Accumulation of newly synthesized docosahexaenoic acid plays an essential role in heart regeneration.

Adult zebrafish and neonatal mice can fully regenerate their hearts after partial amputation through the proliferation of preexisting cardiomyocytes (CMs). However, the adult mammalian heart has limited regenerative capability following cardiac damage. The reason for this phenomenon remains elusive. Here, we find that docosahexaenoic acid (DHA) is accumulated only in the injured hearts of zebrafish and neonatal mice, but not of adult mice, which coincides with the upregulation of DHA synthesis genes in CMs, fibroblasts, and macrophages near the injury areas. Inhibition of Fads2, a DHA synthesi

Cardiac Microanatomy Imaging Using Forward-Viewing Optical Coherence Tomography Endoscope.

OBJECTIVE: Due to limitations in current imaging technologies detecting subtle cardiac microstructural changes that can lead to sudden cardiac death is a significant clinical challenge. To address this problem, we developed a forward-viewing optical coherence tomography (OCT) endoscope for the detection of relevant cardiac microstructures in the subendocardium, including Purkinje fibers, scar tissue, surviving myocytes, and adipose tissue. METHODS: An endoscopic probe based on the scanning fiber principle was developed for OCT measurements in contact. The probe was evaluated in freshly excise

Heart Scar-In-A-Dish: Tissue Culture Platform to Study Myocardial Injury and Mechanics In Vitro.

Myocardial Infarction (MI) occurs when blood flow is blocked to a portion of the left ventricle and leads to necrosis and scar formation. Many therapies are under development to improve infarct healing, and three-dimensional engineered heart tissues (EHTs) offer an in vitro drug screening option to help reduce, refine, and potentially replace animal testing. Unfortunately, existing EHTs oversimplify cardiac mechanics and neglect the spatial variations of the infarcted ventricle in vivo, wherein the passive infarct zone is cyclically stretched under tension as the remote zone cyclically contrac

Cardioprotective Effects of Soluble Guanylate Cyclase and Its α1 Subunit on Myocardial Ischemia/Reperfusion Injury via the PGC-1α/UCP2 Pathway.

This study investigates the cardioprotective potential of soluble guanylate cyclase (sGC) and its α1 subunit in myocardial ischemia/reperfusion (I/R) injury, along with the underlying mechanisms. We used a model involving Sprague Dawley rats undergoing left coronary artery I/R, complemented by H9c2 cell cultures in an anaerobic environment to simulate I/R conditions in vitro. Both loss- and gain-of-function approaches were applied to assess the role of sGC and its α1 subunit in myocardial I/R injury. Immunofluorescence microscopy, western blotting, and RT-PCR were employed to examine how sGC a

Cardioprotective Effects of Apelin in Myocardial Ischemia/Reperfusion Injury: A Systematic Review and Meta-Analysis.

Myocardial ischemia/reperfusion (I/R) injury remains a major clinical challenge, because blood flow restoration can exacerbate tissue damage. Apelin, an endogenous peptide acting through the APJ receptor, has demonstrated cardioprotective effects in experimental models. The APJ receptor, a G-protein-coupled receptor widely expressed in cardiovascular tissues, mediates vasodilation, cardiac contractility, and angiogenesis. This systematic review and meta-analysis evaluates its efficacy in myocardial I/R injury. A systematic search in Medline (PubMed), Embase, Scopus, and Web of Science was cond

A "drug-carrier homologation" cardiac patch for myocardial infarction therapy via month-long controlled H2S release.

Hydrogen sulfide (H2S), a gaseous signaling molecule with multiple cardioprotective effects, has attracted considerable attention for treating myocardial infarction (MI), a leading global cause of death. However, its ultrashort half-life (seconds to minutes) and the extended recovery required for myocardial repair pose substantial challenges to therapeutic efficiency, emphasizing the urgent need for month-level controlled H2S delivery strategies, an endeavor still unresolved. Inspired by natural trisulfide H2S donors, we introduce a novel disulfide/trisulfide cross-linked network derived from

The Ets2 super-enhancer modulates endothelial-mesenchymal transition during cardiac ageing.

AIMS: Cardiac ageing is characterized by endothelial dysfunction and associated cardiovascular pathologies, often involving endothelial-to-mesenchymal transition (EndoMT) in cardiac endothelial cells. While the transcription factor Ets2, a member of the Ets family, is known to regulate endothelial cell survival and function, its role in EndoMT and cardiac ageing remains poorly understood. METHODS AND RESULTS: To investigate this, we utilized single-nucleus RNA sequencing (snRNA-Seq) in Ets2-super-enhancer (Ets2-SE)-deficient mice to examine the regulation of Ets2 expression across various car

MCPPIR promotes cardiomyocyte proliferation and cardiac repair via o8G oxidation of POC1B mRNA.

AIMS: The adult mammalian heart possesses severely limited regenerative capacity. Cardiomyocyte loss during cardiac injury, coupled with this restricted regenerative potential, represents a fundamental cause of heart failure and associated mortality. While PIWI-interacting RNAs (piRNAs) are abundantly expressed in cardiac tissue, their functional roles and molecular mechanisms in cardiomyocyte proliferation and heart regeneration remain largely undefined. In this study, we systematically investigated piRNA-mediated regulation of cardiomyocyte proliferation and cardiac repair processes. METHOD

Precision targeting of FDX1-mediated cuproptosis by a ROS-responsive hydrogel for myocardial ischemia-reperfusion injury treatment.

Rationale: Myocardial ischemia-reperfusion injury (MIRI) poses a critical clinical challenge due to intertwined oxidative stress and cuproptosis-driven cell death. Current therapies inadequately address dual-pathology mechanisms (ROS overproduction and copper dysregulation), while conventional drug delivery lacks spatiotemporal precision. Methods: A ROS-responsive hydrogel (OD@G4CAsi-FDX1) was engineered via dynamic Schiff base crosslinking between oxidized dextran (OD) and polyamidoamine dendrimers (PAMAM G4). The hydrogel co-encapsulates antioxidant caffeic acid (CA) and FDX1-targeted siRNA

Pharmacologically inherited carbon dots from Salvia miltiorrhiza with potent antioxidant activity and multi-pathway modulation for myocardial ischemia-reperfusion injury therapy.

Rationale: Myocardial ischemia-reperfusion (I/R) injury remains a major clinical challenge that limits the efficacy of reperfusion therapy in acute myocardial infarction, mainly due to excessive production of reactive oxygen species (ROS) and the resulting oxidative stress, inflammation, and cardiomyocyte death. However, conventional antioxidant strategies show limited clinical efficacy, highlighting the urgent need for novel redox-regulating therapies. Methods: We synthesized carbon dot nanozymes (SM-CDs) via a green hydrothermal process using Salvia miltiorrhiza, a traditional Chinese medici

Meox1 Promotes Cardiac Fibrosis and Pathological Remodeling following Myocardial Infarction through Cthrc1/p-Smad2/3 Signaling.

Aims: Myocardial infarction (MI) induces pathological cardiac fibrosis and ventricular remodeling, which leads to cardiac dysfunction and heart failure. Mesenchyme homeobox 1 (Meox1) was shown to be an essential transcriptional switch in fibroblasts activation; however, whether Meox1 is involved in the fibrosis process following MI remains unknown. We aimed to explore the role of Meox1 in cardiac fibrosis and remodeling post-MI and its underlying mechanisms. Methods and results: Herein, we identified that Meox1 was highly expressed in activated fibroblasts (myofibroblasts, Myofbs), in response

Activity of Cardiomyocyte Type 3 Deiodinase After Myocardial Infarction Influences Cardiac Recovery in Females.

Thyroid hormone (TH) is essential for cardiovascular function, and women are disproportionately affected by TH disorders and experience worse outcomes following myocardial infarction (MI). However, the role of sex-specific TH regulation in post-MI cardiac recovery remains poorly understood. We investigated TH homeostasis and type 3 deiodinase (D3) activity, an enzyme that inactivates TH, in male and female C57BL/6 mice following MI. Using cardiomyocyte-specific D3-deficient (Dio3ΔHeart) mice, we investigated how impaired TH inactivation influences cardiac function and mitochondrial respiration

Spatial Metabolomics Reveals GLA-Mediated Imbalance of Gal2Cer/GalCer as a Therapeutic Target in Myocardial Ischemia-Reperfusion Injury.

Myocardial ischemia-reperfusion (I/R) injury, a major complication of reperfusion therapies for acute myocardial infarction that exacerbates myocardial necrosis, impairs cardiac function, and worsens patient prognosis. Profound metabolic disturbances drive myocardial I/R injury; however, their spatial heterogeneity remains unclear. Using MALDI mass spectrometry imaging with spatial segmentation, we mapped the spatiotemporal metabolite dynamics in mouse heart tissue across multiple time points (30 min ischemia to 7 days reperfusion), aiming to identify critical metabolic pathways and regulatory

Molecular imaging of immune and fibrosis targets to guide therapy for repair after myocardial infarction.

Heart injury after acute myocardial infarction requires adequate repair to prevent left ventricular remodelling and the development of chronic heart failure. The early immune response to myocardial injury and its tight interconnection to fibroblast activation have a crucial role in determining the outcome after myocardial infarction. Emerging therapies aim to exploit these targets, harnessing the subacute window after cardiac injury to prevent subsequent contractile dysfunction. Given the complexity of the tissue response to myocardial injury and the molecular specificity of targeted therapies

ENPP2 Protects Mouse Myocardium from Ischemia-Reperfusion-Induced Ferroptosis Injury Via the SIRT1/PGC-1α/NRF1 Pathway.

Ferroptosis is a critical contributor to ischemia-reperfusion (I/R) injury and subsequent organ failure. While ENPP2 has been implicated in regulating ferroptosis in cardiomyocytes, its specific role in myocardial I/R injury remains unclear. This study aims to elucidate the function of ENPP2-mediated ferroptosis in myocardial ischemia-reperfusion injury (MI/RI), to provide novel insights into potential therapeutic strategies. A mouse model of MI/RI was established and subjected to interventions with ENPP2 overexpression and/or SIRT1 knockdown. In vitro, cardiomyocytes were treated with palmita

Spatial Transcriptional Heterogeneity in the Infarct Core and Its Surrounding Regions Targeting Piezo1 Signals in Rats With Myocardial Ischemia-Reperfusion Injury.

Myocardial ischemia-reperfusion (MIR) injury is a major cause of cardiac dysfunction, but the spatial heterogeneity of its underlying molecular programs remains unclear. In this study, we applied Visium spatial transcriptomics to generate gene expression maps of rat left ventricles after MIR and identified distinct regional features. The border zones were enriched with phagosome-related genes, incomplete infarct areas showed activation of MAPK, IL-17, and osteoclast differentiation pathways, while the infarct cores were characterized by ferroptosis and mitophagy-related genes. To further resol

Microgel-Based Hierarchical Porous Hydrogel Patch with Adhesion and Resilience for Myocardial Infarction.

While there has been considerable success in the 3D bioprinting of hydrogel scaffolds for tissue engineering, the application of traditional centimeter-scale bulk hydrogels with a dense internal nanoscale network structure remains a particular challenge. Here, we present a microgel-based modular fabrication strategy to engineer programmable hierarchically porous microgel-based hydrogel patches (HPMPs). This strategy generates porous microgels with adjustable porosity and bio/cytocompatibility via gas-shearing microfluidics integrated with an aqueous two-phase system, exhibiting precise model s

Lactylation at the metabolic-epigenetic interface in cardiovascular diseases: context-dependent mechanisms and translational roadmap.

BACKGROUND: Lysine lactylation has redefined lactate's biological role from a metabolic byproduct to a signaling molecule. This post-translational modification directly couples cellular energetics with gene regulation, creating a metabolic-epigenetic axis particularly relevant to cardiovascular pathophysiology. Ischemic and inflammatory stress drive glycolytic reprogramming and lactate accumulation in these diseases. Lactylation modifies both histone and non-histone proteins, enabling metabolic states to reshape chromatin accessibility and protein function. However, therapeutic translation fac
