Versican: A Promising Therapeutic Target for Post-Myocardial Injury Cardiac Repair.

This review re-evaluates the literature from a matrix-centered perspective and determines how strongly it supports versican as a regulator of myocardial renewal rather than merely a marker of tissue remodeling. The evidence is organized around molecular identity, developmental location, temporal behavior after infarction, and causal intervention. During cardiac formation, versican-rich extracellular matrices recur in cushions, valves, septal and outflow structures, where matrix hydration, cell adhesion, and proteolytic processing accompany morphogenesis. In adult injury models, versican abunda

Aberrant Myelopoiesis and Haematopoietic Stem Cell Niche Remodelling Precedes Cancer Cachexia Onset in a Sex-Dependent Manner.

BACKGROUND: Male cancer patients are 30% more likely to develop cachexia compared to females; however, the underlying aetiology remains unknown. Myelopoiesis induces inflammation in cancer cachexia, which contributes to muscle atrophy. Whether differences in myelopoiesis during cancer cachexia initiation contribute to sex-based differences in cachexia incidence remains unknown. METHODS: Male and female C57BL/6 and B6(C)-Ccr2tm1.1Cln/J mice were randomized into sham (PBS) or were inoculated subcutaneously with Lewis lung carcinoma (LLC) to induce cachexia. Mice were sacrificed 2 (14 days), 2.5

Lipid dysregulation in polyglutamine diseases.

Polyglutamine (polyQ) diseases are fatal neurodegenerative disorders caused by pathological CAG repeat expansions in disease-associated genes. They are characterized by progressive motor dysfunction and typically late onset. Although multiple mechanisms have been proposed, accumulating evidence indicates that lipid dysregulation contributes to neuronal dysfunction and degeneration. Lipids are essential for brain development and homeostasis, and disrupted lipid metabolism is implicated across neurodegenerative conditions. Within the polyQ spectrum, Huntington's disease is the best studied and s

GLP-1-Based Pharmacotherapies and Skeletal Muscle Health in Obesity: A Narrative Review of Body Composition, Muscle Function, and Sarcopenia-Related Outcomes.

Background: Obesity is associated with impaired skeletal muscle health and sarcopenic obesity. GLP-1-based pharmacotherapies induce weight loss, but accompanying reductions in fat-free or lean mass raise concerns about skeletal muscle loss. Conversely, reductions in adiposity, inflammation, insulin resistance, and intramuscular fat may be beneficial. Objective: This narrative review evaluated the effects of GLP-1-based pharmacotherapies on body composition, muscle quantity, composition and function, and their potential role in sarcopenia. Methods: PubMed and Google Scholar were searched for cl

Effects of Body-Mass-Scaled Protein and Leucine Supplementation on Body Composition and Cardiometabolic Blood Markers During Weight Loss in Older Men-A Randomized Controlled Trial.

Background: Overweight and obesity are burgeoning health epidemics. Diets including alternate-day fasting (ADF) promote weight loss but risk significant muscle loss, posing health implications to seniors at risk of sarcopenia. Leucine, the key muscle protein synthesis (MPS) driver, effectively stimulates MPS with lower calorie intake than protein. However, the effects of body-mass-scaled protein and leucine intakes are unclear. Objectives: To evaluate body-mass-scaled leucine or protein supplementation on body composition, sarcopenic, and cardiometabolic markers in older men during ADF. Method

Prevalence of Hypovitaminosis D and Its Association with Motor Impairment in Spinal Muscular Atrophy.

Background/Objectives: In recent years, the role of vitamin D in the etiology of central and peripheral neurological diseases has been widely investigated, and evidence suggests benefits of vitamin D replacement therapy. However, few studies have evaluated the relationship between serum vitamin D levels and patients with spinal muscular atrophy (SMA). Therefore, this study aimed to analyze serum vitamin D levels in patients with SMA and their relationship with disease type, degree of motor impairment, and the presence of musculoskeletal deformities. Methods: This was a cross-sectional analytic

In Vitro Modeling of Sarcopenia-Related Pathology and Exercise-Associated Adaptations: Integration with In Silico Approaches.

Sarcopenia is a progressive and generalized skeletal muscle disorder characterized primarily by reduced muscle strength, with low muscle quantity or quality confirming the diagnosis and poor physical performance indicating greater severity. Although human studies and animal models remain essential, their use is constrained by ethical considerations, inter-individual variability, species-specific differences, cost, and the difficulty of isolating individual mechanisms within whole-body systems. This review critically evaluates in vitro models that reproduce selected sarcopenia-related pathologi

Is There a Relationship Between the Gut Microbiome and the Functional Capabilities of Skeletal Muscle? A Narrative Review.

Diet is one of the primary determinants of gut microbiota composition and its metabolic output. That output is emerging as a critical determinant of systemic health and has been linked to most physiological systems in the body, yet its relationship to skeletal muscle physiology has only recently received attention. Evidence from experimental, observational, and clinical studies supports the existence of a gut-muscle axis, in which microbial composition and metabolic activity in the gut, influenced by diet, affect muscle metabolism, strength, function, and physical performance. Fiber, protein i

Association of serum 25-hydroxyvitamin D concentrations with premature mortality in adult U.S. patients with sarcopenia.

BACKGROUND: Lower serum 25-hydroxyvitamin D ([25(OH)D]) levels may be associated with a higher risk of premature mortality in patients with sarcopenia; however, this relationship remains inadequately characterized in U.S. METHODS: Data were from 9,561 participants in NHANES 2011-2018. Sarcopenia was defined using FNIH criteria (appendicular lean mass/BMI). Weighted multivariable logistic regression was used to examine the association between serum 25(OH)D levels and sarcopenia prevalence. Among participants with sarcopenia, premature mortality (death before age 75) was assessed by linking dat

B-Type Natriuretic Peptide Is Associated With Sarcopenia in Older Patients Without Symptomatic Heart Failure.

AIM: B-type natriuretic peptide (BNP) has been identified as a biomarker of sarcopenia in community-dwelling populations and in patients with heart failure. However, it remains unclear whether BNP is associated with sarcopenia in the pre-heart failure stage in older adults with cardiometabolic diseases. METHODS: In this cross-sectional study, BNP was measured in 697 older outpatients with cardiometabolic disease and no history of heart failure or atrial fibrillation. Sarcopenia was assessed using the Asian Working Group for Sarcopenia criteria 2019. The participants were grouped according to

GNMT alleviates heart failure-induced skeletal muscle atrophy by regulating ATP5J2-mediated mitochondrial metabolic reprogramming through succinylation.

Heart failure (HF) is defined as a condition characterized by abnormalities in the heart's structure or function, which lead to the heart's inability to deliver oxygen to tissues in a timely manner, and it may progress to symptoms such as muscle atrophy of skeletal muscles. Our research primarily focuses on Glycine N-methyltransferase (GNMT), an enzyme that utilizes S-adenosylmethionine (SAM) to catalyze glycine, in order to explore its specific role in HF-induced skeletal muscle atrophy. In this study, an HF rat model induced by abdominal aortic constriction (AAC) and a C2C12 muscle atrophy c

An integrative transcriptomic atlas of ALS and aging skeletal muscle defines shared and distinct molecular programs of degeneration.

Skeletal muscle atrophy and weakness resulting from loss of motor neuron innervation are shared hallmarks of amyotrophic lateral sclerosis (ALS) and age-associated sarcopenia. However, muscle wasting in ALS is rapid and severe, whereas sarcopenia progresses gradually over decades, peaking at an advanced age. The molecular mechanisms underlying muscle wasting in these conditions remain incompletely defined. Here, we used the SOD1G93A ALS mouse model and wild-type mice across the lifespan to generate an integrated single-nucleus transcriptomic atlas of skeletal muscle, mapping changes associated

Immobilzation-Induced Muscle Atrophy Exacerbates Vascular Calcification and Abdominal Aortic Aneurysms Through Irisin Downregulation.

BACKGROUND: Sarcopenia is widely recognized that affects multiple organs, including the cardiovascular system. Clinical studies have consistently shown associations between sarcopenia and vascular aging phenotypes such as vascular calcification, atherosclerosis, and abdominal aortic aneurysms (AAA). However, the molecular mechanisms by which muscle deterioration contributes to vascular degeneration remain poorly understood. METHODS: To elucidate muscle-vascular crosstalk, we established a murine model combining inflammation-associated muscle atrophy induced by hindlimb immobilization with CaP

Capillary-selective vasodilation through myosin inhibition halts ischaemic cascades.

In acute myocardial infarction and acute kidney injury, capillary no-reflow denotes that capillary blood flow is still blocked despite successful arterial reperfusion. This is associated with substantially increased mortality and reinfarction risk; nevertheless, no targeted therapy exists so far1,2. Because capillary no-reflow is driven by smooth muscle myosin-mediated hypercontraction of capillary pericytes, we developed R(+)2501, a myosin-specific and capillary-selective vasodilator. In reperfused animal models of acute myocardial infarction and acute kidney injury, direct in vivo imaging sh

Impact of advancing age on diaphragm thickness and maximal inspiratory pressure: Comparison with peripheral skeletal muscle.

Aging is associated with deleterious effects on skeletal muscle size and function, known as sarcopenia. Less understood is the impact of advancing age specific to the size and function of the diaphragm. The purpose of this study was to determine the extent to which structural and functional responses of the diaphragm to advancing age are similar or unique to those observed in peripheral muscle. Ten young (22 ± 1 yr, 5 M, 5 F) and ten old (69 ± 3, 4 M, 6 F) healthy individuals volunteered for this study. Diaphragm thickness (end exhalation, end-tidal inhalation, end-maximal inhalation) and thic

Azilsartan ameliorates angiotensin II-induced skeletal muscle atrophy via AT1R-mediated PI3K/AKT/FOXO3a signaling pathway.

BACKGROUND: Sarcopenia is a progressive age-related skeletal muscle disorder characterized by the loss of muscle mass, strength, and physical performance. Increasing evidence suggests that activation of the renin-angiotensin system (RAS) contributes to muscle wasting through promotion of protein degradation and impairment of anabolic signaling. Azilsartan (AZL), a highly selective angiotensin II type 1 receptor (AT1R) blocker, has demonstrated pleiotropic effects beyond blood pressure control. However, its effects on skeletal muscle atrophy and the underlying mechanisms remain incompletely und

From marrow memory to adverse ventricular remodeling: trained immunity after myocardial infarction.

Myocardial infarction [MI] triggers a sterile inflammatory response that is essential for necrotic-tissue clearance and scar formation and should normally subside as repair progresses. In a subset of patients, however, persistent myeloid activation accompanies progressive fibrosis, ventricular dilatation, and adverse remodeling long after the acute insult. Trained immunity may help explain this temporal mismatch by allowing myocardial injury to leave a durable functional bias in innate immune cells or their hematopoietic progenitors. Damage-associated molecular patterns, inflammatory cytokines
