Dermatomyositis is a rare autoimmune disease characterized by muscle weakness and distinctive skin rashes. Patients typically present with symmetric proximal muscle weakness alongside extramuscular manifestations, including interstitial lung disease and skin infections. While the exact cause remains unknown, researchers believe genetics, infections, and underlying autoimmune processes contribute to its development. The disease likely involves inappropriate complement protein activation targeting the perimysium, which triggers blood vessel inflammation and destruction around muscle fascicles. T

Orbital imaging has undergone a significant transformation over the past few decades, evolving from basic radiographic techniques to sophisticated cross-sectional imaging modalities. Historically, conventional radiography was limited to visualizing only the bony structures of the orbit or radiodense foreign bodies, making it inadequate for detecting soft tissue pathology. Advances in imaging over the last several decades-first with computed tomography (CT) in the 1970s and later with magnetic resonance imaging (MRI) in the 1980s-have revolutionized orbital diagnostics by enabling detailed visu

BACKGROUND AND AIMS: Rapacz pigs with familial hypercholesterolemia (FH pigs) fed with high-fat diet (HFD) develop early atherosclerotic lesions and complex atheromas in coronaries mimicking human coronary atherosclerotic disease (CAD). FH pigs have proven to be an excellent model for basic and pre-clinical atherosclerosis-focused research. However, unlike human atherosclerosis there has been no established classification system for porcine atherosclerosis.
METHODS: We isolated 104 plaque-containing coronary fragments from atherosclerotic FH pigs. A set of indices (features) of vessel and pla

Valvular heart disease (VHD) affects over 209 million people worldwide, with calcific aortic valve disease (CAVD) playing an increasingly dominant role. However, no effective pharmacotherapies are currently available. Existing animal or cellular models are limited by insufficient physiological relevance or lack of scalability. Here, we report for the first time engineered valvular tissues (EVTs) constructed from human induced pluripotent stem cell (hiPSC)-derived valvular interstitial cells (VICs). This model was fabricated using a three-dimension (3D) hydrogel system (fibrinogen/Matrigel/coll

Perivascular epithelioid cell tumors (PEComas) are rare mesenchymal neoplasms characterized by myomelanocytic differentiation and dichotomic molecular pathogenesis (mTOR-activating mutations versus TFE3 gene rearrangements). A recently recognized subset harboring YAP1::TFE3 gene fusion, termed inflammatory spindle cell PEComa, has been described predominantly in the lungs of adults showing significant morphologic and molecular overlap with YAP1::TFE3-fused clear cell stromal tumor of the lung (CCST-L). We report the first pediatric case of YAP1::TFE3-rearranged inflammatory spindle cell PEComa

INTRODUCTION: Stroke leaves many people with upper limb impairments which significantly affect function and Quality Of Life (QOL). Most studies report outcomes at the impairment and activity level. Less attention has been paid to real-life use of the affected upper limb in daily activities. Self-reported measures of performance of activity in daily life using the affected upper limb is important to obtain as people may have minimal deficits on impairment and activity level outcomes yet still have significant difficulties performing self-care and other Activities of Daily Living (ADLs). These d

We performed comprehensive systematic review of orbital and periorbital non-familial amyloidosis, including clinical features, radiology, histopathology, management and outcomes. We included English-language articles describing biopsy-proven orbital and periorbital amyloidosis published from inception to February, 2025. Cases without ocular histopathology and familial amyloidosis were excluded. We included 120 studies detailing 168 cases. Most were primary localized amyloidosis. The clinical presentation was variable, resulting in diagnostic uncertainty. Amyloidosis most commonly affects super

The heart functions as a coordinated four-chambered pump, with four valves ensuring unidirectional blood flow. The tricuspid valve, located between the right atrium and right ventricle, consists of anterior, posterior, and septal leaflets that coapt during systole to prevent retrograde flow. Valvular disorders may result in stenosis or regurgitation, thereby increasing cardiac workload and predisposing to chamber dilation and heart failure. Congenital abnormalities, including partial or complete leaflet absence, are rare but can significantly disrupt normal hemodynamics. Reports of absent tric

BACKGROUND: Tricuspid valve replacement is the preferred method when valve repair is not feasible. Stented xenobioprostheses, which limit the growth of the fibrous ring due to their rigid titanium frame and often have a short service life, are particularly problematic in children. An alternative approach is the implantation of a mitral valve homograft in the tricuspid position, which is more resistant to calcification. However, the limited availability of small-diameter homografts from adult donors restricts their use. To address this, a new technique for downsizing an adult mitral homograft w

BACKGROUND: Rotator cuff tears with concomitant preoperative shoulder stiffness present a clinical challenge because the rehabilitation needed to restore motion may conflict with the protection typically used to support tendon healing after rotator cuff repair. Although recent studies suggest that preoperative stiffness may not be associated with inferior long-term outcomes, whether it is associated with a different pattern of postoperative improvement and distinct inflammatory features in the torn tendon remains unclear.

Dermatomyositis is a rare autoimmune inflammatory disease with a highly heterogeneous clinical presentation and potential for multisystem involvement. Early diagnosis may be challenging, particularly in patients presenting with amyopathic disease and limited initial laboratory abnormalities. We report the case of a 45-year-old woman initially diagnosed with amyopathic dermatomyositis based on characteristic cutaneous manifestations in the absence of objective muscle involvement on laboratory testing and electromyography. After being lost to follow-up for four years, she presented with progress

Omics technologies enable deep profiling of human atherosclerosis but have mostly been applied in small studies lacking integration with other data modalities. AtherOMICS is a biobanking project linking multiomic atherosclerotic plaque phenotyping with blood biobanking, in vivo imaging, and clinical data. Here, we present the study protocol and show pilot results that demonstrate technical feasibility. Since August 2022, 246 patients scheduled to undergo carotid or femoral endarterectomy at Ludwig-Maximilians-Universität Klinikum (Munich, Germany), have been enrolled (median age, 73 years; 32%

Transcription factors from the family of Signal Transducers and Activators of Transcription (STAT) are activated by numerous cytokines. Two members of this family, STAT5A and STAT5B (collectively called STAT5), have gained prominence in that they are activated by a wide variety of cytokines such as interleukins, erythropoietin, growth hormone, and prolactin. Furthermore, constitutive STAT5 activation is observed in the majority of leukemias and many solid tumors. Inactivation studies in mice as well as human mutations have provided insight into many of STAT5's functions. Disruption of cytokine

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l

BACKGROUND: Mobility impairment is a common consequence of multimorbidity in older adults, especially among those with diabetes mellitus and hypertension. Although robotic exoskeletons have shown promising results in neurological rehabilitation, evidence regarding their use in non-neurological multimorbid populations remains scarce.
OBJECTIVE: To evaluate the feasibility and effectiveness of Keeogo exoskeleton-assisted rehabilitation, in combination with conventional therapy, on functional mobility in older adults with multimorbidity.

Metabolic syndrome (MetS) is associated with osteogenic transdifferentiation of vascular smooth muscle cells (VSMCs) and accumulation of arterial calcifications (ACs). Metformin (MET) inhibits this transdifferentiation in vitro. Here, we evaluate the in vivo efficacy of oral MET to reduce AC in a model of MetS. Twenty young male Wistar rats were divided into two groups: one received water and the other received water plus 20% fructose to induce MetS. After 14 days, and for another 4 weeks, MET (100 mg/kg per day) was added to half of each group's drinking source, thus C (water), F (fructose),

Ectopic calcifications occur in tendons, ligaments, entheses, muscles, and fasciae, and are often associated with pain and inflammation. In clinical settings, these calcifications are commonly treated by physical therapy and/or surgical interventions. However, there is not enough understanding of pharmacological treatments as primary cures, supportive therapy to physical or surgical treatment, or even preventive measures to avoid or diminish the development of ectopic calcifications. Here, we summarize preclinical and clinical evidence for pharmacological candidates for treatment/prevention of

Radiomics, the extraction of quantitative features from medical images, has shown great promise in enhancing diagnostic and prognostic models, particularly in CT and MRI. However, its application in ultrasound (US) imaging, especially in musculoskeletal (MSK) imaging, remains underexplored. The inherent variability of ultrasound, influenced by operator dependency and various imaging settings, presents significant challenges to the reproducibility of radiomic features. This study aims to identify whether commonly used image pre-processing methods can increase the reproducibility of radiomics fe

Substrate mapping is an important component of electrophysiological (EP) study for the treatment of reentrant ventricular tachycardia (VT). It is used to detect characteristics of the electrical circuit and, in particular, the location and properties of the central common pathway, aka the isthmus, where multiple circuit loops can coincide. Typically, reentrant circuits are single or double loop, but as the common pathway size increases, 4-loop patterns may emerge, consisting of 2 parallel isthmuses or a single isthmus with 4 loops. Arrhythmogenic substrate contains a mixture of scar, calcifica
