Developing Metal-Polyphenol Network-Based Single-Protein Particle Encapsulation System for Sustained Release of Whey Protein to Mitigate Sarcopenia.

Sarcopenia has emerged as a major health challenge in the aging population. Whey protein (WP)-rich in branched-chain amino acids-is an important nutritional supplement for mitigating sarcopenia. However, the rapid digestion of WP limits its therapeutic efficacy. Developing a sustained-release system for WP may enhance its effectiveness in mitigating sarcopenia. Herein, a novel "single-protein particle encapsulation" (SPPE) system is developed to protect WP and control its release. Specifically, the "molecular glue" metal-polyphenol networks (MPNs) are assembled onto the surface of individual p

Tonic-dominant sleep bruxism in multiple system atrophy: A polysomnographic comparison with Parkinsonian-related disorders.

Sleep bruxism (SB), classified into phasic, tonic, and mixed electromyographic (EMG) patterns, is increasingly recognized as a heterogeneous phenomenon in neurodegenerative disorders. However, its specific characteristics in multiple system atrophy (MSA) relative to Parkinsonian-related disorders (PRD) remain unclear. We retrospectively analyzed overnight polysomnography (PSG) with bilateral masseter EMG in patients with MSA (n = 8) and PRD (n = 7). SB episodes were classified as phasic, tonic, or mixed according to standard criteria. Sleep architecture and bruxism parameters were assessed, an

A metabolic-epigenetic switch governs multicellular cardiac repair following succinate dehydrogenase inhibition.

Cardiac regeneration is limited in the adult mammalian heart because of restricted cardiomyocyte proliferation and persistent fibrosis. We previously demonstrated that transient pharmacological inhibition of succinate dehydrogenase (SDH) with malonate promotes regeneration after myocardial infarction. Here, we integrate single-nucleus RNA sequencing and ATAC-seq to reveal multicellular transcriptional and epigenetic reprogramming underlying this response. Cell-specific Sdhb deletion dissected contributions of SDH inhibition; cardiomyocyte-specific Sdhb deletion transiently increased cardiomyoc

Effects of an eight-week combined exercise program on functional and sarcopenia-related outcomes in institutionalized older adults with and without Alzheimer's disease: a comparative intervention study.

BACKGROUND: Alzheimer's disease (AD) is a major cause of disability in older adults and is frequently accompanied by sarcopenia and functional decline, contributing to loss of independence. To investigate the effects of an eight-week combined exercise program on physical and cognitive function, sarcopenia-related outcomes and nutritional status in institutionalized older adults with and without AD. METHODS: Twenty-three participants aged ≥ 65 years (12 with moderate AD and 11 cognitively healthy controls) completed supervised combined aerobic and resistance exercise three times per week for e

Skeletal muscle histological remodeling precedes functional impairment in a muscle-specific manner following monocrotaline-induced heart failure in rats.

PURPOSE: Skeletal muscle remodeling in heart failure contributes to exercise intolerance and muscle weakness, but the temporal relationship between histological alterations and functional impairment remains poorly understood. We aimed to clarify whether histological changes in skeletal muscle precede the onset of functional impairment. METHODS: Rats were allocated into either a normal control group (n = 21) or a monocrotaline-induced heart failure group (n = 42) and evaluated at 1, 2, or 4 weeks after injection. We assessed motor function (grip strength and exhaustive running), organ weights,

Prevalence and Overlap of Malnutrition, Sarcopenia and Related Nutritional Syndromes in a Large Haemodialysis Population.

BACKGROUND: Chronic kidney disease is a major global health burden linked to malnutrition, sarcopenia and related nutritional syndromes including protein-energy wasting (PEW)/cachexia and sarcopenic obesity (SO). Data from end-stage kidney disease (ESKD) populations in Saudi Arabia and the wider Middle East are scarce. This study assessed the prevalence and factors associated with malnutrition and sarcopenia risk in a large haemodialysis (HD) cohort and the co-occurrence of PEW/cachexia risk and SO. METHODS: Multicentre, cross-sectional study in 40 dialysis clinics across Saudi Arabia (June-A

Inverse correlation between plasma 3-chlorotyrosine and pinch strength in chronic kidney disease patients.

BACKGROUND AND OBJECTIVES: This cross-sectional study aimed to explore whether plasma myeloperoxidase (MPO) and its active product, 3-chlorotyrosine (3-CT), as markers of inflammation and oxidative stress, have any independent pathophysiological association with sarcopenia-related parameters in patients with chronic kidney disease (CKD). METHODS AND STUDY DESIGN: A total of 292 patients, including 147 on maintenance hemodialysis (HD) and 145 non-dialyzed hospitalized patients with CKD stages 1-5, were enrolled from September 2022 to December 2023 and categorized into three groups: CKD stages

Serum uric acid, sarcopenia, and muscle strength: A systematic review of dose-response evidence with exploratory meta-analysis.

BACKGROUND: Serum uric acid (SUA) has antioxidant activity but is also linked to metabolic, renal, and inflammatory pathways that may impair skeletal muscle. We assessed whether outcome-specific dose-response evidence identifies a reproducible SUA range associated with better muscle health. METHODS: We reviewed studies reporting three or more SUA categories, category-specific outcomes, or nonlinear models or thresholds. Muscle outcomes were analysed separately by construct and sex. Restricted cubic spline synthesis required at least five independent compatible clusters; small-sample inference

Imeglimin provides dual hepatic and skeletal muscle protection in experimental MASH-associated sarcopenia.

BACKGROUND & AIMS: Metabolic dysfunction-associated steatohepatitis (MASH) is frequently complicated by sarcopenia, in which mitochondrial dysfunction and inflammatory catabolic signaling are implicated. We investigated whether imeglimin, an antidiabetic mitochondrial modulator, attenuates MASH-associated muscle atrophy. METHODS: MASH-associated sarcopenia was induced in mice using a choline-deficient, L-amino acid-defined high-fat diet. After 6 weeks of feeding, imeglimin (200 mg/kg, twice daily) was administered for 6 weeks. Four groups of 10 mice were examined. Muscle phenotype, mitochondr

Tissue-specific models of spinal muscular atrophy identify muscle functions required for Drosophila neuromuscular junction architecture.

Spinal Muscular Atrophy (SMA) is a neuromuscular disorder associated with motor neuron degeneration, yet the role of the well-conserved Survival of Motor Neuron (SMN) protein in muscle remains insufficiently understood. Using the Drosophila neuromuscular junction (NMJ) as a model, we combined tissue-specific Smn depletion with transcriptomic profiling and prior genetic modifier screens to identify SMN-dependent genes that contribute to synaptic architecture. Functional screening of candidate genes significantly and selectively effected in muscle identified genes whose knockdown phenocopied NMJ

Sarcopenia as a manifestation of systemic energetic failure with ageing.

Sarcopenia remains underdiagnosed and undertreated, partly because it continues to be mainly conceptualised as a disorder intrinsic to skeletal muscle. Although current definitions incorporate muscle strength and physical performance, they are not intended to explain the integrated physiological processes that underlie age-related impairment of muscle function. In this Personal View, we propose that sarcopenia is more appropriately understood as a manifestation of systemic energetic failure. In this framework, muscle function is considered an emergent property of integrated physiology that pro

Roles of GDF-15 in Acute Heart Failure: from Mechanistic Insights to Clinical Applications.

Acute heart failure (AHF) is a clinical syndrome characterized by the rapid or gradual onset of symptoms and signs of heart failure that are severe enough to require urgent evaluation and treatment. It may represent de novo heart failure or acute decompensation of pre-existing chronic heart failure. Natriuretic peptides remain the cornerstone biomarkers for diagnosis and risk assessment, but predominantly reflect hemodynamic stress and incompletely capture inflammation, oxidative injury, cellular damage, and multiorgan dysfunction. Growth differentiation factor-15 (GDF-15), a stress-responsive

Ubiquitin-Dependent Protein Quality Control in Polyglutamine Diseases.

Polyglutamine diseases comprise a family of nine age-dependent disorders caused by CAG triplet-repeat expansions that produce misfolded proteins with elongated glutamine tracts. Although these diseases share proteotoxic stress and engagement of protein quality control pathways as responses to their presence and disruptive activities, they differ in protein context, normal function, localization, interactomes and selective vulnerability. Here, we review how expanded polyglutamine proteins engage ubiquitin-dependent protein quality control across Huntington's disease, dentatorubral-pallidoluysia

Mitochondrial membrane lipid cardiolipin controls fiber-type adaptations in aging muscle via estrogen-related receptor γ.

Aging paradoxically leads to both a decline in skeletal muscle mitochondrial function and a shift in muscle composition that favors fibers rich in mitochondria. Yet the biological rationale and mechanism underlying this phenomenon remain largely unknown. Here we show that synthesis of the mitochondrial membrane lipid, cardiolipin, causally links mitochondrial dysfunction to fiber-type adaptations in aging mouse and human skeletal muscle. By mimicking the aging decline of skeletal muscle cardiolipin levels in young mice using inducible tissue-specific cardiolipin synthase 1 (Crls1) deletion, we

Preserved satellite cell pool in a laminin-α2-deficient model of congenital muscular dystrophy.

Deficiency in the α2 subunit of laminin-211, encoded by LAMA2, causes congenital LAMA2-related muscular dystrophy (LAMA2 MD), characterized by severe and progressive muscle weakness. In this study, we analyzed muscle regeneration and degeneration in the gastrocnemius muscle of dy2J mice, a model of LAMA2 MD, across different ages. We found that the proportion of satellite cells (SCs) per fiber was significantly higher in the dy2J groups compared to that in the wild-type (WT) group, and that SC numbers did not decrease with disease progression. The percentages of activated SCs, and the expressi

CT-defined sarcopenia as a predictor of early mortality following hip fracture: a retrospective cohort study.

PURPOSE: Sarcopenia is increasingly recognised as a marker of physiological vulnerability. Computed tomography permits objective quantification of skeletal muscle mass, yet the prognostic significance of CT-defined sarcopenia after hip fracture remains incompletely characterised. This study aimed to determine sarcopenia prevalence in patients with hip fractures and evaluate associations with early postoperative outcomes. METHODS: This single-centre retrospective cohort study included patients with hip fracture during 2015 to 2025 who had abdominal computed tomography imaging, including the th

[Cell Death of Cardiomyocytes in Heart Failure].

Heart failure, a leading cause of death worldwide, is generally defined as a pathophysiological condition in which the heart is unable to eject the arterial blood required by all tissues in the body. Heart failure is broadly classified into acute heart failure and chronic heart failure. In clinical practice, acute heart failure is often considered synonymous with acute myocardial infarction. In acute heart failure, cardiac contractile dysfunction is induced due to necrosis of cardiomyocytes during ischemia/reperfusion. In contrast, in chronic (congestive) heart failure, cardiac contractile dys

Muscle-brain crosstalk in sarcopenia and cognitive impairment: endocrine signaling, metabolic regulation, and exercise modulation.

Aging is accompanied by progressive changes in endocrine signaling, metabolic regulation, and inter-organ communication that may contribute to functional decline. Sarcopenia and cognitive impairment frequently coexist in older adults, raising the possibility that their association involves disruption of communication between skeletal muscle and the brain in addition to shared age-related processes. This narrative review synthesizes clinical and mechanistic evidence on muscle-brain crosstalk in sarcopenia and cognitive impairment, with emphasis on endocrine and metabolic mechanisms and their mo

Neutrophil-mediated myocardial ischemia-reperfusion injury: mechanisms and potential therapeutic targets.

Although reperfusion therapy improves the prognosis of acute myocardial infarction, myocardial ischemia-reperfusion injury continues to limit its clinical benefits, with neutrophil-driven inflammatory cascades representing a central mechanism of injury amplification. This article delineates the molecular event chain of neutrophils from recruitment to effector execution: activated neutrophils damage cardiomyocytes through three principal pathways-respiratory burst, degranulation, and neutrophil extracellular traps formation-thereby establishing a positive feedback loop that exacerbates inflamma
