[Update on anal dysplasia and anal cancer].

Anal dysplasia and anal cancers are predominantly caused by high-risk types of the human papillomavirus. Lesions are mostly asymptomatic and disproportionately affect high-risk populations such as people living with HIV, women with a history of vulvar (pre)cancer, and long-term immunosuppressed individuals. For these risk groups, the incidence of anal cancer is up to 100 times higher, which is why screening is recommended in the S2k guideline completed in 2024. However, it should be noted that two-thirds of anal cancers occur in older women without HIV, for whom scalable screening strategies d

Association between KSHV genotype and immune response to Kaposi sarcoma.

Kaposi sarcoma (KS) is a locally aggressive endothelial tumor that can be classified into four clinical types: classic, AIDS-related, iatrogenic, and endemic. KS usually affects the skin and soft tissues, but it can develop into mucosal, lymph node, and multiple-organ lesions. KS is caused by Kaposi sarcoma-associated herpes virus (KSHV), which has six genotypes (A, B, C, D, E, and F). However, it remains unclear whether the KSHV genotype affects clinical presentation. In this study, we investigated clinical stage, cellular proliferation, NF-κB activity, and lymphocyte infiltration around the

Epstein-Barr Virus-Positive Primary CNS Lymphoma in a Patient Receiving Mycophenolate Mofetil: Diagnostic and Therapeutic Considerations.

Epstein-Barr virus (EBV)-positive primary central nervous system lymphoma (PCNSL) is a rare entity typically associated with profound immunosuppression, most commonly in transplant recipients or individuals with HIV. We report a case of EBV-positive PCNSL arising in a 75-year-old male with myasthenia gravis receiving chronic mycophenolate mofetil (MMF) therapy outside the transplant setting. The patient presented with progressive neurological deficits, and brain magnetic resonance imaging demonstrated multiple enhancing lesions. Stereotactic biopsy revealed diffuse large B-cell lymphoma of non

Determinants of precancerous cervical lesions among women screened for cervical cancer at Debre Markos town public health facilities, Ethiopia.

BACKGROUND: The potential for precancerous cervical lesions to develop into cervical cancer over time is a significant public health concern in low and middle-income countries, including Ethiopia. The Ethiopian government is supporting the prevention of cervical cancer by acetic acid screening and vaccinations. However, the determinants of precancerous cervical lesions were not well studied. Therefore, this study aimed to identify determinants of pre-cancerous cervical lesions among women screened for cervical cancer at Debre Markos town public health institution, Ethiopia, 2025. METHODS: An

Spatial and bulk transcriptomics reveal distinct molecular signatures in Kaposi sarcoma with and without other KSHV-associated diseases.

BACKGROUND: Kaposi sarcoma (KS) is an angioproliferative tumor caused by Kaposi sarcoma herpesvirus (KSHV) that occurs in people with HIV. Concurrent KSHV-associated diseases (KAD), including multicentric Castleman disease, primary effusion lymphoma, and KSHV-associated inflammatory cytokine syndrome may modify KS biology and impact clinical outcomes. Transcriptomic profiling of archival KS tissue enables investigation of molecular heterogeneity associated with these overlapping disease states. METHODS: Archival formalin-fixed paraffin-embedded (FFPE) KS skin biopsies from 42 patients with HIV

Integrated multi-omics analysis identifies TBC1D10C as an NK- and T cell-associated diagnostic biomarker in childhood obesity.

Childhood obesity is a growing global health crisis associated with an increased risk of metabolic and cardiovascular diseases in adulthood. While accumulating evidence implicates immune dysregulation in obesity pathogenesis, the specific transcriptional alterations of immune cells, particularly NK cells and T cells in the context of childhood obesity, and their potential as non-invasive diagnostic biomarkers, remain largely unexplored. Here, we investigated the transcriptional alterations of NK cells and T cells in childhood obesity and evaluated their potential as non-invasive diagnostic bio

Epidemiology of Monoclonal Gammopathies in Sub-Saharan Africa: A Systematic Review and Meta-Analysis of MGUS and Multiple Myeloma.

BACKGROUND: Monoclonal gammopathy of undetermined significance (MGUS) and multiple myeloma (MM) are reported to occur more frequently in Black populations, particularly African Americans. However, despite Sub-Saharan Africa (SSA) being home to the largest Black population globally, epidemiological data on MGUS and MM in the region remain scarce. METHODS: We conducted a systematic review and meta-analysis of published studies from SSA to estimate the pooled prevalence of MGUS and MM, regional variations, survival outcomes and key clinical complications, including HIV-associated MM, renal failu

Exploring variation in research priorities generated by AI tools.

BACKGROUND: Artificial intelligence (AI) tools based on large language models (LLMs) are being increasingly used by researchers and may play a role in health-related research priority-setting exercises (RPSEs). However, little is known about how these tools may differ in the types of research priorities they generate. METHODS: We examined research priorities aimed at improving treatments for four diseases: cancer, COVID-19, HIV, and Alzheimer. We compared the outputs from five AI tools (DeepSeek, ChatGPT, Claude, Perplexity, and Gemini) using SBERT-BioBERT embeddings and cosine similarity sco

The relational association between multiple sexual partners and HIV testing on cervical cancer screening among women of reproductive age in Ghana: A national population-based study.

BACKGROUND: Cervical cancer screening remains vital in the early detection of precancerous lesions and promotes better treatment outcomes. Though evidence suggested multiple sexual partners and HIV infection as risk factors for cervical cancer, limited studies have investigated how multiple sexual partners and HIV infection impact cervical cancer screening in Ghana. Therefore, this study assessed the association of multiple sexual partnership and HIV testing on cervical cancer screening among Ghanaian women of reproductive age. METHODS: The study utilized data from the 2022 Ghana Demographic

Elevated m6A RNA modifications associate with immune dysregulation and cancer in people with HIV-1.

BACKGROUND: N6-methyladenosine (m6A) modifications of human immunodeficiency virus type 1 (HIV-1) and cellular RNA contribute to regulation of viral and host gene expression. RNA m6A dysregulation has been implicated in multiple cancers. However, the role of m6A modifications in HIV-1-associated cancers remains unclear. Here, we aim to address this important question using clinical samples. METHODS: Using enzyme-linked immunosorbent assay (ELISA), RNA m6A levels were quantified in peripheral blood mononuclear cells (PBMCs) from 43 de-identified people living with HIV-1 (PLWH), comparing those

Lived experiences of cancer care for people living with HIV who are treated for anal cancer: a scoping review.

OBJECTIVE: This scoping review aims to identify existing evidence on the lived experiences of people living with HIV and treated for anal cancer, and to identify what aspects of health and well-being are addressed in clinical guidance. DESIGN: A preregistered protocol (Open Science Framework, 2025) guided the review. We followed the Arksey and O'Malley framework, incorporating Levac et al's refinements around stakeholder consultation. Joanna Briggs Institute (JBI) guidance informed eligibility and data for charting, and reporting adhered to Preferred Reporting Items for Systematic Reviews and

Diagnostic challenges for a case of maxillary carcinoma cuniculatum in the setting of human immunodeficiency virus: a case report.

PURPOSE: Carcinoma Cuniculatum (CC) is an exceptionally rare variant of squamous cell carcinoma with limited description in the literature. Oral Carcinoma Cuniculatum (OCC) continues to be a diagnostic dilemma as it generally presents as an indolent infection, often undergoing multiple biopsies and debridements prior to definitive diagnosis. This study aims to review the diagnostic challenges, clinical features, histopathology, and management of OCC. We specifically present a unique case of OCC in a patient with human immunodeficiency virus (HIV). METHODS: A review of the current literature on

A mixed effect similarity matrix regression model (SMRmix) for integrating multiple microbiome datasets at the community level.

Recent studies have highlighted the importance of the human microbiota in health and disease. However, in many areas of research, individual microbiome studies often provide inconsistent results due to limited sample sizes and the heterogeneity in study populations and experimental procedures. This inconsistency underscores the need for integrative analysis of multiple microbiome datasets. Despite the critical need, statistical methods that incorporate multiple microbiome datasets and account for study heterogeneity are not available in the literature. To address this, we propose a mixed effec

Extended Genotyping as Triage of HPV Positive Screened Women in Low-and-Middle Income Countries.

Cervical cancer screening coverage in low- and middle-income countries is low, and many screened women are lost to follow-up. Point-of-care testing that allows same-day treatment could reduce the incidence of this cancer, but same-day triage testing is required to prevent overtreatment and minimise loss-to-follow-up before treatment. Built-in extended genotyping with the Xpert® human papilloma virus (HPV) assay offers point-of-care triage, but treatment thresholds differ. We calculated test performance of the Xpert® HPV assay for HIV-positive and HIV-negative women. Extended HPV-genotyping was
