Sphenoid Temporal Meningioma With Granular Cell Features and Extracranial Infratemporal Extension.

Meningiomas usually arise from arachnoid cap cells and present as intracranial, dural-based lesions. Extracranial extension into the temporalis muscle, infratemporal fossa, orbit, and skull base foramina is uncommon and can mimic primary soft-tissue or peripheral nerve sheath tumors. We report a 54-year-old female who presented with a slowly progressive right-sided facial and temporal swelling over two years, associated with intermittent pain and mild proptosis but no focal neurological deficit. Computed tomography demonstrated a small extra-axial minimally enhancing right anterior temporal le

Lysosomal dysfunction in vascular smooth muscle cells: Mechanisms and therapeutic targets in vascular disease.

Vascular smooth muscle cell (VSMC) dysfunction is a common feature of atherosclerosis, aortic aneurysms, vascular calcification, and other vascular diseases. In VSMCs, lysosomes play an essential role in autophagic degradation, endocytic cargo processing, intracellular recycling, and cellular stress responses. Under pathological conditions, lysosomal dysfunction promotes foam cell formation, osteogenic differentiation, and extracellular matrix remodeling in VSMCs, thereby contributing to vascular diseases. This review summarizes current evidence on the role of lysosomal dysfunction in VSMC bio

Multifocal Skeletal Muscle Mineralisation Following Severe Trauma and Prolonged Critical Illness: A Case Report.

Soft tissue mineralisation is an uncommon complication of severe trauma and critical illness and may occur in association with metabolic derangements, tissue injury and prolonged immobilisation. We present a case of a 26-year-old male patient who developed extensive skeletal muscle calcification following multiple stab wounds, hypovolemic shock, and surgical interventions, including limb amputation and bowel resections. Initial imaging showed no calcification; however, six weeks post-injury, CT demonstrated extensive amorphous calcification in the abdominal wall, chest, iliopsoas, and gluteus

Incidentally Discovered Pulmonary Hamartoma: A Case Report and Review of the Literature.

Pulmonary hamartoma is a rare benign tumor of the lung. It predominantly affects older males and is frequently peripherally located. On chest radiography, it classically appears as a "coin lesion" exhibiting characteristic popcorn-like calcifications. We report the case of a 62-year-old male patient, a former smoker, being treated for multiple myeloma. A computed tomography (CT) scan revealed a solid, bilobed pulmonary nodule in the lingula measuring 8 mm in the long axis, for which he underwent surgical resection. Macroscopically, the tumor appeared whitish, multilobulated, well-defined, and

Calcified Angioleiomyoma on the Dorsum of the Hand of a Patient With End-Stage Renal Disease: A Case Report.

BACKGROUND Angioleiomyoma is an uncommon benign soft-tissue tumor, typically found in the lower extremities. Angioleiomyoma is infrequently observed in the hand, and occurrences involving calcification are even less common. In patient with end-stage renal disease (ESRD) who receive hemodialysis, calcified angioleiomyoma can be challenging to distinguish diagnostically due to its resemblance to tumoral calcinosis. We report a unique case of calcified angioleiomyoma on the dorsum of the left hand in an ESRD patient undergoing long-term hemodialysis, focusing on clinical, imaging, and histopathol

Simultaneous High-Grade Upper-Tract Urothelial Carcinoma and Xanthogranulomatous Pyelonephritis.

A 67-year-old female with a previous history of superficial urothelial carcinoma of the bladder, discharged from surveillance follow-up, presented to her general practitioner with nausea and unintentional weight loss. The patient had radiological findings of a poor nephrogram with significant perirenal inflammatory stranding and hydronephrosis secondary to conglomerate pelvic calcification. This was suggestive of xanthogranulomatous pyelonephritis (XGP) and possible malignant changes. The patient ultimately underwent a radical nephrectomy; subsequently, the specimen revealed high-grade T4 urot

Specific serotonin receptor antagonist reduces murine calcific atherosclerosis.

High levels of peripheral serotonin, produced in the gut, are associated with increased cardiovascular disease risk and bone loss. We previously found that vascular smooth muscle and valvular cells express serotonin receptors, predominantly type 2A (HTR-2A) at baseline and type 2B (HTR-2B) upon TNF-a stimulation. Serotonin treatment augmented TNF-a-induced matrix calcification, whereas the inhibitor of gut serotonin, LP533401, blunted the initiation, but not the progression, of cardiovascular calcification in Apoe-/- mice. In this study, we tested which receptor subtypes mediated the effects o

Precision RNA interference of Smad3 and Runx2 via dual targeting nanocarriers mitigates aortic valve disease.

BACKGROUND: Calcific aortic valve disease (CAVD) is a common malady with few treatment options other than valve replacement by surgery or transcatheter aortic valve implantation (TAVI). Endothelial-to-mesenchymal transition (EndMT) of valvular endothelial cells and osteogenic differentiation of valvular interstitial cells are crucial processes of CAVD. Smad3 and Runx2 are key transcription factors (TFs) that drive these processes by regulating gene expression and cellular functions. We hypothesize that downregulation of these TFs with nanoparticle-mediated RNA interference could mitigate aorti

Proteomics reveals spatial and molecular heterogeneities in advanced atherosclerotic carotid artery plaques.

Atherosclerotic plaque rupture is a major cause of cerebrovascular events, yet the molecular determinants underlying vulnerability-related plaque morphology, including fibrous-cap thickness, remain incompletely defined. Using histomorphology-guided spatial proteomics, here we delineate molecular programs associated with plaque cap phenotype across discrete plaque subregions. In 112 carotid endarterectomy specimens, differences between thin-cap and thick-cap plaques were predominantly localized to the necrotic core and fibrous cap. These differences were enriched for processes related to inflam

CTC1 mutation causing cerebro-retinal microangiopathy with calcifications and cysts type 1, masquerading as TORCH Infection.

An early adolescent male from a consanguineous family presented with progressive neurological deterioration, including developmental delay, seizures, left hemiparesis and cognitive decline from early childhood. He subsequently developed gastrointestinal bleeding, chronic liver disease with oesophageal varices, severe anaemia and thrombocytopenia. Initial investigations suggested TORCH (Toxoplasmosis, Other, Rubella, Cytomegalovirus, Herpes simplex) infection due to intracranial calcifications and multisystem involvement. However, detailed neuroimaging revealed extensive periventricular calcifi

Endogenous α-Klotho is a common target in atherosclerosis and calcific aortic valve disease.

Although atherosclerotic vascular disease (ASVD) and calcific aortic stenosis (AS) are distinct clinical entities, inflammation is a common driving force. The contribution of endogenous α-Klotho, an anti-aging protein, in the vasculature and aortic valve (AV) as an inflammatory mediator of such diseases remains unclear. We investigated α-Klotho expression using qPCR, ELISA, and immunohistochemistry in vascular (aorta, carotid, and femoral) and AV tissue samples from patients with ASVD (n = 112) and calcific AS (n = 172), respectively. We also determined circulating levels of soluble α-Klotho.

In vivo mouse model of calcific myonecrosis induced by injury.

Skeletal muscle is the major tissue that provides a space and source for bone, which is formed by osteoblasts through the secretion of extracellular matrices, such as type I collagen, and the deposition of hydroxyapatite calcium phosphate crystals. In the present study, we injected notexin, a snake venom, into skeletal muscle to induce local injury. Notexin injection caused local ectopic calcification in muscle fibers within a couple of days, which remained for more than 16 months. In contrast to ectopic bone formation, the damaged muscles after injection of notexin contains neither bone marro

Dystrophin-Deficient Muscular Dystrophy in a Jack Russell Terrier With a Large Deletion in the Canine DMD Gene.

A 6-month-old male Jack Russell Terrier presented with failure to thrive, lethargy, muscle atrophy, hypersalivation, and exercise intolerance. Similar signs were reported for the only other male puppy in the litter that died prematurely, whereas all the female puppies developed normally. An electromyography (EMG) test revealed abnormal electrical activity, such as complex repetitive discharges and fibrillation potentials. Cryosections of skeletal muscles revealed degenerative and regenerative myopathy with calcific deposits, consistent with a form of muscular dystrophy. Immunohistochemical tes

More Than Skin Deep: Diffuse Calcinosis Mimicking Malignancy in Refractory Adult-Onset Dermatomyositis With Anti-transcription Intermediary Factor 1-Gamma (Anti-TIF1-γ) Positivity.

Dermatomyositis is a systemic autoimmune condition characterized by proximal muscle weakness and characteristic cutaneous manifestations, and it carries an increased risk of malignancy. Calcinosis cutis represents a recognized complication, more commonly seen in juvenile disease but occasionally occurring in adults, where it may present with atypical or extensive involvement. We present a patient with dermatomyositis who developed extensive calcinosis with atypical clinical features that raised concern for an underlying malignancy. Hypopharyngeal Venous Malformation Mimicking Malignancy: A Case Report.

Hypopharyngeal venous malformations are rare and may mimic malignancy on imaging, creating diagnostic and management challenges, particularly when tissue diagnosis is required. A 61-year-old woman presented with progressive dysphagia and odynophagia. Contrast-enhanced computed tomography (CT) of the neck demonstrated an enhancing lesion involving the left tongue base, aryepiglottic fold, and hypopharynx, with associated coarse calcifications, raising concern for neoplasia. Flexible laryngoscopy revealed a compressible, purplish, vascular-appearing lesion. The patient underwent direct laryngosc

Senescence-related myocardial dysfunction: keeping a young heart.

The heart, a vital organ, works without interruption and constantly adjusts to the ever-changing demands on our body. It adapts to physiological and pathological changes, including exercise and emotional state, as well as metabolic, respiratory, and vascular abnormalities. The pumping action of the heart is determined by the health of the myocardium, which undergoes changes with ageing that are both under-investigated and incompletely understood, potentially impacting our approach to pathological conditions. Here, the alterations in cellular, tissue, and gross physiological function of the hea

Chronic Electronic Cigarette Exposure Promotes Atherosclerosis and Chondrogenic Modulation of Smooth Muscle Cells.

BACKGROUND: Electronic cigarette (E-cig) use has reached epidemic proportions worldwide, yet its cardiovascular consequences remain poorly defined. While several lines of evidences in human epidemiological and animal studies suggest chronic aerosol exposure accelerates atherosclerosis; the cellular and molecular mechanisms underlying this pathological remain unknown. METHODS: We exposed hyperlipidemic mice to chronic e-cigarette aerosol inhalation and characterized the plaque cellular landscape by coupling SMC lineage tracing with single-cell transcriptomic/epigenomic profiling and histologic

The potential role of sex related differences of the microbiome and its impact on calcific aortic stenosis: a narrative review.

Aortic stenosis (AS) presents with distinct sex-related differences in clinical manifestation, pathophysiology, and response to treatment. Women typically present at an older age, with greater frailty, more pronounced symptoms, and paradoxical low-flow AS, often associated with concentric left ventricular remodeling and fibrotic valve changes. In contrast, men show a predominance of calcific AS, eccentric remodeling, and more extensive aortic valve calcification. These differences are not solely anatomical or hemodynamic; they extend to molecular pathways and emerging contributors such as the
