BACKGROUND: Long-term exercise is recognized as one of the most effective means of maintaining health after aging, but the relationship between moderate exercise and health in the older population is often overlooked.
OBJECTIVE: The present study was conducted to investigate the effects of excessive endurance exercise on the old mice musculoskeletal system.
RESULTS: The 8 weeks of normal endurance exercise significantly improved skeletal muscle mitochondrial biogenesis and increased femoral osteogenesis in young and old mice. However, the continued accumulation of total exercise volume as th

Accessory or supernumerary bones are bones that are not normally present in the body but can be found as an anatomical variant. While hallucal sesamoids are common, lesser metatarsal sesamoids are uncommon and often underreported. These ossicles develop through endochondral ossification and aid biomechanics by enhancing muscle function and reducing tendon friction. This report presents the first documented case of bilateral multiple lesser metatarsal sesamoids involving several metatarsophalangeal joints, supported by radiologic and clinical correlation. A 48-year-old woman presented with bila

BACKGROUND: Fibrodysplasia ossificans progressiva (FOP) is an autosomal dominant ultra-rare disease characterized by progressive episodic multi-focal heterotopic ossification of skeletal muscle, ligaments, tendons, and fascia. Comprehensive characterization and understanding of the natural disease history of FOP, including mortality, comorbidities, and medication use, is currently limited.
OBJECTIVE: This systematic review, which we believe is the first of its kind, aims to synthesize current knowledge on the morbidity, mortality, and medication use associated with FOP.
METHODS: A systematic

INTRODUCTION: Many of the shoulder pathologies present with pain and stiffness; the principal diagnosis of this condition includes calcific tendonitis or rotator cuff tears. Among these, myositis ossificans is very rare.
CASE REPORT: A 22-year-old female presented to us with a complaint of pain in the shoulder for 6 months. On clinical examination, external rotation and internal rotation were restricted. On evaluation, X-ray was normal, while magnetic resonance imaging and computed tomography scan showed 1.5 cm × 1 cm lesion in the subscapularis extending in the shoulder joint. First an ultra

Myositis ossificans (MO) is a self-limiting benign ossification disorder, characterized by heterotopic ossification within skeletal muscles. Pediatric MO cases are rarer, easy to be misdiagnosed. The diagnosis of traumatic MO is based on a thorough history, physical examination, and imaging studies. Early and proactive conservative treatment can often be effective. We report a case of a 6-year-old patient with a femoral fracture combined with severe MO. He was misdiagnosed at a community hospital as having a bone tumor combined with a pathological femoral fracture and was referred to our hospi

Myositis ossificans (MO) is a rare benign condition characterized by heterotopic bone formation within soft tissues, with masticatory muscle involvement being exceptionally uncommon. This case report describes a 45-year-old male patient who developed progressive trismus and a preauricular mass six months after right frontotemporal craniotomy for meningioma resection. Imaging revealed an ossified lesion in the temporalis muscle, consistent with post-traumatic MO. Surgical management involved zygomatic arch osteotomy and coronoidectomy, restoring intraoperative mouth opening to 23 mm. Histopatho

BACKGROUND: The intact tendon-bone interface (TBI) consists of four histological layers-tendon, fibrocartilage, calcified fibrocartilage, and bone-that gradually merge into each other, making complete structural restoration after injury challenging. Osteoporosis poses a significant risk for rotator cuff tears (RCTs) and re-tears after arthroscopic rotator cuff repair (ARCR). Activating Leptin receptor (Lepr) mediated Stat3 signaling transduction facilitates the transcription of Runx2 and Sox9, respectively, and promotes osteogenesis and chondrogenesis.

Genome-wide association studies have identified multiple loci associated with bone mineral density, a major determinant of osteoporotic fracture risk. At one such locus, genetic, bioinformatic, and zebrafish knockout data strongly prioritize membrane palmitoylated protein 7 (MPP7) as a candidate gene, although its precise role in bone biology remains poorly defined. MPP7 encodes a member of the p55 Stardust family of membrane-associated guanylate kinase proteins, which are key regulators of epithelial cell polarity and junctional organization. Here, we investigated the functional role of MPP7

BACKGROUND: Vascular calcification is an active process driven by osteogenic phenotypic transition of vascular smooth muscle cells (VSMCs) and regulated by a bone-related gene regulatory network. Recent studies showed that FAK (focal adhesion kinase) regulates bone formation by affecting the cellular localization of HDAC (histone deacetylase) 4 and HDAC5. However, it is not known whether FAK exerts effects on vascular calcification in VSMCs through regulating HDACs.
METHODS: We used perturbational assays to assess the role of HDAC4, HDAC5, and FAK in VSMC calcification. Pharmacological inhibi

The reconstruction of large segmental bone defects remains a formidable challenge in orthopedic surgery. Benefiting from the rapid advancement of three-dimensional (3D) printing technology, growth factor-loaded artificial bone scaffolds have been extensively studied and have emerged as one of the pivotal strategies for bone defect repair. However, the development of an ideal scaffold system that meets comprehensive clinical requirements still requires sustained efforts. This study designed a biomimetic bone repair scaffold with bioactive properties using low-temperature 3D bioprinting technolo

Gnathodiaphyseal Dysplasia is a rare autosomal-dominant disease caused by a mutation in GDD1. Patients typically present with pathological fractures, muscle weakness, and jawbone deformities. This genetic disorder has long been misdiagnosed until 2004 when the genetic basis of this disorder was identified, differentiating it from Osteogenesis Imperfecta and Fibrous Dysplasia. However, clinicians are still largely unaware of this entity and continue to label and treat it as a variation of Osteogenesis Imperfecta. This report describes a 21-year-old male with GDD presenting with numerous fractur

Introduction Swelling in the short external rotator muscles (SERMs) in patients with cerebrovascular disease may regress to non-traumatic myositis ossificans. This study investigates the temporal changes in CT findings of myositis ossificans, aiming to provide exploratory insights for diagnostic accuracy and clinical management. Methods We retrospectively reviewed 1,469 CT examinations of the pelvis conducted at our institution between 2001 and 2023, totaling 436 cases of cerebrovascular disease. We selected 20 patients showing swelling of SERMs, including six patients with bilateral swelling.

B-cell lymphoma 9 (BCL9) is a regulatory protein that plays a key role within the Wnt/β-catenin signaling pathway. In recent years, it has garnered significant attention across the domains of tissue repair and tumour biology. The Wnt/β-catenin signaling pathway is of great importance in regulating bone tissue formation and osteoblast differentiation. As a co-activator of β-catenin, BCL9 is capable of governing bone development and regeneration by augmenting the transcriptional activity of the Wnt signaling. In addition, BCL9 is also closely related to diverse biological processes, such as angi

The comparative and functional anatomy of the cleaver-like jaws of the Late Devonian arthrodire Dunkleosteus terrelli remains poorly understood, despite significant advances in our understanding of arthrodire anatomy. Here, we re-investigate the jaw morphology of D. terrelli from a comparative anatomical perspective. Rare cartilaginous elements, preserved attachment sites, and comparisons with other arthrodires suggest the skull of Dunkleosteus contained significantly more cartilage than previously assumed, with many major jaw muscle attachments and cranial articulations located on cartilages.

BACKGROUND: The associations between spine-specific muscle sarcopenia and clinical outcomes after cervical and lumbar spinal surgery have been investigated. However, no study has investigated the associations between cervical multifidus sarcopenia (CMS) and outcomes after laminoplasty for cervical ossification of the posterior longitudinal ligament (OPLL). The aim of the present study was to investigate whether CMS affects cervical sagittal alignment (CSA) and patient-reported outcomes (PROs) in patients with cervical OPLL who underwent laminoplasty.

Knee osteoarthritis (KOA) is a prevalent degenerative joint disease characterized by pain, dysfunction, and stiffness, significantly impairing quality of life. While various interventions exist, aerobic exercise stands out as a safe and effective core treatment. This review synthesizes current evidence on the therapeutic benefits and underlying mechanisms of aerobic exercise for KOA. We recommend low-to-moderate intensity aerobic training (RPE 11-14) for KOA patients, performed 3-4 times per week for 30-60 min, for at least 6 weeks. Recommended modalities include gentle exercises like Wuqinxi,

BACKGROUND CONTEXT: Segmental motor paralysis is a well-recognized complication following anterior cervical spine surgery. While risk factors have been identified, little is known about the factors that influence recovery outcomes, and there has been no systematic analysis of the relationship between onset timing and recovery patterns.
PURPOSE: This study aimed to investigate the clinical course of segmental motor paralysis after anterior cervical spine surgery, identify risk factors for persistent paralysis, and determine the relationship between paralysis onset timing and recovery patterns.

Multinucleated osteoclasts, generated by fusion of mononucleated precursors, play an essential role in the lifelong remodeling of our bones. Since within the physiological range of osteoclast sizes, their bone-resorbing activity grows with successive fusion events, both initiation of this fusion reaction and its switch off are tightly controlled. In this review, we discuss the mechanisms and proteins that facilitate and regulate this fusion process. The pathway of membrane rearrangements in osteoclast fusion shares many mechanistic motifs with other physiologically important cell-cell fusion p

OBJECTIVE: Cervical spinal stenosis predominantly affects the elderly. After 40 years of age, aging induces progressive loss and rupture of elastic fibers in the ligamentum flavum, accompanied by abnormal proliferation and cross-linking of collagen fibers, as well as calcium salt deposition and even ossification. This study investigates the clinical efficacy and safety of large-channel endoscopic unilateral laminotomy decompression for the treatment of multilevel cervical spinal canal stenosis.
METHODS: A retrospective study was conducted on 36 Cervical spinal canal stenosis patients with rad

Osteoporosis (OP) is a major public health challenge affecting over 200 million individuals worldwide. While pharmacological interventions represent mainstream treatment, concerns regarding long-term side effects and patient compliance have driven the exploration of alternative therapies. Exercise interventions have emerged as promising complementary approaches due to their excellent safety profiles, cost-effectiveness, and high adherence rates. Diverse exercise modalities, including aerobic, resistance, and balance exercises, effectively modulate bone homeostasis by promoting osteogenesis, in
