Inhibition of Annexin A2 Facilitates PHB2-Mediated Mitophagy in Cardiomyocytes to Alleviate Cardiac Injury and Remodeling After Infarction.

BACKGROUND: Mitophagy is critically involved in cardiac injury and repair after myocardial infarction (MI), whereas the annexin A family plays an important role in mitophagy. However, the intrinsic molecular underpinnings that orchestrate the homeostasis of mitophagy in the infarcted heart remain to be fully characterized. Here, we aimed to evaluate the role of ANXA2 (annexin A2) in cardiac mitophagy in response to MI. METHODS: Transcriptome analyses were conducted to identify differentially expressed genes and enriched pathways. Mitophagy, mitochondrial function, and cardiac injury and remod

Enhancing MRI contrast between sub-chronic myocardial infarct, remote and healthy myocardium using periodic irradiation in the rotating frames.

Relaxations Along a Fictitious Field in the rotating frame of rank 2 (RAFF2) was used to characterize fibrosis-related myocardial diseases with MRI. RAFF2 can be sensitized to molecular dynamics processes by changing the durations of the RAFF2 pulses. Here, we studied the effect of altering the RAFF2 pulse durations for characterization of myocardial infarction (MI). 10 mice underwent myocardial infarct surgery, and the hearts were collected 7 days after the operation. 3 mice hearts were served as healthy control. We tested the influence of stretching factor (TL) on relaxations, where TL was b

Extracellular BRICK1 drives heart repair after myocardial infarction in mice.

Tissue repair after myocardial infarction entails a vigorous angiogenic response that mitigates scarring and worsening of heart function. Angiogenesis in the infarct wound is guided by incompletely defined myeloid cell-endothelial cell interactions. Here, we identify the 75-amino acid microprotein BRICK1 (BRK1) as an indispensable driver of postinfarction angiogenesis in a mouse model of reperfused myocardial infarction. We show that BRK1 is preferentially expressed by myeloid cells and translocates to the extracellular space after myocardial infarction in mice and humans. As a subunit of the

Adaptive hydrogel platform for nitric oxide release and post-infarction cardiac microenvironment modulation.

After myocardial infarction (MI), oxidative stress, inflammatory response and myocardial fibrosis severely impair tissue regeneration. Aiming at the problem that multifunctional hydrogel systems are difficult to achieve precise therapeutic regulation due to limited response characteristics and insufficient synergistic effect. Here, we developed an adaptive hydrogel platform for targeted nitric oxide (NO) release and post-infarction cardiac microenvironment modulation. A small molecule phenylboronic acid-based cross-linker (N(BA)4) with a four-arm topology was reported for the first time, and i

Mechanical characterization of infarcted porcine hearts using left anterior descending and left circumflex coronary artery models.

This study comprehensively analyzes passive mechanical and structural changes of cardiac tissue 6 weeks after myocardial infarction (MI) by biaxial, simple triaxial shear, and confined compression characterization. We considered a porcine model, comparing two MI types: Left Anterior Descending (LAD) artery and Left Circumflex (LCX) artery occlusions. LAD model generated apical transmurally infarcted hearts, while the LCX model generated medial heterogeneous infarctions ranging from mild locally infarcted tissue to transmurally infarcted tissue. Infarcted tissue exhibited significantly higher s

Inhibiting L-type calcium channel promotes cardiomyocyte proliferation through activating the canonical Wnt signaling pathway.

The adult human heart is incapable of regeneration after myocardial infarction (MI) injury. One potential therapeutic strategy is to enhance the proliferation of resident cardiomyocytes (CMs). In this study, we developed a high-content screening assay based on DNA synthesis in human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) to identify small molecules that could promote CM proliferation. In the primary screening, we found that L-type calcium channel (LTCC) blockers induced DNA synthesis of hPSC-CMs. Among the 6 clinically approved calcium channel blockers tested in secondary scre

Piezo1 activation in endothelial cells aggravates microvascular ischemia-reperfusion injury in limbs by enhancing ferroptosis.

Acute limb ischemia-reperfusion injury (ALIRI) prominently involves microvascular dysfunction, with notable contributions from damage to microvascular endothelial cells (MECs). Previous research suggests that the mechanosensitive ion channel Piezo1 becomes active in response to mechanical stress conditions, including ischemia and trauma. However, its precise function within the ALIRI context remains elusive. Notably, the expression of Piezo1 was markedly elevated postreperfusion in mouse hind limb ischemia/reperfusion (I/R) models, implicating its crucial involvement in limb survival. Employin

Lactate regulates the YTHDF2-FTH1 axis to promote cardiomyocyte ferroptosis and aggravate myocardial ischemia-reperfusion injury.

Myocardial ischemia–reperfusion (MI/R) injury remains a major clinical challenge, and ferroptosis has recently emerged as a crucial contributor to its pathogenesis. However, the regulatory mechanisms underlying ferroptosis in MI/R remain incompletely understood. Here, we investigated the role of lactate-mediated YTHDF2 regulation in cardiomyocyte ferroptosis. A murine ischemia–reperfusion (I/R) model and an H9C2 hypoxia/reoxygenation (H/R) model were established. Biochemical assays revealed elevated lactate levels in MI/R hearts, accompanied by increased infarct size, enhanced structural damag

Bioinspired Spatiotemporal-Responsive Nanoparticles Synergistically Regulate Mitochondrial Repair and Inhibit Fibrosis in Acute Myocardial Infarction Treatment.

Cardiovascular diseases, particularly acute myocardial infarction (AMI), have posed a significant global health burden due to the high morbidity and mortality rates. Current treatment methods are compromised by a narrow therapeutic window, risk of ischemia-reperfusion injury, rapid systemic drug clearance, and inadequate targeting efficiency. To address these limitations, we developed an innovative multifunctional biomimetic nano-therapeutic system (PDMC/CHP@S/V), which comprised a triblock copolymer synthesized through controlled polymerization of dopamine methacrylamide (DMA), 2-methacryloyl

BM-164 (an H₂S-donor) modulates microRNA expression associated with reperfusion-induced ventricular fibrillation in rat hearts.

Ventricular arrhythmias are a leading cause of sudden cardiac death in myocardial infarction and heart failure. Reperfusion following ischemia can trigger severe arrhythmias, including ventricular fibrillation (VF), and microRNAs (miRNAs) which are increasingly recognized as critical regulators of these processes. This study aimed to identify miRNAs associated with ischemia/reperfusion (I/R)-induced ventricular fibrillation by comparing expression profiles between fibrillated (VF) and non-fibrillated (No VF) rat hearts. Nanostring-based screening (n = 12; 4 in each group: No I/R control, I/R n

NGR1 Pretreatment Enhances the Therapeutic Efficacy of Transplanting Cardiomyocytes Derived from Human Induced Pluripotent Stem Cells for Myocardial Infarction.

Human induced pluripotent stem cells (hiPSCs) offer significant potential for differentiation and research applications in cardiovascular diseases. When induced differentiated hiPSC-derived cardiomyocytes (hiPSC-CMs) are transplanted into the infarcted myocardial region, they exhibit extremely low survival rates and unsatisfactory therapeutic effects due to ischemia, hypoxia, and immune inflammation in the surrounding environment. To address this issue, we used Panax notoginseng saponin R1 (NGR1), which has demonstrated significant protective effects in prior research, to pretreat hiPSC-CMs be

Anacardic acid mitigates post‑MI cardiac fibrosis via suppressing p38/JNK-GATA3 pathway and H3K9ac‑dependent epigenetic regulation.

BACKGROUND: Myocardial fibrosis (MF), a hallmark of cardiovascular diseases (CVDs) such as myocardial infarction (MI), drives progressive cardiac dysfunction and adverse remodeling. Histone acetylation is a critical epigenetic regulator in cardiovascular pathology. Anacardic acid (AA), a histone acetyltransferase inhibitor (HATi) with pleiotropic bioactivities, has been studied in various disease contexts; however, its antifibrotic efficacy and mechanisms in MF remain unclear. METHODS: In vivo, a mouse model of post-infarction MF was established by permanent left anterior descending (LAD) lig

High-intensity versus very-high-intensity statin, both on top of ezetimibe, in acute myocardial infarction: A pragmatic randomized study.

BACKGROUND: Early lipid-lowering therapy with "high-intensity" statins is recommended post-acute myocardial infarction (AMI), yet the optimal dose within such regimens remains uncertain. We compared efficacy and safety of very high-intensity versus high-intensity statin regimens, both combined with ezetimibe, initiated during AMI hospitalization. METHODS: In a stepped-wedge cluster randomized controlled single-center trial, patients with ST- or non-ST-elevation AMI were assigned in sequential 6-month blocks to either very high-intensity statin (Group A: atorvastatin 80 mg or rosuvastatin 40 m

1,3-Butanediol enhances autophagy via PI3K/Akt/FOXO3 pathway to ameliorate cardiac remodeling post-myocardial infarction.

BACKGROUND: Myocardial infarction (MI) remains a leading cause of cardiovascular mortality. While ketone bodies show cardioprotective potential, their role in regulating cardiomyocyte autophagy post-MI is unclear. METHODS: A rat MI model was established and treated with 1,3-butanediol (1,3-BD, 10 mg/100 g/day), a ketone precursor. Cardiac structure and function were assessed alongside autophagy and apoptosis levels. In vitro, hypoxia-induced cardiomyocytes were treated with β-hydroxybutyrate (β-HB) and phosphatidylinositol 3-kinase (PI3K) inhibitor. Mechanisms were explored via transcriptomic

Temporal inhibition of ADAM17 in fibroblasts reduces stiffness and promotes vascularization following myocardial infarction.

AIMS: Myocardial infarction (MI) triggers a complex remodeling that, if uncontrolled, leads to heart failure. Increased levels of ADAM17 (disintegrin and metalloproteinase-17) in ischemic injury has been reported, but its direct role in scar formation and subsequent recovery from MI has not been identified. We investigated the role of ADAM17 in the function of homeostatic fibroblasts (FBs) vs. activated myofibroblasts (myoFBs) in scar formation, and recovery following MI. METHODS AND RESULTS: Human myocardial specimens showed upregulated ADAM17 in the infarct tissue, colocalized to myofibrobl

MAG-PSIR mismatch in a patient with multi-territory myocardial infarcts with and without acute ischemic edema.

Cardiac MRI (cMRI) is crucial for characterizing acute myocardial infarction (MI) complications. We present a case where acute ischemic edema demonstrated a magnitude (MAG) and phase-sensitive inversion recovery (PSIR) mismatch. This highlights that the MAG-PSIR mismatch can arise from the very long T1 times of acute edema, a key finding confirmed by T1 and T2 mapping [1].

Letter to the Editor. Effect of encephalomyosynangiosis procedure on post stroke recovery in a permanent model of ischemic stroke.

This correspondence comments on the methodological issues of Anthony et al. on encephalomyosynangiosis (EMS) for acute ischemic stroke in mice. The central issue is the clinically unrealistic intervention time of 4 h post-occlusion, which limits the translational relevance of the reported benefits in infarct reduction and recovery. Furthermore, the surgical design-specifically, the differential handling of the temporalis muscle between groups-introduces a potential confounder, as the excision in controls may alter baseline intracranial pressure compared to the preserved muscle in the EMS group
