Myositis Ossificans of the Masseter Muscle: A Rare Post-traumatic Case.

Post-traumatic myositis ossificans of the masseter is an uncommon cause of severe trismus without a standardized management pathway. A 23-year-old man developed progressive jaw stiffness six months after facial trauma, with maximal interincisal opening <10 mm. Imaging demonstrated mature heterotopic ossification bridging the mandibular ramus and zygomatic arch. Excision of the ossified masseter was performed via a combined intraoral and external approach. Trismus recurred within three months, and re-operation through the prior incision removed residual ossification with coronoidectomy. No adju

Does Lateral Pterygoid Muscle Reattach After Ramus-Condyle Unit Reconstruction in Temporomandibular Joint Ankylosis Patients?

BACKGROUND: The removal of ankylotic bone requires detachment of lateral pterygoid muscle (LPM) from the ankylosed condyle. Whether spontaneous reattachment of LPM occurs or not following autogenous ramus-condyle unit (RCU) reconstruction in temporomandibular joint ankylosis (TMJA) is not yet known. PURPOSE: The study purpose was to measure and compare the incidence of spontaneous LPM reattachment following 2 RCU reconstruction techniques-costochondral graft (CCG) and transport disc distraction osteogenesis (TDDO) and its implications on postoperative jaw motion. Matrix-directed therapy losartan to identify the effect on the bone resorption marker carboxy-terminal crosslink of type I collagen telopeptide (CTX) in older adolescents and adults with osteogenesis imperfecta recruited from secondary care sites: the 'MOI-A' study; a randomised, phase 2/pilot, dose-escalating trial.

INTRODUCTION: Osteogenesis imperfecta (OI) is the most common inherited cause of bone fragility (approximately 1 in 16 000). People with OI suffer bone fragility causing fractures, pain and deformity; sarcopenia causing fatigue and poor endurance; aortic root dilatation and hearing loss. No drug currently has market authorisation to treat OI in Europe. Current standard-of-care is multidisciplinary, with pharmacological interventions-primarily bisphosphonates-directed at increasing bone mass; however, such interventions are of equivocal efficacy. The structural damage that can accumulate as a r

Carcass weight, meat quality and economic impact of liver fluke infection on cattle.

Liver fluke infection in cattle is typically sub-clinical, with animals lacking in visible clinical signs. Consequently, farmers are often dubious about investing in control measures as they are often unaware of its deleterious effects on meat quality and weight gain. The aim of this study was to quantify the effect of liver fluke infection on carcass weight and meat quality in Australian cattle, and, in turn, to estimate the economic loss to the producer. This study utilised abattoir data obtained from an Australian meat processor. The data encompassed observations of over 4 million cattle pr

Malignant metastatising solitary fibrous tumor of the ovary with additional dedifferentiation and osteoid deposition: an unusual presentation, with a brief description of a diagnostic alghoritm.

Solitary fibrous tumor (SFT) is a rare spindle cell neoplasm showing fibroblastic differentiation, initially observed in the pleura, but now currently recognized to develop in any extrapleuric location 1,2. In the female genital tract, SFTs are extremely rare and have a predilection for the vulva, vagina and cervix 2,3. There are very few cases of ovarian SFTs having been reported in the literature. Malignant SFTs of the ovary are exceedingly rare neoplasms characterized by their mesenchymal origin and distinctive histopathological features. First identified as a separate entity in soft tissue

Lactylation's role in bone health and disease: mechanistic insights and therapeutic potential.

This article reviews the mechanisms and research progress of lactylation, an emerging post-translational modification (PTM) of proteins, in bone metabolism and related diseases. Lactate-derived lactylation modifies lysine residues on histones, affecting chromatin structure and gene expression. Studies indicate that lactylation plays a significant role in bone metabolism, with mechanisms including the regulation of osteoblast differentiation, potential influence on osteoclast activity, and indirect effects on bone homeostasis through the modulation of immune cell functions such as macrophages a

Alpha-smooth muscle actin-expressing dermal sheath cells are a major cellular contributor to heterotopic subcutaneous ossifications in a mouse model of Albright hereditary osteodystrophy.

Heterotopic ossifications (HOs) are the pathologic process by which bone inappropriately forms outside of the skeletal system. Despite HOs being a persistent clinical problem in the general population, there are no definitive strategies for their prevention and treatment due to a limited understanding of the cellular and molecular mechanisms contributing to lesion development. One disease in which the development of heterotopic subcutaneous ossifications (SCOs) leads to morbidity is Albright hereditary osteodystrophy (AHO). Albright hereditary osteodystrophy is caused by heterozygous inactivat

Rapidly Resolving Myositis Ossificans in an Adolescent Female.

Myositis ossificans (MO) is a rare benign tumor characterized by heterotopic ossification of soft tissues. While the term "myositis" suggests that the lesion commonly affects muscles, it can also occur in rare locations such as tendons or subcutaneous tissues. MO is infrequent in children, primarily affecting young adults and adolescents. It presents distinct radiological and histological features. However, it is crucial to exclude malignancy when investigating heterotopic ossification. Surgical excision of the lesion following an appropriate biopsy is an option, but cases of self-limiting MO

Bone-fat linkage via interleukin-11 in response to mechanical loading.

Positive regulators of bone formation, such as mechanical loading and PTH, stimulate and negative regulators, such as aging and glucocorticoid excess, suppress IL-11 gene transcription in osteoblastic cells. Signal transduction from mechanical loading and PTH stimulation involves two pathways: one is Ca2+-ERK-CREB pathway which facilitates binding of ∆FosB/JunD to the AP-1 site to enhance IL-11 gene transcription, and the other is Smad1/5 phosphorylation that promotes IL-11 gene transcription via SBE binding and complex formation with ∆FosB/JunD. The increased IL-11 suppresses Sost expression

Cutaneous burn injury represents a major risk factor for the development of traumatic ectopic bone formation following blast-related extremity injury.

Blast-related traumatic heterotopic ossification (tHO) impacts clinical outcomes in combat-injured patients, leading to delayed wound healing, inflammatory complications, and reduced quality of life. Blast injured patients often have significant burns. This study investigated whether a partial thickness thermal burn injury exacerbates blast-related tHO in a clinically relevant polytrauma animal model. Adult male Sprague Dawley rats were subjected to an established model involving a whole-body blast overpressure exposure (BOP), complex extremity trauma followed by hind limb amputation (CET) fol

Novel Sheep Model to Assess Critical-Sized Bone Regeneration with Periosteum for In Vivo Bioreactors.

Considerable research is being undertaken to develop novel biomaterials-based approaches for surgical reconstruction of bone defects. This extends to three-dimensional (3D) printed materials that provide stable, structural, and functional support in vivo. However, few preclinical models can simulate in vivo human biological conditions for clinically relevant testing. In this study we describe a novel ovine model that allows evaluation of in vivo osteogenesis via contact with bone and/or periosteum interfaced with printed polymer bioreactors loaded with biomaterial bone substitutes. The HIF-1α and mTOR Pathways Amplify Heterotopic Ossification.

Fibrodysplasia ossificans progressiva (FOP; MIM# 135100) is an ultra-rare congenital disorder caused by gain-of-function point mutations in the Activin receptor A type I (ACVR1, also known as ALK2) gene. FOP is characterized by episodic heterotopic ossification (HO) in skeletal muscles, tendons, ligaments, or other soft tissues that progressively causes irreversible loss of mobility. FOP mutations cause mild ligand-independent constitutive activation as well as ligand-dependent bone morphogenetic protein (BMP) pathway hypersensitivity of mutant ACVR1. BMP signaling is also a key pathway for me

SiONx Coating Regulates Mesenchymal Stem Cell Antioxidant Capacity via Nuclear Erythroid Factor 2 Activity under Toxic Oxidative Stress Conditions.

Healing in compromised and complicated bone defects is often prolonged and delayed due to the lack of bioactivity of the fixation device, secondary infections, and associated oxidative stress. Here, we propose amorphous silicon oxynitride (SiONx) as a coating for the fixation devices to improve both bioactivity and bacteriostatic activity and reduce oxidative stress. We aimed to study the effect of increasing the N/O ratio in the SiONx to fine-tune the cellular activity and the antioxidant effect via the NRF2 pathway under oxidative stress conditions. The in vitro studies involved using human

Trauma patient heterotopic ossification diagnosis is associated with increased hospital length of stay.

BACKGROUND: Traumatic heterotopic ossification (tHO) refers to the development of extra-skeletal bone in muscle and soft tissues following tissue insult secondary to surgery or trauma. This presents a persistent clinical concern associated with significant patient morbidity and expense to diagnose and treat. Traumatic HO is a substantial barrier to rehabilitation for trauma-injured patients. As such, the development of tHO after burn and other trauma is hypothesised to prolong inpatient length of stay (LOS) and thus increase health care costs. OBJECTIVE: To investigate the association between

Matrix metalloproteinase-9 deficiency confers resilience in fibrodysplasia ossificans progressiva in a man and mice.

Single case studies of extraordinary disease resilience may provide therapeutic insight into conditions for which no definitive treatments exist. An otherwise healthy 35-year-old man (patient-R) with the canonical pathogenic ACVR1R206H variant and the classic congenital great toe malformation of fibrodysplasia ossificans progressiva (FOP) had extreme paucity of post-natal heterotopic ossification (HO) and nearly normal mobility. We hypothesized that patient-R lacked a sufficient post-natal inflammatory trigger for HO. A plasma biomarker survey revealed a reduction in total matrix metalloprotei
