bone defect repair. However, the development of an ideal scaffold system that meets comprehensive clinical requirements still requires sustained efforts. This study designed a biomimetic bone repair scaffold with bioactive properties using low-temperature 3D bioprinting technolo

Double jaw surgery for a patient with Gnathodiaphyseal dysplasia (GDD): A case report and literature review.

Gnathodiaphyseal Dysplasia is a rare autosomal-dominant disease caused by a mutation in GDD1. Patients typically present with pathological fractures, muscle weakness, and jawbone deformities. This genetic disorder has long been misdiagnosed until 2004 when the genetic basis of this disorder was identified, differentiating it from Osteogenesis Imperfecta and Fibrous Dysplasia. However, clinicians are still largely unaware of this entity and continue to label and treat it as a variation of Osteogenesis Imperfecta. This report describes a 21-year-old male with GDD presenting with numerous fractur

BCL9 as a Key Player in Wnt/β-catenin Signaling: Implications for Osteogenesis, Tissue Repair, and Oncology.

B-cell lymphoma 9 (BCL9) is a regulatory protein that plays a key role within the Wnt/β-catenin signaling pathway. In recent years, it has garnered significant attention across the domains of tissue repair and tumour biology. The Wnt/β-catenin signaling pathway is of great importance in regulating bone tissue formation and osteoblast differentiation. As a co-activator of β-catenin, BCL9 is capable of governing bone development and regeneration by augmenting the transcriptional activity of the Wnt signaling. In addition, BCL9 is also closely related to diverse biological processes, such as angi

Functional anatomy, jaw mechanisms, and feeding behavior of Dunkleosteus terrelli (Placodermi, Arthrodira).

The comparative and functional anatomy of the cleaver-like jaws of the Late Devonian arthrodire Dunkleosteus terrelli remains poorly understood, despite significant advances in our understanding of arthrodire anatomy. Here, we re-investigate the jaw morphology of D. terrelli from a comparative anatomical perspective. Rare cartilaginous elements, preserved attachment sites, and comparisons with other arthrodires suggest the skull of Dunkleosteus contained significantly more cartilage than previously assumed, with many major jaw muscle attachments and cranial articulations located on cartilages.

Cell fusion in osteoclastogenesis.

Multinucleated osteoclasts, generated by fusion of mononucleated precursors, play an essential role in the lifelong remodeling of our bones. Since within the physiological range of osteoclast sizes, their bone-resorbing activity grows with successive fusion events, both initiation of this fusion reaction and its switch off are tightly controlled. In this review, we discuss the mechanisms and proteins that facilitate and regulate this fusion process. The pathway of membrane rearrangements in osteoclast fusion shares many mechanistic motifs with other physiologically important cell-cell fusion p

Efficacy analysis of large-channel spinal endoscope unilateral laminotomy decompression for the treatment of multilevel cervical spinal stenosis with ligamentum flavum hypertrophy.

OBJECTIVE: Cervical spinal stenosis predominantly affects the elderly. After 40 years of age, aging induces progressive loss and rupture of elastic fibers in the ligamentum flavum, accompanied by abnormal proliferation and cross-linking of collagen fibers, as well as calcium salt deposition and even ossification. This study investigates the clinical efficacy and safety of large-channel endoscopic unilateral laminotomy decompression for the treatment of multilevel cervical spinal canal stenosis. METHODS: A retrospective study was conducted on 36 Cervical spinal canal stenosis patients with rad

Brachial Plexus Neuropraxia Post Open Reduction and Internal Fixation of Left Acetabulum Fracture.

INTRODUCTION: Chronic osteomyelitis is a debilitating bone infection that can lead to significant morbidity and functional impairment. This condition is often challenging to treat due to its complex nature and the ability of bacteria to form biofilms, which make them resistant to antibiotics. Early diagnosis and a multidisciplinary approach are crucial for successful management. Orthopedic surgeons often encounter chronic osteomyelitis as a difficult and persistent problem. The key to effective treatment lies in prompt diagnosis and a combination of surgical intervention and antimicrobial ther

A rare case of heterotopic mesenteric ossification after remote abdominal gunshot wounds.

This case documents a rare occurrence of heterotopic mesenteric ossification (HMO) identified at autopsy, highlighting the long-term sequelae of prior abdominal trauma and expanding awareness of this uncommon entity in forensic pathology. HMO, also called intraabdominal myositis ossificans, is a subset of myositis ossificans traumatica and is characterized by the abnormal formation of osseous, cartilaginous, and sometimes bone marrow elements. It is generally associated with trauma or previous abdominal surgeries and can lead to potential life-threatening complications. Autopsy findings may hi
