Clinical Implications of the AWGS 2025 Sarcopenia Criteria in a Large Cohort of Community-Dwelling Thai Older Adults.

BACKGROUND: The Asian Working Group for Sarcopenia (AWGS) 2025 criteria introduced revised age-specific cutoffs and appendicular skeletal muscle mass adjusted for body mass index (ASM/ BMI) as an alternative muscle mass index. We evaluated their effects on sarcopenia classification among Thai older adults. METHODS: This cross-sectional study included 8,073 community-dwelling adults aged ≥60 years attending a tertiary geriatric outpatient clinic. Low muscle mass and sarcopenia were classified using AWGS 2019 and 2025. Discordant ASM/height²- and ASM/BMIbased phenotypes were characterized, and

Polydeoxyribonucleotide attenuates dexamethasone-induced skeletal muscle atrophy by suppressing oxidative stress and regulating muscle protein turnover.

Glucocorticoid-induced muscle atrophy is characterized by excessive oxidative stress and disrupted muscle protein homeostasis. Polydeoxyribonucleotide (PDRN) exerts tissue-protective and regenerative effects; however, its therapeutic potential in muscle atrophy remains unclear. This study investigated the protective effects of PDRN against dexamethasone (DEX)-induced muscle atrophy in vitro and in vivo. Differentiated C2C12 myotubes were treated with PDRN in the presence or absence of DEX (100 µM). PDRN showed no cytotoxicity, and concentrations of 10, 25, and 50 µg/mL were selected for subseq

Beyond reduced intake: tumor metabolism and immune dysregulation drive cancer-related malnutrition in head and neck squamous cell carcinoma (HNSCC).

Cancer-related malnutrition is highly prevalent in patients with head and neck squamous cell carcinoma (HNSCC) and represents a major determinant of treatment tolerance, quality of life, and survival. Although traditionally attributed to reduced oral intake caused by dysphagia, mucositis, and treatment-related toxicity, increasing evidence indicates that malnutrition in HNSCC is largely driven by tumor-induced metabolic and inflammatory alterations. Enhanced aerobic glycolysis, systemic inflammation, and tumor-derived factors promote hypermetabolism, substrate redistribution, and progressive s

Cyclic azapeptide CD36 ligand attenuates cardiac injury and reduces long-chain fatty acid accumulation after myocardial ischemia-reperfusion in mice.

Ischemic heart disease remains a leading global cause of death. We investigated the cardioprotective effects of the selective cluster of differentiation-36 receptor (CD36) modulator azapeptide MPE-298 in a mouse model of myocardial ischemia-reperfusion. Given before reperfusion, a single intravenous dose of azapeptide MPE-298 reduced infarct size by 44% of the area-at-risk and transiently decreased left ventricular long-chain fatty acids (LCFA) accumulation, independently of saturation status. Metabolomic profiling identified changes in amino acids that may fuel the tricarboxylic acid cycle an

Empagliflozin-pretreated mesenchymal stem cell-derived extracellular vesicles facilitate cardiac repair via AGGF1.

BACKGROUND: Current treatment modalities can only reap a limited efficacy in reversing myocardial damage and promoting functional recovery in patients with myocardial infarction. Therefore, there is an urgent need to explore new therapeutic approaches to enhance cardiac repair. METHODS: MSCs were pretreated with empagliflozin to obtain EMPA-EVs, with non-pretreated MSC-EVs serving as controls. In vitro experiments were conducted to evaluate the effects of the two types of EVs on macrophage phenotypic transformation, cardiomyocyte apoptosis, and vascular endothelial cell lumen formation. Psoralen enhances direct cardiac reprogramming via activating PPARα and driving mitochondrial fission and improves cardiac function after myocardial infarction.

BACKGROUND: Direct cardiac reprogramming offers a promising strategy to regenerate cardiomyocytes for heart repair, but its efficiency and maturation remain limited. Psoralen, a natural compound isolated from Psoralea corylifolia L., has been widely studied for its bioactive properties. This study aimed to determine whether psoralen enhances chemical cardiac reprogramming and improves functional maturation, and to evaluate its therapeutic potential in myocardial infarction. METHODS: Mouse embryonic fibroblasts were treated with a chemical reprogramming cocktail containing RepSox and Forskolin

P2X4 receptor limits adverse cardiac remodeling following ischemia-reperfusion through regulation of autophagy in cardiac fibroblasts.

BACKGROUND: Pathological ventricular remodeling is an adverse tissue response following acute myocardial infarction (AMI), characterized by an exaggerated fibrosis leading to tissue disorganization and chronic heart failure. Cardiac fibroblasts have a prominent role in the healing process, but also in the fibrosis development, through their differentiation into myofibroblasts, which produce the extracellular matrix, in response to ischemia-reperfusion. Recently, the ATP-gated P2X4 ionotropic receptor has been identified as a key regulator of macro-autophagy, activated under metabolic challenge

PHLDA3 hypomethylation at the mercy of PTBP1-mediated DNMT3a decay prompts ferroptosis of cardiomyocytes to accelerate microvascular endothelial cell senescence following ischemia/reperfusion injury.

Myocardial ischemia-reperfusion (I/R) injury is a complex condition characterized by oxidative stress, inflammation, and mitochondrial dysfunction. Ferroptosis, an iron-dependent form of regulated cell death, plays a critical role in cardiomyocyte damage during I/R. Inhibiting ferroptosis has been shown to reduce myocardial injury and improve cardiac function, making it a promising therapeutic target for enhancing clinical outcomes. An in vivo I/R model was established, and infarct size was assessed using triphenyltetrazolium chloride (TTC) staining. Histological changes were analyzed using he

Pseudocapacitive Charge-Driven Adhesive-Electrocoupling Hydrogels for Myocardial Infarction Repair via Interstitial Microenvironment Remodeling.

Myocardial infarction (MI) creates a hostile microenvironment characterized by oxidative stress, inflammation, fibrosis, and electrical conduction block, posing a multifaceted challenge for tissue repair. To address this, we developed a multifunctional hydrogel (Gel-PDZC), through an artificial neural network (ANN)-guided optimization process, incorporating nanoparticles comprising a ZIF-8@CaO2 core for sustained oxygen release, a polydopamine (PDA) interlayer for adhesion and antioxidant activity, and a conductive poly(3,4-ethylenedioxythiophene) (PEDOT) shell. The hydrogel exhibits a unique

Aging-, obesity-, and diabetes-related sarcopenia: Pathogenesis and interventions.

As global aging accelerates, sarcopenia has become an increasingly important public health concern. Although typically recognized as an age-related condition, sarcopenia can develop and progress more rapidly under certain pathological conditions, such as obesity and type 2 diabetes. With the ongoing advancement of related research, the mechanisms underlying sarcopenia are being progressively elucidated. Management strategies include both nonpharmacological and pharmacological interventions. Resistance exercise is currently regarded as one of the most effective and foundational nonpharmacologic

Cardiac nanomedicine for ischemic injury and remodeling: imaging and therapy.

Cardiovascular disease remains a major cause of morbidity and mortality worldwide, with myocardial infarction and pressure overload driving progressive cardiac injury and maladaptive remodeling that involve dynamic changes in vasculature, cells, and extracellular matrix. Current therapies do not fully prevent this process, creating a need for targeted interventions that can image and treat distinct pathogenic phases. This Review examines nanocarrier strategies for imaging and therapy across ischemic injury, inflammatory repair, fibrosis, and pressure-overload remodeling. Imaging systems exploi

Reframing obesity in aging people with HIV: sarcopenic obesity, muscle quality, and frailty.

PURPOSE OF REVIEW: As people with HIV (PWH) age, obesity is increasingly prevalent, yet body weight alone inadequately captures the functional and metabolic consequences of adverse body composition remodeling. This review examines emerging evidence supporting a shift from weight-centric approaches toward a framework centered on sarcopenic obesity, muscle quality, and frailty in aging PWH. RECENT FINDINGS: Weight gain in the contemporary antiretroviral therapy era often reflects increased visceral and ectopic adiposity. Sarcopenic obesity is a clinically relevant phenotype characterized by the

Transcription factor activity divergence across muscle atrophy conditions: A comparative analysis of spaceflight, aging, and disuse using decoupleR.

Skeletal muscle atrophy arises from heterogeneous insults including spaceflight, aging, and immobilization. Prior studies have catalogued differentially expressed genes (DEGs) in each context, but the transcription factors (TFs) coordinating these programs remain inconsistently characterized across conditions, and gene-level overlap between conditions is small. Three mouse transcriptomic datasets were analyzed: NASA OSD-576 (Rodent Research-23) tibialis anterior muscle from spaceflight versus ground control animals; GSE145480 gastrocnemius muscle from 28-month-old versus 8-month-old mice as a

Establishment of an Animal Model of Type 2 Cardiorenal Syndrome.

Type 2 cardiorenal syndrome (CRS) is a complex clinical syndrome characterized by chronic kidney injury resulting from chronic cardiac dysfunction. Its underlying pathophysiological mechanisms remain incompletely understood, highlighting the urgent need for disease models that better mimic human pathology and accurately simulate the progression of cardiac-induced renal injury. Existing models possess significant limitations and often fail to adequately replicate the onset and progression observed in clinical Type 2 CRS patients. We therefore hypothesized that a double-hit approach combining le
