A model for development and evolution of complex morphological structures.

How 'complex' or composite morphological structures like the mammalian craniomandibular region arise during development and how they are altered during evolution are two major unresolved questions in biology. Herein, we have described a model for the development and evolution of complex morphological structures. The model assumes that natural selection acts upon an array of phenotypes generated by variation in a variety of underlying genetic and epigenetic controlling factors. Selection refines the integration of the various morphogenetic components during ontogeny in order to produce a functi

Ossifying fibromyxoid tumor of soft parts.

A histologically uncommon soft-tissue tumor of the extremities and neck of a 54-year-old male is reported. The solid, bony-hard tumors occurred at the inner region of the right thigh at 44 years of age with additional tumor formation at the posterior region of the same thigh, at the inner region of the right upper arm and at the neck during the following 10 years. All tumors were located in the deep muscle layer. The neck tumor directly invaded the fifth cervical vertebra and later the upper mediastinum. Histologically, all three tumors of the extremities contained mixed lobular growths of rou

Clonal osteogenic cell lines express myogenic and adipocytic developmental potential.

Clonal osteoblastic cell lines were isolated from neonatal rat calvariae and characterized with regard to a number of features associated with authentic osteoblasts. These included elevated alkaline phosphatase activity (relative to fibroblasts), PTH and PGE2-stimulated increases in cAMP, the predominant synthesis of type 1 collagen, and the production of a mineralized matrix in vitro. By these criteria, five clones with osteoblast-like phenotypes were identified (ROB-C8a, C11, C20, C23, and C26) which varied somewhat in shape, levels of alkaline phosphatase activity, and in responsiveness to

Noninflammatory knee joint effusions in spinal cord-injured and other paralyzed patients. Four case studies.

Four patients with knee effusions are described. Three had spinal cord injuries and the fourth had Guillain-Barrè syndrome. Of the spinal cord-injured patients, the effusions were unilateral in one case and bilateral in another. The third patient developed effusions on two separate occasions with bilateral involvement once and unilateral involvement once. The patient with Guillain-Barrè syndrome had bilateral knee joint effusions. All effusions were tapped and revealed type I synovial fluid except for one joint that contained some blood cells. Two of the three spinal cord-injured patients had

Stapedius tendon ossification: a rare cause of congenital conductive hearing loss.

Two cases are presented of bilateral conductive deafness in siblings. An initial presumptive diagnosis of otosclerosis was made in the first case, that of a nine-year-old girl with a bilateral hearing loss of 45 dB. Exploratory tympanotomy revealed normal ossicles but the stapedius 'tendon' was found to be a solid bony bar fixing the stapes. Division of the 'tendon' resulted in normal ossicular mobility and return of hearing with closure of the air-bone gap. Identical ossification was found in her other ear, and some years later in both ears of one of her brothers. Similar surgery restored hea

The urohyal: Development and homology within osteichthyans.

The formation of the unpaired structure ventral to the basibranchial region, the so-called urohyal, differs within osteichthyans. A cartilaginous preformed, unpaired "urohyal" is present in sarcopterygians. A three-tendon ossification is present in Polypterus. An "urohyal" or urohyal is absent in both Amia and Lepisosteus. The urohyal formed as an unpaired ossification of the tendon of the sternohyoideus muscle is a feature of teleosts. A new structure, the parurohyal, arises as a double ossification of the tendon of the sternohyoideus muscle in siluroids; during ontogeny an anterodorsal crest

Distribution of insulin-like growth factor peptides in the developing chick embryo.

Growth factors are likely to be of major significance in developmental biology. Here, the distribution of insulin-like growth factor (IGF) peptides is described during development of the chick embryo. IGF was immunolocalised using a polyclonal antibody to human IGF I detected with a modified Vectastain ABC procedure. Under the conditions used, the antibody binds strongly to IGF I and weakly to IGF II; thus the distribution of IGF peptide, rather than the individual factors, is described. Muscle, peripheral nerve and the notochord were labelled whenever present. Muscle label was associated with

Investigation of a hydroxyapatite and calcium sulfate composite supplemented with an osteoinductive factor.

In the search for a suitable bone graft substitute, a study was conducted using a material that combined a proven osteoconductive composite, hydroxyapatite-calcium sulfate (HA/CS), with an osteoinductive factor, bovine osteogenic factor (OF). The initial study demonstrated the osteoinductive potential of OF in the rabbit muscle model. Once satisfied that the OF was active, it was added to the HA/CS composite and placed in 8-mm trephine defects in the rabbit cranium. This HA/CS/OF was directly compared to HA/CS augmented with a control protein, rabbit serum albumin (RSA). Animals were sacrifice

[Ossification after transplantation of model cartilage in the rat patella].

The cartilage model of the rat's patella constitutes a formation susceptible of inducing ossification, following its transplantation in a variety of sites (muscle, thyroïd, testis, ovary, anterior chamber of the eye, etc.), including those considered inappropriate in other experiments of induced ossification (liver, kidney). So we have an experimental model allowing the influence on the osteogenesis of various factors, local or general, natural or experimental, to be studied. Ossification occurs in this model only in cases of histocompatibility between donor and recipient. It is constant after

Immunohistochemical studies using BRL 12, a monoclonal antibody reacting specifically with osteogenic tissues.

A monoclonal antibody of immunoglobulin class G1 has been produced which reacts with a high molecular weight antigen apparently present exclusively in osteogenic tissues. Immunohistochemical studies have shown that the antigen is present throughout the mineralized matrix and in osteoid. None of the other tissues examined namely liver, intestine, kidney, spleen, thymus, heart, lung, skin, cartilage and skeletal muscle showed evidence of specific antibody binding. Immunohistochemical staining was also demonstrated in tissues developing from rabbit marrow cultured in vitro and in diffusion chambe

Mechanism of the reduction of Meckel's cartilage in man.

The mechanism of reduction of the anterior end of Meckel's cartilage was studied in human embryos, with the following findings: 1. Meckel's cartilage is surrounded, from the outside and from below, by newly formed mandibular bone over the extent of the insertion of the musculus mylohyoideus. 2. Blood vessels from the newly formed bone penetrate Meckel's cartilage and break it down in the same way as in enchondral ossification of cartilaginous models of other bones. 3. The anlagen of the musculus mylohyoideus and musculus genioglossus are at first inserted on Meckel's cartilage; further muscle

The tension-stress effect on the genesis and growth of tissues: Part II. The influence of the rate and frequency of distraction.

To assess the influence of both the rate and the frequency of distraction on osteogenesis during limb elongation, a canine tibia was used with various combinations of distraction rates (0.5 mm, 1.0 mm, or 2.0 mm per day) and distraction frequencies (one step per day, four steps per day, 60 steps per day). The distractions were performed after both open osteotomy and closed osteoclasis. Histomorphic and biochemical studies were conducted on the elongated osseous tissue, fascia, skeletal muscle, smooth muscle, blood vessels, nerves, and skin. It was determined that distraction at a rate of 0.5 m

Bone formation following intrarenal transplantation of isolated murine chondrocytes: chondrocyte-bone cell transdifferentiation?

Isolated syngeneic epiphyseal chondrocytes transplanted into a muscle formed cartilage in which matrix resorption and endochondral ossification began at the end of the second week after transplantation. After 56 days cartilage was converted into an ossicle. In 7-day-old intrarenal transplants, epiphyseal chondrocytes formed nodules of cartilage. In 10-day-old transplants, islands of bone appeared. Slight resorption of cartilage was first noted in 14-day-old transplants of chondrocytes. After eight weeks, transplants contained mainly bone. Intramuscularly transplanted rib chondrocytes formed ca

The enthesopathic changes of hypophosphatemic osteomalacia in adults: radiologic findings.

The enthesopathy (bone proliferation at sites of ligament, tendon, joint capsule, and interosseous membrane attachment) that may be present in adults who have longstanding hypophosphatemic osteomalacia, the most common type of osteomalacia, has received little attention in the radiologic literature. These proliferative enthetic changes may be associated with various musculoskeletal symptoms. The spectrum of proliferative enthesopathic changes, involving the axial and appendicular skeleton, is described in six patients 18-63 years old. Proliferative changes at the attachment of the annulus fibr

Immunohistochemical localization of the matrix glycoprotein tenascin in the skull of the growing rat.

Tenascin is an extracellular matrix glycoprotein which interacts with other matrix molecules and with cells, and which appears to play important roles in growth and differentiation. The immunohistochemistry of sections of newborn, 5-day and 20-day-old rats showed that accumulation of tenascin was largely restricted to bones, cartilages and teeth. It was also present in the periosteal and endosteal surfaces of membrane bones, in perichondrium, and ion the dental pulp, but was absent from mature bone, cartilage and dentine. In nasal cartilage, tenascin was present only in the perichondrium, wher

Cardiovascular pathology in osteogenesis imperfecta type IIA with a review of the literature.

Lethal perinatal osteogenesis imperfecta (OI Type II) is a biochemically diverse collagen disorder characterized by short, crumpled long bones, beaded ribs, blue sclerae and thin, fragile skin. Cardiovascular abnormalities are rarely described. Using morphometry and light and electron (SEM and TEM) microscopy, we analyzed the hearts and great vessels from 2 fetuses with OI Type IIA and compared the findings with age-matched controls. The heart weights and atrioventricular valve (AVV) circumferences were reduced in OI. The chordae tendineae were short and fragile; both the AVVs and the chordae

Anatomical studies in the hyoid region of the mouse in term of evolutional importance.

The mouse hyoid region was studied in detail in term of its evolutional importance. A pair of small pyramidal-shaped "epiphyseal cornua" wedged between the body and the caudal cornua. It is suggested that the epiphyseal cornu is an independent element of the hyoid bone, and that it is not homologous with the typical hypohyal in other mammals. Just ventral of the tympanic bulla, there was a stylohyal, on which the "paroccipito-styloideus" and "paroccipitostylohyoideus" muscles (appellations by the authors) were attached. Ossification of the hyoid bone began at the attachments of the hyoid muscl

Bovine bone morphogenetic protein (bBMP) induced repair of skull trephine defects in sheep.

An aggregate of partially purified bovine bone morphogenetic protein (bBMP) and bone matrix insoluble noncollagenous proteins (iNCP), weighing a total of 100 mg of lyophilized BMP/iNCP, was implanted using ultra thin gelatin capsules in skull trephine defects in adult sheep. One hundred milligram samples of freeze-dried bovine serum albumin (BSA) were similarly implanted for controls. In five sheep, the capsules were implanted in 18-20 mm trephine skull defects and also in posterior cervical muscle pouches for heterotopic controls. In two out of five sheep, the trephines were repaired with bon

Pattern of the insulin-like growth factor II gene expression during rat embryogenesis.

The rat insulin-like growth factor II (IGF-II) gene, encoding a fetal somatomedin, expresses a family of transcripts in embryonic/fetal tissues, and also in the adult choroid plexus and the leptomeninges. We have localized IGF-II gene transcripts in sections of rat embryos of embryonic days 10-16 by performing in situ hybridization. These transcripts are present in the head mesenchyme, formed from both the mesoderm and the cephalic portion of the neural crest, and also in the majority of other tissues of mesodermal origin, predominantly those derived from the somites and the lateral mesoderm.

Quantitative computation of induced heterotopic bone formation by an image analysis system.

The formation of heterotopic bone (HO) is an unsolved problem and occurs in response to a variety of conditions including muscle contusions, injuries, surgical procedures, and burns. A sensitive method for quantification of HO would be valuable for experimental work and research on bone morphogenetic protein (BMP). The hardware and software used in an image analysis system are presented. This system provides a rapid and highly accurate method for determining the amount of HO in a muscle. Using the mouse thigh pouch as an experimental system, this method demonstrated that quantities of new bone
