Low-Field Magnetic Resonance Imaging Features of Axial Proximal Sesamoid Osteomyelitis and Intersesamoidean Ligament Rupture in a Horse.

A 7-year-old Warmblood mare used for show jumping was evaluated for progressive left hindlimb lameness of 5 weeks' duration, localized clinically to the metatarsophalangeal region. Radiography revealed ill-defined osteolysis of the axial apical regions of both proximal sesamoid bones (PSBs). Due to rapid clinical deterioration, low-field magnetic resonance imaging was performed and demonstrated aggressive axial osteomyelitis of both PSBs, partial rupture of the intersesamoidean ligament and proximal scutum with herniation of synovium of the plantar joint recess, and an occult penetrating tract

Synovial fibroblast responses to different types of injury resulting in cartilage repair or osteoarthritis.

OBJECTIVE: Single-cell RNA-sequencing (scRNA-seq) has advanced our understanding of the heterogeneity of synovial fibroblasts and their roles in tissue repair and osteoarthritis. Here, we compared the fibroblast responses to different types and duration of joint injury in mice. DESIGN: Published scRNA-seq data of synovial fibroblasts from a model of joint surface injury (JSI, day 6) and two post-traumatic osteoarthritis models, destabilisation of the medial meniscus (DMM, day 7 and 2 months) and anterior cruciate ligament rupture (ACLR, day 7 and day 28), were integrated and analysed using cl

Comparison of innate and adaptive immune responses to joint injury during the onset and progression of post-traumatic osteoarthritis in common murine models.

Post-traumatic osteoarthritis (ptOA) following joint injury accounts for at least 12% of cases of symptomatic OA. Surgical restoration of joint biomechanics does not prevent ptOA development, suggesting other factors influence individual OA risk. One suggested factor is the immuno-inflammatory response in the joint, with both adaptive and innate immune responses implicated. Whether different joint injuries incite different acute inflammatory responses, potentially explaining variable ptOA risk, has not been explored. Male 10-12-week-old C57BL/6 mice were randomly assigned to one of 6 intervent

Lipid droplet-induced T cell death sustains autoimmune tissue inflammation.

Autoimmunity leading to rheumatoid arthritis (RA) involves CD4+ T cell recruitment into synovial tissue. However, metabolic conditions supporting the survival and pro-inflammatory effector functions of these tissue-invading T cells remain poorly understood. Lipidomic analysis identified the inflamed synovium as a lipid-rich environment. In functional studies, administration of the free fatty acid oleic acid exacerbated synovitis. Tissue-invading CD4+ T cells responded to fatty acid with rapid cell lysis, releasing cytoplasmic and nuclear content into the extracellular space. This T cell lytic

Tetranectin is significantly elevated in the synovial fluid of patients with full-thickness rotator cuff tears.

Rotator cuff tears are one of the most common shoulder disorders and can lead to pain, weakness, and limited mobility. Although the clinical condition has been widely studied, its molecular mechanisms are still not fully understood. In this exploratory study, we analyzed the proteomic profile of synovial fluid in patients with full-thickness rotator cuff tears and compared the results to those from a control group. Proteomic analysis was performed on synovial fluid samples from seven patients with full-thickness tears and three patients with partial-thickness tears, who served as the control g

Cellular and molecular landscapes of inflammation in anterior cruciate ligament rupture patients are independent on concurrent meniscal injury.

BACKGROUND: Anterior cruciate ligament (ACL) rupture greatly increases the risk of developing knee osteoarthritis (OA), especially when accompanied by meniscal injury. Therefore, we examined biological processes in the knee joint of ACL rupture patients with or without concurrent meniscal injury. The aim of this study was to explore whether concurrent meniscal injury leads to distinct inflammatory cellular or molecular landscapes and patient reported outcome measures (PROMs), like knee function and pain, compared to an isolated ACL rupture with intact menisci. METHODS: Synovium, synovial fluid

Stage-dependent analysis of IL-6, TGF-β, osteocalcin, and sRANKL in the synovial fluid of dogs with osteoarthritis.

Osteoarthritis (OA) is traditionally considered a degenerative joint disease. However, morphological similarities between OA progression and fracture healing suggest that OA may be a dysregulated bone repair process. This study investigated the morphological and biochemical parallels between OA and bone healing in dogs following cranial cruciate ligament rupture. To differentiate between biochemical profiles at different stages of the disease, a structured OA scoring system was developed based on radiographic and additional intraoperative findings. Synovial fluid samples from the OA cohort wer

Synovial Fluid Biomarkers Reflect Pre- and Postoperative Clinical Features in Rotator Cuff Tears, Independent of Tear Size.

Synovitis is a major contributor to pain and dysfunction in rotator cuff tears (RCTs), yet it remains unclear whether routine synovial fluid (SF) parameters reflect clinical features independent of tear size, operation method, and patient age. This prospective cohort study aimed to evaluate the associations between routine SF parameters and clinical features before and after rotator cuff repair. Preoperative glenohumeral SF was aspirated from 109 patients undergoing arthroscopic repair, and interleukin-6, total protein levels, and differential white blood cell counts were analyzed. Clinical fe

Clinical Outcomes of Hidden Lesions of the Medial Meniscus Treated With a Flexible All-Inside Device.

Purpose Ramp lesions are characteristic injuries associated with anterior cruciate ligament (ACL) rupture and involve disruption of the meniscus-synovium interface near the posterior horn of the medial meniscus. Among these, tibiofibular side-only tears at the bursal attachment, so-called hidden lesions, are difficult to visualize from the standard anterior portal, making their diagnosis and treatment challenging. This study examined the outcomes of suturing hidden lesions using an all-inside technique with the FAST-FIX device from the anterior portal, with a focus on clinical results and the

Novel Biomaterial-Based Synovial Fluid Analysis Reveals Protective microRNA Signatures in a Mouse Model of Acute Synovitis-Driven Osteoarthritis.

Acute synovitis and continuous mechanical stress are critical factors of Post-traumatic Osteoarthritis (PTOA); however, the effect on cartilage degeneration and its underlying mechanisms remains unclear. Here, we established a novel biomaterial-based method for synovial fluid analysis in mice to uncover the mechanisms underlying the onset of PTOA, focusing on synovitis and mechanical stress. Twelve-week-old C57BL/6J males were divided into the ACL-Transection (ACL-T), ACL-rupture (ACL-R), and Intact groups. We performed a joint instability test and histological analysis for cartilage degenerat

Spontaneous flexor digitorum profundus tendon rupture of the little finger in patients with rheumatoid arthritis: A report of two cases with different mechanisms.

Subcutaneous flexor tendon rupture in patients with rheumatoid arthritis is rare compared with extensor tendon rupture, with a reported incidence of approximately one-tenth of that of extensor tendons. The pathophysiology of flexor tendon rupture is not fully understood. We report two cases of spontaneous flexor digitorum profundus tendon rupture of the little finger in patients with rheumatoid arthritis despite low disease activity and the absence of advanced joint destruction. Both patients presented with an inability to actively flex their little fingers without any history of significant t

Knee Synovial Fluid Biomarker Type and Concentration With Long-Term Outcomes After ACL Reconstruction With Concomitant Meniscal Injury.

BACKGROUND: Anterior cruciate ligament (ACL) rupture frequently occurs alongside meniscal injury, together altering the intra-articular environment. While ACL injury triggers an inflammatory cascade within synovial fluid, meniscal injury may intensify these responses, contributing to joint degeneration. However, little is known about how combined ACL and meniscal injuries shape the synovial inflammatory milieu or whether these profiles influence long-term outcomes. PURPOSE/HYPOTHESIS: The purpose was to characterize synovial fluid inflammatory phenotypes at the time of ACL reconstruction with

Acute local and systemic effects of intra-articular administration of liposomal Ido-1 following anterior cruciate ligament rupture.

Joint injuries, such as rupture of the anterior cruciate ligament (ACL), are associated with the development of post-traumatic osteoarthritis (PTOA). ACL rupture can lead to disruption of metabolic pathways, including the conversion of tryptophan to kynurenine, which is associated with a sustained inflammatory response. An in vivo study was undertaken to determine the acute effects of intra-articular administration of liposomes loaded with the tryptophan-catabolizing enzyme indoleamine 2,3-dioxygenase-1 (IDO-1) following ACL rupture. Using a rat model of non-surgical ACL injury, male and femal

Early synovial regulatory T cell engagement in posttraumatic osteoarthritis.

OBJECTIVE: Posttraumatic osteoarthritis (PTOA) develops despite restoration of joint biomechanics, suggesting that early post-injury biological processes within the synovium shape disease trajectories. We investigated whether synovial regulatory T (Treg) cells are engaged during the early phase following joint injury and whether experimental enrichment of this compartment is associated with altered local inflammation and PTOA-related pathology. METHODS: Using surgically induced destabilization of the medial meniscus (DMM) and a non-invasive anterior cruciate ligament (ACL) rupture model in mi

Intra-articular Genicular Artery Pseudoaneurysm Mimicking Pigmented Villonodular Synovitis: A Case Report.

Pigmented villonodular synovitis (PVNS) is a non-aggressive entity involving the synovium. Magnetic resonance imaging (MRI) is regarded as the preferred imaging modality. MRI can occasionally be deceptive, with many benign conditions having a propensity for repeated bleeding intimately resembling PVNS. We present a case of an intra-articular pseudoaneurysm of the knee joint resembling PVNS on MRI. This 80-year-old male had multiple comorbidities, including a ruptured abdominal aortic aneurysm presented with recurrent knee swelling. The MRI showed complex hemorrhagic knee joint effusion with an

Thrombospondin-2 deficiency primes the synovial joint for aberrant tissue remodeling and injury response.

OBJECTIVE: This study investigates joint injury-induced angiogenesis and the effects of genetic deficiency of thrombospondin-2 (TSP2), an anti-angiogenic factor, in joint homeostasis and post-traumatic osteoarthritis (PTOA). METHOD: We utilized a murine non-invasive anterior cruciate ligament rupture (ACLR) model of PTOA and mined published synovial transcriptomics datasets to investigate injury-induced synovial angiogenesis. Spatial transcriptomics and flow cytometry of TSP2-GFP reporter mice were used to assess injury-induced thrombospondin-2 and its cellular origins in synovium. Global TSP

Synovial biomarkers in the diagnosis of post-traumatic osteoarthritis following anterior cruciate ligament and meniscus injuries: protocol for a systematic review.

INTRODUCTION: This review aims to synthesise research evidence regarding biomarkers in the synovial fluid that may predict the risk of post-traumatic osteoarthritis (PTOA) in young adults. Considering the high prevalence of knee joint injuries, particularly among youth sports athletes, this review will focus on anterior cruciate ligament and/or meniscal ruptures. These injuries are highly associated with PTOA, with studies indicating that even with surgical reconstruction, 50%-80% of affected individuals develop knee PTOA within a 10-year follow-up. METHODS AND ANALYSIS: The results of this s

Synovial macrophage diversity and activation of M-CSF signaling in post-traumatic osteoarthritis.

Synovium is home to immune and stromal cell types that orchestrate inflammation following a joint injury; in particular, macrophages are central protagonists in this process. We sought to define the cellular and temporal dynamics of the synovial immune niche in a mouse model of post-traumatic osteoarthritis (PTOA), and to identify stromal-immune crosstalk mechanisms that coordinate macrophage function and phenotype. We induced PTOA in mice using a non-invasive tibial compression model of anterior cruciate ligament rupture (ACLR). Single-cell RNA-sequencing and flow cytometry were used to asses

A bilinear model for the elastic response of hydrated lipid bilayers under normal pressure difference.

The elasticity of phospholipid membranes as a function of hydration was investigated using coarse-grained molecular simulations. Multilamellar membranes consist of two or more lipid bilayers separated by a thin layer of water, a system commonly found in cell membranes that provides surface tension in the alveoli of the lungs and on cartilaginous surfaces of synovial joints. The objective was to quantify the response of such systems to compression in the direction perpendicular to the membranes as a function of the amount of water between the bilayers or hydration of the system. The present stu

Loosening the Lid on Shoulder Osteoarthritis: How the Transcriptome and Metabolic Syndrome Correlate with End-Stage Disease.

Metabolic syndrome (MetS) associated with Osteoarthritis (OA) is an increasingly recognised entity. Whilst the degenerative pattern in cuff-tear arthropathy (CTA) has been well documented, the biological processes behind primary shoulder OA and CTA remain less understood. This study investigates transcriptomic differences in these conditions, alongside the impact of MetS in patients undergoing total shoulder replacement. In a multi-centre study, 20 OA patients undergoing total shoulder replacement were included based on specific treatment indications for OA and cuff-tear arthropathy as well as
